The Effects of Cinnamomum cassia Blume, Aconitum carmichaeli Debx, and Pueraria lobata Benth on Glucose and Energy Metabolism in C2C12 Myotubes (C2C12 골격근 세포에서 육계, 부자, 갈근 물 추출물의 당대사 및 에너지 조절 효과)

  • Objectives: The prevalence of obesity and metabolic syndrome is increasing worldwide. Regulation of cellular energy metabolis has the potential to be manipulated therapeutically to serve as a target for obesity and insulin resistance. Skeletal muscle is regarded as a target for regulation of energy metabolism and insulin resistance. In this study, the authors investigated the regulatory effect of (Cinnamomum cassia Blume, CCB), Aconitum carmichaeli Debx (ACD), and Benth (Pueraria lobata Benth, PLB) on energy and glucose metabolism in C2C12 myotubes. Methods: The water extracts of CCB, ACD, and PLB (0.5 mg/ml) were treated in differentiated C2C12 myotubes. The expressions of adenosine monophosphate-activated protein kinase (AMPK) and phosphorylation AMPK were detected with western blotting. Glucose metabolism was investigated with glucose uptake assay and glucose consumption assay, total adenosine triphosphate (ATP) content was also analyzed. Results: CCB, ACD, and PLB activated the phophorylation of AMPK, they also increased the glucose metabolism and total ATP contents in C2C12 myotubes. Conclusions: This study suggests that CCB, ACD, and PLB have the potential to increase energy and glucose metabolism in skeletal muscle.

Ethanol Effect of Nelumbo nucifera Root on Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨 흰쥐에 대한 연근 에탄올 추출물의 당대사 효소활성과 항산화 작용에 미치는 영향)

  • This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

The Mixed Herbal Extract, CAPA, Prevents Obesity and Glucose Intolerance in Obese Mice (비만형 동물모델에서 복합 한약 추출물 CAPA의 비만 및 당대사 개선 효능)

  • Objectives: This study investigated the effects of the mixed herbal extract from Cinnamomum cassia Blume, Atractylodes macrocephala Koidzumi, Pueraria lobata Benth, and Aconitum carmichaeli Debx (CAPA) on obesity and glucose tolerance in obese mice. Methods: Animals were divided in 6 groups, normal diet, high fat diet (HFD), HFD with CAPA 100 mg/kg (CAPA 100), CAPA 300 mg/kg (CAPA 300), and metformin 200 mg/kg or lorcaserin 10 mg/kg as positive controls, and treated for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after treatment. The oral glucose tolerance test was also carried out after treatment. Results: Compared to HFD, CAPA extract treated mice showed significant decreases in body weight, adipose tissue weight, lipid accumulations in liver and serum lipid levels without a reduction of food intake. And fasting glucose and glucose tolerance were all improved in the CAPA treated mice. Conclusions: Our results suggest that CAPA extract can prevent diet induced obesity and glucose intolerance without a reduction of energy intake in obese mice.

The Effect of Atypical Anti-psychotic Agents on Obesity and Glucose Metabolism (비정형 항정신병약제가 비만과 당대사에 미치는 영향)

  • Atypical antipsychotics are more effective than typical antipsychotics and have fewer side effects such as tardive dyskinesia and extrapyramidal symptoms; therefore, prescriptions of atypical antipsychotics are increasing. However, recently, it has been reported that atypical antipsychotics have a higher incidence of diabetes, hyperglycemia, and obesity than typical antipsychotics. Atypical antipsychotics induce obesity-inhibiting appetite-related receptors such as serotonin and dopamine. Decreased exercise due to improving psychotic symptoms, and genetic characterictics can also cause weight gain. Hyperglycemia and hypoglycemia were another metabolic problem related to treatment with atypical antipsychotics. The mechanisms of hyperglycemia were mainly related obesity, decreased anorexigenic hormones, and increased insulin resistance in multiple organs. There are also reports that genes related to diabetes have an effect on the incidence of diabetes mellitus treated with atypical antipsychotics. On the other hand, although it is not clear why hypoglycemia occurs, it documented in case reports all over the world. There are more reports of atypical antipsychotics than typical antipsychotics and these are frequently reported in Asians. Further research on the mechanism of hypoglycemia related to atypical antipsychotics is strongly recommended.

A study of Nan xian(南軒) Zhangshi(張?)'s cultivation - Focusing on the mutual influence of Huxiangxue and Zhuzixue (남헌(南軒)장식(張?)의 수양론 연구 - 호상학과 주자학의 상호 영향을 중심으로 -)

  • Zhangshi established his own ideal system in the process of self-cultivation in the Southern Song dynasty period, where academic exchanges were active. A common feature in the process of self-cultivation is the transformation of ideas. In the case of Zhangshi, his academic exchanges with contemporary mainstream scholars had a major impact on them, and Zhangshi was also able to systematize his own ideas. However, while there have been a lot of researches on the mainstream schools of the Southern Song period, there has been little research on the ideological trends and trends of the time when mainstream schools could have occurred. This paper attempts to understand the ideological trends of the Southern Song era by examining the formation process of the his theory of self-cultivation, and to examine the process of exchanging of mainstream schools through the field of self-cultivation. This work will be a meaningful process for understanding the philosophy of the Song era and for examining the tendency objectively. This paper examines the formation process of self-cultivation in the first half and the second half. In the first half, this paper refers to the ideological impact of schools that had influenced Zhangshi's thought, in the second half, this paper deals with the process of change of his theory of self-cultivation and then examines how his ideas had changed. This work will help to understand Zhangshi's own ideas and to understand how Zhu Xi's theory of Zhu xi, who had an ideological exchange with him, could be established.

An Understanding of the Archival Management in Early Joseon Dynasty (조선전기 기록관리 체계의 이해)

  • In this article, I outlined the archival management system in Early Joseon Dynasty and examined the characteristics of the system. At first, I explained the three aspects of the archival management; the memory of the past, the documentation of the contemporary, and the vision of the future through the documentation. Secondly, I tried to understand the character of the Veritable Record and its compilation by the concepts of archival science such as 'authenticity', 'reliability'. In the memory of the past, the History of a Dynasty(Koryo-Sa) and the Comprehensive History of Eastern Kingdom were included. The arrangement of the past was accompanied with the systematic study of the domestic and foreign histories. At the beginning of the state building, there was many practical need to the experiences of government and social re-construction. It was also the process of the legitimacy establishment of the new dynasty. And the Bureaucracy promoted the development of the records and archival management system because it needed the continuity and evidence of business. The dualistic structure of the records and archival management system was the most unique character of this age. The management of general administrative records was not different from the modern one. But the historical drafts and the compilation of Veritable record were different. Here, I had to examine the characteristics of these procedures by the concept authenticity, reliability, and custodianship. In doing so, I suggested the need of conceptualization of the historical terms such as 'the primary sources' and 'the secondary sources' in historical study. The archival concepts will be the most useful means to that issue. Through the memory of the past and the documentation of the contemporary, they made visions of the future, new vision of the Literati Governance. In this tradition, in spite of the revision of the Veritable records by the new changed political party, both the orignal and the revised remained as the comparative evidence for the future generation in the name of the Black-Red Revised History.

Correlation between Serum Osteocalcin and Hemoglobin A1c in Gwangju General Hospital Patients (광주 소재 한 종합병원을 방문한 성인에서 혈중 Osteocalcin과 HbA1c의 상관관계)

  • Recently, there have been many studies on the relationship between the bone metabolism and the mechanism of diabetes. In those studies, it was shown that osteocalcin can be involved in the treatment of type 2 diabetes. Therefore, this study examined the correlation between osteocalcin and HbA1c. From January 1, 2015 to December 31, 2016, a total of 714 adults aged 40 years or older ($70.8{\pm}10.4years$, 452 males), in whom both osteocalcin and HbA1c checked in a general hospital in Gwangju, were enrolled in the study. The serum calcium, total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, and 25-(OH) vitamin D were measured and basic information, such as the height and weight, were recorded. There was a weak negative correlation (r=-0.183, P<0.001) between the osteocalcin and HbA1c levels in diabetic patients but a negative correlation (r=-0.251, P<0.001) when adjusted by all other study variables. The present study showed that there was a negative correlation between the osteocalcin and HbA1c levels in diabetic patients. The relationship between the bone metabolism and the incidence of diabetes mellitus should be studied based on the influence of biological mechanisms and associated factors of bone and glucose metabolism.

An Hwak's Recognition of 'Joseon' and 'Joseon Cheolhak' (안확의 '조선' 인식과 '조선철학')

  • The full-scaled study of Joseon conducted by Japan in the 1910s was part of its colonial policy, while the native Joseon studies against it contained political aspiration to recover the national rights and independence. Accordingly, the conceptual meaning of 'Joseon' varied according to its subject of speech. The establishment of modern nation-state failed along with the extinction of Korean Empire, but 'Joseon' was newly discovered within national ideology. It became a historical concept in which the experience of the past and the expectation toward the future could be united. The so-called 'Joseon Studies' was only limited to intellectuals in the academic circle, but 'Joseon' embraced the articulations from more various social agents. Furthermore, it is only natural that 'Joseon Studies' should be interpreted within the historical semantics of 'Joseon', considering the connection between concept and discourse. In his The History of Joseon Civilization, An Hwak encompassed the history from the times of ancient mythology to the contemporary times under the banner of 'Joseon'. Opposing Japanese distortion of history carried out in the name of historical positivism, he idealized Joseon history as comparable to that of the Western democracy. He extended the study of 'Joseon' into culture at large, foreshadowing a kind of Joseon philosophy. In his An Overview of Joseon Philosophical Ideas, the first description of 'Joseon philosophy' as an independent field, he proposed philosophy as one of three sources of pride in Joseon and asserted its uniqueness and originality compared to the West. It was an attempt to grasp the peculiarity of Joseon ideas from a perspective of the history of universal human civilization. He considered 'Jong'(倧) as an ideological foundation held from the ancient to the modern times, and the acceptance of Buddhism and Confucianism as beneficial to 'Joseon philosophy'. The birth of 'Joseon philosophy', the modern transformation of the traditional knowledge system, was an intellectual experiment to apply traditional knowledge to the modern disciplinary classification system.

Hypoglycemic Action of Components from Red Ginseng : (I) Investigation of the Effect of Ginsenosides from Red Ginseng on Enzymes related to Glucose Metabolism in Cultured Rat Hepatocytes (홍삼 성분의 혈당강하작용 연구 (I) :쥐의 배양 간세포의 당대사 관련 효소 활성에 미치는 홍삼 사포닌 성분의 영향 조사)

  • In this study, rat hepatocytes known to have active carbohydrate metabolism were obtained by using the liver perfusion technique to examine the hypoglycemic action of red ginseng saponin components [ginsenoside (mixture, $Rb_1$, and $Rg_1$)] and incubated in two different media-one containing insulin and glucagon (control group), and the other containing glucagon only, The specific activities of some regulatory enzymes such as glucokinase, glucose 6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, and glucose 6-phosphatase, in main pathways which were directly related to the glucose metabolism were compared between these two kinds of hepatocytes cultured in two different media. The effects of red ginseng saponin components [ginsenoside (mixture, $Rb_1$, and $Rg_1$)] under the concentration of $10^3$~$10^6$% on these enzymes In hepatocytes were also investigated, when they were added to these two media. The results were as follows. The specific activity of enzymes such as glucokinase, glucose 6-phosphate dehydrogenase, and 6-phosphogluconate dehydrogenase related to glucose-consuming pathways of insulin-deficient group was much less than control one, however, their decreased activity was recovered after the addition of ginseng components at all range of concentrations. The increased specific activity of these on - zymes was shown by the addition of ginseng components to the control group. On the other hand, the specific activity of glucose 6-phosphatase related to glucose-producing pathway of insulin-deficient group was much higher than control one, but their Increased activity was decreased after the addition of ginseng components at all range of concentrations. The same results were obtained after the addition of ginseng components to the control group. These results suggest that the red ginseng saponin components might better diabetic hyperglycemia by regulating the activity of enzymes related to glucose metabolism directly and/or Indirectly though more detailed studies were needed.

Effects of $CV_{12}$ and $ST_{36}$ Acupuncture on Glucose Metabolism and Protection of Pancreatic Beta Cells in Neonatal-streptozotocin Model (중완과 족삼리 자침이 Neonatal-streptozotocin모델 실험동물에서 당대사 및 베타세포 보호작용에 미치는 영향)

  • 목적 : 중완과 족삼리 자침이 neonatal-streptozotocin 모델 실험동물에서 혈당과 베타세포 손상에 대한 보호효과를 알아보고자 하였다. 방법 : 비-비만 제2형 당뇨모델인 neonatal STZ 실험동물을 무처치 대조군, 임의혈 대조군, 중완 자침군, 족삼리 자침군으로 나누어 90일 동안 실험하였다. 체중과 혈당을 3일에 한 번씩 측정하였으며, 90일이 경과 후 혈중 인슐린을 측정하고 당부하 검사를 실시한 후 동물을 희생시켜 이자 조직을 적출하고 표본을 제작하였다. 면역세포화학법을 통하여 이자의 베타세포 손상에 대한 세포보호효과를 관찰하였다. 결과 : 중완 자침군과 족삼리 자침군은 대조군에 비해 유의한 혈당 강하 효과가 관찰되었다. 인슐린 농도는 모든 군에서 상승하였으며 당부하 검사에서는 임의혈 자침군과 중완 자침군 및 족삼리 자침군에서 내당능이 있는 것으로 나타났다. 조직 검사에서는 무처치 대조군과 임의혈 대조군에서는 베타세포 손상이 심했으나, 중완 자침군과 족삼리 자침군에서는 베타세포 손상에 대한 세포보호 효과가 현저하게 관찰되었다. 결론 : 중완과 족삼리 자침은 비-비만 제2형 당뇨모델에서 손상된 베타세포를 회복하고 인슐린 분비를 촉진하여 혈당을 강하시키는 효과가 있는 것으로 사료된다.
