Development of an Analytical Method for the Determination of Pyriofenone residue in Agricultural Products using HPLC-UVD (HPLC-UVD를 이용한 농산물 중 살균제 pyriofenone 분석법 확립)

  • Pyriofenone is an aryl phenyl ketone fungicide that is newly registered in Korea in 2013 to control powdery mildew on food. The objective of this study was to develop reliable and sensitive analytical method for determination of pyriofenone residue in agricultural products for ensuring the food safety. The pyriofenone residues in all samples(Korean melon, pepper, potato, mandarin, soybean, and hulled rice) were extracted with acetonitrile, partitioned with dichloromethane, and then purified with a silica cartridge. The purified samples were analyzed by HPLC-UVD and confirmed with LC-MS. The linear range of pyriofenone was 0.05~5 mg/kg with the correlation coefficient ($r^2$) > 0.999. Average recoveries of pyriofenone ranged from 72.8% to 99.5% at the spiked level of 0.05 and 0.5 mg/kg, while the relative standard deviation was 2.3%~6.4%. In addition, the limit of detection and limit of quantification were 0.01 and 0.05 mg/kg, respectively. The results revealed that the developed and validated analytical method was suitable for pyriofenone determination in agricultural products.

Weed control and herbicide usage by seed potato growers in Kangwon alpine area (강원도 고냉지 씨감자 경작자들의 잡초방제 및 제초제 사용 실태)

  • The objective was to know if seed potato farmers in Kangwon alpine area control weeds effectively and use herbicides properly to control weeds. Seed potato farmers in Pyongcbang, Hoengsong, and Kangnung have noxious weeds, such as marsh pepper, lambsquarters, barnyard grass, mugwort, asiatic dayflower, hairy carbgrass, ladysthumb, common chickweed, field horsetail, and common purslain, in their farmyard. These are controlled by a combination of physical and chemical weed control methods. However, several weeds such as marsh pepper, field horsetail and, mugwort are poorly controlled by 2-fold amount of several herbicides, including paraquat, pendimethalin, and metribuzin (58% of the answered). Herbicides are not selected and used in a proper manner by seed potato farmers: most of farmers selected herbicides by their experience (51% of the answered) and dealers recommendation (34% of the answered), while only a few farmers of the answered took advantage of the Farmers Handbook for Herbicide Application. In addition, the selected herbicides, including non-registered ones such as paraquat and fluazifop-butyl, are applying three to five years in a succession. With these improper selection and usage of herbicides, sixty-seven percent of the answered farmers have experienced crop damage and seventy-five percent have poisoning. We conclude that a systemic educational program for the proper selection and use of herbicides is needed for seed potato farmers in Kangwon alpine area.

Selection and Validation of an Analytical Method for Trifludimoxazin in Agricultural Products with LC-MS/MS (LC-MS/MS를 이용한 농산물 중 Trifludimoxazin의 시험법 선정 및 검증)

  • Trifludimoxazin is a triazinone herbicide that inhibits the synthesis of protoporphyrinogen oxidase (PPO). The lack of PPO damages the cell membranes, leading to plant cell death. An official analytical method for the safety management of trifludimoxazin is necessary because it is a newly registered herbicide in Korea. Therefore, this study aimed to develop a residual analysis method to detect trifludimoxazin in five representative agricultural products. The EN method was established as the final extraction method by comparing the recovery test and matrix effect with those of the QuEChERS method. Various sorbent agents were used to establish the clean-up method, and no differences were observed among them. MgSO4 and PSA were selected as the final clean-up conditions. We used LC-MS/MS considering the selectivity and sensitivity of the target pesticide and analyzed the samples in the MRM mode. The recovery test results using the established analysis method and inter-laboratory validation showed a valid range of 73.5-100.7%, with a relative standard deviation and coefficient of variation less than 12.6% and 14.5%, respectively. Therefore, the presence of trifludimoxazin can be analyzed using a modified QuEChERS method, which is widely available in Korea to ensure the safety of residual insecticides.

Development of Analytical Method for Detection of Fungicide Validamycin A Residues in Agricultural Products Using LC-MS/MS (LC-MS/MS를 이용한 농산물 중 살균제 Validamycin A의 시험법 개발)

  • Validamycin A is an aminoglycoside fungicide produced by Streptomyces hygroscopicus that inhibits trehalase. The purpose of this study was to develop a method for detecting validamycin A in agricultural samples to establish MRL values for use in Korea. The validamycin A residues in samples were extracted using methanol/water (50/50, v/v) and purified with a hydrophilic-lipophilic balance (HLB) cartridges. The analyte was quantified and confirmed by liquid chromatograph-tandem mass spectrometer (LC-MS/MS) in positive ion mode using multiple reaction monitoring (MRM). Matrix-matched calibration curves were linear over the calibration ranges (0.005~0.5 ng) into a blank extract with $R^2$ > 0.99. The limits of detection and quantification were 0.005 and 0.01 mg/kg, respectively. For validation validamycin A, recovery studies were carried out three different concentration levels (LOQ, $LOQ{\times}10$, $LOQ{\times}50$, n = 5) with five replicates at each level. The average recovery range was from 72.5~118.3%, with relative standard deviation (RSD) less than 10.3%. All values were consistent with the criteria ranges requested in the Codex guidelines (CAC/GL 40-1993, 2003) and the NIFDS (National Institute of Food and Drug Safety) guideline (2016). Therefore, the proposed analytical method is accurate, effective and sensitive for validamycin A determination in agricultural commodities.

Alternative Fungicide Spraying for the Control of Powdery Mildew Caused by Sphaerotheca fusca on Greenhouse Watermelon (Citrullus lanatus) (시설 수박 흰가루병 방제를 위한 살균제 교차방제 체계)

  • Alternative spraying was developed using fungicides and potassium phosphonate to control watermelon powdery mildew (WPM) caused by Sphaerotheca fusca under greenhouse condition. In 2010 trial, a total of 9 combinations of 6 fungicides which are legally registered in Korea and currently used on watermelon farms were tested for the control efficacy against WPM. Though the combination of chlorothalonil (Chl)-myclobutanil (My)-fenarimol (Fen)-hexaconazole (Hex) was the most effective (82.6%) in the first-year trial, it had the problem that My, Fen and Hex have the same mode of action. Therefore, in the second-year trial to improve the fungicide combination and to reduce fungicide application, fungicides and potassium phosphonate (P) combinations were tested for WPM control using 500 ppm aqueous potassium phosphonate solution which is both eco-friendly and showing different mode of action from the fungicides tested. The P-My-Fen-Hex applied at 10-day intervals provided relatively high control (75.6%), whereas Chl-My-Fen-P provided 31.8% control. To improve the control, fungicides and P were applied at 7-day intervals in the third-year trial. Though Chl-My-Fen-Hex provided the highest control (89.6%), P-My-Fen-Hex also provided relatively high control (83.5%) compared to 65.9 % control with Chl-My-Fen-P, which suggests that Chl-My-Fen-Hex could be replaced with P-My-Fen-Hex for WPM control.

Enhancement of Analytical Method for Thidiazuron Residues and Monitoring of its Residues in Agricultural Commodities (농산물 중 thidiazuron 잔류분석법 개선 및 잔류실태 조사)

  • BACKGROUND: This study was conducted to develop analytical method with reproducibility, accuracy and applicability to agricultural products than the existing methods. METHODS AND RESULTS: Mean recoveries of thidiazuron ranged from 89.2 to 91.2 in hulled rices, 87.2 to 92.1 in peppers, from 76.4 to 86.9 in potatoes, from 91.2 to 95.7 in watermelons, from 86.5 to 88.5 in kiwi fruits, and from 89.5 to 94.0 in grapes, with less than 10% of relative standard deviations. In addition, the limit of quantitation was set to be 0.05 mg/kg and there were no interfering peaks in integrating the thidiazuron peak. CONCLUSION(S): These results represent that the enhanced analytical method has reliable accuracy, precision, selectivity, and sensitivity.

Effective Fungicides on Control of Stem and Root Rot of Paprika Caused by Nectria haematococca (Nectria haematococca에 의한 착색단고추 줄기 및 과실썩음병의 약제방제)

  • A previous study reported that the stem and root rot of paprika (Capsicum annum L. var, grossum) caused by Nectria haematococca became a threat to safe cultivation of the plant in the country. However, no strategies for control the disease have been suggested. In this study, fungicides registered for pepper were screened to evaluate their control effects on the disease. Among fungicides tested, prochloraz manganase complex com pletely suppressed mycelial growth of the pathogen at 10 ppm a.i. tebuconazole, benomyl, and carbendazim $\cdot$kasugamycin also effectively inhibited mycelial growth of the fungus. However, kresoxim-methyl and triflox ystrobin did not suppress mycelial growth but significantly suppressed conidial germination of the fungus. Azoxystrobin, benomyl, prochloraz, tebuconazol, and carbendazim$\cdot$ kasugamycin were also effective to retard conidial germination. In vivo tests, tebuconazole strongly inhibited the plant growth even at 16,000x (15.6 ppm a.i.), while others did not induce chemical injury at 4,000x or 8,000x when drenched into a rockwool cube. In a greenhouse test, prochloraz manganase complex at 125 ppm a.i. (4,000x) showed highest control value by $89.9\%$. Other fungicides thiophanate-methylthiram, axozystrobin, trifloxystrobin, and benomyl presented $60-80\%$ control value in the hydroponic cultivation system. However, application time and interval remained to be investigated for identify maximum residue limit.

  • In here, we investigated the quantitative analysis method of indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) with liquid chromatography-mass/mass spectrometry (LC-MS/MS) or gas chromatography-MS. Two ways of clean-up process were investigated for LC-MS/MS instrumental analysis of IAA, but both a simple dilution and hydrophile-lipophile balance (HLB) solid phase extraction (SPE) were not met the optimal recovery rates for quantitative analysis. On the other hand, the clean-up method for GC-MS was finally optimized through HLB-SPE from 250-folds diluted sample and methylation with trimethylsilyl chloride in methanol for 4 h. The limit of detection for methyl ester of IAA and IBA were both 1.4 mg/L, and recovery rates showed 93-107% from the concentrated liquid fertilizer.

Susceptibility of Myzus persicae on Potato field and Riptortus clavatus on Soybean field to Insecticides treated by Multi-copter (농업용 멀티콥터를 활용한 감자의 복숭아혹진딧물과 콩의 톱다리개미허리노린재의 약제방제 효율)

  • The Aphid, Myzus persicae, and the bean bug, Riptortus clavatus, are major insects in crops. This study examined the insecticide susceptibility and phytotoxicity of insecticides dispersed using an Unmanned Aerial Vehicle (UAV, multi-copter) against the insects. Sulfoxaflor suspension concentrate (SC, 16X) on potato fields and etofenprox, methoxyfenzide suspo-emulsion(SE, 8X) on soybean fields were dispersed after deploying water-sensitive paper within the field to measure the distribution pattern and coverage index of the falling insecticide. Both insecticides showed a controlled mortality of 76.4% against aphids and 97.5% and 94.4% against the 2nd nymphal, and 5th nymphal stage of the bugs, respectively. The droplet distribution was less than 0.5mm, and coverage analysis revealed an inside and outside coverage of 3.1 and 1.6, respectively. The surrounding area was affected by insecticide spraying using a multi-copter. This study is expected to help expand UAV control and use it safely in the future.

Evaluation of Environment-friendly Control Agents for the Management of Powdery Mildew Infection during Seedling Stage of Three Cucurbitaceae Vegetables (친환경 육묘시 세 가지 박과채소의 흰가루병에 대한 친환경 제제의 방제효과)

  • The purpose of this study was to evaluate the control efficacy of major environment-friendly control agents against powdery mildew, as affected by the application condition such as disease-developing stage and microclimate, as compared with the efficacy of pesticides in plug seedling of three Cucurbitaceae vegetables, including cucumber, melon, and oriental melon. Single or combined application of major six environment-friendly control agents was used in the experiment: two biofungicide (Ampelomyces quisqualis 94013 and Bacillus subtilis Y1336), two plant extracts (neem oil and extracts of Rheum undulatum), and two mineral materials (wettable sulfur powder and lime bordeaux mixture). These control agents were treated to the plug seedlings for preventing powdery mildew and curative applications for managing powdery mildew. In all treatments, the disease incidence declined as daily average temperatures increased to $30^{\circ}C$ for consecutive 6~8 days with maximum temperature over $40^{\circ}C$. In preventative application, the control efficacy against powdery mildew was the highest in the treatment of wettable sulfur powder, and lowest in the B. subtilis Y1336, with values of 20~40%. In cucumber seedlings, the preventive single application of neem oil or wettable sulfur powder was more effective than curative application of fungicides, while the control efficacy of these agents was similar to those of fungicides in melon seedlings. The single application of R. undulatum extracts was also effective in preventing the disease for both cucumber and melon seedlings, showing a higher control efficacy than those of biofungicides during seedling stage. The treatment of water spray was not effective and showed a higher disease incidence than the untreated control plot in the oriental melon and melon seedlings. The curative application with environment friendly control agents, after powdery mildew was first detected, could not successfully controlled the disease at the middle stage (5~10% of disease incidence) of disease development. The curative combined application of [R. undulatum extracts (1st application) + wettable sulfur powder (2nd) + neem oil (3rd)] showed the highest control efficacy among the other treatments, with control value over 80% at the early stage (less than 1% of disease incidence) of disease development.
