Aspergillus kawachii S-27에 의한 Rice-Koji의 제조에 관한연구

  • 현재 우리나라의 탁약주 용 Rice-Koji 제조 용종국균으로 널리 분양되고 있는 분양되고 있는 fawachi, S-27 균주가 생성하는 Amylase의 효소학적 특성과 Rice-Koji i제조 시 유기산 및 당화 효소의 생성 조건을 검토했다. 1. 본 균은 유기산 생성력이 강하고 또 생성하는 당화효소 및 액화효소는 내산성이 극히 강하며 pH3~5의 범위내에서 최고 활성을 나타내었으므로 탁약주 제조 용 종국균으로 아주 적합하였다. 2. 본 균의 발아 최적온도는 36$^{\circ}C$이었으며 Rice -Koji제조시 제국시간을 40시간으로 볼 때 유기산 생성의 최적온도는 32$^{\circ}C$이었고 당화효소 생성의 최적온도는 36$^{\circ}C$이었으며 고온인 4$0^{\circ}C$에서는 유기산 생성이 극히 불량하였다. 3. Rice-Kojiw제조시 당화효소의 생성은 배지의 수분함량 35%일 때 최고치를 보였으며 유기산 생성은 이 보다 높은 조건인 수분함량 40%일 때 이었고 비교적 건조한 조건인 수분함량 30%일 때는 유기산 생성이 특히 불량하였다. 4. 배지의 두께가 3cm이상이 되면 유기산 및 당화효소 생성에 않은 지장을 주었다. 5. 종국균의 점종량은 유기산 및 당화효소의 생성에 별로 영향을 미치지 못했다.

  • 누룩은 생소맥을 조분쇄하여 물로 혼합시켜 자연적으로 공지중의 여러 종류의 미생물을 배양시켜 만들었다. 그러므로 누룩에서는 많은 종류의 사상균, 호모와 세균이 증식하였다. 누룩의 당화력은 원료 생소맥의 당화력과 대부분의 사상균의 당화력과 알붕의 세균성 당화력에 기인하여, 발효력은 누룩효모와 극소수의 사상균애 의하여 이루어지고 있다. 1945년 이전까지 누룩으로부터 12속 59종의 사상균이, 8속 29종의 효모와 4속 16종의 세균이 분리 되었다. 누룩사상균은 Aspergillus 속이 주종을 이루었고 Rhizopus속, Absidia속, Nucor속의 순으로 분포하고 있었고, Aspergillus속과 Rhizopus속은 중요한 누룩 당화균으로 작용하였다. 누룩의 대표적 효모는 Saccharomyces cerevisiae 이며 , 이균이 알코올 발효에 가장 큰 역할을 담당하며, Saccharomyces cerevisiae도 전통 민속주의 탁주와 약주의 발효역에 깊이관여한다고 사료된다. 누룩 중에 증식하는 세균은 양조학상 특별한 역할을 담당하지않지만 Bacillus 속 과 젖산세균이 많이 분리되었으며, 젖산세균은 담금초기에 pH의 안정화에 기여할 수 있다고 추측할 수 있다.

Acid Production and Phytate Degradation using a Leuconostoc mesenteroides KC5l Strain in Saccharified-Rice Suspension (현미 당화액에서 Leuconostoc mesenteroides KC51 균주에 의한 산의 생성과 Phytate의 분해)

  • A saccharified-rice was fermented using Leuconostoc(Ln.) mesenteroides KC51 strain in various dry matter (DM) contents (4%, 8%, and 12%) at $30^{\circ}C$ for 18 h. The changes of viable cell number, acid production and phytate degradation in saccharified-rice during fermentation were investigated. The viable cell population of Ln. mesenteroides KC51 was increased rapidly in proportion to DM contents during the 9 h of cultivation. The changes of pH and titratable acidity in saccharified-rice were dependent on DM contents. At high DM content (12%), the viable cell number of Ln. mesenteroides KC51 increased to 9.56 log CFU/g after 6 h of fermentation. The pH and titratable acidity reached to pH 3.38 and 0.93% after 18 h of fermentation, respectively. The phytate, known as an antinutrient factor, in saccharified-rice was degraded by Ln. mesenteroides KC51 cultivation. The decrease of phytate during fermentation approximately coincided with the increase of Ln. mesenteroides KC51 population observed in fermented saccharified-rice. Regardless of DM contents, the levels of phytate were reduced to around 50% of initial concentration.

Preparation and characteristics of yogurt added with enzymatically saccharified Korean rice wine lees powder (효소로 당화시킨 주박 분해물을 첨가한 요구르트의 발효 특성)

  • This study was carried out to determine the effect of enzymatically saccharified Korean rice wine lees powder (eKRWLP) on the quality characteristics and storage stability of curd yogurt. Yogurt with different contents [0.5-2.0% (w/w)] of eKRWLP was incubated with commercially available mixed lactic acid bacteria (Lactobacillus acidophilus, Streptococcus thermophilus, Bifidobacterium longum) at $40^{\circ}C$ for 18 h. The production of acid measured at pH and titratable acidity of yogurts increased with increasing eKRWLP content. After 12 h fermentation, titratable acidity of eKRWLP yogurt was 0.77-0.90% and was higher than that (0.72%) of yogurt made without eKRWLP. The viable cell counts of lactic acid bacteria in eKRWLP yogurts were increased in proportion to the addition of eKRWLP, and increased up to 8.01-8.13 log CFU/g after 12 h incubation. The repressive effect of whey separation in eKRWLP yogurt curd significantly decreased than that in Korean rice wine lees powder (KRWLP) yogurt. With sensory evaluation, yogurt with 0.5% eKRWLP obtained the highest scores among all eKRWLP yogurts. When eKRWLP yogurts and the control preparations fermented for 12 h were incubated at $4^{\circ}C$, their pHs and titratable acidities were slightly changed and the number of viable lactic acid bacteria were well maintained above $10^7CFU/g$ for 16 days.

Quality Characteristics of Fermentation Gastrodia elata Blume by Saccharifying Methods (당화방법에 따른 유산균 발효천마의 품질특성)

  • This study aimed to evaluate the quality characteristics of Gastrodia elata Blume fermented by lactic acid bacteria after saccharifying by 3 methods including enzyme, malt, and rice-nuruk. The lactic acid bacteria (LAB), Pediococcus inopinatus BK-3, isolated from kimchi could reduce the unpleasant taste and odor of Gastrodia elata Blume. The total acidity value of Gastrodia elata fermented by LAB on the malt and rice-nuruk extract solution for 3 days was 2.23% and 2.33%, respectively. After saccharification by malt and rice-nuruk extract solution for 3 days, the viable cell number of fermented Gastrodia elata was 9.14 log cfu/mL and 9.27 log cfu/mL, respectively. The total acidity values were increased above 3.35% by malt and rice-nuruk extract solution for 8 days. Thus, the viable cell number was the highest by malt and rice-nuruk extract solution fermentation for 3 days. The amino acid content of Gastrodia elata fermented by LAB after saccharification by malt extract solution was higher than that of other saccharifying methods. The free sugar content and p-hydroxybenzyl derivatives induced by the enzyme method were higher than those of other saccharifying methods. The overall acceptability was the highest at 4.2 point in Gastrodia elata fermented by malt extract solution.

Substitution effects of enzymatically saccharified Korean rice wine lees powder on skim milk in yogurt fermentation (요구르트 발효에서 효소로 당화시킨 주박 분해물의 탈지분유 대체 효과)

  • Yogurt was prepared with different substitution ratio [10, 20, 30, and 50% (w/w)] of skim milk with enzymatically saccharified Korean rice wine lees powder (eKRWLP) and fermented with commercially available mixed lactic acid bacteria (Bifidobacterium longum, Lactobacillus acidophilus, Streptococcus thermophilus) at $40^{\circ}C$ for 18 h. Fermentation characteristics were evaluated in terms of acid production (pH and titratable acidity) and viable cell counts of lactic acid bacteria. The pH of yogurts decreased with increasing eKRWLP substitution ratio of skim milk. After 12 h fermentation, titratable acidities of eKRWLP substitution and control (yogurt made without eKRWLP) were 0.84~1.04% and 0.93%, respectively. The titratable acidities of yogurts prepared with 10 and 20% substitution ratio increased than that of the control yogurt, but titratable acidities of yogurts of 30 and 50% substitution ratio decreased. After 9 h fermentation, the number of viable lactic acid bacterial cell were increased to 8.18~8.24 log CFU/g in all yogurts. In sensory evaluation, there were similar preference for eKRWLP yogurts prepared with 10 and 20% substitution ratio and the control. When eKRWLP substitution and control yogurts fermented for 9 h were incubated at $4^{\circ}C$, their pHs and titratable acidities were slightly changed but the number of viable lactic acid bacteria were well maintained above $10^7CFU/g$ for 11 days in yogurts prepared with 10 and 20% substitution ratio among eKRWLP substitution yogurts. These results suggest that eKRWLP can be used as substituent of skim milk and the optimum substitution ratio is around 10~20%.

Studies on Preparation of Lactic Acid Fermented Beverages from a Malt Syrup (맥아 당화액을 이용한 유산균음료의 제조에 관한 연구)

  • The possibility of developing new kinds of lactic acid beverage from a malt syrup was studied. The optimum sugar concentration of malt syrup for the cultivation of lactic acid bacteria was $10^{\circ}Bx$. The acidity of the fermented malt syrup was improved by the supplement of yeast extract(0.5%) or sodium citrate(0.08%). Though the activity of Lactobacillus lactis in malt syrup was superior to other strains, sensory test indicated that the mixed culture of Lactobacillus lactis and Streptococcus diacetilactis was better because of masking malt flavour. The changes in acidity and viable cells of malt syrup during the lactic fermentation were not so good as skim milk medium, but malt syrup medium containing milk(50 : 50) was nearly similar to skim milk medium. In the sensory scores among samples, no significant differences(P<0.05) were noted between fermented milk and fermented malt syrup containing milk, but fermented malt syrup showed a poor quality. However fermented malt syrup was not inferior to marketing lactic fermented fruit juices with regards to the lactic acid fermented beverage type.

Bioethanol Production from Seaweed Kappaphycus alvarezii by Simultaneous Saccharification and Fermentation (홍조류(Kappaphycus alvarezii)의 동시 당화 발효를 이용한 바이오에탄올의 생산)

  • Thermal acid hydrolysis pretreatment of Kappaphycus alvarezii was carried out with 12% (w/v) seaweed slurry and 180 mM H2SO4 at 140°C for 5 min. Utility of the thermotolerant yeast Kluyveromyces marxianus KCTC7150 was evaluated with respect to cell growth and ethanol fermentation at 40°C was close to optimal for enzymatic hydrolysis. This could lead to the integration of both the saccharification and fermentation processes. The levels of ethanol production by simultaneous saccharification and fermentation (SSF) with non-adapted and adapted K. marxianus KCTC7150 were 9.1 g/l with an ethanol yield (YEtOH) of 0.24 and 10.2 g/l with an ethanol yield (YEtOH) of 0.27 at 156 h, respectively. The two-phase SSF process was employed in this study to improve the efficiency of ethanol fermentation. Adapted K. marxianus KCTC7150 using the two-phase SSF process produced 13.5 g/l with an ethanol yield (YEtOH) of 0.35 at 96 h. Development of the two-phase SSF process could enhance the overall ethanol fermentation yields of the seaweed K. alvarezii.

Enzymatic saccarification of cellulosic wastes by pectinase

  • The study was aimed to saccharify callulosic waste by pectinase produced from strain KL34 isolated from soil. The enzyme activity in the culture using 1%(w/v) fruit waste as carbon source reached to 3.8 U/ml. In the enzymatic hydrolysis of cellulosic waste, we obtained 9.5g/L reducing sugar in the condition of supernatant containing 5 U/ml enzyme and 10%(w/v) apple rind as substrate. Additionally, in enzymatic hydrolysis of food waste using pectinase from KL34, reducing sugar of 12.7g/L was obtained, indicating enhancement of 1.6 fold compared with that of only cellulase employment.

Screening and Identification of a Potent Fungus for Producing Raw Corn Meal Saccharifying Enzyme (옥수수 생 전분 당화 효소 생산 곰팡이의 분리 및 동정)

  • We have been searching microorganisms which produce highly active raw starch saccharifying enzyme and also have a good cultivation characters in submerged culture. About 170 strains of molds isolated from soil and compost were tested for their amylase productivity on plate contained 2% raw corn meal. Thirty-four strains out of 170 strains produced clearance on the plates, and were tested for their raw starch saccharifying activity. Then, 4 strains which had shown relatively high levels of saccharifying activity were selected. Among them, Strain No. 55 was found to have highest level of raw starch saccharifying activity, and selected for the further studies. In this paper, the morphological, physiological and cultural characteristics of Strain No. 55 were described. Based on the results obtained in these experiments, Strain No. 55 was identified to be a similar species to Aspergillus niger.
