Physiological Effects of Curcumin Extracted by Supercritical Fluid from Turmeric (Curcuma longa L.) (강황(Curcuma longa L.)으로부터 초임계 유체 추출한 curcumin의 생리활성)

  • Physiological effects of curcumin, major yellow-colored pigment in tumeric (Curcuma longa L.), extracted by traditional extracting methods, ethanol and hot-water extractions, and supercritical fluid extraction (SFE) using supercritical carbon dioxide as new extracting method. Antioxidative activity of ethanol extract was higher than those of SFE and hot-water extracts. Results of Ames mutagenicity test on SFE, ethanol, and hot-water extracts revealed no mutagen in the extracts. Antimutagenicity rates of SFE, ethanol, and hot-water extracts against direct mutagen, 2-nitrofluorene (2-NF), were 20.1, 9.3, and 15.2%, respectively. Antimutagenicity rate of SFE extract against TA98 derived from indirect mutagen, 2-acetamidofluorene (2-AF), was 12.2%, whereas none was observed in ethanol and hot-water extracts. Nitrite-scavenging ability of SFE extract was higher than those of ethanol and not-water extracts.

  • Genetic polymorphisms due to variation in the number of tandem repeats of DNA sequences(VNTRs) provides a useful means for discrimination between individuals. Allele and genotype frequencies of the highly polymorphic Human Thyroid Peroxidase(TPO) gene were determined in Korean population samples by using PCR followed by polyacrylamide gel electrophoresis, a procedure called the amplified fragment length polymorphism(Amp-FLP) technique. In 123 unrelated Korean individuals 10 different alleles and 29 genotypes were observed. The TPO gene demonstrated a heterozygosity of 0.707 and the power of exclusion(POE) was 0.945. The probability of having the same DNA band within two unrelated individuals was 14.6$\times10^{-2}$. The distribution of observed genotypes conformed to Hardy-Weinberg equilibrium($x^2$=4.48, 0.05

Short Culm and Early Maturing Mutants Induced by Gamma Irradiation in Rice I . Mutation Rate and Variability (감마선 조사에 의한 수도의 단간 및 조숙돌연변이체 I. 변이체의 출현빈도 및 변이분포)

  • For selection of mutants two rice varieties, Sangpungbyeo and Seomjinbyeo, were irradiated with 20 kR and 25 kR of gamma rays. One panicle per each plant was harvested in Ml, and single needling per hill was planted for M2 and M3 generations. Mutations for short culm, earliness and the major traits were examined. 1. The mutation rates were varied with the rice varieties and the doses of radiation, higher in Sangpungbyeo and 25kR than in Seomjinbyeo and 20 kR, respectively. 2. The rates in Sangpungbyeo with the 20 kR and 25 kR were 1.10% and 1.47%. respectively and those of Seomjinbyeo were 0.51% and 1.25%, respectively. 3. The culm lengths of short-culm mutants derived from Sangpungbyeo and Seomjinbyeo were reduced about 10% compared to their mother varieties, especially it was possible to select the dwarf mutants reducing 57% and 40% of culm length in Sangpungbyeo and Seomjinbyeo, respectively with the dose of 25 kR irradiation. 4. The range of heading date of the mutant ion in the M3 generation was comparatively wide. Many earliness mutants shortened about 7 days were selected as compared with their mother varieties, some mutants of the irradiated group had early matured by 20 days and 30 days, respectively in both Seomjinbyeo and Sangpungbyeo.

In order to obtain useful mutants such as early maturity, resistance to lodging, high protein and oil content, and capability of high yield, dormant seeds of two soybean varieties, Jang Dan Baik Mok and Clark, were treated with ${\gamma}$-ray, Ethyl Methane Sulfonate(EMS), Ethylene Imine(EI)and combinations of ${\gamma}$-ray and EMS or EI. 2. The germination rate and survival rate in a variety Jang Dan Baik Mok were significantly decreased with ${\gamma}$-ray treatment while it was not the same in the Clark variety. A significant decrease for seedling height measured at 14 and 21 days after sowing was found with the increase of ${\gamma}$-ray dose in both varieties. 3. Germination rates in both varieties were significantly decreased as EI concentration increases, particularly severe damage in germination was observed at 0.008 Mo. concentration. Germination rate damages were found with EMS concentration increases in the variety Jang Dan Baik Mok while no regular responses in seedling height were observed in the variety Clark. 4. Germination rate was significantly lowered with the combined treatment of EMS and ${\gamma}$-ray 24KR than that of EMS alone. In the treatments of ${\gamma}$-ray with three levels of EI concentration, the combined treatments except 24KR+EI 0.002 Mol. resulted in better germination than of EI alone. In both varieties, significant reduction in seedling height was observed in the combined treatments of ${\gamma}$-ray with various concentrations of EMS, whereas stimulation effect on seedling height was found with treatment of ${\gamma}$-ray EI +0.08Mol. 5. Germination rate, survival rate, and plant height as criteria of radio sensitivity, variety Jang Dan Baik Mok is moze sensitive to ${\gamma}$-ray, EMS, and EI than the variety Clark, and the varietal difference in responsibility to mutagen may be due to the genetic-constitution of the varieties.

  • 쌀바구미의 유충을 3기로 구분하여 각종 농도의 Apholate를 처리하여 그의 치사율$\cdot$발육속도$\cdot$산란수$\cdot$불임률 및 그의 지속성 여부를 조사하여 다음과 같은 결과를 얻었다. (1) 당대 치사율은 농도가 높아짐에 따라 높아 켰으며, 유령기 처리시의 치사율은 노령기 처리시보다 높았다. (2) 차대 치사율은 처리된 성에 따라 차가 있었고 전체적으로 자충 처리시가 자충 처리시보다 높아 치사돌연변이 유기율이 웅충 처리시에 높아짐을 알 수 있었다. (3) $0.5\%$ 이상의 처리구에서는 우화 성충 중에 각종 기형이 출현하였으며, 그 수는 농도가 높아짐에 따라 컸다. (4) 산란수의 감소는 자충 처리시가 웅충 처리시보다 심하였으며, 이것은 Apholate의 난자형성에 미치는 직접적인 영향이라고 생각된다. (5) 발육속도는 노령기 처리시가 유령기 처리시보다 늦어지며, 그 효과는 농도의 증가에 따라 높아진다. 차대의 생육기간은 자충 처리시가 웅충 처리시보다 길어졌다. (6) 화학적 불임제는 당대의 영향뿐 아니라 차대에도 그 영향력이 지속된다.

  • 서울의 지하철은 하루 450만 명이 이용하는 중요한 대중교통수단임에도 불구하고 역구내의 먼지에 대한 단순 측정ㆍ분석만이 이뤄지고 있을 뿐 그 먼지의 생물학적 영향에 대한 연구는 미흡한 실정이다. 본 연구에서는 지하철역 구내에서 포집한 먼지가 돌연변이원을 함유하고 있는 지를 평가하였다. 신촌역 지하철입구, 대합실, 승강장에서 포집한 총부유분진 (TSP; total suspended particulates)을 용매에 용출시켜 공시재료로 사용하였다.(중략)

  • Trimorphomyces papilionaceus dikaryon 으로부터 자영적인 돌연변이에 의해 분리한 이배체를 대상으로 생장속도, U. V. 광 조사 후의 생존율, U. V. 분광광도계와 Hoechst 33258 핵염색에 의한 DNA 함량등을 monokaryon 및 dikaryon 과 비교하여 결정하였다. 이배체는 dikaryon 핵의 DNA 양과 비슷하였고 monokaryon 핵의 DNA 양의 2배 정도가 되었다. Glyoxalase II 의 band 양상에서 monokayon은 한 개의 isozyme band 만을 보인 반면, 이배체와 dikaryon은 같은 위치에서 두개의 isozyme band 가 관찰되었다. 이원전개한 전기영동실험에서 dikaryon 특이 단백질은 분자량이 66.000 보다 컸으며 분자량 24,000-29,000 사이에서 이배체 특이 단백질이 존재한다.

  • Haematococcus pluvialis is a great producer of astaxanthin (3,3'-dihydroxy-$\beta$,$\beta$-carotene-4,4'-dione). The activities of astaxanthin include potential cancer prevention, immune response enhancement, antioxidant activity, and so on. Nevertheless, it tried to manipulate by mutation for overcoming low growth rate of wild type and limited production of astaxanthin. Mutated colony that is lager and more reddish one than wild type was selected by attempting to expose strains to UV irradiation and to treat chemical such as EMS and colchicines as mutagen. Selected mutants were further screened using inhibitors of the carotenoid biosynthetic pathway. Inhibitors used were nicotine and diphenylamine and both had decreased the survival rate by 40-50%. Among over 50,000 mutant colonies screened, two strains were selected. One selected mutant strain (U15-5) from UV treatment showed 1.68-fold higher total carotenoid contents per cell than that of the wild type strain. On the other hand, the other selected mutant strains (DS, M4-3) from colchicine treatment showed 20$\sim$30% faster cell growth than the wild type strain.

Enhanced Production of Astaxanthin in Paracoccus haeundaensis Strain by Physical and Chemical Mutagenesis (물리·화학적 돌연변이 유도를 통한 Paracoccus haeundaensis의 astaxanthin 생산량 증대)

  • Carotenoids are natural lipid-soluble pigments, which are produced primarily by bacteria, algae, and plants. Many studies have focused on the identification, production, and utilization of natural sources of astaxanthin from algae, yeast, and crustacean byproducts as an alternative to the synthetic pigment, which is mostly used today. The aim of the present study was to identify a mutant of Paracoccus haeundaensis by exposure to UV and ethyl methanesulfonate (EMS). The mutant was then exposed to nutrient stress conditions to isolate an astaxanthin-hyperproducing strain, followed by characterization of the mutant. The survival rate decreased in accordance with an increase in the UV exposure time and an increase in the EMS concentration. A mutant of the original P. haeundaensis strain was identified that showed hyperproduction of astaxanthin following exposure to UV irradiation (20 min) and EMS treatment (0.4 M concentration). The optimal culture conditions for the PUE mutant were $25^{\circ}C$, pH 7-8, and 3% NaCl. The effects of various carbon and nitrogen sources on the growth and astaxanthin production of PUE were examined. The addition of 1% raffinose and 3% potassium nitrate influenced cell growth and astaxanthin production. The selected mutant exhibited an increase of 1.58 folds in astaxanthin content compared to initial wild type strain. A genetically stable mutant strain obtained using mutagen (UV irradiation and EMS treatment) may be a suitable candidate for further industrial scale production of astaxanthin.
