Physiological activity of Flammulina velutipes sp. ethanol extract (팽이버섯 에탄올 추출물의 생리활성)

  • This study was carried out to investigate the physiological activities of the ethanol extracts from 11 strains of Flammulina velutipes. In addition, the ${\beta}$-glucan and polyphenol contents were also measured. The contents of polyphenol and ${\beta}$-glucan of Flammulina velutipes were found to be more than 100 mg% and 20%, respectively, one of which showed 244.74 mg% and 27.37%, respectively. Further, The highest inhibitory activities on DPPH radical, ${\alpha}$-amyloglucosidase activity and nitric oxide production were 91.74%, 63.57% and 61.44%, respectively. However, ethanol extracts from all the strains have little effects on Angiotensin I converting enzyme(ACE)-inhibitory effects. Finally, the highest cytotoxic activities of ethanol extracts from all the strains on A549 and HT-29 cell were 76.07% and 67.05%, respectively.

Immune Enhancing Effects of Intracellular and Extracellular Polysaccharides Extracted from Mycelial Cultivate of Agaricus blazei Murill (신령버섯(Agaricus blazei Murill) 균사체내외 다당체의 면역활성효과)

  • This study was performed to compare in vitro immune enhancing effects of polysaccharides extracted from cultivated mycelia of Agaricus blazei Murill. Carbohydrate contents of semi-purified polysaccharides were 85.6% and 95.3%, while ${\beta}$-glucan conents were 67.9% and 88.1% for intracellular and extracellular polysaccharide, respectively. Samples were adjusted to the same in their carbohydrate contents before efficacy tests. Both intracellular and extracellular polysaccharide increased nitric oxide (NO) synthesis of macrophage RAW 264.7 in dose dependent manner, and the maximum increase rate was 53.9 and 53.1% in intracellular and extraceltular polysaccharide, respectively. The polysaccharides also increased synthesis of cytokines such as interleukin (IL)-$1{\beta}$, IL-6 and tumor necrosis factor (TNF)-${\alpha}$ in RAW 264.7. For all the 3 cytokines, the increase rate of synthesis was much higher in extracellular polysaccharide compared to intracellular polysaccharide, especially at low concentration. Both polysaccarides increased the proliferation of splenocytes in vitro, intracellular polysaccharide showed increase in dose dependent manner while extraceltular polysaccharide showed increase untill medium concentration ($250\;{\mu}g/ml$). They did not show direct cytotoxicity against cancer cells such as B16F0 melanoma. As results, it was regarded that the both intracellular and extracellular polysaccharide from A. blazei showed immune enhancing effects in vitro, but the activity is higher in extracellular polysaccharide compared to intracellular polysaccharide.

Pharmacological Effects of Proteoglycans Extracted from Fruiting Bodies of Fomitella fraxinea (장수버섯 자실체의 열탕추출액으로부터 분리한 단백다당체의 약리적 효과)

  • Anti-complementary assay for immuno-stimulating polysaccharide screening, human tumor colony-forming assay for discovering anti-tumor drugs, and toxic assay against mouse were performed to examine pharmacological activities of polysaccharides extracted from fruiting bodies of Jang-soo mushroom (Fomitella fraxinea). Hot water $(100^{\circ}C,\;FF-I)$, 1% ammonium oxalate solution $(80^{\circ}C,\;FF-II)$, and 5% sodium hydroxide solution $(80^{\circ}C,\;FF-III)$ were used for extraction of three polysaccharides from fruiting bodies of it. Anti-complementary activity of FF-I was more effective than the others. FF-I was further fractionated into three groups of polysaccharide by DEAE-Sephadex A25 column chromatography (FF-NP, FF-AP1, and FF-AP2). FF-AP1 showed not only the highest anti-complementary activity but also the growth-inhibitory activity against Snu-I (human stomach cancer cell) among 9 kinds of human tumor cell lines. But FF-AP2 exhibited its activity against Hep-2(larynx cancer) and KB(mouth epidermal cancer) cell lines at $500\;{\mu}g/ml$ although its anti-complementary activity was lower than it of FF-AP1. When FF-I was orally administrated to mice with dosage of 5000 mg/kg, no remarkable changes were observed in viewpoint of tissue-pathology.

Acute Oral Toxicity Studies of Extract of Sanghwang Mushroom (Phellinus linteus) (재배 상황버섯 추출물의 경구투여 급성독성 연구)

  • The current study was performed to determine the acute oral toxicity of a crude extract of sanghwang mushroom (Phellinus linteus), in SD rats. 5 rats of each sex were orally treated with a single dose of extract of sanghwang mushroom at doses of 0, 500, 1,000, 2,000 mg/kg, respectively. After the treatment, clinical signs and body weight change, the food and water consumption were observed for 14 days. All animals survived during the study and did not show any clinical signs. Body weight gain showed no significant difference between the control and treated rats. However, body weight gain delayed in high dose group (2,000 mg/kg) on day 1~3 after administration. Another 5 rats of each sex were orally treated with a single dose of extract of sanghwang mushroom at dosages 4,000, 5,000 mg/kg respectively, but all animals survived during the study and did not show any clinical signs. It is suggested that LD$_{50}$ of extract of sanghwang mushroom by oral administration was estimated to be over 5,000 mg/kg in both sexes of rats.s.

Studies on Toxic Components of Auricularia polytricha (털목이버섯의 독성(毒性)에 관한 연구(硏究))

  • To screen biologically active components of the higher fungi of Korea, the dried carpophores of Auricularia polytricha were extracted with water. The extract was examined for acute toxicity in ICR mice. A low molecular weight toxin of this fungus was purified by acetone precipitation followed by cellulose, silica gel and LH-20 Sephadex column chromatography. Major symptoms of this toxin were eye extrusion, hair erection, trembling of head, paralysis, rapid running or moving before death and depression of respiration. The median lethal doses of the total extract were 1.25 g/kg and 4.18 g/kg by i.p. and p.o. administrations, respectively. The amounts of one mouse lethal unit (MLU) of the total extract and final fraction that killed a 20-g mouse within 30 minutes were 28.5 mg/mouse and 12 mg/mouse, respectively.

담자균류의 약효성분의 개발에 관한 연구

  • 영지와 구름버섯의 원형질체 융합 균주 F-1의 배양 균사체로 부터 열수추출한 분획 Fr.I을 DEAE-cellulose ion chromatography와 gel filtration chromatography를 통하여 분획 Fr.II, III, IV,로 분리 정제하였다. Sarcoma 180에 대한 종양억제율을 검사한 결과 Fr.IV는 68.73%로 가장 우수하였으며 동계 복수암에 대한 수명 연장 효과도 140 %의 유의적인 결과를 얻었다. 면역 관련 장기의 중량에 대한 영향을 실험한 결과 정상군에 비해 간, 비장 및 흉선의 중량을 증가시켰고, 마우스 암세포에 대한 직접적인 세포독성 작용을 보이지 않았으나 면역 실험을 실시한 결과, 마우스에서 용혈반 형성세포수를 1,36배 증가시켰으며 암이식군에서 감소된 T lymphocyte활성을 정상수준까지 회복시켰고 또, macrophage의 superoxide anion 분비를 2.25배 증가시켰다. 이로써 이 항암 성분은 면역세포를 활성화시켜 항암 효과를 나타냄을 의미한다. Fr.IV히 분자량은 7.9$\times$$10^4$ dalton이고 75,57%의 다당체와 4.47%의 단백질로 이루어졌으며 그 다당체는 주로 glucase, xylose와 mannose로 구성된 heteropolysaccharide이었으며 그 단백질은 Alanine과 Valine을 위시한 15종의 아미노산을 함유하고 있음이 확인되었다.

Protective Effects of $\beta$-Immunan Isolated from the Mycelium of Ganoderma lucidum IY009 against Cisplatin-induced Nephrotoxicity (영지버섯 균사체(Ganoderma lucidum IY009)로부터 추출한 $\beta$-Immunan의 시스플라틴 유발 신독성 보호효과)

  • $\beta$-Immunan was proteoglycan obtained from mycelium of Ganoderma lucidum IY009. In this study, the protective effects of $\beta$-Immunan, against the CDDP induced in vitro cytotoxicity and in vivo renal toxicity, was measured. Concentration dependent cytotoxicities of CDDP in normal kidney cells (Vero, TCMK-l) were reduced by $\beta$-Immunan treatment. Increased renal toxicity factors, such as elevation of blood urea nitrogen (BUN) and serum creatinine, reduction of kidney weight and malonidialdehyde (MDA), by intraperitoneal administration of CDDP in rats was improved. These results indicated that $\beta$-Immunan have a protective effects against the CDDP induced renal toxicity, however, it needed to confirm the detailed mechanism for therapeutic effects.

  • 식품에서의 갈변현상은, 외관을 손상시킴으로 인해 상품가치를 저하시킬 뿐만 아니라 경우에 따라서는 독성물질을 생성하는 등의 문제를 일으킨다. 대표적인 것으로 사과ㆍ바나나의 껍질을 벗긴 후에 진행되는 갈색화 또는 수산물에서의 새우ㆍ가재류의 흑변 현상이 잘 알려져 있다. 이러한 현상은, polyphenoloxidase(PPO, EC 1.14.18.1)에 의해 유리 tyrosine등의 phenol성 화합물이 quinone체로 산화하고, 생성 된 quinone 또는 유도체들은 중합되어 melanine색소를 형성하는 것이 원인으로 알려져 있다. 현재, 화학적 합성품인 4-hexylresorcinol의 침가가 흑변 현상의 방지책으로서 인정되어 있으나, 최근 소비자들의 사고를 고려할 때, 안전성이 높은 천연물 유래의 유효한 첨가물의 개발이 요망되고 있다. Studies on the toxicity of auricularia polytricha (털목이버섯의 독성에 관한 연구)

  • To screen biologically active components of the higher fungi of Korea, the carpophores of Auricularia polytricha, a well-known edible mushroom, were extracted with 0.14M NaCl solution. The extractive was successively fractionated by adding ammonium sulfate in various amounts, the respective precipitates being weparated by centrifugation, dialyzed and freeze-dried. When a dose of 60mg/kg of each was, i.p., injected into ICR mice, the fraction shich was precopitated at 20% (NH/sub 4/)/sub 2/SO/sub 4/ showed the highest toxicity, killing seven mice within two days. The fraction obtained at 40% (NH/sub 4/)/sub 2/SO/sub 4/ showed the second highest toxicity. The two fractions were named auratoxin I and II after the genus name. The symptoms of the intoxication were convulsion during the first 30 minutes after the injection, then sleeping within an hour, and tremor, lacrimation, nasal and ophthalmic bleeding, congestion and death in 24 hours. Particularly the spleen of the mice was fonud to be enlarged remarkably. The chemical analysis of the toxins showed that auratoxin I consisted of 4.4% protein and 84.5% polysaccharide and that auratoxin II 35.8% protein and 48.0% polysaccharide.

Lipolytic Effect of Sparassis crispa Extracts in Differentiated 3T3-L1 Cells and High Fat Diet-induced Obese Mice (분화된 3T3-L1 세포와 비만유도 쥐에 꽃송이버섯 추출물의 지질분해 효과)

  • The purpose of this study was to investigate whether water extracts of Sparassis crispa (SC) have anti-obesity effects. Treatment of mature adipocytes with SC caused a decrease in lipid accumulation (assessed by Oil Red O staining) and an increase in glycerol release. Mice were induced to obesity by a high fat diet (45% fat in total kcal) and experimental groups were treated with two different dosages of SC extracts, a low SC (LSC, 100 mg/kg/day) or high SC (HSC, 300 mg/kg/day). SC extracts were administered by gavages for 10 weeks in the experimental groups, while the control group was fed with distilled water. The body weight gain of mice fed SC was significantly reduced (11.88% lower in LSC, 14.54% lower in HSC) compared to the control group. Additionally, there were significantly reduced serum levels of triglycerides (13.57% lower in LSC, 19.46% lower in HSC), total cholesterol (32.22% lower in LSC, 24.67% lower in HSC) and glucose (28.85% lower in LSC, 25.82% lower in HSC) in mice fed SC compared to the control group. Hepatic triglycerides in mice fed SC were lower (9.68% lower in LSC, 14.24% lower in HSC) than the control group and total cholesterol levels were also lower in mice fed SC (38.72% lower in LSC, 35.20% in HSC). These results demonstrate that the water extract of SC may enhance lipolysis and up-regulate the expression of lipolytic enzymes in vitro and reduce body weight in vivo. These significant effects were found for both low and high doses of SC treatment, and suggest SC can be used as potential therapeutic substances for the prevention and treatment of obesity.
