  • 모드 동기된 아르곤 이온 레이저를 여기광원으로 사용한 시간상관 단일 광자 계수장치를 이용하여 에탄올 용액 내에서 Rhodamine 6G에서 Rhodamine B로의 에너지 전달에 관하여 연구하였다. 측정된 형광 소멸 곡선은 본 실험실에서 개발한 full-fit program으로 deconvolution 하여 reduced concentration 과 critical transfer distance를 구했다. donor 의 농도가 acceptor의 농도보다 작을 경우에는 Foster 모델이 클 경우에는 Huber 모델이 잘 맞음을 확인하였다.

  • 1.이온토포레시스에 의한 ISP의 경피촙수증가는 단순투과의 약 13배로서 그 증가정도는 전류세기와 약물농도에 비례하였다. 2. 가해주는 $Na^{+}$ 농도가 커질수록 ISP의 flux는 감소하였다. 3. ion-pairing agent률 가하면 ISP의 flux는 감소하는데, 그 감소정도는 TU>SAL>BEN 로서 이는 이 물질들이 ISP와 ion-pair를 형성하는 능력순서와 같았다. 4. ISP용액의 pH증가시 ISP의 flux는 대체적으로 증가하며 그 pattern은 피부의 pKa를 3.5로 가정할 때의 피부해리곡선과 유사하였다. ISP가 광범위한 pH에서 완벽하게 해리된다고 가정할 때 pH증가시 flux증가는 피부해리 증가에 따른 permselectivity 증가에 기인한 것으로 생각되었다.

Transport and Fate of Benzene in a Sandy Soil (사질토양에서의 Benzene의 이동성에 관한 연구)

  • Hydrocarbon compounds in vadose zone soils caused by adsorption onto the surfaces of solid particles are generally considered to show retardation effect. In this study, we investigated the retardation effect on the transport of Benzene in a sandy soil by conducting batch and column tests. The batch test was conducted by equilibrating dry soil mass with Benzene solutions of various initial concentrations. and by analyzing the concentrations of Benzene in initial and equilibrated solutions using HPLC. The column test consisted of monitoring the concentrations of effluent versus time known as a breakthrough curve (BTC). We used KCl and Benzene solutions with the concentration of 10 g/L and 0.88 g/L as a tracer, and injected them into the inlet boundary of the soil sample as a square pulse type respectively, and monitored the effluent concentrations at the exit boundary under a steady state condition using an EC-meter and HPLC. From the batch test, we obtained a distribution coefficient assuming that a linear adsorption isotherm exists and calculated the retardation factor based on the bulk density and porosity of the column sample. We also predicted the column BTC curve using the retardation factor obtained from the distribution coefficient and compared with the measured BTC of Benzene. The results of the column test showed that i) the peak concentration of Benzene was much smaller than that of KCl and ⅱ) the travel times of peak concentrations for the two tracers were more or less identical. These results indicate that adsorption of Benzene onto the sand panicles occurred during the pulse propagation but the retardation of Benzene caused by adsorption was not present in the studied soil. Comparison of the predicted with the measured BTC of Benzene resulted in a poor agreement due to the absence of the retardation phenomenon. The only way to describe the absolute decrease of Benzene concentration in the column leaching experiment was to introduce a decay or sink coefficient in the convection-dispersion equation (CDE) model to account for an irreversible sorption of Benzene in the aqueous phase.

Pharmacokinetics of oxolinic acid in cultured olive flounder Paralichthys olivaceus by oral administration, injection and dipping (Oxolinic acid의 경구투여, 주사 및 약욕에 따른 넙치, Paralichthys olivaceus 체내 약물동태학적 특성)

  • The pharmacokinetic properties of oxolinic acid (OA) were studied after oral administration, intraperitoneal injection and dipping to cultured olive flounder, Paralichthys olivaceus (average 90 g, $23{\pm}1{^{\circ}C}$). Plasma samples were taken at 3, 5, 10, 15, 24, 30, 48, 96 and 144 h post-dose. In oral dosage at 15, 30 and 60 ㎎/㎏, the peak plasma concentrations of OA, which attained at 10~15 h post-dose, were 1.92, 2.45 and 3.72 $\mu{g}/m\ell$, respectively. In intraperitoneal injection with 10 and 20 ㎎/㎏, the peak plasma concentrations of OA, which attained at 10 h post-dose, were 4.1 and 4.8 $\mu{g}/m\ell$, respectively. In dipping in 30 and 50 ppm for 1 h, peak concentrations were observed at 5 h and 30 h post-dose, were 0.22 and 0.38 $\mu{g}/m\ell$, respectively. The kinetic profile of absorption, distribution and elimination of OA in plasma were analyzed fitting to a one-compartment model by WinNonlin program. Calculated parameters for a single oral dosage of 15, 30 and 60 ㎎/㎏, respectively, were: AUC (the area under the concentration-time curve)=70.93, 120.0 and 141.86 $\mu{g}$ $h/m\ell$ $T_{max}$ (time for maximum concentration)=16.22, 20.39 and 17.33 h; $C_{max}$ (maximum concentration)=���D1.61, 2.40 and 3.01 $\mu{g}/m\ell$. Following intraperitoneal injection of 10 and 20 ㎎/㎏, these parameters were AUC=184.7 and 315.92 $\mu{g}$ $h/m\ell$ $T_{max}$=5.91 and 6.26 h; $C_{max}$=4.19 and 4.45 $\mu{g}/m\ell$. Following dipping at 30 and 50 ppm, these parameters were AUC=17.58 and 21.69 $\mu{g}$ $h/m\ell$ $T_{max}$=19.08 and 31.43 h; $C_{max}$x=0.22 and 0.25 $\mu{g}/m\ell$.

Microfluidic System for the Measurement of Cupric Ion Concentration using Bilayer Lipid Membrane on Silver Surface (은 표면의 이중층 지질막에 의한 구리 이온 농도 측정용 마이크로플루이딕 시스템)

  • A microfluidic system has been developed using biomaterial for the measurement of cupric ion concentration. The cell-membrane-mimicking bilayer lipid membrane(BLM)-coated silver electrode was used for the sensing of cupric ion concentration. The silver-supported BLM could increase its stability. A silver-supported bilayer lipid membrane(s-BLM) was easily obtained using its self-assembling characteristics by immersing silver wire into lipid(phosphatidylcholine; PC) solution and then dipping into aqueous KCl solution. These s-BLMs were used to determine the relationship between $Cu^{2+}$ concentration and current crossing s-BLM. Their relationship showed high linearity and reproducibility. The calibration curve was constructed to express the relationship between $Cu^{2+}$ concentration and current in the $Cu^{2+}$ concentration range of 10 and $130{\mu}M$. This calibration curve was used to measure $Cu^{2+}$ concentration in an unknown sample. Microfluidic system with s-BLM was made of PDMS(polydimethyl siloxane) using typical soft photolithography and molding technique. This integrated system has various functions such as activation of the silver surface without cutting silver wire, coating of BLM on silver surface, injection of KCl buffer solution, injection of $Cu^{2+}$ sample and measurement of $Cu^{2+}$ concentration in the sample.

카드뮴에 노출된 수생식물 5종의 엽록소형광 분석

  • 개구리밥, 생이가래, 은행이끼, 수련, 부들 등 5종의 식물을 대상으로 카드뮴 $(Cd^{2+})$의 농도를 달리하여 처리한 후 시간별로 식물의 광합성효율을 분석하였다. 그 결과, 개구리밥, 생이가래, 은행이끼, 부들 등 4종은 카드뮴에 민감한 것으로 조사되었고, 수련은 다른 식물종에 비해 덜 민감한 것으로 나타났다. O-J-I-P 곡선의 패턴을 살펴보면, 종에 따라 다소의 차이는 있으나 개구리밥, 생이가래, 은행이끼, 부들 등 4종의 식물은 카드뮴 농도가 증가할 수록 J, I, P값이 크게 감소하는 것을 볼 수 있었다. 특히 P-단계의 감소는 개구리밥에서 현저하게 나타나 다른 3종 보다 카드뮴에 민감한 것으로 보인다. 광계II의 광합성효율(Fv/Fm) 변화양상을 보면, 개구리밥과 생이가래는 카드뮴 농도가 50uM 이상에서는 배양 1일부터 Fv/Fm값이 급격하게 감소하였으며, 은행이끼와 부들은 3일 이후부터 50uM 이상의 카드뮴 농도하에서 Fv/Fm값이 감소하였다. 수련은 큰 변화를 보이지 않았다.

  • Czochralski pulling method에 의해 성장된 고농도 도핑 Nd:LSB의 광학적 특성을 조사하였다. 결정 성장조건에 의해 달라지는 space group은 지금까지 C2/c, R32, Cc와 C2등 4가지가 보고되었으며, 기본적으로 6방정계의 구조를 가지고 비선형 광학 결정으로서의 특성을 보였다. x선 회절 분석계를 이용하여 구조를 분석하였고, 도핑 농도에 따른 형광 수명의 변화, 분산곡선 특성을 조사하였다. 또한 흡수 및 저온 형광 스펙트럼을 이용하여 도핑 농도 및 space group의 변화에 따른 에너지 준위의 변화를 관찰하고 비교하였다.

Rheological Properties of Citrus Pectin Solutions in the Presence of NaCl (감귤류 펙틴의 리올로지 특성에 대한 NaCl 첨가효과)

  • 농후한 점도영역에서 감귤류 펙틴용액에 NaCl을 첨가하였을 때 점도 및 점탄성 변화를 연구하였다. NaCl의 농도가 증가함에 따라 펙틴용액의 점도도 점차 증가하였으며, 1.5 M의 NaCl을 첨가한 경우 점도가 약 3배 증가하였다. NaCl의 첨가에 따른 점도특성의 일반화 곡선으로서 ${\eta}/{\eta}_0$$\dot{\gamma}/\dot{\gamma}_{0.8}$ 관계는 상대적으로 낮은 중첩도를 보였다. Zero-shear viscosity $({\eta}_0)$와 NaCl 농도(C)와는 ${\eta}_0\;{\propto}\;10^{0.32C}$의 관계를 보였으며, 이때 상관계수(R2)는 0.995였다. 한편, NaCl 농도가 증가함에 따라 저장탄성률(G#)과 손실탄성률(G@)이 모두 증가하였다.

Assessment of Rainfall-Sediment Yield-Runoff Prediction Uncertainty Using a Multi-objective Optimization Method (다중최적화기법을 이용한 강우-유사-유출 예측 불확실성 평가)

  • In hydrologic modeling, prediction uncertainty generally stems from various uncertainty sources associated with model structure, data, and parameters, etc. This study aims to assess the parameter uncertainty effect on hydrologic prediction results. For this objective, a distributed rainfall-sediment yield-runoff model, which consists of rainfall-runoff module for simulation of surface and subsurface flows and sediment yield module based on unit stream power theory, was applied to the mesoscale mountainous area (Cheoncheon catchment; 289.9 $km^2$). For parameter uncertainty evaluation, the model was calibrated by a multi-objective optimization algorithm (MOSCEM) with two different objective functions (RMSE and HMLE) and Pareto optimal solutions of each case were then estimated. In Case I, the rainfall-runoff module was calibrated to investigate the effect of parameter uncertainty on hydrograph reproduction whereas in Case II, sediment yield module was calibrated to show the propagation of parameter uncertainty into sedigraph estimation. Additionally, in Case III, all parameters of both modules were simultaneously calibrated in order to take account of prediction uncertainty in rainfall-sediment yield-runoff modeling. The results showed that hydrograph prediction uncertainty of Case I was observed over the low-flow periods while the sedigraph of high-flow periods was sensitive to uncertainty of the sediment yield module parameters in Case II. In Case III, prediction uncertainty ranges of both hydrograph and sedigraph were larger than the other cases. Furthermore, prediction uncertainty in terms of spatial distribution of erosion and deposition drastically varied with the applied model parameters for all cases.

Quenching Curves for VOCs in the Analysis of Groundwater $^{14}C$ using Liquid Scintillation Counter (액체섬광계수기를 이용한 지하수의 $^{14}C$ 측정에서 휘발성유기화합물(VOC)에 대한 소광곡선)

  • Optimal quenching curves have been studied for the accurate analysis of $^{14}C$ in groundwater polluted by reducing efficiency of volatile organic compounds (VOCs) in liquid scintillation counter (LSC). The quenching parameters (SQP(E)) were measured for ten VOCs such as benzene, toluene, ethylbenzene, o-(m-,p-)xylene, trichloroethylene (TCE), tetrachloroethylene(PCE), carbon tetrachloride and chloroform. The quenching curves were plotted using $^{14}C$ standard solution and chloroform as a quenching agent. Optimal plotting conditions were determined for standard solution, LSC measuring time and the concentration of chloroform. The quenching effects of chlorinated organic compounds such as TCE, PCE, carbon tetrachloride and chloroform were greater than those of BTEX (benzene, toluene, ethylbenzene and xylene). Optimum measuring time was 100 minutes far 7,000 dpm/mL standard solution. A few mL of chloroform should be added for good quenching curves. These quenching curves have good correlation coefficients (> 0.99) and the curves could be applied to accurate analysis of $^{14}C$ in groundwater and tap water.
