Analysis of junction site between T-DNA and plant genome in Lissorhoptrus oryzophilus resistance GM rice (벼물바구미 (Lissorhoptrus oryzophilus) 내충성 GM 벼에서 T-DNA와 게놈의 인접부위 분석)

  • Four transgenic rice lines harboring insect-resistant gene cry3A showed ideal field performances characterized by high considerable resistance to rice water weevil (Lissorhoptrus oryzophilus Kuschel). In this study, we estimated the insertion number of foreign genes, and analyzed the flanking sequences of T-DNAs in rice genome. As a result, T-DNA of BT12R1 line was inserted in exon region of rice chromosome 10. Two copies of T-DNAs were inserted in line BT12R2. BT12R3 line was analyzed at only left border flanking sequence. BT12R4 line was confirmed one copy of foreign gene insertion at the position 24,516,607 ~ 24,516,636 of rice chromosome 5, accompanied by a deletion of 30 bp known genomic sequences. This intergenic position was confirmed none of expressed gene and any deletion/addition of T-DNA sequence. In conclusion, these molecular data of rice water weevil resistant Bt rice would be used to conduct the biosafety and environment risk assessment for GM crop commercialization.

  • Bacterial diseases in soybean are bacterial pustule by Xanthomonas axonopodis pv. glycines, wildfire by Pseudomonas syringae pv. tabaci, bacterial blight by Pseudomonas savastanoi pv. glycines and bacterial brown spot by Pseudomonas syringae pv. syringae in Korea. It is difficult to identify each disease by early symptoms in fields, because the initial symptoms of these diseases are very similar to each other. In this study, we developed multiplex PCR detection method for rapid and accurate diagnosis of bacterial diseases. The glycinecin A of X. axonopodis pv. glycines, the tabtoxin of P. syringae pv. tabaci, the coronatine of P. savastanoi pv. glycines and the syringopeptin of P. syringae pv. syringae have been reported previously. These bacteriocin or phytotoxin producing genes were targeted to design the specific diagnostic primers. The primer pairs for diagnosis of each bacterial diseases were selected without nonspecific reactions. The studies on simultaneous diagnosis method were also conducted with primarily selected 21 primers. As a result, we selected PCR primer sets for multiplex PCR. Sizes of the amplified PCR products using the multiplex PCR primer set consist of 280, 355, 563 and 815 bp, respectively. This multiplex PCR method provides a efficient, sensitive and rapid tool for the diagnosis of the bacterial diseases in soybean.

Evaluate of SSRs for Heat Tolerance using Korean Major Wheat Cultivars and Heat Resistant Turkey Resources (주요 국산밀 품종과 내고온성 터키 유전자원을 이용한 내고온성 관련 SSR 마커 평가)

  • High temperature is one of major environmental stress. Heat tolerance managing is difficult through the phenotypic selection, so marker assistant selection (MAS) using molecular markers like as RAPD, SSR etc. was tried to select useful traits for heat tolerance. Fourteen SSR markers reported by previous research were selected for this research. We tried to evaluate 14 SSR markers for MAS using 31 useful wheat resources including 24 crossing line from Turkey, six Korean wheat cultivars and Chinese spring. The average of the number of alleles and PIC values in this study were 6.14 and 0.64, respectively. Two major clades and four sub clades were grouped by phylogenetic tree using UPGMA. Four Korean wheat cultivars were distinct from other Turkey resources in the phylogenetic dendrogram. From the results, we expected that these markers were able to adapt to screening wheat genotyping for heat tolerance.

Some Aspects of High Lysine Maize Breeding using Opaque-2 Gene (Opaque-2 인자를 이용한 고라이신 옥수수의 육종)

  • Several field and sweet corn varietiea from several sources were crossed with a variety carrying the opaque-2 gene to determine the phenotypic interactions in the breeding of high lysine maize. Although opaque-2 lines showed lower protein content than the corresponding normal varieties, there was no correlation between the protein levels of the two types. opaque-2 maize contained more lysine, but no relationship was found between the protein content and the lysine content of either normal or opaque-2 types, suggesting that high lysine corn using the opaque-2 gene may be developed independently from the protein content. The F2 segregation ratios for normal and opaque-2, 100-kernel weights, percentage seed set, opaque-2 phenotype, disease susceptibility, and the relationship between protein and lysine content of normal and opaque-2 were investigated. The determinations and observations were made on the F2, F3, and BC1 Lysine content was determined by the ion exchangeresin combined with paper chromatography method. Most crosses segregated in a 1-opaque-2 : 3-normal ratio as expected. Opaque-2 segregates were lighter than the normal type and smaller in size. A mottled phenotype of opaque-2 maize observed in the Philippines yellow endersperm. In some varieties opaque-2 maize was very susceptible to the ear and kernel rot disease. No. 5(female) and opaque-2(male). Selectlon of a double mutant of waxy and opaque-2 by using the iodine technique and electric lamp was discussed. opaque-2 and floruy-2 were not allels. Different percentage of seed set were observed in the segregation of aewx crossed with opaque-2. An unusual gametophytie relationship was involved in a cross between Glutinous.

Growth and Flowering of Orostachys japonicus A. Berger by Controlling Daylengths (일장조절에 따른 바위솔의 생장과 개화)

  • Orostachys japonicus, called Wasong as herbal medicine and a short day plant, should be artificially kept in long daylength to control anthesis of its florets. The study was done to clarify the effect of daylength (10, 13, 16 hours a day) on growth, morphological characters, and flowering. The treatments were done on August 25 and afterward samples were taken every 2 weeks. Growth, morphological and flowering related characters were measured from each sample. With longer daylength plant height and inflorescence length were increased but number of leaves including bracts and stem diameter were decreased. Leaves and bracts, stem, root, shoot and total dry weights were increased with longer daylength from late September while floret dry weight showed reverse result. Florets formed were the greatest in daylength of 10 hours during September but the least during the other period. Anthesis of the florets was observed only in daylength of 10 hours during October and all the plants from the treatment were flowered.

Comparison of biological activity according to extracting solvents of four Acanthopanax root bark (추출 용매에 따른 오갈피속 근피의 생리활성 기능 탐색 및 비교)

  • The biological activities of water, ethanol and 50% ethanol extracts from Acanthopanax root bark were compared. 94% of Hep3B cell growth was inhibited by adding 1.0g/L of 50% ethanol extracts from A. senticosus root bark. It was also showed that above 90% of A549 cell growth was inhibited by adding 1.0g/L of 50% ethanol extracts. The 50% ethanol extracts of A. sessiliflorum root bark showed that the extracts selectivity were from 1.5 to 3.4 by adding all samples. For screening immunomodulating activities, Jurkat(T-cell) was showed that the cell growth and viability were more increased and activited 275% by adding the 50% ethanol extracts from A. senticosus root bark. The result of anti-mutagenicity of 50% ethanol extracts of A. senticosus root bark was most effective than any other samples. The enhancement of glutathione-S-transferase activity was increased 241% by adding 1.0g/L 1 : 1 extracts of A. senticosus root bark. 72% of oxidation was inhibited by adding 1.0g/L of 50% ethanol extracts from A. senticosus root bark.

Effect of Temperature and Daylength on Flowering and Growth Characteristics (온도와 일장조건에 따른 참깨 개화 및 생육특성 영향)

  • Sesame is typically a temperature- and day length-sensitive plant, as its flowering is promoted under high temperature and shorter day length conditions. The experiment carried out in the present study revealed that day length influenced sesame flowering to a much greater extent than temperature. The degree of day length influence differed depending on the sesame variety. Days from sowing to flowering was negatively correlated with yield-related characteristics such as stem length, capsule number per plant, 1,000 seed weight, and seed weight per 10a. In cases where the period from sowing to flowering was longer and flowering date was later, the reproductive growth period was shorter during maturity. Analysis of the effects of temperature and day length on seed yield production revealed that temperature was a much more influential factor than day length. However, day length was more influential than temperature under low temperatures, such as 22℃. All varieties apart from Ansan were shown to be primarily influenced by temperature. Sesame varieties were grouped in view of optimal seed sowing time through principal components analysis. The Poongsung and Sungboon varieties favored early sowing under low temperatures and short day lengths. The DT45, Poongsung, 90 days, Ansan, and Sungboon varieties favored late sowing under high temperatures and long day lengths. The Yangbaek and Arum varieties could be sown both early and late.

Growth and Biomass Productivity of Seedlings from Seeds in Jerusalem Artichoke (Helianthus tuberosus L.) (돼지감자 실생묘의 생장특성과 바이오매스 생산성)

  • In order to examine the possibility of seed propagation of jerusalem artichoke (Helianthus tuberosus L.) which has been propagated vegatatively so far, it was investigated the growth characteristics of seedlings and the biomass productivity by seed propagation in jerusalem artichoke. The results indicated that the seedlings from seeds exhibited the physiological dwarfing as a rosette habit. The physiological dwarfing as a rosette habit in jerusalem artichoke seedlings from seeds was a kind of appearance affected by photoperiod. This abnormal habit in seedlings from JA6 seeds during the growing period was persisted in short photoperiod of 10 hrs but was disappeared in long photoperiod of 14 hrs. In the condition of long photoperiod, seedlings from seeds grew normally and the average tuber yields obtained from seed propagation was finally 3.51 ton/10a, which can be the satisfying level to the use of seed propagation despite its complexity such as the needs of specific seedling time because it gives a high biomass productivity. This potential utilization of seed propagation in jerusalem artichoke can be basically applied to the collection or preservation of germ plasm and the improvement of jerusalem artichoke.

Fibrinolytic Enzyme Activity of Extract from Camellia japonica L. (동백나무 추출물의 혈전용해 효소활성)

  • The fibrinolytic activities of soluble proteins extracted from young leaves of Camellia japonica L. were studied. Fibrinolvity activity of extract from partitions of C. japonica L. showed 1.6-2.0 times higher than plasmin used as positive control. The fibrinolytic enzyme was confirmed directly from young leaves of C. japonica L. by a fibrin Plate and fibrin zymography. The protein was composed of a single polypeptide and its apparent molecular weight was found to be 45 kDa, as judged by SDS-polyacrylamide gel electrophoresis. The optimum pH and temperature for the fibrinolytic activity were pH 5.5 and $30^{\circ}C$, respectively. Also, the fibrinolytic activity was clearly inhibited by PMSF and TLCK, suggesting that it is a member of the trypsin-like serine protease. All these results suggest the protease is a fibrinolytic enzyme belong to a family of trypsin-like serine protease.

Biological Activities in roots of Glycyrrhiza uralensis Fisch (감초 세근의 생리활성 탐색)

  • The biological activities of ethanol, ethanol: water(1 : 1v/v) and water extracts from Glycyrrhiza uralensis Fisch, glycyrrhizin and enzymatically hydrolyzed glycyrrhizin were compared. About 50% of the growth of MCF7, A549, Hep3B and AGS cells were inhibited in adding 1.0 g/L of the crude extracts, glycyrrhizin and enzymatically hydrolyzed glycyrrhizin. For example, the ethanol extract inhibited 76%, 66% in MCF7 and Hep3B cells by adding 1.0 g/L. For cytotoxicity on human normal liver cell(WRL-68), the crude extracts were scored as above 26%. For the result of antimutagenecity using CHO V79 cell, the crude extracts proved more effective than other samples. The growth of human immune B and T cells were enhanced up to $1.2{\sim}1.3$ times by adding the crude extracts. In inhibitory effect of ${\alpha}-glucosidase$ activity was showed that the ethanol extract, water extract and ethanol: water (1 : 1v/v) extract were appeared 65%, 68%, 62% in adding 1.0 g/L. The higher enhancement of glutathione -S-transferase activity was observed in the ethanol extract as 257% compared to the control in adding 1.0 g/L. From the results, the biological activities of the crude extracts were equivalent or higher than glycyrrhizin and enzymatically hydrolyzed glycyrrhizin.
