Isolation and Morphological Characterization of Ttichoderma harzianum SJG-99721, a Powerful Biocontrol Agent (길항작용을 나타내는 Trichoderma harzianum SJG-99721의 분리 및 형태학적 특징)

  • Species of Genus Trichoderma are commercially applied as biological control agents against fungal Pathogens. A powerful biocontrol agent, Trichoderma sp. SJG-99721 was isolated from 305 isolates by morphological characters, chitinase activities and antifungal activities against Phytophthora capsiei. The isolate was identified as Trichoderma harzianum from various features such as growth rate at $27^\circ{C}$, significant growth ratio of $27^\circ{C}$ to $17^\circ{C}$, amount of aerial mycelium, types of branching: system, and disposition patterns of phialide and phialospore. Trichoderma harzianum SJG-99721 have been shown to act as a powerful biological agent against fungal phytopathogens; Botrytis cinerea, Rhizoctonia solani, Phytophthora cryptogea, Phytophthora capsiei, Sclerotinia sclerotiorum, Mycoshaerella melonis, Alternaria sotani, Fusarium oxysporum, Collectotrichum gloesporioodes, Alternaria alternata, Phythium ultimum, Phytophthora drechsleri, Pyricularia grisea.

Separation and growth monitoring of oyster mushroom spores using gravitational field-flow fractionation (GrFFF) (중력 장-흐름 분획법을 이용한 느타리버섯 포자의 분리 및 성장 모니터링)

  • Gravitational field-flow fractionation (GrFFF) is a separation technique that utilizes earth's gravity as the external field. GrFFF is a convenient tool for the size and/or density-based separation of micron-sized particles of various origins. In this study, GrFFF was employed for size-based separation of oyster mushroom spores. Oyster mushroom spores have smooth surface and are of cylindrical to narrow kidney-shapes with 5 to 12 im in longer dimension and 3 to 4 im in shorter dimension, as was confirmed by optical microscope (OM). GrFFF conditions were optimized for separation and characterization of spores by varying the channel flow rate from 0.5 to 1 mL/min. During the GrFFF elution of the spores, 3 fractions were collected to confirm the growth of oyster mushroom spore. The collected fractions were incubated for 30 days in water to examine the influence of the size on the growth of the spores. Results suggested that the oyster mushroom spores collected at the middle part of the GrFFF fractogram grew faster than those collected at the beginning or at the end of the fractogram.

Studies on the Compositional Change of Media During Oyster Muchroom Cultivation (느타리버섯 재배기간중 배지의 성분변화에 관한 연구)

  • In order to investigate the compositional change of the lauan and pine sawdust media during the coltivation of oyster mushroom (Pleurotus ostreatus) chemical components of the two media and mushroom were analized periodically from spawning to ending. The results are summerized as follows: 1) The total yield of mushroom from the lauan sawdust media was 6.39 kg/$m^2$, and from the Pine saw-dust media 6.31 kg/$m^2$, 94% of which was produced from the first and second cropping period. 2) In both media ash content increased gradually, while organic matter content decreased as the cultivation progressed. Moisture content, which was about 79% at the time of spawning, decreased a little during the priod of mycelial propagation, after which no change was observed. 3) Total nitrogen content of the two media decreased gradually as the cultivation progressed, and total loss of insoluble nitrogen was greater than that of soluble nitrogen. Amino nitrogen content tended to increase continually. 4) In both media pentosan, $\alpha$-cellulose, lignin and C/N ratio decreased gradually, while the content of reduced sugar, trehalose and mannitol continued to increase. 5) In both media phosphorus, potassium, manganese and zinc decreased, and magnecium, calcium, iron and copper showed irregular changes. 6) CMC-saccharifying and liquefying activity gradually increased from after mycelial propagation to the second cropping, after which it decreased in both media. Xylanase activity rapidly and greatly increased during the second cropping period rather than the first period. At the start of the third cropping period the activity decreased slowly. Protease activity was highest after mycelial propagati, after which it gradually decreased. The pH of the two media decreased as the cultivation progressed. 7) In both media little change was observed in the content of the components of mushroom cropped from the first to the third period, but slight decrease was noticed at the fourth cropping.

Studies on the Benomyl Resistance of Oyster Mushroom (Pleurotus spp.) (느타리버섯의 Benomyl 저항성(抵抗性)에 관한 연구(硏究))

  • The mycelial growth of some mushrooms was inhibited by benomyl treatment. The $ED_{50}$ of benomyl to that of Pleurotus spp., Agaricus bisporus and Flammulina velutipes was 25ppm, 50ppm and 200ppm, respectively, which indicates the former was the most sensitive to the fungicide. The mycelial growth of mushrooms growing on artificial media amended by benomyl was increased when they were cultured successively 5 times and 10 times on the media. The increasing rate of that of each mushroom was the highest at the concentration of $ED_{50}$ of benomyl. The mycelial growth of P. ostreatus was increased progressively as the number of successive culturing increased, while that of P. florida and A. bisporus was increased until they were cultured successively up to 5 times and 7 times, respectively, but they were decreased after that. Mutant sectors of mycelia were induced by successive treatment of benomyl. Mutant sectors of P. ostreatus appeared earlier than those of P. florida and all of them were induced earlier on the media of low contration of benomyl than on that of high concentration. The mycelia of mutant sectors induced by benomyl treatment grow faster than those of mother colony treated with benomyl successively, but there was no difference in resistance against the fungicide between them. The increase of mycelial growth of the mushrooms culturing successively on media containing benomyl indicated that they might obtain the resistance against benomyl.

Effects of Oyster Mushroom on Quality of Sulgidduk and Gyeongdan (느타리버섯이 설기떡과 경단의 노화억제에 미치는 영향)

  • To investigate the effects of oyster mushroom on the quality of Sulgidduk and Gyeongdan, freeze-dried mushroom power and mushroom extract were prepared. Dough for Gyeongdan was held for 0~6 hours at room temperature before cooking in order to allow for enzyme reaction. Upon addition of 2~5% mushroom power or 1~2% mushroom extract, the hardness of Sulgidduk decreased while the storage periods could be extended 1~2 days. Avrami rate constant (k) was 0.0111 for 2% power-added rice cake and 0.044 for the control. In Gyeongdan, hardness also decreased by addition of mushroom. Avrami rate constant (k) was 0.0178 for 10% mushroom power-added Gyeongdan and 0.0652 for the control. In a sensory test, Sulgidduk with 1% mushroom powder showed the highest score. Overall, addition of 1% mushroom powder to rice was appropriate for Sulgidduk. For Gyeongdan, addition of 1~2% mushroom powder was appropriate.

Study on the characteristics of fruit body growth according to incubation temperatures and period for oyster mushroom (느타리버섯 병재배 배양온도 및 배양기간에 따른 생육반응 연구)

  • This study was carried out to determine the proper pin-heading induction time(spawn running time) when different incubation temperature were applied to Pleurotus ostreatus(Chunchu 2ho, Suhan 1ho, Heukpyung). Incubation period was 17 days at 23 and 21 days at 17 for Chunchu 2ho, 17 days at 23 and 26 for Heukpyung, and 22~23 days at 17~23 for Suhan 1ho. Incubation period for Suhan 1ho was not significantly affected by incubation temperature. The time required for initial pin-heading was 4~5 days at 17, 20, 23 and 26 for Chunchu 2ho as well as Heukpyung, and was 3 days at 17, 20, 23 and 5~6 days at 26 for Suhan 1ho. As the incubation period became longer, the available fruit-bodies at Chunchu 2ho were made more but they were short. The yield of Chunchu 2ho and Heukpyung increased when incubated for 22~27 days at 20~23 and that of Suhan 1ho also increased when incubated for 22~23 days at 17~23.

Additive effect of tobacco waste in substrates on yield and mycelial growth of oyster mushroom, Pleurotus spp. and Trichoderma disease (느타리버섯 배지에 담배가루의 처리가 버섯균과 푸른 곰팡이병원균의 균사생장과 수량에 미치는 영향)

  • This study was carried out to investigate effect of adding tobacco waste to promote oyster mushroom, Pleurotus ostreatus, mycelial growth and control of Trichoderma disease. Tobacco waste was treated on the surface of rice straw inoculated with oyster mushroom strains. The mycelial growth treated with 0.1g tobacco waste was somewhat faster than non-treatment. In the case of mixed media with 1.0g tobacco waste, mycelial growth was increased compare to control, but adding 2.0 g tobacco waste decreased the mycelial growth. We also treated the tobacco waste on Trichoderma disease. All of the tobacco waste treatment force down the growth of Trichoderma disease. We treated tobacco waste on the surface of rice straw substrates. ASI 2072 mycelial growth was decreased regardless of amount of tobacco waster and method of treatment. Mycelial growth of ASI 2180 was somewhat decreased with treatment of 0.1g and 0.2g In the case of media mixed, mycelial growth of ASI 2072 was decreased in the overall treatment. Mycelial growth of other strains were decreased as only 2.0g treatment. Field test of mycelial growth was conducted using rice straw and waste cotton with substrates. Rice straw culture with treated 3% tobacco waste and cotton waste culture with treated 6% tobacco waste showed the highest yield. There was no effect to Trichoderma disease.

Trends of commercial strain development and spawn industry in mushrooms (버섯의 품종 육성과 종균 산업의 동향)

  • According to UPOV (International Union for Protection of New Varieties of Plants), mushroom spawn market have to be opened by the year 2009. Number of commercial strains distributed by the year 2005 were 179 of 24 species of edible and medicinal mushrooms. Only nine strains of oyster mushrooms were registered as protected variety, which is not compatible with those recorded in other advanced countries. Meaning of spawn in broad sense contains commercial strains. Development of commercial strains faces two main problems in Korea : One is the complicated genetic patterns and sexuality of mushroom species, and the other is expensive experimental equipments and fruiting body growing houses. Resolution of these problems leads to development of mushroom strains. This could be achieved as follows; genetic resources collection and assessment, molecular characterization of useful genetic characters, development of new commercial strains by hybridization using typical genetic resources, strengthening of breeding research using "Mushroom Breeding Group", management of spawn research company by consortium, foundation of mushroom general industry, promotion of consumption, and upgrade of competition ability for other countries. These points are under discussion.

Growth characteristics and productivity of oyster mushroom according to adding of Salvia miltiorrhiza (단삼 첨가에 따른 느타리버섯의 생육 특성 및 수량성)

  • This study was carried out to investigated optimum mixing ratio of Korean cultivation Salvia miltiorrhiza for production of functional oyster mushroom. Mycerial growth was slow at addition of Salvia miltiorrhiza, and significant difference by increase of Salvia miltiorrhiza. Yields of fruiting body show the highest to 139.5 g/850 ml of medium which are addition 5 g/bottle of Salvia miltiorrhiza but rapid decrease by increase of Salvia miltiorrhiza. Diameter of pileus and thick of stipes were higher at addition Salvia miltiorrhiza than those of the controls. Thick of pileus and length of stipes were the highest at addition 30g, and 20 g and 30 g of Salvia miltiorrhiza. The L value of stipes were the highest at addition 20 g and 50 g of Salvia miltiorrhiza and the L value of pileus were the highest at addition 30 g of Salvia miltiorrhiza, but there was no significant difference in the a-value and the b-value.

Nutritional contents and physiological activity of Pleurotus eryngii by extraction solvents (추출용매에 따른 큰느타리 버섯의 영양성분 및 생리활성)

  • Physiological activities of 70% methanol, fermented ethanol and hot-water extracts of Pleurotus eryngii were investigated. Free radical scavenging activities of P. eryngii extracts were determined according to the elimination of DPPH radicals. Their nitrite scavenging activity and total polyphenol content were also determined. Amino acid analysis showed that phenylalanine (Phe) and glutamic acid (Glu) are most abundant essential and non-essential amino acids in the analyzed extracts. The hot-water extract of ASI 2394 represented the highest antioxidant activity with the DPPH radical scavenging rate value of $40.97{\pm}1.65%$. ASI 2820 displayed the superior capacity to eliminate nitrate regardless of extraction solvents. The hot-water extract of ASI 2887 had the highest content of polyphenol. Our results showed that P. eryngii is well qualified as a functional food.
