Process Development of Algae Culture for Livestock Wastewater Treatment Using Fiber-Optic Photobioreactor (축산폐수 처리를 위한 광섬유 생물반응기를 이용한 조류 배양 공정 개발)

  • In this study, algae cultivation using the photobioreactor has been applied to remove the nitrogen and phosphorus compounds in the wastewater of the livestock industry. The optimal ratio of nitrate and ortho-phosphate concentration was found for the enhancement of removal efficiency. To achieve the high density culture of algae, the photobioreactor consisted of optical fibers wes developed to get the sufficient light intensity. The light could be illuminated uniformly from light source to the entire reactor by the optical fibers. The structured kinetic model was proposed to describe the growth rate, consumption rate of nitrates and ortho-phosphates in algae culture. The self-organizing fuzzy logic controller incorporated with genetic algorithm was constructed to control the semi-continuous wastewater treatment system. The proposed fuzzy logic controller was applied to maintain the nitrated concentration at the given set-point with the control of wastewater feeding rate. The experimental results showed that the self-organizing fuzzy logic controller could keep the nitrate concentration and enhance algae growth.

Rapid and Simple Method for Isolating Plasmid DNA from Lactic acid Bacteria (유산균 Plasmid DNA의 신속 간편한 분리방법)

  • A simple procedure for rapid isolation of plasmid DNA from lactobacillus species and streptococcus species is described. Lactic acid bacteria were cultured in the TCM broth containing 0.5% glycine and plasmid DNA was isolated from cells treated with mutanolysin by alkaline-detergent lysis method. Good results for releasing and isolating plasmid DNA from lactobacillus species were obtained by treatment of cells with 30$\mu\textrm{g}$ of mutanolysin per ml at 37$^{\circ}C$ for 5 to 10 min. For the streptococcus species, the optimum conditions were slightly different. The procedure could be used for rapid characterization of plasmid DNA in Lactobacillus casei, Lactobacillus acidophilus, Lactobacillus helveticus, Streptococcus lactis, Streptococcus faecalis, Streptococcus faecium, and Streptococcus cremoris strains. Using this procedure, plasmids isolated from $1.5m\ell$ cultures could readily be visualized in agarose gel.

  • The DGPS reference station is a national infrastructure generating GPS correctional information and transmitting the signal for Differential GPS. Currently, Korea has applied and operated the software-based DGPS reference station as a standard of the next generation proposed by the USCG in order to improve the hardware-based DGPS reference system. However, software-based DGPS reference station proposed by USCG was changed in software method, only for form. There is no advantage to changing software-based because the most critical part of architecture has not been improved. In this paper, we have designed a new software-based marine DGPS station architecture that a reference station software and a monitor station were integrated. The new marine DGPS station architecture based on software is a more simplified structure than it used to be and can be utilized in the DGPS reference station.

  • Objectives: This clinical study reports on the effects of Yukgunja-tang on aspiration pneumonia.Method: This case examined a comatose male patient who suffers from continuous regurgitation. He showed symptoms of aspiration pneumonia with accompanying gastroesophageal reflux. We gave the patient Yukgunja-tang for 28 days and observed any changes in symptoms.Results: During the period when the patient was given Yukgunja-tang, he defecated on his own without glycerin enema, regurgitation occurred less frequently, and the aspiration pneumonia symptoms improved.Conclusion: Yukgunja-tang helped the patient's bowel movements move more quickly and smoothly through his colon. As a result, Yukgunja-tang appears to have prevented regurgitation and aspiration pneumonia.

  • This paper describes the procedure to analyze the performance of the end-plated propeller(EPP) by a boundary integral method. The screw blade(SB) and end-plate(EP) are represented by a set of quadrilateral panels, where the source and normal dipole of uniform strength are distributed. The perturbation velocity potential, being the only unknown via the potential-based formulation, is determined by satisfying the flow tangency condition on the blade and the end-plate at the same time. The Kutta condition is satisfied through an iterative process by requiring the null pressure jump across the upper and lower sides of the trailing edges of both the SH and the EP. Sample calculations indicate that the EP increases the loading near the tip of the SB while spreading the trailing vortices along the trailing edge of the EP, thus avoiding the strong tip-vortex formation. Predicted performance of the EPP shows good correlations with the experimental results. The method is therefore considered applicable in designing and analyzing the EPP which may be an alternative for energy-saving propulsive devices.

  • Aconitine, found in the Aconitum species, is highly extremely toxic, and has been known to cause fatal cardiac arrhythmias and cardiovascular collapse. Although several reports have described treatment of aconitine intoxication, management strategy for the patient in a hemodynamically compromised state who experienced cardiopulmonary collapse is unknown. We report here on a case of a successful cardiopulmonary resuscitation and therapeutic hypothermia in an aconitine-induced cardiovascular collapsed patient. A 73-year-old male who presented with nausea, vomiting, chest discomfort, and drowsy mental state after eating an herbal decoction made from aconite roots was admitted to the emergency department. He showed hemodynamic compromise with monomorphic ventricular tachycardia resistant to amiodarone and lidocaine. After 3 minutes on admission, he collapsed, and cardiopulmonary resuscitation was initiated. We treated him with repeated cardioversion/defibrillation of 51 times, 10,150 joules and cardiopulmonary resuscitation of 12 times, 69 minutes for 14 hours and therapeutic hypothermia for 36 hours. He recovered fully in 7 days.

Dose-dependent suppression of tolaasin-induced hemolysis by gadolinium ion (가돌리니움 이온에 의한 톨라신 용혈활성의 농도의존적 억제)

  • Brown blotch disease of oyster mushrooms is caused by tolaasin and its analog peptide toxins which are produced by Pseudomonas tolaasii. Tolaasin peptides form pores in the plasma membrane and destroy the fruiting body structure of mushroom. Lysis of red blood cells, hemolysis, can be occurred by cytotoxic activity of tolaasin. The hemolytic activity of tolaasin is inhibited by metal ions, such as Zn2+ and Ni2+. When Gadolinium ion was added, a biphasic effect was observed on tolaasin-induced hemolysis, an increase in hemolysis at submillimolar concentrations and an inhibition at millimolar concentrations. The mechanism of gadolinium ion-induced inhibition of tolaasin activity may not be similar to those of the inhibitions by other metal ions. Since gadolinium ion has been reported to change a lateral pressure of lipid membrane by binding to the negative charges of membrane lipids, it may not directly work on the tolaasin channel gating, but rather decrease the stability of tolaasin channel by increasing firmness of membrane.

  • Innovation is essential for the survival of a company in the fierce competitive environment of the global era of borders along with rapid technological development and changes from the environment. It is very important to secure your own competitive advantage. In this study, the relationship between internal R&D capability and organizational support as a company internal factor and external R&D capability, external knowledge search, and absorptive capability as a company external factor on performance was examined. As a result of the analysis, it was found that internal R&D capability, organizational support, and external knowledge search had a significant effect on absorptive capability, and innovation performance had a positive effect on new product performance. However, the mediating effect of absorptive capacity has not been verified, suggesting that absorptive capacity has a low correlation between innovation performance and new product performance, or that other important factors act between absorptive capacity and performance variables. The results of this study are expected to be utilized as useful guidelines when establishing R&D strategies in companies.

  • This study analyzes the effects of the absorptive capacity on the technology innovation and commercialization capacities and the management performance. Based on previous studies, the absorptive capacity is sub-classified into the exploratory absorptive capacity and the exploitative absorptive capacity, which are set as independent variables, to analyze their effects on the technology innovation and commercialization capacities. In short, the absorptive capacity has positive effects on the technology innovation and commercialization capacities. In addition, the technology innovation capacity and the technology commercialization capacity have positive effects on the management performance. The findings confirm that the absorptive capacity is an important factor impacting on the corporate innovation, and suggest that businesses should recognize the importance of the absorptive capacity as a guideline for developing their strategies for technological innovation.

Enzyme-linked Immunosorbent Assay Strip Sensor for Rapid Detection of Staphylococcus aureus (Staphylococcus aureus 신속 검출을 위한 효소면역측정 스트립 센서)

  • In this study, an established enzyme-linked immunosorbent assay and immuno-chromatography technique are combined to fabricate an immuno-strip sensor for the detection of S. aureus. The immuno-strip is manufactured by using four different functional membranes. The capture antibody is immobilized on the nitrocellulose membrane due to the high affinity and the capillary action through porous membranes induces a flow of sample. A colorimetric signal is appeared according to the enzyme reaction and is analyzed by the digital camera (qualitative analysis) and home-made image analysis software (quantitative analysis). Under the optimal conditions, samples with S. aureus in the range of $2.7{\times}10^4{\sim}2.7{\times}10^7CFU/mL$ can be detected by the colorimetric method within 30 min.
