Physiological Effects of TOPE, a Photo - independent Diphenylether, on Higher Plants (비광요구형 디페닐에테르계 TOPE 의 생리적 작용에 관한 연구)

  • Several physiological responses were investigated in plants treated with TOPE as a preliminary step to know its action site. Unlike photo-dependent diphenylethers, herbicidal activity of TOPE appeared slowly and its typical symptoms were both burning of leaf blades and abnormal division of meristem in grasses, Similarly, both leakage of cell electrolytes and the curling of cotyledon margin were also shown in cucumber(Cucumis sativus L.). Biosynthesis of chlorophyll in etiolated cucumber cotyledon was not inhibited directly by treatment of TOPE at low light intensity(5.5${\mu}$ mol $m^{-2}s^{-1}$ PAR) and protoporphyrin IX was not also accumulated. The contents of phytoene, phytofluene and ${\beta}$-carotene were abnormaly increased. Photosynthesis was inhibited only at high concentration. Mitochondrial respiration was inhibited at high concentration but rather increased significantly at 10${\mu}$M of TOPE. However, respiration inhibitors did not alleviate the two symptoms of TOPE in cucumber cotyledon. In the same experiments, using inhibitors of protein or nucleic acid biosynthesis, only one of the two symptoms was alleviated by chloramphenicol and cycloheximide. In contrast, both symptoms were alleviated by actinomycin-D and hydroxyurea, suggesting that nucleic acid metabolism might be preferentially related to the mode of action of TOPE. DNA, RNA and protein contents were accumulated in both cucumber cotyledon and rice (Oryza sativa L.) routs treated with TOPE, and the DNA of them was increased at first. Thus, it is conjectured that TOPE increase nucleic acid metabolism directly or indirectly, and then disturb various metabolic pathways causing abnormal physiological and morphological effects followed by final death.

Adsorption Pattern of the Herbicide, Bentazon and Its Metabolites on Soil (제초제 Bentazon과 그 대사산물들의 토양 중 흡착양상)

  • In order to elucidate the adsorption mechanism of the herbicide, bentazon and its metabolites on soil, their adsorption patterns on soil and six adsorbents were investigated with Freundlich, Langmuir and linear isotherm. Freundlich constants ($K_f$) and maximum adsorption amount($Q^0$) of bentazon on soil was 0.55 and 3.97. Kd and Koc values of it were 0.18 and 18. The all of metabolites used except deisopropylbentazon amounts sorbed on the soil were much higher than bentazon. The most of 8-hydroxybentazon was adsorbed on soil showing $K_f$ = 316.6, $Q^0$ = 3,488, Kd = 29.7 and Koc = 2,970. Bentazon, deisopropylbentazon and 8-hydroxybentazon were shown high affinity to anion exchange regardless of pH and $NH_2$ in low pH range. Reversed phase $C_{18}$ resulted in 100% retention of N-methylbentazon regardless of pH and other metabolites were retained below 40%. The AIBA was strongly adsorbed in silica gel, COOH and cation exchange phase but poor retention was on anion exchange sorbent. 2-Aminobenzoic acid showed an amphipathic nature which had high affinity for COOH and cation exchange phase at pH 7.0 as well as $NH_2$ and anion exchange sorbent at pH 3.0.

Metabolomic analysis of perilla seeds harvested from Korea and China (국내산 및 중국산 들깨 종자의 대사체 분석)

  • A comprehensive comparison of fatty acid and metabolite profiles in 8 species of perilla seeds harvested from Korea and China was carried out to understand the correlation between cultivation conditions and the quality of perilla seeds. Metabolomic studies revealed that the perilla seeds were distinct from each other, based on the partial least squares -discriminant analysis (PLS-DA) score plots; 23 compounds including metabolites, fatty acids, and phytosterols that predominantly contributed to this interspecies difference were identified. Changes in the growth environment affected these compounds. In particular, phenolic compounds were affected by rainfall and temperature. Moreover, temperature and altitude showed negative and positive correlations, respectively, to oleic acid. Although more studies on the effect of various environmental factors on cultivation of perilla plants as well as their seeds are needed, the present results provide information that would be useful in identifying perilla seeds produced in specific regions and in the cultivation of good quality perilla.

Effect of phenobarbital sodium and 3-methylcholanthrene on metabolism in vitro and toxicity of $^{14}C$-carbofuran in rat (쥐에서 phenobarbital sodium 및 3-methylcholanthrene이 $^{14}C$-carbofuran의 독성과 in vitro 대사에 미치는 영향)

  • In order to elucidate the effect of phenobarbital sodium(PB) and 3-methylcholanthrene(3-MC) on metabolism in vitro and toxicity of $^{14}C$-carbofuran in rat, they were administered by the chemicals, alone or in combination, and their survival ratios and metabolites were investigated. The $LD_{50}$(96 hrs) value of carbofuran to rats was 6.9 mg/kg. The toxicities of the major metabolites were in the decreasing order of 3-hydroxycarbofuran, 3-ketocarbofuran, 3-hydroxycarbofuran phenol and were much lower than that of the parent compound. When the rats were orally administered by the dose of carbofuran alone, 8.4 mg/kg, the survival ratio was 0%, whereas that was raised up to $60{\sim}80%$ with 20 mg/kg of PB or 3-MC, and 100% with 60 mg/kg of PB or 3-MC. Their metabolism in vitro occurred in the microsomal fraction. In case of carbofuran alone, the major metabolite was 3-hydroxycarbofuran. When carbofuran with PB or 3-MC, on the other hand, was treated, it was 3-ketocarbofuran. In addition, when the co-factor(NADP+G-6-P+G-6-P-DG) was added to the microsomal fraction(phase I system), and a mixture of NADPH+GSH to the 105,000g supernatant(phase II system) taken by carbofuran alone, each metabolites were produced by the maximum levels, respectively. In case of the carbofuran treatment with PB or 3-MC, the microsomal fraction of phase I system produced the maximum levels of metabolites, as in the treatment of carbofuran alone, whereas the 105,000g supernatant supplemented with the co-factor NADPH+FAD(phase II system) was brought about the maximum production of metabolites. The ratio of the formation of metabolites was 2 to 3 times higher in the combined treatment of carbofuran with PB or 3-MC than in the treatment of carbofuran alone.

Comparison of the Effects of Cyclodextrin-Naringin Inclusion Complex with Naringin on Lipid Metabolism in Mice Fed a High-Fat Diet (고지방식이를 섭취한 마우스에서 나린진과 나린진-사이클로텍스트린 포접화합물의 지질대사에 대한 영향 비교)

  • Naringin has antioxidant and antihyperlipidemic properties, however, phenolic compounds including naringin are unstable in the presence of light, heat and oxygen. Beta-cyclodextrin ($\beta$-CD) is a cyclic heptamer composed of seven glucose units that enhances the stability and solubility of molecules through the formation of inclusion complexes. This study was conducted out to compare the effects of CD-naringin (CD-N) inclusion complexes with naringin on lipid metabolism in high fat-fed animals. Male C57BL/6 mice were fed either CD-N (0.048%, w/w) or naringin (N, 0.02%, w/w) in a 20% high-fat (HFC, 15% lard, 5% corn oil, w/w) diet for 10 weeks. Orlistat (Xenical, 0.01%, w/w) was used as a positive control (PC). There were no differences in body weight, food intake, liver and heart weights, plasma triglyceride(TG), leptin, adiponectin, resistin, IL-$1{\beta}$ and IL-6 concentrations, and hepatic $\beta$-oxidation, carnitine palmitoyl transferase(CPT), glucose-6-phosphate dehydrogenase (G6PD) and malic enzyme activities between the HFC and CD-N groups or between the HFC and N groups. However, both CD-naringin and naringin supplementation les to a significant reduction in the epididymal and perirenal white adipose tissue weights, plasma free fatty acid, insulin and blood glucose concentrations, hepatic cholesterol and TG contents and hepatic fatty acid synthase (FAS), phosphatidate phosphohydrolase (PAP) and HMG-CoA reductase activities compared to the HFC group. The plasma HDL-cholesterol concentration was significantly higher in CD-N and N groups than in HF and PC groups. These results indicate that both CD-naringin and naringin supplementation effectively improved plasma and hepatic lipid metabolism without differences between CD-N and naringin groups.

Analysis of Free Amino Acids and Polyphenol Compounds from Lycopene Variety of Cherry Tomatoes (방울토마토 라이코펜 품종의 유리아미노산 및 폴리페놀 화합물의 분석)

  • In order to elucidate the usefulness of Lycopene, a cherry tomato variety, as a food material, the compositions of free amino acids, amino acid metabolites and polyphenol compounds were analyzed using HPLC and LC-MS/MS method. Lycopene contained eighteen free amino acids except for L-Cys and L-Try. L-Glu was the most abundant free amino acid, followed by L-Gln and L-Asp. The percentages of L-Glu, L-Gln and L-Asp of total free amino acid were 55.5%, 15.9% and 9.9% respectively. Lycopene contained essential amino acids with the exception of tryptophan. The following amino acid metabolites were found : ${\gamma}$-aminobutyric acid(GABA), carnitine(L-Car), o-phosphoethanolamine(o-Pea), hydroxylysine(Hyl) phosphoserine (p-Ser), N-methyl-histidine(Me-His), ethanolamine($EtNH_2$). Especially, GABA known as a neurotransmitter was present at a high level(305.99 mg/100 g dry weight). We identified the following polyphenol compounds in the cherry tomatoes : caffeic acid-hexose isomer I (CH I), caffeic acid-hexose isomer II (CH II), 3-caffeoylquinic acid(3-CQA), 5-caffeoylquinic acid(5-CQA), caffeoylquinic acid isomer(CQAI), quercetin-hexose-deoxyhexose-pentose(QTS), quercetin-3-rutinoside(Q-3-R), di-caffeoylquinic acid(di-CQA), tri-caffeoylquinic acid(tri-CQA), naringenin chalcone(NGC). Large quantities of Q-3-R and NGC known as bioactive compounds were found. These results revealed that Lycopene variety contained various nutritional and bioactive compounds and would be a potent functional food material.

In vitro Response of Cytochrome P450 (CYP) and EROD Activity in Bivalve (Coelomactra antiquata) by DDT, PCB-153 and TCDD (유기염소화합물인 DDT, PCB-153 및 TCDD에 in vitro 노출시킨 명주조개 미크로좀의 CYP및 EROD활성 반응)

  • The effect of xenobiotics on cytochrome P450 (CYP) and 7-ethoxyresorufin-O-deethylase (EROD) in hepatopancreatic microsome of surf clam, Coelomactra anti-quata, were investigated. The microsome isolated from the digestive gland of the surf clam, collected from the east coast of Korea, was in vitro exposed to p, p -DDT (0.1,0.4 and 1.0 mM) for 30 min and 2,3,7,8-TCDD (0.01, 0.04 and 0.1 ppb) and PCB-153 (0.01, 0.04 and 0.1 ppb) for 7 hr. In the case of DDT exposure, the CYP content and EROD activity of 1.0 mM exposure group increased up to about 117% and 120% of the DMSO solvent control group after 10 min. exposure, respectively. After 2 hr exposure of TCDD, the CYP content and EROD activity were also induced to the range of 103∼110% and 121∼139%, respectively. The PCB-153 exposure group showed 107∼115% of CYP content and 129∼140% of EROD activity after 2 hr exposure. Three test chemicals apparently induced CYP and EROD activity in the microsome of surf clam. The inducing potentials depend en the test chemicals.

Antioxidative Activities and Whitening Effects of Ethyl Acetate Fractions from The Immature Seeds of Abeliophyllum distichum (미선나무 미성숙 종자의 항산화 및 미백 활성)

  • Abeliophyllum distichum Nakai is deciduous shrubs of flowering plant in Oleaceae. It is important plant resources and consists of one species in the world. Also the endemic plant of A. distichum has been protected and designed endangered plant in Korea. For this reason, study on the immature seeds of A. distichum (ADS) hasn't progressed. In the present study, we evaluated the antioxidant activity and inhibitory effects on proteins and mRNA levels were related in the whitening effect in B16F10 cells. ADS was effective for reaction oxygen species (ROS). ROS causes various diseases such as aging, inflammation, cancer, and etc. Antioxidant properties were evaluated DPPH, ABTS radical scavenging activity and Reducing power. Plants were known that contained phenolic compounds were related in antioxidant activity. Phenolic compounds were phytochemicals commonly named natural polyphenols. These are secondary metabolites of plants involved in the defense against different types of stresses. In results, ADS suppressed the expression and transcription of Tyrosinase, TRP-1, TRP-2, and Microphthalmia-associated transcription factor (MITF). Tyrosinase, tyrosinase-related protein 1 (TRP-1), tyrosinase-related protein 1 (TRP-2) are known to play an important role in melanin biosynthesis. MITF regulated the expression and transcription of Tyrosinase, TRP-1, and TRP-2. In conclusion, ADS was effective in both antioxidant activities and whitening effects. Also, they were associated with the content of phenolic compounds. We suggested that ADS can be use antioxidants and skin-whitening functional cosmetics material derived from natural plant resources.

Phylogenetic characteristics of bacterial populations and isolation of aromatic compounds utilizing bacteria from humus layer of oak forest (상수리림 부식층으로부터 방향족 화합물 분해세균의 분리 및 세균군집의 계통학적 특성)

  • In this study, we isolated aromatic compounds (lignin polymers) utilizing bacteria in humus layer of oak forest and investigated phylogenetic characteristics and correlation with major bacterial populations in the humus layer by pyrosequencing. Forty-two isolates using aromatic compounds such as p-anisic acid, benzoic acid, ferulic acid and p-coumaric acid were isolated and phylogentic analyses based on 16S rRNA gene sequences showed that the isolates belonged to the genus Rhizobium, Sphingomonas, Burkhorlderia, and Pseudomonas. Among these, Burkhorlderia species which belong to Betaproteobacteria class occupied 83% among the isolates. The bacterial populations in humus layer of oak forest were characterized by next generation pyrosequencing based on 16S rRNA gene sequences. The humus sample produced 7,862 reads, 1,821 OTUs and 6.76 variability index with 97% of significance level, respectively. Bacterial populations consist of 22 phyla and Betaproteobacteria were the major phylum consisting of 15 genera including Burkholderia, Polaromonas, Ralstoria, Zoogloea, and Variovorax. Approximately fifty percentage of them was Burkholderia. Burkholderia as the majority of population in the humus was considered to play a role in degrading lignin in humus layer of oak forest.

Investigation of Medicinal Substances from in vitro Cultured Cells and Leaves of Artemisia princeps var. Orientalis (쑥의 잎과 기내 배양세포로부터 약용물질의 탐색)

  • The young leaves of A. princeps have been a well known for a crude medicine and used in treatment of colic pain, vomiting and menstrual irregularity. Based on TLC and HPLC and used an artemisinin, an anti-malarial compounds which is believed to be detected only in A. annuaup so far can be biosynthesized in A. princeps. To investigate the production of secondary metabolites like artemisinin in cultured cells, the cell culture of A. princeps was established. Callus and suspension cultured cells of A. princeps were induced and grown highest in MS media containing $0.2\;mg/{\ell}$ 2,4-D, $0.1\;mg/{\ell}$ BAP and 2% sucrose. Different metabolites from in vitro cultured cells (callus and suspension cultured cell) and intact plants were analyzed by TLC analysis. As a result, we can confirm that in vitro culture has a potential for mass production of secondary metabolites from A. princeps.
