Effect of Light and Scale Explant Conditions on Propagation Efficiency in Lilium callosum Scale Culture (땅나리 기내 인편 배양시 광환경과 배양 절편체의 조건이 증식 효율성에 미치는 영향)

  • Series of in vitro experiments in Lilium callosum were conducted to investigate efficient multiplication through finding the optimal cultural environment, and organogenic capability of cultural explants, and then to determine the progressive method for enhancing bulblet growth in Lilium callosum scale culture. Twenty-four hr photoperiod was most effective for the growth of bulblet and the formation of other organs. Optimum light intensity for bulblet growth was 2,500~5,000 Lux. When bulbets were subcultured, growth of bulblets were enhanced by removing excessive leaf blade. Number of bulblets per scale increased as mother scale size increased, whereas diameter of bulblet from the small size mother scale increased. Bulblet formation and development was induced when explants were placed above the medium to be exposed to more light.

The Chemical and Microbial Characteristics of Northern Sand Lance, Ammodytes personatus, Sauce Manufactured with Fermentation Accelerating Agents (발효촉진제로 속성 발효한 까나리 어간장의 화학 및 미생물적 특성)

  • Chemical and microbial properties of Northern sand lance, Ammodytes personatus, sauce fermented at $15^{\circ}C$ with fermentation accelerating agents, koji, enzyme, and squid viscera, were analyzed. Total creatine content of Northern sand lance sauce with squid viscera increased up to 2 months of ripening and decreased gradually thereafter, while that with koji or enzyme, and control, increased up to 3 months of ripening and then decreased slowly. TBA values of all samples increased sharply during early ripening, followed by slight decrease. Free amino acid content with all agents increased continuously as fermentation progressed. Major free amino acids of Northern sand lance sauce were glutamic acid, alanine, valine, leucine, and lysine. Total viable cell counts with all agents also increased during early ripening and then decreased. Total viable cell count of Northern sand lance sauce with squid viscera was the highest followed that with koji, enzyme, and control. Northern sand land sauce manufactured with koji showed the most acceptable sensory evaluation result, followed by that with squid viscera.

Effect of Low Temperature for the Breaking Dormancy of Primula sieboldii, P. modesta var. fauriae and Disporum smilacinum (저온처리 기간이 앵초, 설앵초 및 애기나리의 휴면타파에 미치는 영향)

  • This study has been conducted to find out the possibility of the winter production for pot cultured plants, such as P. sieboldii, P. modesta var. fauriae and D. smilacinum in highland, 800 meter above sea level by early breaking dormancy. Low temperatures for breaking dormancy were treated for 300, 500, 700 and 900 hours under $5^{\circ}C$ in incubators and, then these plants were moved to greenhouse controlled at $10^{\circ}C$ of night temperature. Results showed that P. modesta var. fauriae and D. smilacinum, and P. smilacinum required more than 500 hours and 700 hours of low temperature $5^{\circ}C$ for breaking dormancy, respectively.

The Flora of Vascular Plants around Tuman River in China (두만강 접경지역 일대의 관속식물상)

  • This study was conducted by China along with the Tumen River, which cuts between of North Korea and China. As a result 92 family, 287 genera, 470 species, 57 variety 4 formas, total 531 taxa of vascular plants were identified. The surveyed area was include the region that had various vegetation such as upper swampy land, seashore dune, a river, a high mountain grassy plain, deciduous tree zone and evergreen coniferous forest zone. The specfic plants which is protected by The Natural Environmental Preseration Act appeared to 6 taxa, and rare and endangered plants conserved by Forestry Administration were found 21 taxa in these areas. Among them Lilium cernum, Jeffersonia dubia, Acanthopanax senticosus could be regarded as very important one for chinese medicine. Picea koraiensis, Larix gmelini var. principis-ruprechtii, Populus koreana, Betula fusenensis, Ulmus pumila, Ledum palustre var. diversipilosum, Rhododendron parvifolium, Vaccinium uliginosum, Astragalus membranaceus, Aquilegia flabellata var. pumila, Carex gotoi, Carex capricornis etc. are not distributed in the South Korea.

Breeding of Lilium × formolonga F1 hybrid 'Eorayeon 1ho' for Cut Flower (절화용 신나팔나리 F1품종 '어라연 1호' 육성)

  • The white flowered 'Eorayeon 1ho' is the first Kangwon National University cultivar of L. ${\times}$ formolonga, commonly known as seed propagating lily. Crosses were conducted from 1999 to 2003 among the selected selfing lines($S_3$) of L. longiflorum 'Lorina', L. ${\times}$ formolonga 'Augusta' and 'Raizan Herald'. A new cultivar was preliminarily selected as 'E-No.1' in 2003 for its stable upward flowering habit. The evaluation of characteristics was investigated from 2003 to 2004 in Chuncheon and Yungweol, Kangwon province. 'Eorayeon 1ho' blooms in early July at outside, with high resistance to Botrytis and having upward flowering habit. The plant height is 112.4 cm and the percentage of seed germination is 96.4%. This seed propagating cultivar shows good quality as a cut flower and low blasting rate.

Dormancy Breaking by Hot-water Treatment of Lilium Oriental Hybrids Bulblets Regenerated in Vitro (온탕처리에 의한 오리엔탈나리 소인경의 휴면 타파)

  • The objective of this study was to investigate the effect of hot water and dipping times, and hot- PGR's solution on dormancy breaking of Lilium Oriental Hybrid 'Casa Blanca' and 'Siberia' bulblets regenerated in vitro. Hot-water treatments with distilled water were effective not sprouting rate but shortening of days to sprouting. While $100mg{\cdot}L^{-1}\;GA_{4+7}$ was effective in the dormancy breaking process of bulblets. The content of mitochondrial protein and fumarase activity were highest in bulblet of $100mg{\cdot}L^{-1}\;GA_{4+7}$ addition in hot water treatment. Therefore $GA_{4+7}$ addition in hot-water treatment seem to be the alternative dormancy breaking method to low-temperature treatment in Lilium Oriental Hybrid bulblet regenerated in vitro.

Flow Path of Choosan Spring in Nari Basin, Ulleung Island, South Korea (울릉도 나리분지 추산용천수의 유동 경로)

  • This study clarified the flow path of Choosan Spring, Nari Basin, Ulleung Island, South Korea. The orientations of faults and fractures developed on the inner edge of the caldera were identified as major factors affecting the flow path. The main flow paths include fracture zones oriented N-S and E-W. The spring also flows in a NE or NNE direction under the influence of the irregular shape of the caldera, which slopes to the NNE. Using Entrobacteriaceae species as tracers, it was found that Nari groundwater flows toward Choosan Yongchulso. However, the small number of water samples used in the analysis limits our understanding of the flow path from Sungin Valley to Nari Basin and Choosan Yongchulso.

Taste Compounds and Antioxidant Properties in Extracts of Angelica keiskei and Oenanthe javanica Juice By-Products According to Extraction Methods (추출 방법에 따른 명일엽과 돌미나리 착즙박의 정미성분 및 항산화 특성)

  • This study aimed to examine the possibility of upcycling extracts of Angelica keiskei and Oenanthe javanica juice by-products through comparing enzyme extraction (EE) and complex extraction (CE) methods to increase the extraction yield and flavor of materials. A higher extraction yield was obtained for free amino acid content with EE and CE for A. keiskei and O. javanica juice by-products, respectively, and a higher extraction efficiency was achieved with juice by-products than with extracts prepared from raw materials before juice production. The content of major amino acids varied depending on the extraction method used. When used according to the characteristics of the extract, their use as a functional material was confirmed along with improvement in the flavor of the food. Consistently high extraction yields for organic acid and sugar levels were obtained with CE in A. keiskei and O. javanica juice by-products. The DPPH radical scavenging ability and TPC were consistently high with CE in A. keiskei and O. javanica juice by-products; the increase in extracted content was likely because of the reaction between the ethanol used for CE and the phenolic compounds. However, because the antioxidant capacity of the juice by-product extracts was somewhat lower than that of the extracts from raw materials before juice production, the amount used should be reviewed. The TFC was found to be higher in extracts obtained with EE than with CE for A. keiskei juice by-products; however, no significant difference was observed between EE and CE in the O. javanica juice by-products. Through this study, the taste compounds and antioxidant properties of extracts obtained from juice by-products produced after the production of A. keiskei and O. javanica green juice were analyzed, and the availability of high value-added materials was confirmed. Based on these research results, expanding specific R&D for practical use should be explored.

희나리 태양초 고추로부터 분리한 곰팡이의 Mycotoxin 안전성

  • 2002년 8월부터 11월까지 전국 27개 지역에서 희나리 고추를 포함하고 있는 태양초 시료 40군을 수집하여 총 197주의 곰팡이를 분리하였다. 이 곰팡이들을 고체배지상에서의 번식속도, 분생포자의 모양 및 배양특성의 특성에 따라 6개의 group오로 나누었고 각 group에서 대표가 되는 30 균주를 임의로 선택하여 18S rRNA gene 염기서열을 분석하여 동정하였다. 희나리 곰팡이 중 에서 Colletotrichum 속 곰팡이가 66.5% (131/197)를 차지하여 가장 많이 분리되었고, 기타 Diaporthe phaseolorum var. sojae (28주, 14.2%), Alternaria alternata (17주, 8.6%), Botryosphaeria ribis (9주, 4.6%) Aspergillus oryzae var. oryzae (3주 1.5%) 및 Fusarium incarnatum (9주, 4.6%)이 동정되었다. 각 group에서 임의적으로 한 균주씩을 선택하여 현미에 배양한 뒤 쥐(rat)에 투여시험한 결과 A. alternata를 접종한 사료를 먹인 실험동물이 2주내에 모두 죽었으며 다른 곰팡이를 배양한 사료에는 특이한 영향이 없었다. A. alternata 곰팡이를 현미와 고추즙에 배양하여 주요 독소들을 분석한 결과 17주의 곰팡이 중 8주가 현미와 고추즙에서 많은 양 (현미: $488{\sim}1572\;{\mu}g/g$, 고추즙: $115{\sim}1050\;{\mu}g/g$)의 tenuazonic acid (TeA)를 생성하였다. alternariol(AOH)독소와 alternariol monomethyl ether (AME)는 현미에 배양했을 때만 흔적량 내지 소량씩이 관찰되었다. Alternaria 독소 중 altenuene는 현미와 고추즙 배지 모두에서 검출되지 않았다.

Effects of Diniconazole Application on Anatomical and Biochemical Characteristics Related to Stress Tolerance in Lilum davuricum (날개하늘나리에 있어서 디니코나졸 처리가 스트레스 내성과 관련된 해부학적 및 생화학적 특성에 미치는 영향)

  • This study was carried out to examine the effects of foliar-sprayed diniconazole on the morphological characteristics and the contents of polyamines (PA) and proline, and to investigate their relationship with stress tolerance in Lilium davuricum native to Korea. Plants with 5 cm mean height were sprayed with $50mg{\cdot}L^{-1}$ diniconazole or distilled water (control) and grown in a greenhouse maintained at 23/$18^{\circ}C$ (day/night) for 30 days. Diniconazole decreased plant height, leaf length, diameter and length of the pith and cortical cell of the stem, while it increased the thickness and epicuticular wax of leaves measured at 30 days after treatment. In polyamines contents, diniconazole increased spermidine content at 5-10 days after treatment and spermine content after the 10th day of treatment as compared with the control. Diniconazole decreased proline content after the 10th day of treatment as compared with the control. From these results, it was concluded that foliar-sprayed diniconazole might morphologically and biochemically improve the stress tolerance of this plant species.
