Optimization of Tyrosinase Inhibitory Activity in the Fermented Milk by Lactobacillus plantarum M23 (Lactobacillus plantarum M23 균주를 이용한 Tyrosinase 저해 활성 발효유 생산의 최적화)

  • The melanin pigment in human skin is a major defense mechanism against ultraviolet light to the skin, but darken skin color. Tyrosinase is mainly responsible for melanin biosynthesis (melanogenesis) in animals and enzymatic browning (melanosis) in plants. The purpose of this study was to optimize the fermented milk process for the melanin formation inhibition by using Lactobacillus plantarum M23 with tyrosinase inhibitory activity. We used 4-factor-3-level central composite design combining with response surface methodology. Yeast extract concentration (%, $X_1$), addition of grape (%, $X_2$), incubation temperature ($^{\circ}C$, $X_3$) and incubation time (h, $X_4$) was used as an independent factor, on the other hand, pH (pH, $Y_1$), overall palatability (score, $Y_2$) and tyrosinase inhibitory activity (%, $Y_3$) was used as a dependant factor. Based on the optimization for the highest tyrosinase inhibitory activity with pH 4.4, the expected data of pH, palatability and tyrosinase inhibitory activity with 14.8 h incubation at $37.1^{\circ}C$ by the addition of 0.127% of yeast extract, 2.95% of grape was 4.42, 7.06 and 86.65%, but the real data was 4.35, 6.86 and 84.05%, respectively. Based on the previous results, fermented milk using Lactobacillus plantarum M23 with the tyrosinase inhibitory activity could contribute for the whitening and antiaging of human skin.

Functions of Various Hydrocolloids as Anticaking Agents in Korean Rice Cakes (친수성콜로이드류의 떡 응고방지에 관한 연구)

  • This study was carried out to investigate the influences of hydrocolloids on suppression of retrogradation in the Korean rice cake. Hydrocolloids were influenced to the textural characteristics of the Korean rice cake. In particular arabic gum and carrageenan had the effect of suppressing the retrogradation. Cohesiveness of the Korean rice cake added with hydrocolloids was shown to be high compared to control in organoleptic characteristics. Hydrocolloids exhibited slightly effective in retarding surface color change of the Korean rice cake. Onset temperature of gelatinization in thermal characteristics showed somewhat low in case of addition of arabic gum into the Korean rice cake and was followed by carrageenan, guar gum, gelatin and locust bean gum in onset temperature of gelatinization order. All samples added with hydrocolloids had low values in change of melting enthalphy and their range was 12.8 ∼ 17.7 J/g. The Korean rice cake added with arabic gum was lowest in melting enthalphy. The recrystallinity of the Korean rice cake added with arabic gum was greatly low compared to rice cake without hydrocolloids. Melting spreadability of the Korean rice cake added with arabic gum or carrageenan was superior in thermal characteristics. The value of exponent of Avrami was 0.97 ∼ 1.12 in study of Avrami equation. In conclusion carrageenan and arabic gum would have a suppressive effect of retrogradation in the Korean rice cake.

  • White jelly fungus (Tremella fuciformis Berk; TF) has been used as a traditional medicine in Asia; it is known to prevent hypertension, aging, cancer, and arteriosclerosis. This study aimed to investigate the anti-diabetic effects of fermented Tremella fuciformis Berk (FTF) ethanol extracts fermented with L. rhamnosus BHN-LAB 76. We show that FTF increases the ${\alpha}$-glucosidase inhibitory activity and suppress the adipogenesis of 3T3-L1 adipocytes. These inhibitory effects of FTF are accompanied by the regulation of the phosphorylation of AMPK, JNK, and Akt. These data demonstrate that FTF not only inhibits adipogenesis by affecting the adipogenic signaling, but also increases the anti-diabetic effects by regulating the insulin signaling pathway. Therefore, we suggest that the FTF can be used for developing functional food and cosmetics materials.

Phenolic compounds from the flowers of Cosmos bipinnatus and their anti-atopic activity (코스모스(Cosmos bipinnatus) 꽃으로부터 phenolic 화합물의 분리 동정과 항아토피 효과)

  • The flowers of Cosmos bipinnatus were extracted with solvent made with methanol:water (4:1) and the concentrates were partitioned into ethyl acetate (EtOAc), n-butanol (n-BuOH), and water (H2O) fractions. The octadecyl silica gel (ODS) and silica gel (SiO2) column chromatographies were repeated for the EtOAc fraction to isolated of two phenolic compounds. The chemical structure of the isolated compounds were identified as benzyl O-β-ᴅ-glucopyranoside (1), and 2-phenylethyl O-β-ᴅ-glucopyranoside (2) through spectroscopic datas such as nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy. These two compounds were first isolated from C. bipinnatus flowers through this study. To evaluate the anti-atopic activity of the two isolated compounds using a HaCaT cell line induced by ultraviolet light, several experiments were conducted and neither both compounds showed toxicity in the concentration range of 1 to 1,000 ㎍/mL. In the results of anti-atopic activity through Thymus and activation regualted chemokine (TARC) assay, both compounds showed dose-dependent TARC inhibitory activity. In particular, compound 1 showed significant activity even in a low concentration range of 10 ㎍/mL, and in different concentration ranges. Also compound 1 showed higher inhibitory activity than other compound, confirming that the anti-atopic activity was the most excellent. Based on these results, it is considered that it can be used as a functional cosmetic material.

The Effect of Collagen Peptide Intake on UVB-induced Skin Damage in Hairless mice (콜라겐 펩타이드 섭취 무모생쥐에서 UVB 조사에 의한 피부 손상에 미치는 효과 규명)

  • This study examined the effects of collagen peptide intake extracted from the tilapia fish (TFCP) on UVB-induced skin in hairless mice. To cause photoaging, the hairless mice were exposed to UVB irradiation three times a week and TFCP was administered with 545 and 1090 mg/kg once a day for 12 weeks. The level of wrinkle formation, thickness of the skin, acute erythema, skin hydration, amount of hydroxyproline, expression of MMPs and filaggrin were measured. Wrinkle formation, thickness of the skin, and acute erythema were significantly lower and skin hydration was higher in the TFCP intake group compared to the UVB-control mice group. In addition, the amount of the collagen and hydroxyproline of the collagen peptide intake group were much higher than the UVB-control group. Oral administration of TFCP inhibited the expression of the MMP-3 and MMP-13 mRNA level as well as activated MMP-2 and MMP-9 induced by UV irradiation, and increased the protein expression of filaggrin in hairless mice skin. In summary, the oral administration of TFCP can inhibit wrinkle formation and protect the skin from UV-damage. Therefore, this study suggests that TFCP is a potential candidate material for skin functional foods.

Whitening Effects of Lactobacillus rhamnosus Associated with Its Antioxidative Activities (Lactobacillus rhamnosus 파쇄물의 항산화 및 미백효과)

  • This study was performed to investigate the whitening effects of Lactobacillus rhamnosus in addition to its antioxidative activities. The cytotoxicity of the Lactobacillus rhamnosus was 7.6% at 10.0% (v/v) concentration. Its cytotoxicity was lower than 3.2% of Lactobacillus casei when adding the same concentration. Lactobacillus rhamnosus exhibited high antioxidative activities at 14.9% of DPPH radical scavenging activity, and a lower reducing power was measured. Lactobacillus casei exhibited relatively lower antioxidative activities at 13.4%. The tyrosinase inhibition activity of Lactobacillus rhamnosus was observed at 31.3% when adding 10.0% (v/v), as compared to 17.7% for Lactobacillus casei. Lactobacillus rhamnosus demonstrated strong inhibition activity for melanin synthesis at 58.6% when adding 10.0% (v/v), while Lactobacillus casei increased to 80.6%. It was also observed that the high antioxidative activities of Lactobacillus rhamnosus were strongly correlated to whitening activities, due to the inhibition of both tyrosinase and melanin synthesis. These results support the expanded use of lactic acid bacteria as a functional bioresources in the cosmetics industry.

Antioxidant, Inhibitory on NO Production and In-vitro Cell Regeneration Effects of Pink-aloe (핑크-알로에의 항산화, NO 생성 억제 및 세포 재생 효과)

  • Aloe vera (Aloe barbadensis Miller) has been used since ancient times to improve various skin diseases such as burns, wounds, and eczema. It has been reported that Aloe vera contains vitamin, enzyme, mineral, sugar, phenolic compound, fatty acid and amino acid. Aloe vera changes its color from green to red under the extreme thermal and arid climate to protect itself. These morphological changes induce variation of composition such as increasing of aloe-emodin content. Aloe-emodin is one of the major anthraquinone in aloe family plants. Since aloe-emodin contains a polyphenolic structure, this compound may be responsible for the reported antioxidant and anti-inflammatory effects of aloe. However, there is no research on the process of increasing the compounds of Aloe vera. Therefore, the purpose of this study is to develop a pink aloe manufacturing process that increases the aloe-emodin content and enhances the antioxidant and anti-inflammatory activities of aloe. As a result of heating aloe under appropriate conditions, pink aloe increased aloe-emodin content compared to general aloe, and exhibited effects such as increasing antioxidant activity, inhibiting NO production, and promoting cell regeneration. Through this study, the applicability of pink aloe as a new anti-aging material in the cosmetic field was confirmed.

Protective Effect of Dietary Buchu (Allium tuberosum Rottler) on Oxidative Stress and Lipofuscin Formation in Streptozotocin-Induced Diabetic Rats (Streptozotocin-유발 당뇨쥐에서 부추식이의 산화적 스트레스 및 Lipofuscin 생성 억제 효과)

  • Diabetes mellitus has been known to be a state of increased oxidative stress. Free radical formation and lipid peroxidation are accelerated in this metabolic disorder. Buchu (Allium tuberosum Rottler) contains lots of antioxidative nutrients such as chlorophyll, vitamin C, $\beta$-carotene, phenolic compounds and sulfur compounds. To investigate the protective effects of buchu, 10% lyophilized buchu diet was fed to streptozotocin (STZ)-induced diabetic rats for 14 weeks and lipid peroxidation, protein oxidation, contents of reactive oxygen species, activities of antioxidative enzymes and contents of accumulated lipofuscin were measured as indicators of oxidative stress. Hepatic MDA and carbonyl contents tended to decrease in 10% buchu diet group compared with control group. Dietary buchu significantly suppressed lipid and protein oxidation in the skin of rats (p<0.05). Contents of hepatic hydroxyl radicals, which exert the highest toxicity among the reactive oxygen species, were significantly decreased in rats fed 10% buchu diet (P<0.05). Activities of antioxidative enzyme, such as superoxide dismutase, catalase, and glutathione peroxidase, tended to increase in liver and skin of rats fed 10% buchu diet, while hepatic catalase activity was significantly increased in buchu group compared with control group. Buchu supplementation significantly inhibited the accumulation of lipofuscin, an end-product of lipid peroxidation reactions induced by reactive oxygen radicals, in eye tissues compared with control diet (p<0.001). In conclusion, buchu supplementation diminished the oxidative stress, so dietary buchu could help to attenuate diabetes complications.

Improvement of Microwave Heating Properties of Frozen Starch by Spray of Surface Materials (Maltodextrin류의 표면도포에 의한 냉동전분의 Microwave 가열특성 개선)

  • This study was conducted to investigate the surface spraying effect of materials on the rheological properties of frozen starch with microwave heating. Microwave helps reduce cooking time by high temperature, but swiftly snatches moisture from foods such as frozen starches (buns and noodles etc) and makes surface of foods harder. Four types of maltodextrin materials have been prepared for different concentration solutions and sprayed on surface doughs of sheet type. Sprayed dough samples were Quickly frozen at $-70^{\circ}C$ and wrapped with polypropylene film. All the treated samples were kept at 0, -20 and $-50^{\circ}C$, and then taken out periodically for measurement of the quality during storage. The quality attributes evaluated after heating with microwave energy include sensory quality retrogradation, texture, surface color and microstructure. The quality of frozen starches deteriorated with long term storage even at low temperatures of -20 and $-50^{\circ}C$, and the spray materials were found to improve the textural and physical properties of frozen starches in the microwave heating. Particularly, maltodextrin with D.E value of $9\~12$ had the most desirable effects of quality improvement.

Inhibitory Effects of Rosa rugosa Crude Extract and Solvent Fractions on Adipogenic Differentiation of 3T3-L1 Preadipocytes (해당화 추출물이 3T3-L1 지방세포 분화에 미치는 영향)

  • Halophytes have been reported to possess a variety of physiological activities, such as anti-cancer, anti-oxidant, anti-diabetes, anti-inflammatory, and anti-obesity. Studies on the roots of the halophyte Rosa rugosa, in particular, have shown a variety of physiological activities and are known to be effective for nursing diabetic complications in traditional Korean medicine. In this study, the effect of R. rugosa on adipogenesis was investigated in 3T3-L1 pre-adipocytes treated with crude extract and solvent fractions (H2O, n-BuOH, 85% aq. MeOH, and n-Hex) obtained from R. rugosa roots. Treatment with extract and the solvent fractions inhibited the formation of intracellular lipid droplets in differentiated 3T3-L1 adipocytes compared to the untreated group. In particular, n-BuOH and 85% aq. MeOH fractions effectively decreased the expression of adipogenic transcription factors: peroxisome proliferator activated receptor-γ (PPARγ), CCAAT/enhancer-binding protein α (C/EBPα), and sterol regulatory element-binding protein 1c (SREBP1c) in both mRNA and protein levels. In conclusion, these results suggest that R. rugosa contains anti-adipogenic molecules that can be utilized as a nutraceutical against obesity. Further refining of n-BuOH and 85% aq. MeOH fractions and analysis of their action mechanisms could yield potential therapeutic agents with anti-adipogenic effects.
