Production of Sedum Extract Adding Jelly and Assessment of Its Physicochemical Properties (돌나물 즙을 첨가한 젤라틴 젤리의 제조 및 품질특성)

  • Sedum sarmentosum, also blown as stonecrop (dolnamul), is a widely consumed herb, and is used as an ingredient in salads in Korea. Unfortunately, sedum is perishable and vulnerable to tissue damage during preservation. Therefore, this feasibility study was performed in order to increase the availability of sedum and increase its value. Various concentrations of sedum extract (0.5-3%) were added to gelatin jelly, and their physicochemical properties were determined. The ascorbic acid content of the sedum jelly increased in proportion to the sedum extract concentration. Calcium content of the sedum jelly was 4 to 28 times higher than that of the control. Contrary to the control, iron was detected in the sedum jelly (0.023-1.031 mg/100 g dry weight). Furthermore, magnesium and potassium levels were higher in the sedum extract groups. There were significant differences (p < 0.05) in greenness (a value) and yellowness (b value) between the control and the sedum extract groups. However, significant differences between the 2% and 3% sedum extract groups were not detected. As sedum extract concentration increased, the pH level of soft jellies (solid state) decreased. Therefore, hardness and gumminess were decreased significantly. These results are in agreement with the sensory evaluation. According to sensory tests, the values for palatability, appearance, and color in the 2% sedum extract group were higher than those of the 0.5-1% and 3% sedum extract groups.

Taxonomic Study of genus Sedum and Phedimus (Crassulaceae) in Korea Based on External Morphology (외부형태 형질에 근거한 한국산 돌나물과내 돌나물속과 기린초속의 분류학적 고찰)

  • The genus Sedum comprises about 400 taxa with worldwide distribution, and about 20 taxa of the genus Phedimus distributed from Asia to Europe. According to some taxonomists, Phedimus and Sedum are two separate genera of the family Crassulaceae; however, in Korea Phedimus has been treated as a subgenus Aizoon within the genus Sedum. This ambiguous taxonomic treatment of the genus Phedimus needs further confirmation. In the study, we investigated morphological characteristics such as the whole plant height, leaf morphology, and seed coat features of these two genera. We believe that these characters are very important to distinguish Phedimus and Sedum. The result indicated that the seed surface pattern was the most useful diagnostic character. The Sedum had a hexagonal pattern with a single nipple protrusion, whereas the Phedimus had a rectangular pattern with papilla on both ends. The results of this study strongly support the separation of Phedimus from Sedum and thus Phedimus need to be treated as an independent genus.

The Effects of Sedum sarmentosum Bunge on Collagen Content of Connective Tissues in Ovariectomized Rats (돌나물이 난소를 절제한 흰쥐 결합조직 중의 Collagen 함량 변화에 미치는 영향)

  • The aim of this study was to evaluate the effects of Sedum sarmentosum Bunge (SS) on the collagen content of the connective tissues in ovariectomized rats. From day 2 until day 37 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with SS at 50 mg/kg BW/day (OVX-SS50), 100 mg/kg bw/day (OVX-SS100) and 200 mg/kg bw/day (OVX-SS200), and an ethyl ether fraction of SS at 10 mg/kg bw/day (OVX-EE) and an ethyl acetate fraction of SS at 10 mg/kg bw/day (OVX-EA). The SS ethanol extracts and SS fractions were orally administrated 1 mL per day. The OVX rats were significantly heavier than the sham-operated rats at all time points, but supplementation with the SS extracts tended to gain weight less than OVX-control. Supplementation with the SS extracts prevented a decrease in the collagen level in bone and cartilage tissues. These results are consistent with the conclusions based on the estrogenic activities of SS. Therefore, it may be used to possibly improve the quality of life in menopausal women.

An unrecorded species in Korean flora: Sedum tosaense Makino (Crassulaceae) (한국산 미기록 식물: 주걱비름 (돌나물과))

  • Sedum tosaense Makino (Crassulaceae), an unrecorded species in Korean flora, was collected from parasitic crater, Sangumburi in Jocheon-eup Bukjeju-gun, Jeju-do. This species is most similar to S. bulbiferum and S. orizifolium, but differs in some substantial characters. S. tosaense is perennial and stoloniferous herb with bulbils on stems, while S. bulbiferum is annual or biennial herb without bulbils in leaf axil. In addition, S. tosaense has leaves emarginate at apex whereas S. bulbiferum has leaves somewhat uneven at apex. the leaves of S. tosaense are conspicuously flattened and spatulate in shape at the stem without flowers, but those of S. orizifolium, are terete or slightly flattened and linear to narrowly oblong in shape.

A test of the hybrid origin of Korean endemic Sedum latiovalifolium (Crassulaceae) (한국특산 태백기린초(돌나물과)의 교잡 기원에 대한 검증)

  • Eighteen morphological characters from 40 populations and ten isozyme loci from 35 populations of Sedum latiovalifolium and related species were examined to investigate the degree of morphological and genetic variation. The high-frequency marker alleles $MDH-2^a$ and $PGI-1^a$ in S. aizoon, S. kamtschaticum, and S. zokuriense did not appear in the populations of S. latiovalifolium. In addition, the high-frequency allele ($MDH-2^c$) in S. latiovalifolium appeared at a very low frequency in other subg. aizoon species. Thus, the isozyme data did not support a hybrid origin of S. latiovalifolium from S. aizoon with S. kamtschaticum. The results of an unweighted pair-group method using the arithmetic average method and a principal components analysis using morphological data also did not support a hybrid origin of S. latiovalifolium. However, our data strongly suggest that some individuals from the populations found in the Gumdaebong area were most likely hybrids due to introgression between S. latiovalifolium and S. kamtschaticum or S. aizoon and S. kamtschaticum.

The Effects of Water Stress on C$_3$ Plant and CAM Plant (C$_3$ 식물과 CAM 식물에서 수분 스트레스의 효과)

  • The differences of several kinds of physiological responses between Commelina communis (C$_3$ plant) and Sedum sarmentosum (CAM plant: Crassulacean Acid metabolism) when both plants were exposed to water stress for 3 weeks were investigated. In case of Commelina it was shown a clear loss of water to 12% in three weeks, but no changes were observed in Sedum. Total chlorophyll content was also reduced to 57% in Commelina but not clear changes of chlorophyll content in Sedum. were observed for three weeks. In chlorophyll fluorescence experiments Fv/Fm ratios were reduced to 19% in Commelina, but no changes were observed in Sedum. There were very sensitive responses according to the different KCl concentrations and the stomatal aperture of epidermal strips was 12.8 ${\mu}m$ at 200 mM KCl in Commelina, but less than 3 ${\mu}m$ was observed at the same KCl concentration in Sedum. In addition, there were no chloroplasts in guard cells of Sedum, but most plants had chloroplasts including Commelina. From the above results, the ability of water stress resistance in Sedum. could be come from slow physiological metabolism including growth and less loss of water through unique stomatal characteristics.

The Effects of Sedum sarmentosum Bunge on Serum Lipid Concentration in Ovariectomized Rats (돌나물이 난소 절제한 흰쥐의 혈 중 지질 함량에 미치는 영향)

  • The aim of this study was to investigate in an in vivo model the effects of ethanol extract and ethyl ether and ethyl acetate fractions of Sedum sarmentosum Bunge (SS). To investigate the effects of ethanol extract and ethyl ether and ethyl acetate fractions, Sprage-Dawley female rats were randomly assigned to groups as follows : Sham, OVX-control and ovariectomized rats supplemented with SS at 50 mg/kg bw/day, with SS at 100 mg/kg bw/day, with SS at 200 mg/kg bw/day, with ethyl ether fraction of SS at 10 mg/ka bw/day and with ethyl acetate fraction of SS at 10 mg/kg bw/day. Food intake and food efficiency ratio were not significantly different in groups. But body weight gain was decreased by supplementation of ethyl acetate fraction of SS compared to OVX-control. The levels of total cholesterol and triglyceride in serum were higher in ovariectomized control rats than Sham-operated rats, but supplementation of SS ethanol extracts at 200 mg/kg bw/day decreased the level of the lipid in serum. The level of HDL-cholesterol in serum increased by supplementation of SS ethanol extracts at 200 mg/kg bw/day (p<0.01). The levels of total cholesterol and triglyceride in serum were decreased by supplementation of ethyl acetate fraction of SS (p<0.01). On the other hand, the level of HDL-cholesterol in serum was increased by supplementation of ethyl acetate fraction of SS (p<0.05). On conclusion, it might be expected that ethanol extract and ethyl acetate fraction of SS is believed to be a possible protective effects for the fatty serum increasing serum lipid.

Notes on genetic variation in Sedum sarmentosum (Crassulaceae): Implications for the origin of southern Korean populations (돌나물(돌나물과)집단의 유전적 변이: 남부지방 집단의 기원에 대한 암시)

  • The succulent herbaceous perennial Sedum sarmentosum commonly grows in the southern part of the Korean Peninsula. It is a species native to China, most likely introduced into Korea due to its edible and medicinal uses. If plants were introduced from a single source, we would expect no or low levels of genetic variation in Korean populations. Alternatively, if plants were introduced from multiple sources, we would expect, in contrast, high levels of genetic diversity. To test which is more likely, we surveyed the degree of allozyme variation in ten populations of this species from southern Korea. We found that S. sarmentosum was monomorphic at all fifteen allozyme loci. However, two congeners (S. polytrichoides and S. kamtschaticum) and two related species (Hylotelephium ussuriense and H. verticillatum) maintain moderate to high levels of genetic diversity ($H_e=0.144$, 0.203, 0.201, and 0.204, respectively). We suggest that southern Korean populations of S. sarmentosum likely descended from a single introduction of a few plants and then became naturalized exclusively via vegetative spreading (as plants in Korea, but also as occurs in other parts of its native and naturalized range, are sterile).
