Characteristics and Virulence Assay of Entomopathogenic Fungus Nomuraea rileyi for the Microbial Control of Spodoptera exigua (Lepidoptera: Noctuidae) (파밤나방의 미생물적 방제를 위한 병원성 곰팡이 Nomuraea rileyi의 특성 및 병원성 검정)

  • To date, chemical control remains the most common way to reduce beet armyworm (Spodoptera exigua) populations. However, this insect has become more tolerant or resistant to many chemical insecticides and the insect larvae usually hide inside hollow, tube-like leaves of host plant so they were difficult to kill by spraying insecticides. The use of viral and bacterial insecticide to solve these problems has not been successful because of their novel feeding habit. To overcome these problems, in this study, the biological characteristics and virulence of an entomopathogenic fungus isolated from the cadaver of larvae beet armyworm were investigated. Isolated entomopathogenic fungus was identified as Nomeraea rileyi (Farlow) Samson by morphological examinations and genetic identification using sequences of the ITS, ${\beta}$-tubulin gene and EF1-${\alpha}$ regions. This fungus was named as N. rileyi SDSe. Virulence tests against 3rd larvae of beet armyworm were conducted with various conidial suspensions from $1{\times}10^4$ to $10^8$ conidia/ml of N. rileyi SDSe in laboratory conditions. Mortality rate of beet armyworm showed from 20 to 54% and the virulence increased with increasing conidial concentrations. Although N. rileyi SDSe showed low mortality rate against beet armyworm, it is expected that N. rileyi SDSe will be used effectively in the integrated pest management programs against the beet armyworm.

Inheritance Study of Male Sterile Transformants Containing Pollen-specific Promoter and Diphtheria Toxin A Gene (수술특이프로모터와 디프테리아 독성 유전자에 의한 웅성불임 형질전환체의 후대 유전분석)

  • The objectives of this study were to investigate the genetic and phenotypic features of male sterile transformants by pollen-specific expression of diphtheria toxin gene and to find out inheritance patterns of transgene to the next generation. When backcrossed (BC) progenies were tested for expression of kanamycin resistance ($Km^R$), 9 lines out of 13 lines, except 4 lines ($BC_{1}5-13,\;BC_{1}5-23,\;BC_{1}5-28,\;BC_{1}5-32$), showed the ratio of $Km^R$ to kanamycin sensitive ($Km^S$), from 1:30 to all $Km^S$. As a result, they were much lower than Mendelian segregation of a dominant gene. To determine whether male sterility is a heritable and stable trait, 5 male sterile plants ($BC_{1}5-13,\;BC_{1}5-14,\;BC_{1}5-23,\;BC_{1}5-32,\;BC_{1}5-33$ lines) which had different transgene copy numbers were backcrossed as female parents with pollens from wild type. To confirm the existence of the DTx-A gene in the genome of the progenies, PCR was conducted using specific primers of the DTx-A coding region. A PCR band of 428 bp was obtained from each generation, which is the predicted size of the DTx-A gene fragment. Trangenes were inherited to the next $BC_4T_0$ progenies and showed male sterility, however, based on the copy numbers of DTx-A gene male sterile plants did not show predicted ratio. When male sterile plants were backcrossed with fertile plants, fruit capsule sizes and seed settings were relatively reduced from those of selfing wild type plants. The fruit sizes and seed settings were reduced in proportion to the increase in the copy number of DTx-A gene.

Metamorphism of the Buncheon and Hongjeas Granitic Gneisses (분천과 홍제사 화강암질 편마암체의 변성작용)

  • On the basis of lithology, the Precambrian Hongjesa Granitic Gneiss can be locally zoned into granoblastic granitic gneiss, porphyroblastic granitic gneiss, migmatitic gneiss from its center to the marginal part. There are no distinct differences in mineral assemblages by lithologic zoning, but it partly shows the change of mineral assemblage in the adjacent with migmatitic gneiss, thus mineral assemblage can be subdivided into Zone I and Zone II. In terms of mineral compositions, the characteristics of Zone I are coexisting K-feldspar+muscovite+sillimanite. The characteristics of Zone II are (1) breakdown of muscovite, (2) coexisting garnetScordierite, (3) coexisting garnet+cordierite + orthoamphibole. The Buncheon Granitic Gneiss is mainly composed of augen gneiss. In the adjacent area with Honjesa Granitic Gneisses, Buncheon Granitic Gneiss has the mineral assemblage of sillimanite+biotite+K-feldspar+(kyanite). Kyanite occurs as relict grains in the Buncheon and Hongjesa Granitic Gneissess. Kyanite shows anhedral to subhedral form and coexists with sillimanite in only one of these samples. Garnet from a migmatitic gneiss (Zone 11) has relatively high $X_{Fe}$ value in core and rim. Garnet from a porphyroblastic granitic gneiss(Zone I) has relatively homogemeous core but compositionally-zoned rim. Biotites show various colour from greenish-brown, brown to reddish brown at maximum adsorption. Also, the Ti, and Mg content in biotites increases from Zone I to Zone II. The plagioclases shows the chemical composition of $Ab_{84}An_{16}$ -$Ab_{70}An_{30}$ (oligoclase) in Zone I and $Ab_{70}An_{30}$ -$Ab_{50}An_{50}$(andesine) in Zone 11. These variations indicate that the gneisses in the study area experienced a upperamphibolite facies. The presence of kyanite as relict grains indicates that the metamorphic rocks in this area exprienced a high-temperature/medium-pressure type metamorphism, followed by high-temperaturellow-pressure metamorphism. Metamorphic P-T conditions for each gneiss estimated from various geothermobarometers and phase equilibria are 698-$729^{\circ}C$/6.3-11.3 kbar in augen gneiss, 621-$667^{\circ}C$/1.0-5.4 kbar in migmatitic gneiss, and 602-$624^{\circ}C$/1.9-3.4 kbar in porphyroblastic granitic gneiss. These data suggest that the study area was subjected to a clockwise P-T path with isothermal decompression (dP/dT=about 60 bar/$^{\circ}C$).

  • The Seongsan mine is one of the largest dickite deposits in the southwestern part of the Korean Peninsula. The main constithent minerals of the ore are dickite and quartz with accessory alunite, kaolinite and sericite. The geology around the Seongsan mine consists mainly of the late Cretaceous felsic volcanic rocks. In the studied area, these rocks make a synclinal structure with an axis of E-W direction plunging to the east. Most of the felsic volcanic rocks have undergone extensive hydrothermal alteration. The hydrothermally altered rocks can be classified into the following zones: Dickite, Dickite-Quartz, Quartz, Sericite, Albite and Chlorite zones, from the center to the margin of the alteration mass. Such zonal arrangement of altered rocks suggests that the country rocks, most of which are upper part of the rhyolite and welded tuff, were altered by strongly acid hydrothermal solutions. It is reasonable to consider that initial gas and solution containing $H_2S$ and other compounds were oxidized near the surface, and formed hydrothermal sulfuric acid solutions. The mineralogical and chemical changes of the altered rocks were investigated using various methods, and chemical composition of fifty-six samples of the altered rocks were obtained by wet chemical analysis and X.R.F. methods. On the basis of these analyses, it was found that some components such as $SiO_2$, $Al_2O_3$, $Fe_2O_3$, CaO, MgO, $K_2O$, $Na_2O$ and $TiO_2$ were mobilized considerably from the original rocks. The formation temperature of the deposits was estimated as higher than $200^{\circ}C$ from fluid inclusion study of samples taken from the Quartz zone. On the basis of the chemical composition data on rocks and minerals and estimated temperatures, the hydrothermal solutions responsible for the formation of the Seongsan dickite deposits were estimated to have the composition: $m_{K^+}=0.003$, $m_{Na^+}=0.097$, $m_{SiO_2(aq.)}=0.008$ and pH=5.0, here "m" represents the molality (mole/kg $H_2O$).

Mineralogy and Genetic Environments of the Seongdo Pb-Zn deposit, Goesan (괴산 성도 연-아연 광상의 산출광물과 생성환경)

  • The Seongdo Pb-Zn deposit, located in the northwestern part of the Ogcheon Metamorphic Belt, consists of skarn ore replacing limestone within the Hwajeonri Formation of Ogcheon Group and hydrothermal vein ore filling the fracture of host rock. Skarn minerals comprise mostly hedenbergitic pyroxene, garnet displaying oscillatory zonal texture composed of grossular and andradite, and a small amount of wollastonite, tremolite, and epidote, indicating reducing condition of formation. Ore minerals of skarn ore include sphalerite and galena with a small amount of pyrite, pyrrhotite, and chalcopyrite. In hydrothermal vein ore, arsenopyrite, sphalerite, chalcopyrite, and pyrite occur with a small amount of galena, native Bi, and stannite. Chemical compositions of sphalerite vary from 17.4 mole% FeS in average for dark grey sphalerite, 3.6 mole% for reddish brown sphalerite in skarn ore, and to 10.3 mole% FeS in hydrothermal vein ore. In comparison with representative metallic deposits in South Korea on the FeS-MnS-CdS diagram, skarn and hydrothermal vein ore plot close to the field of Pb-Zn deposits and Au-Ag deposits, respectively. Arsenic contents of arsenopyrite in hydrothermal vein ore decrease from 31.93~33.00 at.% in early stage to 29.58~30.21 at.% in middle stage, and their corresponding mineralizing temperature and sulfur fugacity are $441{\sim}490^{\circ}C$, $10^{-6}{\sim}10^{-4.5}atm$. and $330{\sim}364^{\circ}C$, <$10^{-8}atm$. respectively. Phase equilibrium temperatures calculated from Fe and Zn contents for coexisting sphalerite and stannite in hydrothermal vein are $236{\sim}254^{\circ}C$. Sulfur isotope compositions are 5.4~7.2‰ for skarn ore and 5.4~8.4‰ for hydrothermal vein ore, being similar or slightly higher to magmatic sulfur, suggesting that ore sulfur was mostly of magmatic origin with partial derivation from host rocks. However, much higher sulfur isotope equilibrium temperatures of $549^{\circ}C$$487^{\circ}C$, respectively for skarn ore and hydrothermal ore, than those estimated from phase equilibria imply that isotopic equilibrium has not been fully established.

Development Characteristics and Life Cycle of a Sciarid Fly(Lycoriella sp.) in Indoor Rearing (실내 사육에 의한 버섯파리(Lycoriella sp.)의 발육 단계별 특성 및 생활사)

  • This study was carried out to investigate the developmental characteristics and life cycle of sciarid fly (Lycoriella sp.), an important mushroom pest. The sciarid fly was reared ina growth chamber at 21$\pm$$1^{\circ}C$ and 55$\pm$5% of relative humidity with a 16h high, 8h dark cycle and Flammulina velutipes was used at diet. The egg was oval with 0.17 mm in diameter and 0.27 mm in length. Average egg period was 4 days. Body length of each instar from the 1st to 3rd was 0.7, 1.5, and 4.5, mm. respectively. Average weight of each instar was 1.9, 15.4 and 93.6 $\mu\textrm{g}$, respectively. In the 4th instar larvae, however, developmental characteristics of female and male became different by body length, weight and segment size. Body length and weight of the 4th instar female larvae were 5 mm and 162$\mu\textrm{g}$, and those of the male larvae were 3.5 mm and 90 $\mu\textrm{g}$, respectively. The larval periods of female and male were respectively 13.5 and13 days. Pupal length of female and male was 3.5 and 2.7 mm, and average weight was 136 $\mu\textrm{g}$ and 6.5$\mu\textrm{g}$, respectively. Adults were emerged from female and male pupae on 5.5 and on 5 days after pupation, respectively. Average longevity of male and female were 7 and 5 days, respectively. The weight of female was two-fold heavier than the male. After copulation, female laid approximately 130 to 150 eggs on/in the sporophore of mushroom. In conclusion, total lifespan of male and female were 29 and 28 days, respectively.

Petrology of the Tertiary Basaltic Rocks in the Yeonil and Eoil Basins, Southeastern Korea (한반도 동남부 제3기 연일, 어일분지에 나타나는 현무암질암의 암석학적 연구)

  • Eoil basalt in the Eoil basin and Yeonil basalt and its related volcanic rocks in Guryongpo and Daebo area were researched and analyzed to purse the tectonic settings and magma characteristics of those Tertiary volcanic rocks in the south-east Korean peninsula. It is highly suggested that zoning, resorption and sieve texture in plagioclase and reaction rim in pyroxene indicate unstable tectonic environments and complex volcanism in the study area. Volcanic rocks from Janggi basin are identified as basalt and basaltic andesite in TAS diagram and sub-alkaline series in terms of magma differentiation. $Na_2O$ and $K_2O$ show positive trend however FeO, CaO, MgO and $P_2O_5$ indicate negative trend in Harker variation diagram with $SiO_2$. Basaltic rocks from Eoil area are identified as calc-alkaline series in AFM diagram and show medium K series calc-alkaline in $K_2O-SiO_2$ diagram. Compatible trace elements of Co, Ni, V, Zn, and Sc in Yeonil basalt show negative trend with crystallization but incompatible trace element of Ba, Rb show positive trend with $SiO_2$ 0.81~1.00 of $Eu/Eu^*$ value suggests minor effect of plagioclase fractionation in Yeonil basaltic rocks. Plagioclase composition of Eoil basalt ranges from $An_{63.46-98.38}\;Ab_{1.62-32.96}\;Or_{0-3.58}$ (anorthite-labradorite) in core to $An_{40.89-82.44}\;Ab_{17.10-46.43}\;Or_{0-12.68}$ (bytownite-labradorite) in rim. $^{87}Sr/^{86}Sr$ and 143Nd;t44Nd ranges 0.704090~0.704717 and 0.512705~0.512822 respectively. Negative linear trends in 87Sr/86Sr and $^{143}Nd/^{144}Nd$ correlation diagram indicate that magma produced Yeonil basalt and basaltic andesite has been originated as partial melting product of mantle wedge by subducting Pacific plate affected by oceanic crust with less effect of continental crust indicating calc-alkaline magma characteristics.

Chemical and Optical Absorption Spectroscopic Study of Colored Tourmalines (유색 전기석의 화학적 및 광학흡수 분광학적 연구)

  • The chemical and optical absorption spectroscopic characters of pink and colorless tourmalines from San Diego mine in California, U.S.A., blue/green tourmalines from anonymous mine, Brazil, and brownis black tourmalines from Uncheon and Haksan mines in Korea have been studied using X-ray diffractometer, electron microprobe, optical absorption spectroscopy, and heat treatment. Least-squares refinements give unit cell diminsions : a = 15.96-16.01 ${\AA}$, c = 7.15-7.16 ${\AA}$ for the brownish black tourmalines, a = 15.82 - 15.87 ${\AA}$, c = 7.09 - 7.10 ${\AA}$ for pink tourmalines, and a = 15.88 - 15.94 ${\AA}$, c = 7.12 - 7.15 ${\AA}$ for blue green tourmalines. The colors of tourmalines are responsible for the transition elements. The pink color is attributed to the $Mn^{3+}$ ions, the blue-green to $Fe^{2+}$ and $Mn^{2+}$, bluish green to $Cu^{2+}$, and the brownish black to $Fe^{2+}$, $Fe^{2+}$ - $Fe^{3+}$, and $Fe^{2+}$ - $Ti^{4+}$. The $Mn^{3+}$ ions of pink color tourmalines are stabilized in the Y sites compressed along the O(1)H-O(3)H axis by Jahn-Teller distortion. Heating removes the pink or red component from tourmalines, producing the colorless stones from the pink and red ones. The bluish green samples change into the greenish blue ones and a certain yellowish green samples change into the light green ones by heat treatment. In the elbaite-schorl series, the concentration of Fe and Mn are variable depending on the color zones. The green zone is characterrized by the high content of Fe and Mn are variable depending on the color zones. The green zone is characterized by the high content of Fe, whereas the pink zone by the high content of Mn. Mn increases in deep yellow zone compared with yellow or colorless zones.

A Study on Gardening Theories of Li, Yu in 'Xian Qing Ou Ji' (한정우기(閑情偶寄)를 통해 본 이어(李漁)의 조원(造園) 이론)

  • Li Yu's Xian Qing Ou Ji was written during the late Ming and early Qing Dynasty when material culture flourished due to the development of the commercial economy. Xian Qing Ou Ji is considered a retrospective work that reflects the rich material society at that time. This study focuses on his gardening theories through the interpretation and analysis of these two texts of 'Jushibu(居室部)' and 'Zhongzhibu(種植部)' in Xian Qing Ou Ji. The results are summarized as follows. First, he explained the method of garden configuration and orientation according to the topography utilizing the theory of 'suiting one's measures to local conditions(因地制宜)'. He emphasized the detailed understanding of topography as the starting point for all the gardening theories. It stated that he tried to adjust the height of the terrain artificially to create an ideal form of 'anterior high and posterior low(前低後高)'. The successful arrangement of houses, pavilions, rockery and waterways et al. was able to be achieved because of his accurate understanding of topography. Second, 'borrowing scenery(取景在借)' means 'borrowing the outside scenery to the inside'. He applied this theory by 'viewing in motion(動觀)' and 'viewing in repose(靜觀)'. 'Viewing in motion' is seen as a positive methodology for landscape enjoyment. For example, the view through a window changes with movements of the boat, and it can portray thousands of landscape paintings which can be seen through the windows. In the case of 'viewing in repose', the window is regarded as a means of attracting outside garden views to the inside. Third, he emphasized 'the firmness of objects(制體宜堅)', where the design of windows and railings must be considered ahead of others. Fourth, he interpreted the contents of 'the pursuit of novelty(創異標新)'. This generalizes the characteristics of his gardening theory. The contents included 'hall wall(廳壁)', 'variable sun visor(活?)', 'hanging boards and couplets(聯?)' and 'method of supporting vines with bamboo screen(竹屛扶植法)'.

Alteration Zoning, Mineral Assemblage and Geochemistry of the Hydrothermal Clay Deposits Related to Cretaceous Felsic Magmatism in the Haenam Area, Southwest Korea (한국 서남부, 해남지역에서 백악기 산성마그마티즘에 관련된 열수점토광상의 누대분배, 광물조합의 지구화학적 연구)

  • In the present study, three clay deposits, named the Seongsan, Ogmaesan and Haenam deposits, were investigated. The altered zones are recognized in the hydrothermally altered rocks of the clay deposits from the center of the alteration to the margin: Kaolin, Kaolin-Quartz, Quartz, Sericite and Chlorite zones in the Seongsan deposits; Quartz zone, Alunite zone, Kaolin zone, Sericite zone and Chlorite zone in the Ogmaesan deposits; Quartz zone, Pyrophyllite zone, Sericite zone and Chlorite zone in the Haenam deposits. These zones can be grouped into two types of alteration: Acidic alteration such as Pyrophyllite zone, Alunite zone, Quartz zone, Kaolin zone, Kaolin-Quartz zone and a part of Sericite zone; Propylitic alteration such as Chlorite zone and a part of Sericite zone. All clay deposits belong to high-sulfidation (acid-sulfate) system. The rocks of the acidic alterations are composed of pyrophyllite, alunite, kaolin minerals, sericite, quartz and pyrite. On the basis of bulk chemical compositions, it was found that some components such as $SiO_2$, $TiO_2$, $Fe_2O_3$, FeO, MgO, CaO, $K_2O$ and $Na_2O$ were mobilized considerably from the original rocks. The mobility of these major elements is related to, and controls, mineral assemblages in each altered zone. Polytypes of sericite are determined as $2M_1$ and 1M by X-ray diffraction method. The amount of $2M_1$ is nearly equal to that of 1M in the Seongsan deposits whereas $2M_1$ is less and higher than that of 1M in the Ogmaesan and the Haenam deposits. These facts indicate that formation temperature of sericite is relatively high in the Haenam deposits, moderate in the Seongsan deposits, and low in the Ogmaesan deposits. The ratios of Na/(K+Na) for alunite in the Ogmaesan deposits determined by electron microprobe analyzer (EPMA) are higher than those in the Seongsan deposits. Thus, the alunite of the Ogmaesan deposits must have been formed from the solutions with relatively high aqueous Na/(K+Na) ratios and low pH at a high temperature rather than that of the Seongsan deposits. From all data, it is clarified that alunite is hypogene in origin, and has been formed by oxidation of hydrogen sulfide in the steam-heated environment, and that alunite has been produced by the spectacular solfataric alteration observed at the surface of some present-day hydrothermal systems.
