Toxicity and Behavioral Changes of Medaka (Oryzias latipes) by Brine Exposure (송사리(Oryzias latipes)를 이용한 고염해수의 생태독성 및 단기적 행동변화에 관한 연구)

  • Acute toxicity test and behavioral change analysis of seawater acclimated Japanese medaka were conducted to identify the brine effects on fish by seawater desalination. 7 day acute toxicity test of brine revealed linear concentration-response relationship from 40.0~80.0 psu treatment groups. There was no significant brine effect for 30-40 psu groups and mass mortality was observed from >50 psu exposure (7-day $LC_{50}$=51.4 psu). Images from the real time camera system were analyzed to observe the changes in behavioral patterns of medaka exposed to various salinity. 40.0 and 50.0 psu exposed groups were stabilized in behavioral patterns after 3.1 and 4.6 hours, respectively and 60.0 psu group showed sharp increase in activity during first 12 hours and 50% mortality thereafter. Similar patterns were observed to 70 and 80 psu groups and both experimental groups showed 100% mortality within 12 hours. Acute toxicity test and behavioral patterns showed very similar toxicity results which revealed the increases in mortality and behavioral activities from 50.0 psu. This critical salinity for fish impacts must be implemented to brine discharge strategy by seawater desalination into the coastal area. Also, we recommend that real time camera monitoring system must be a useful tool for early warning of fish toxicity for other applications. Analysis of Toxicity in Endometrial Cells Exposed Phthalate (자궁내막세포에 노출된 프탈레이트의 독성연구)

  • Di-ethylhexyl phthalate (DEHP) is an environmental contaminant that is used as a plasticizer. Endometriosis is a complex disease with an unknown etiology that is believed to be associated with exposure to DEHP. The present study examined the potential toxicity of DEHP by exposing endometrial adenocarcinoma cells (Ishikawa cells) to DEHP. In the experiments, the cells were treated with stepwise DEHP concentrations (0, 0.01, 0.1, 1, and $5{\mu}M$) for different exposure times (24, 48, and 72 h). When the relationship between the resulting survival rate of the cells and initial inflammation was examined, the cell viability, expression of $TNF-{\alpha}$, an inflammatory mediator, and the expression of MMP-9, an ECM degradation protein, were increased remarkably when the cells were exposed to $5{\mu}M$ DEHP for 48 and 72 hours. These results suggest that the exposure of Ishikawa cells to certain concentrations of DEHP, estrogen mimics, may cause time-dependent toxicity that affects the cell viability and inflammation, implying a potential role in the etiology of endometriosis. Further research on the effects of endocrine disruptors on the pathogenesis of endometriosis may reveal strategies to prevent this disease.

  • 무선 센서 네트워크 환경에서의 구성요소간의 상호 인증은 중요하다. $\mu$-TESLA에서의 브로드캐스트 인증은 인증시간의 지연이 발생한다. 지연의 발생은 현재 받은 메시지의 인증이 메시지를 받은 즉시 이루어지지 않고 다음 메시지를 받았을 때 이전의 메시지에 대한 인증이 이루어지기 때문이다. 이는 실시간 처리를 필요로 하는 통신에는 적합하지 않다. 또한 대칭키를 기반으로 인증하는 방법에서는 인증시간의 지연이 발생하지 않지만 인증을 위한 키가 노출되었을 때, 키를 재생성, 분배해야하는 문제가 있다. 본 논문에서 제안하는 인증방안은 식별자와 해쉬 함수를 이용하여 인증시간의 지연이 없는 실시간 인증과 인증에 사용하는 식별자가 노출되었더라도 키의 재생성 및 분배없이 인증을 달성하는 방법을 제시한다.

Outside -15℃ Exposure Time Impact on Early Frost Damage (외기온 -15℃에 노출시간 변화가 콘크리트의 초기동해 피해에 미치는 영향)

  • In this study, when the normal concrete became a $20^{\circ}C$ image after the exposure time at an external temperature of $-15^{\circ}C$, the limit point of the early frost damage was analyzed. As a result, it was confirmed that the degree of concretion was higher than the external level after carrying in and after exposure, and that the initial Tokai damage was observed after 12 hours of exposure.

  • This study investigated the effect of 2-butoxyethanol (BE) as a nonsolvent additive, relative humidity and air contact time on the structure formation of microfiltration membranes, permeation and morphology properties in phase inversion process. The membranes were prepared by using polyethersulfone (PES)/Dimethyl formamide (DMF)/p-toluenesulfonic acid (TSA)/Polyvinylpyrrolidone (PVP)/BE casting solution and water coagulant. Casting solutions containing various concentration of BE were exposed to a water vapor, under 60 and 80% of relative humidity for 40 and 90 sec, which would be absorbed on. The correlations between the membrane permeation properties and surface/inner structures of membrane were investigated. The characterization of membranes was carried out by a capillary flow porometer, a FE-SEM and a water permeation test apparatus. The surface structure of PES membranes was affected by the exposure time as well as the relative humidity strongly. Furthermore, the addition of BE helped control surface and inner structure at certain humidity and exposure time.

Effect of Combined Environmental Factors on Adhesive Shear Strengths and Chemical Structures of Adhesives (복합적 환경인자의 영향에 의한 접착제의 접착전단강도 및 화학구조 변화)

  • Adhesive shear strengths of the established adhesives and the alternative adhesives were evaluated and their chemical structures were analyzed in order to investigate the possibility of replacing the established adhesives with the alternative adhesives applicable to the seeker for the guided missiles. Two types of the adhesives such as the structural adhesives and the sealant adhesives were considered. Those adhesives were exposed to the combined environmental factors consisting of temperature, moisture and ultraviolet over 1000 hours. Adhesive shear test was conducted to evaluate adhesive shear strengths and ATR FT-IR was utilized to investigate chemical structures. According to the results, the adhesive shear strengths of the alternative adhesives revealed higher than those of the established adhesives. Also the alternative adhesives were more stable to the combined environmental condition than the established adhesives. Therefore, it is found that the established adhesives were able to be replaced by the alternative adhesives.

Denaturation and Inactivation of Antioxidative Enzymes due to Repeated Exposure to UV-B and Inhibitory Effect of RGP Lens (UV-B 반복노출에 따른 항산화효소의 변성 및 활성저하와 RGP렌즈의 차단효과)

  • Purpose: The present study was conducted to reveal the correlation of structural denaturation and decrease of enzyme activity when the antioxidative enzymes, superoxide dismutase (SOD) and catalase (CAT) were repeatedly exposed to UV-B, and further investigate whether the denaturation and inactivation of those enzymes can be effectively blocked by using UV-inhibitory RGP lens. Methods: Each enzyme solution was prepared from the standardized SOD and CAT, and repeatedly exposed to UV-B of 312 nm for 30 minutes, 1 hour and 2 hours a day over 1, 2, 3, 4 and 5 days. Structural denaturation of SOD and CAT induced by repeat UV-B irradiation was confirmed by the electrophoretic analysis, and their enzyme activity was determined by the colorimetric assay using the proper assay kit. At that time, the change in structure and activity of the antioxidant enzymes directly exposed to UV-B was compared to the case that UV-B was blocked by UV-inhibitory RGP lens. Results: SOD exposed repeatedly to UV-B showed the polymerization pattern in the electrophoretic analysis when it repeatedly exposed for 30 min a day, however, the change of its activity was less than 10%. On the other hand, CAT repeatedly exposed to UV-B reduced size and density of the electrophoretic band which indicated a structure denaturation, and its activity was significantly decreased. In the case that the repeat exposure time was longer, CAT activity was completely lost even though some enzyme band occurred in the electrphoretic analysis. In addition, the degeneration of CAT due to UV-B irradiation was inhibited to some extent by using RGP lens with a UV-B blocking of 63.7%, however, it was not completely inhibited. Conclusions: From these results, it was revealed that the structural denaturation of antioxidative enzymes was not perfectly correlated with the reduction in enzyme activity according to the type of enzyme. It is recommended to minimize the exposure time to UV when wearing contact lens, or wear the contact lenses having UV blocking rate of the FDA Class I blocker or the sunglasses having equivalent UV-blocking rate for reducing the damage of antioxidative enzymes induced by UV.

The Relationship between Pesticide Exposure and Central Nervous System Symptoms (농약 노출과 중추신경 증상과의 관련성)

  • Objectives: The acute toxic effects of pesticide are well known. Concern has also been expressed that long-term exposure may result in damage to the central nervous system. This study was conducted to test the hypothesis that central nervous system symptoms might occur due to pesticide exposure. Methods: In a cross-sectional study, first, cumulative exposure index (CEI) was estimated. Neurologic symptoms (Q-16 questionnaire) for 541 farmers (exposed to pesticides) were compared with 119 non-exposed persons in spraying season nine rural areas in Korea. Results: The pesticides poisoning rates for last 3 months were 67.2% for orchard farmers, 55.3%for dry field farmers, and 20.5% for husbandry farmers, respectively, showing significant difference (p<0.001). Compared with non-exposure group, exposure groups (especially, orchard farmers) reported significantly more neurologic symptoms and had a higher overall neurological symptoms score (p<0.001). Factors related to the positive neurological symptoms (answers "yes" to six or more of Q-16 questionnaire) adjusted for age, sex, education level, smoking and alcohol drinking were type of farming (OR 3.08, 95% CI 1.50-6.30 in orchard farmers vs non-exposure group), CEI (OR 2.75, 95% CI 1.12-6.78 in Q3 vs Q1), past poisoning (OR 1.97, 95% CI 1.21-3.20 vs normal), current mild poisoning (OR 3.03, 9500 CI 1.47-6.22 vs normal) and current moderate poisoning (OR 6.34, 95% CI 3.03-13.25 vs normal), respectively. Conclusions: These results suggest that long-term exposure to pesticides appears to be associated with subtle changes in the central nervous system.

Effect of growth hormone on neuronal death in hippocampal slice cultures of neonatal rats exposed to oxygen-glucose deprivation (신생 흰쥐 해마 절편 배양에서 산소-포도당 박탈에 의한 신경 세포 사망에 대한 성장호르몬의 효과)

  • Purpose : To investigate whether growth hormone (GH) has a protective effect on neurons in hippocampal slice cultures of neonatal rats exposed to oxygen-glucose deprivation (OGD). Methods : Cultured hippocampal slices of 7-day-old rats were exposed to OGD for 60 min. Then, the slices were immediately treated with three doses of GH (5, 50, or $500{\mu}M$) in media. The relative fluorescent densities of propidium iodide (PI) uptake in the slices and relative lactate dehydrogenase (LDH) activities in the media were determined and compared between each GH- treated group of slices and untreated slices (control) at 12 and 24 h after OGD. Immunofluorescent staining for caspase-3 and TUNEL staining were performed to observe the effect of GH on apoptotic neuronal death. Results : The relative fluorescent densities of PI uptake in CA1 and dentate gyrus (DG) of the hippocampal slices in each GH-treated group were not significantly different from those in the untreated slices at 12 and 24 h after OGD (P>0.05). Treatment with GH could reduce the relative LDH activities in the media of the GH-treated groups only at 12 h after OGD (P<0.05). Expression of caspase-3 and TUNEL positivity in CA1 and DG of the slices treated with 50-iM GH were not different from those of the untreated slices at 12 and 24 h after OGD. Conclusion : Treatment of hippocampal slice cultures with GH after OGD does not show a definitive protective effect on neuronal death but can reduce the LDH efflux of the slices in media at 12 h after OGD.

Delayed Mortality of Benthic Amphipods Monocorophium acherusicum Exposed to Various Pollutants in Seawater(Cd, Cu, Hg, TBT, Ammonia and Phenanthrene) (유해오염물질에 급성 노출된 단각류 Monocorophium acherusicum의 지연 사망률에 관한 연구)

  • 다양한 유해오염물질에 급성 노출된 단각류 Monocorphium acherusicum의 노출 기간 이후에 발생하는 지연 사망(latent mortality)이 반수치사농도(LC5O)산출에 어떤 영향을 미치는 지를 규명하기 위한 일련의 실험이 수행되었다. 본 연구에서는 실험생물을 카드뮴, 구리, 수은과 같은 중금속, tributyltin(TBT), 암모니아 그리고 방향성탄화수소인 phenanthrene에 각각 96시간 동안 노출시킨 후 깨끗한 해수에 옮겨 다시 6일 동안 배양하면서 사망률을 조사하였다. 실험결과 구리, TBT, 암모니아, phenanthrene과 같은 물질에 노출된 M. acherusicum의 사망률은 노출이 끝난 이후에도 계속적으로 증가하는 지연 사망이 관찰되었으며, 이에 따라 기존의 방법_으로 산출된 96-h LC50보다 지연 사망을 고려한 새로운 LC50이 크게 낮아지는 경향이 관찰되었다. 지연사망률을 고려하지 않은 기존의 독성시험 결과는 지연 사망의 영향을 반영하지 못하므로 실제 현장에서 발생할 수 있는 오염물질의 영향을 과소평가 할 가능성이 있다. 따라서 지연 사망률에 대한 고려는 실제 현장 개체군에 대한 유해오염물질의 영향을 보다 정화하게 예측하는 데에 활용될 수 있을 것이다.
