Effects of Liquid Culture of Agaricus blazei Murill on Lipid Metabolism in Rats Fed Cholesterol Diet. (신령버섯 균사체 액체배양액이 고콜레스테롤혈증 흰쥐의 지질대사에 미치는 영향)

  • The effects of liquid culture of Agaricus blazei Murill on the weight gains, serum and hepatic lipid concentrations were studied in male rats. Sprague-Dawley rats (7 weeks old) were given four different types of diets for 6 weeks, respectively: a normal diet group, a high fat control diet group (normal diet+15% lard +0.5% cholesterol), a 30% or 40% A. blazei diet groups (high fat control diet+30% or 40% A. blazei in water) according to the levels of A. blazei supplementation. The body weight gains, food intake, food efficiency ratios, and the liver, kidney, and epididymal fat pad weights of the rats fed 30% or 40% A. blazei diets were similar to those of the rats fed high fat control diet. The concentrations of serum total cholesterol, LDL-cholesterol and atherogenic index in rats fed the 30% or 40% A. blazei diets were significantly decreased compared with those of rats fed the high fat control diet. The HDL-cholesterol/total-cholesterol ratios of the rats fed 30% or 40% A. blazei diet were significantly increased compared with those of rats fed the high fat control diet. The fecal excretion of total lipid . in the rats fed 40% A. blazei diet was significantly increased compared with those of rats fed the high fat control diet. The concentrations of serum total lipid in the rats fed 40% A. blazei diet was significantly lower than that in the rats fed high fat control diet. But the concentrations of serum HDL-cholesterol, hepatic total cholesterol and triglyceride of rats fed the 30 or 40% A. blazei diets were similar to those of rats fed the high fat control diet. These results showed that the 30 or 40% A. blazei diets feeding decreased the total cholesterol, LDL-cholesterol and atherogenic index, and increased the HDL-cholesterol/total-cholesterol ratio in serum of rats.

  • The effects of cheonggukjang (CK) on the weight gains, serum and hepatic lipids levels were investigated in adult female rats fed the cholesterol diets. Sprague-Dawley strain rats were given four different types of diet for 4 weeks, respectively: a normal diet, a control diet (10% fat + 0.5% cholesterol), a 5% or 10% CK diets (control diet + 5% or 10% CK powder). The body weights, food intake and food efficiency ratios, and the uterine fat pad weights of 5% CK and 10% CK groups were more significantly decreased than those of the control group. The liver cholesterol and triglyceride levels were significantly lower of the 10% CK group than those of the control group. The concentrations of serum triglyceride, total cholesterol, LDL+VLDL-cholesterol, and atherogenic index ratios were significantly decreased in the 10% CK group compared with those in the control group. The HDL-cholesterol/total-cholesterol ratios was significantly increased in the 10% CK group compared with those in the control group. The fecal total lipid excretion of the 5% and 10% CK groups were more increased than those in the control group. These results showed that the 10% cheonggukjang feeding decreased the triglyceride, total cholesterol, LDL+VLDL-cholesterol and atherogenic index ratios in serum as well as the triglyceride and cholesterol in liver, and increased the HDL-cholesterol/total-cholesterol ratio in serum.

  • This study was performed to know the hypocholesterolemic effect of depolymerized alginate obtained by hydrolysis of alginate through a heating process at $121^{\circ}C$. The changes of total cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides and phospholipid levels in serum, as well as the atherogenic index were elucidated in rats for 35 days. In order to evaluate single-dose toxicity of HAG-50 in rats via oral routes, induction any mortalities and abnormal signs in clinical finding, body weight, gross findings and histopathological finding, and estimation of $LD_{50}$ were elucidated. Total cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides and phospholipid levels in the serum significantly decreased and resulted in the decrease of atherogenic index in rats fed diets containing $5\%\;and\;10\%$ HAG-50 (p<0.01). HAG-50 did not induce any mortalities and abnormal signs in clinical findings, body weights, gross findings and histopathological findings in single-dose toxicity test via oral routes of rats, after which it was impossible to estimate $LD_{50}$ values.

Effect of Okcheonsan on Blood Glucose, Lipid and Protein bevels in Streptozotocin-Induced Diabetic Female Rats (옥천산이 당뇨 쥐의 혈당, 지질 및 단백질 농도에 미치는 영향)

  • The effects of Okcheonsan powder on the body weights, the organ weights, the blood glucose level, the lipid and protein concentrations of serum and liver in diabetic rats were studied. Female rats (Sprague-Dawley, mean weight 313.6$\pm$18.5 g) were randomly assigned to one normal and two diabetic groups. They were fed experimental diets for 5 weeks. The diabetic groups were divided into the diabetic control (D-control group) and 3% Ok-cheonsan groups (D-Okcheonsan group). Rats were injected with streptozotocin intraperitoneally to induce diabetes. The body weights, the concentrations of total lipid and triglyceride of liver, the concentrations of total protein and albumin of serum in tole D-control and the D-Okcheonsan groups were significantly decreased compared with those in the normal group. The pancreatic weight in the D-control group was significantly more increased than that in the D-Okcheonsan group, but in the D-Okcheonsan group it was similar to that in the normal group. The fasting blood glucose levels and the atherogenic index in all the diabetic groups were significantly higher than those in the normal group. The concentrations of triglyceride, total cholesterol and LDL-cholesterol of serum, and the cholesterol of fiver in the D-Okcheonsan group were significantly lower than those in the D-control group. The concentration of HDL-cholesterol and HDL-cholestrol/total cholesterol ratio of serum were similar to those in all the groups. The concentrations of phospholipid of serum and liver in the D-Okcheonsan group were significantly decreased compared with those of the normal group, In conclusion, the Okcheonsan powder feeding could decrease the pancreatic weight, the concentrations of the triglyceride, the total cholesterol and LDL-cholesterol of serum, and the cholesterol of liver in the diabetic rats. But the concentrations of the blood glucose, the hepatic triglyceride and the atherogenic index seems to be not affected by it.

Effects of Pleurotus eryngii on Lipid Metabolism in Rats Fed High Fat Diet (새송이버섯이 고지방 식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • The effects of Pleurotus (P.) eryngii on the weight gains, food efficiency ratios, serum and hepatic lipid concentrations were investigated in male rats fed the high fat diets. Twenty one week-old Sprague-Dawley rats were given three different types of diet for 10 weeks, respectively: a control diet (high fat; $20\%$ lard), two kinds of P. eryngii diet supplemented with $3\%\;or\;5\%$ of dry P. eryngii powder in substitutes for the same amounts of cellulose in the control diets. The body weight gains and food efficiency ratios of the rats fed $3\%\;and\;5\%$ P. eryngii diets were significantly lower than those of the rats fed the control diet. The concentrations of hepatic total lipid and triglyceride in the rats fed the P. eryngii diets were significantly lower than those in the rats fed the control diet. But the hepatic cholesterol contents of the rats fed the P. eryngii diets were similar to those of the rats fed the control diet. The concentrations in serum total cholesterol, LDL-cholesterol, and atherogenic index ratios were significantly lower in the rats fed the P. eryngii diets compared to those fed the control diet. The HDL-cholesterol/total- cholesterol ratio was significantly higher in the rats fed the P. eryngii diets compared to those fed the control diet. There were no significant difference found in the serum triglyceride, phospholipid and HDL-cholesterol concentrations among the experimental groups. These results showed that feeding of P. eryngii powder feeding decreased the total cholesterol, LDL-cholesterol and atherogenic index and increased the HDL-cholesterol/total-cholesterol ratio in serum of the rats fed high fat diet.

Effects of Germinated-Buckwheat on Blood Pressure , Plasma Glucose and Lipid levels of Spontaneously Hypertensive Rats (발아메밀이 본태성 고혈압쥐의 혈압, 혈당 및 혈중 지질수준에 미치는 영향)

  • The purpose of this study was to investigate the effect of germinated buckwheat on blood pressure, plasma glucose and lipid levels of spontaneously hypertensive rats(SHR). The male and female SHR were divided into two groups. After feeding the germinated buckwheat diet(50%, w/w) ad libitum for 6 weeks, the weight gain of the germinated buckwheat group in male rats was higher than those of control group. Systolic blood pressure of the germinated buckwheat group in male rats was significantly decreased at 6 weeks by 3%. The fasting glucose levels of the germinated buckwheat group had a tendency to be lower compared with those of the control group. Total cholesterol, HDL-cholesterol and HTR of female SHR fed the germinated buckwheat diet were significantly increased compared to the control diet. Atherogenic index of the germinated buckwheat group was decreased in males(ns) and females(p<0.05) than those of the control group. From these results the germinated buckwheat might be useful for anti-hypertensive and anti-hyperlipidemic agents.

  • The effects of liquid culture of Agaricus blazei Murill on the body weight gain, lipid metabolism, protein levels, and enzyme activities were studied in growing male rats. Sprague-Dawley rats were divided into three groups ; control group (A. blazei free water), 20 or 30% A. blazei groups (20 or 30% A. blazei in water) according to the levels of A. blazei supplementation. The rats were fed ad libitum each of the experimental beverage for 5 weeks. The body weight gain, food intake, food efficiency ratio, and organ weight of the 20 or 30% A. blazei groups were similar to those of the control group. There were no differences in the concentrations of triglyceride, total cholesterol, HDL-cholesterol, total protein and albumin, and the activities of GOT, GPT, LDH and alkaline phosphatase of the serum among the experimental groups. The calcium and phosphorus concentrations of serum in the 20 or 30% A. blazei group were significantly decreased compared with those of the control group. The concentrations of LDL-cholesterol and the atherogenic index of the 20 or 30% A. blazei groups were significantly lower than those of the control group, but the serum HDL-cholesterol/total cholesterol ratio was higher. These results showed that the 20 or 30% A. blazei feeding decreased the LDL-cholesterol and the atherogenic index, and increased HDL-cholesterol/total cholesterol ratio in serum of rats.

Effects of Citrus Peel Ethanol Extract on the Serum Lipid and Body Fat of High-fat-diet-fed Rats (고지방식이를 섭취한 흰쥐에서 진피 에탄올추출물이 혈청지질 및 체지방에 미치는 영향)

  • In this work, the major flavonoid content of citrus peel ethanol extract and the changes in the serum lipid and body fat of high-fat-diet-fed rats were examined. The subjects were divided into four groups: the normal, control, O1 (0.1% citrus peel ethanol extract), and O5 (0.5% citrus peel ethanol extract) groups. The experimental groups (O1 and O5) were fed a high-fat diet mixed with 0.1 and 0.5% citrus peel ethanol extract, respectively, for 10 weeks. The results of the analysis showed that the main ingredients of the flavonoid in the citrus peel ethanol extract (60 brix) were $38.21{\pm}1.37$ mg/g hesperidin and $15.02{\pm}0.48$ mg/g narirutin. With regard to the changes in the serum total-cholesterol level, compared with the control group's $79.1{\pm}4.6$ mg/dL, the serum total-cholesterol levels of the O1 and O5 groups were significantly lowered to $64.6{\pm}5.4$ and $59.5{\pm}4.7$ mg/dL, respectively. As for the changes in the LDL-cholesterol level, compared with the control group's $18.4{\pm}3.7$ mg/dL, the LDL-cholesterol levels of the O1 and O5 groups were significantly lowered to $13.0{\pm}3.6$ and $13.0{\pm}1.9$ mg/dL, respectively. For the changes in the serum triglyceride level, compared with the control group's $74.5{\pm}13.2$ mg/dL, the serum triglyceride level of the O5 group was significantly lowered to $48.6{\pm}11.2$ mg/dL. Finally, with regard to the changes in body fat, compared with the control group, the 05 group showed a significant decrease. In conclusion, it was observed that citrus peel ethanol extract was effective in lowering the total-cholesterol, LDL-cholesterol, and triglyceride levels of high-fat-diet-fed rats.

Carotid Intima-Media Thickness Measurement in Obese Children (비만 소아들에서의 경동맥 내막중막 두께 측정)

  • Purpose : The aim of the present study was to ascertain whether the increase of carotid intima-media thickeness(cIMT) as one of premature pathologic changes of atherosclerosis, was present in obese children compared to normal weight children. Methods : The obese group consisted of 21 obese/overweight(body mass index(BMI) above 85 percentile of age, sex standards) children and the control group of 11 normal weight children. None of the children had any chronic illnesses or previous medication history. We investigated the age, sex, height, weight, and systolic/diastolic blood pressure. We measured cIMT by ultrasonogram. In 19 of the obese group, we tested the serum glucose level, liver transaminase level, and cholesterol level etc. Results : The increase of cIMT in obese group did not achieved statistical significance(obese group vs. control group; 0.42 vs. 0.40 mm, P=0.0592). In addition, cIMT showed no significant correlation with any physical/laboratory variables including BMI(P=0.0585). Conclusion : To our knowledge, this is the first study to measure the cIMT in Korean children. Though the results approached statistical significance, we could not prove an increase of cIMT in obese children or an association between cIMT and BMI, due to the study's small sample size. In the future, larger and more extensive trials are needed.

Assessment of Carotid Geometry by Using the Contrast-enhanced MR Angiography (조영증강 MR 혈관 조영술을 이용한 경동맥 기하학의 평가)

  • Purpose : To evaluate the geometry of carotid artery by assessing the images of contrast-enhanced MR angiography (CE-MRA) and interrelationships between the geometry of carotid artery and clinical factors. Materials and Methods : 216 consecutive patients who performed supraaortic CE-MRA with fast spoiled gradient-echo imaging were included. Their medical records were reviewed for variable information including risk factors predictive of generalized atherosclerotic disease (age, hypertension (HTN), diabetes mellitus, hyperlipidema, and smoking), sex, body weight, height, and body mass index (BMI). We reviewed the CE-MRA with carotid origin (3 types), carotid artery tortuosity, angle of internal carotid artery bifurcation, the type of aortic arch branching, and the presence of the coiling of carotid artery. Results : Multinomial logistic regression analysis showed that significantly contributed clinical backgrounds for carotid origin were the age and the BMI. With an increase of age at 1, the probability that the type of carotid origin become from type 1 to type 2 was 0.9 times (p=0.004) in right carotid artery (RCA), 0.9 times (p = 0.031) in left carotid artery (LCA), 0.9 times that are likely to be type3 from type 2 (p<0.001) in RCA and 0.9 times in LCA (p=0.009). Increase in BMI at 1 increased odds of becoming type 2 as 1.1 times (p = 0.067) in RCA, 1.1 times (p=0.009) in LCA and increased chance of becoming type 3 as 1.2 times (p = 0.001) in RCA, 1.2 times (p=0.003) in LCA. Mean value of right and left carotid tortuosity were $240.9{\pm}69.0^{\circ}$and $154.4{\pm}55.0^{\circ}$, respectively. Conclusion : The BMI, age, sex and presence of HTN affects the geometry of carotid arteries, the site of origin and tortuosity of carotid artery specifically.
