Immuno-enhancing Effect of Seed Extracts on a RAW 264.7 Macrophage Cell Line (종자 추출물의 RAW 264.7 세포에 대한 면역증강 효과)

  • In this study, the immuno-enhancing activity of seed extracts were studied on the macrophage cell lines. We examined the effect of nine seed extracts on nitric oxide (NO) production in RAW 264.7 cells and selected four highly-effective seed candidates (Fagopyrum esculentum, Taraxacum platycarpum, Impatiens balsamina, Helianthus annuus) for further immune-related studies. The effects of the four seed extracts on the production of immune-related cytokines in the RAW 264.7 macrophage cell line and proliferation of Molt-4 as a T cell line were investigated. The secretion of NO from the RAW 264.7 cells was increased up to 39 ${\mu}M$ by adding the seed extracts (25 ${\mu}g/mL$) compared to the control. Also, the secretion of tumor necrosis factor-alpha (TNF-${\alpha}$) was also increased up to 32 times by adding the seed extracts (25 ${\mu}g/mL$). Secretion of cytokines such as interleukin-1 beta (IL-$1{\beta}$), interleukin-6 (IL-6), and interleukin-10 (IL-10) was also increased and induced the proliferation of T cells compared to the control. In conclusion, these results suggest that four seed extracts provide beneficial immuno-enhancing effects for human health.

Role of obioactin on toxoplasmacidal activity within mouse peritoneal macrophages (마우스 복강 macrophages내(內)의 살(殺)톡소플라즈마 활성에 있어서 obioactin의 역할)

  • The present study was undertaken to examine the effects of obioactin, lonomycin A, and MDP on toxoplasmacidal activities in glycogen-induced mouse peritoneal macrophages. The killing effect of obioactin on Toxoplasma multiplication was increased significantly in proportion to its concentrations. $O_2{^-}$ generation in obioactin-treated macrophages was also increased from twofold to threefold when compared with that of untreated control. Similarly, $H_2O_2$ continued to rise in parallel with increase of the concentration of obioactin. Lonomycin A-treated macrophages also exhibited a good effect of dose-response on toxoplasmacidal activities. However, $O_2{^-}$ and $H_2O_2$ were not generated significantly in lonomycin A-treated macrophages. Macrophages treated with muramyl dipeptide (MDP) were not found to inhibit the prolifi:ration of Toxoplasma but showed the enhancement of $O_2{^-}$ and $H_2O_2$, generation. The released lysozyme levels from macrophages into cultured media were decreased tn dose-dependent fashion by in vitro treatment of obioactin, lonomycin A, and MDP. The intracellular lysozyme levels appeared to be a constant value regardless of increasing the concentrations of obioactin, lonomycin A, and MDP. Therefore, these results suggest that Toxoplasma multiplication within macrophages treated with obioactin was inhibited by the generation of $O_2{^-}$ and $H_2O_2$ and that lysozyme per se within or released from macrophages had no effect on toxoplasmacidal activity.

Effect of the Extruded Ginseng on Antioxidant Activity (압출 성형 인삼의 항산화 활성 증강 효과)

  • This study was conducted in order to evaluate the antioxidant activity of extruded ginseng in different extracted fractions. Each of the fractions obtained from extruded ginseng and ginseng (control) were extracted with 80% ethanol, and then the lipophilic components were removed with ether while the hydrophilic components were separated with water-saturated butanol. Each of the 80% ethanol/butanol/water layers were collected and evaporated to acquire samples for tests of saponin content and antioxidant activity. The antioxidant activity of extruded ginseng fractions and ginseng fractions were determined via the oxygen radical absorbance capacity (ORAC) assay. Overall, the extruded ginseng samples harbored saponin contents of 2.2 (Rg1), 2.3 (Re), 1.2 (Rc), 1.3 (Rb2), and 2.2 (Rd) times that measured in the ginseng prior to extrusion. Antioxidant capacity was also higher, not only in the 80% ethanol/butanol which harbor a significant quantity of saponin, but also in the water fractions, which harbor relatively low quantities of saponin as compared to the control samples.   • Chrysanthemum indicum L. (Asteraceae) is a traditional herbal medicine that has been used for the treatment of inflammation, hypertension, and respiratory diseases due to its strong antagonistic activity against inflammatory cytokines. The effects of Chrysanthemum indicum L. Extract (CIE) for increasing cell growth, alkaline phosphatase (ALP) activity, and collagen content were totally inhibited, suggesting that the effect of CIE might be partly involved with estrogen activity. Furthermore, the protective effects of CIE on the response of osteoblasts to oxidative stress were evaluated. Osteoblastic MC3T3-E1 cells were incubated with hydrogen peroxide and/or CIE, and markers of osteoblast function and oxidative damage were examined. CIE significantly increased cell survival, ALP activity, and calcium deposition, and decreased the production of Reactive Oxygen Species (ROS) and Tumor Necrosis Factor-${\alpha}$ (TNF-${\alpha}$) in osteoblasts. Taken together, these results indicate that the enhancement of osteoblast function by CIE may prevent osteoporosis and inflammatory bone diseases.

Signal Change of Normal Saline by Oxygen Injection in FLAIR Image (산소주입에 의한 FLAIR 영상에서 생리식염수의 신호 변화)

  • It was reported that there were some cases in which signal was not inhibited but high signal appeared in cerebrospinal fluid on FLAIR(fluid attenuated inversion recovery) of MRI(Magnetic Resonance Imaging) in case a person inhales high-concentration oxygen. This study was to prepare basic database. We produced a phantom fixed with agar gel and by using it, obtained the images of the signals of normal saline into which oxygen was injected and normal saline diluted with contrast media by changing the TI(Inversion Time) of FLAIR technique and analyzed them. In the result of FLAIR technique of MRI using Philips Achieva MR 3.0T in Busan P Hospital, the SNR(Signal to Noise Ratio) of normal saline into which oxygen was injected was higher than the SNR of normal saline into which oxygen was not injected. However, it was not higher than the SNR of normal saline diluted with contrast media. In the TI 1,800ms, we could obtain the images which do not have the rise of the signal due to oxygen. In the CNR(Contrast to Noise Ratio) of normal saline into which oxygen was injected and normal saline diluted with contrast media as well, it was higher in the TI 1,800ms than in the TI 2,800ms that is mainly used clinically. It is thought that the result of this study could be basic database for studies on change of signal of cerebrospinal fluid as a result of injection of oxygen in FLAIR technique of MRI.

Balthazar Computed Tomographic Severity Index Application for Experimental Acute Necrotizing Pancreatitis in Dogs (개의 실험적 급성 괴사성 췌장염 평가에서 Balthazar Computed Tomographic Severity Index의 적용)

  • Computed tomography (CT) is considered as gold standard in evaluating pancreatitis in human, but there have been only a few studies in veterinary field. Balthazar CT severity index (CTSI) used to assess the severity of acute necrotizing pancreatitis in human could be applicable to dogs, because the severity of acute pancreatitis depends on the area of pancreatic necrosis in dogs more than in human. In this study, 25 adult, clinically healthy beagle dogs were used. CT examinations was performed in normal pancreas, positive control group and acute necrotizing pancreatitis induced by autologous bile injection. Balthazar CTSI was applied to canine acute necrotizing pancreatitis in contrastenhanced CT image and compared with the result of histopathologic examination. The Hounsfield unit (HU) of normal canine pancreas was $52.44{\pm}4.58$ and the density was significantly decreased in acute necrotizing pancreatitis (P < .05). In contrast-enhanced CT examination, pancreatic density was decreased significantly and this area was compatible to pancreatic necrosis. Balthazar CTSI showed positive correlation with histopathologic evaluation with a sensitivity of 100% and a specificity of 88.89%. Balthazar CTSI can be applied to evaluate the severity of acute necrotizing pancreatitis in dogs.

The response of peripheral blood lymphocytes against in vivo stimulation with mitogen in carp, Cyprinus carpio (Mitogen 투여에 대한 잉어 순환혈액 림프구의 반응)

  • This work was carried out to investigate the functional heterogeneity of peripheral blood lymphocytes(PBLs) in carp, Cyprinus carpio. PHA, Con A, LPS and BCG were injected intraperitoneally into carp to determine the blastogenic response and rosette formation activity. In each group of fish treated with stimulators, the cell numbers and DNA contents of lymphocytes were higher than those of untreated control group and reached the highest level between 1 week and 2 weeks after injection with mitogens. These results showed that BCG and Con A were strong stimulators of proliferation compared to PHA and LPS. However, PHA-treated fish twice showed the highest rosette formation response among the consecutive stimulations with the same mitogen. Alase, the results on consecutive mitogen stimulation revealed that carps reinjected by different mitogens led to an increased stimulation higher than the one reinjected after 1 week with same mitogen. It seems that different mitogens may stimulate different cell populations and implies functionally separated subpopulations of lymphocytes in carp.

Transformation of Stereoscopic Images for 3D Perception Improvement (입체영상의 3D 증강을 위한 입체영상 변환)

  • Recently, 3DTV and 3D displays have been released in the market. Accordingly, the production of stereoscopic images has gained much interest. Stereoscopic image being composed of left and right images are currently delivered to viewers without any modifications. The researches on the enhancement of depth perception using high-frequency components and the re-production of natural color by color compensation have been carried out for 2D images. The application of such 2D technologies to 3D stereoscopic images is an aim of this paper. This paper proposes the enhancement of 3D perception by color transformation. For this, we propose a stereo matching method for obtaining a depth map and two color transformation methods such as contrast transformation and background darkening. The effectiveness of the proposed method was verified through experiments.

Effect of Tea Polyphenols on Anticancer Activity and Cytokines Production (차 폴리페놀화합물의 사이토카인 생성 및 항암능에 대한 영향)

  • Theaflavins (TF) and thearubigins (TR) are constituents of tea pigments which are polyphenols derived from Korean fermentation tea. After TF, TR and [(-) epigallocatechin-3-gallate](EGCG) have been applied to macrophage cell line (RAW264.7) nitric oxide (NO) synthesis and cytokines production were estimated. Cytokines production by enzyme linked immune-sorbent assay (ELISA) determined. NO production was increased by about 1.5-folds at the dose of $80\;{\mu}g/ml$ compared to control and lipopolysaccharide (LPS) stimulation when TF, TR and EGCG were applied to a RAW264.7 cell. Interleukin-6 (IL-6), Tumor necrosis factor ($TNF-{\alpha}$) and granulocyte-macrophage colony stimulating factor (GM-CSF) increased depended on concentrations of TF, TR and EGCG. The production of tumor necrosis $factor-{\alpha}$ increased highly in TR, TF and EGCG group with LPS. These results suggest that TF, TR and EGCG have immune-enhancement effect through the cytokine production. TF, TR and EGCG inhibited cancer cell viability, the anticancer effect of these polyphenols may explain the anti-tumor promotion action and antioxidant activity of these tea constituents.

Effects of Eucalyptus and Geranium on Production of IL-2 and IL-4 in Mouse Splenocytes (Eucalyptus와 geranium이 마우스 splenocytes에서 IL-2 및 IL-4 생성에 대한 효과)

  • Aromatherpy is the controlled use of essential oils to promote health and well-being. In this work we have investigated the effect of eucalyptus and geranium on the production of interleukin-2 (IL-2) and interleukin4 (IL-4). Mouse splenocytes were incubated with essential oils. The culture supernatants of mouse splenocytes exposed with these oils were harvested to assay IL-2 and IL-4 production. The quantitative changes of IL-2 in splenocytes culture supernatants after exposure with these oils were decreased at high doses, but increased at low doses. But its of IL-4 were increased generally at high doses of eucalyptus. In case of the exposure of geranium, its of IL-4 were dose-dependently increased. These kinds of essential oils showed the probability to improve IL-2- and IL-4-related immune responses at the optimum exposure.
