Pronucleus Formation and Embryonic Development of the Human Oocytes Fertilized by Intracytoplasmic Sperm Injection (세포질내 정자주입을 시행한 인간 난자의 전핵 형성 및 배아 발생에 관한 연구)

  • Mammalian, including human, spermatozoa undergo morphological and physiological changes during sperm maturation. There were, these changes may affect the fertilization and embryonic development. In this study, we examined the pronucleus formation, pronucleus disappearance and embryonic development in the human oocytes fertilized by intracytoplasmic sperm injection (ICSI). The injected spermatozoa were grouped into ejaculated, epididymal and testicular by the collecting region. Among 363 metaphase II injected oocytes, 287(79.1%) oocytes were normally fertilized and displayed two pronuclei. There were no difference in the fertilization rates and in the pronucleus formation and pronucleus disappearance at 16, 20 and 24 hr after ICSI, among the each spermatozoa group. Also, at 64 hr, the appearance of embryonic development was similar. From these results it can be concluded that there was no difference of maturity among the sperm collected from ejaculated, epididymis and testis in the pronucleus formation and embryonic development. Therefore, testicular spermatozoa are successfully used with ICSI in IVF-ET program.

The Effect of Progesterone on Differentiation of Implantation Sites during Early embrynic development of the Rat (흰쥐 초기배아 발생과정 중 착상부위 분화에 미치는 Progesterone의 영향)

  • 포유류 배아의 착상기작을 규명하고자 본 연구에서는 착상시기에 수정된 배아가 착상부위를 어떻게 인식하는 지 그리고 착상부위의 분화유발요인이 무엇인지를 조사하였다. 착상시기(임신 제6일)와 착상후 초기배아 발생기간(임신 제9일)중 혈청내 estradiol (E2)과 Progesterone(P4)의 농도를 측정하였으며, 자궁내막 조직을 착상부위 (antimesome trium)와 비 착상부위(mesometrium)로 분리하여 P4의 수용체 농도를 측정하였다. 혈청내 E2의 농도는 임신 제6일군에서 E2처리군에서 가장 높았으며, 임신 제9일에서는 intact 대조군에서 가장 높고 제9일의 모든 실험군의 농도는 임신 제6일군보다 높았다. 혈청내 P4 농도는 임신 제6일과 9일에서 다같이 대조군에서 가장 높으며 처리군 중에서는 P4처리군에서 높아지고 있다. P4 농도 역시 임신 제9일의 모든 실험군에서 제6일의 실험군보다 높았다. P4수용체 농도는 착상부위가 비 착상부위보다 높으며, 대조군(P < 0.01)과 P처리 군(P < 0.05)에서는 유의한 차이로 비 착상부위보다 높았다. 자궁내막조직 착상을 위한 분화에는 P4의 영향이 크며 P4의 혈청내 농도와 핵의 수용체 농도는 모든 실험군에서 상응하는 관계를 나타내었다. 이 결과는 앞서 일차적으로 발표한 알카리성 phosphatase (ALPase) 활성과도 상응하는 것이다.

Change of hCG Binding Capacity on the Granulosa Cell of Porcine Ovary During Follicular Atresia (돼지 난소내 여포의 폐쇄과정중 과립세포의 결합능의 변화)

  • In order to study the mechanism of follicular atresia, follicles were classified into the normal groups and the atretic ones, according to the criteria with or without corpus luteum, size of follicles, vascularization, status of granulose cells and the hypertrophy of theca layers in the porcine ovary. To estimate the binding capacity of human chorionic gonadotropin (hCG) receptor on the granulosa cells during atresia, hCG were iodinated by the chloramine-T method and then purified through the column chromatography. The concentration" of hCG receptor in each group were measured by the hCG receptor binding assay. Binding capacity in large normal follicles were 1.16%, but atretic ones were 0.45%. But in medium and small follicles (below 6mm in diameter), the binding capacity in normal follicles were 0.09%, but atretic ones were 0.05%, which was lower than those of large follicles. The present ( ) that the concentrations of hCG receptors on granulosa cells is decreased when the follicles become atretic and be used as a sort of creteria for the identification of follicles atresia.

Study on Pregnancy and Viability of Frozen-Thawed Human Embryos by Cryopreservation : DMSO as Cryoprotectant (동결보존에 의한 인간배아의 생존률과 임신에 관한 연구)

  • This study was done to verify factors affecting viability after cryopreservation and pregnancy rate after frozen-thawed embryo transfer into uterus. Embryos were cryopreserved slow freezing and slow thawing and used DMSO as cryoprotectant. The results were to follows. 1. Viability of frozen-thawed embryos were 75.5% (94/105), which compared with viability of embryos according to cell stage, $2{\sim}5$ cell was 68.4% and $6{\sim}16$ cell 80.4% were significant differences (p<0.05). 2. No significant difference in duration of cryopreservation on effects affecting pregnancy rate was observed. 3. Number of embryo transfered into uterus was significant differences (p<0.05). 4. Four pregnancies resulted following replacement of 35 frozen-thawed.

A Study on the Pattern of Proteins on Electrophoresis in the Porcine Follicular Fluid During Atresia (폐쇄에 따른 돼지 여포액내 단백질의 전기영동 양상에 관한 연구)

  • This experiment has been done to evaluate the relationship between the follicular atresia and the protein patterns on electrophoreais of the follicular fluids in porcine ovary. The protein concentration of the follicular fluids was lower than that of serum, and gradually decreased as the follicle siae became larger. The number of protein bands of follicular fluid on electrophoresis was less than that of serum, and gradually increased as follicle size became larger. Three specific bands were detected on disc PAGE and one band(M W. 75,000) on SDS PAGE in the follicular fluids, while not in serum. One band (A) at ${\beta}$-globulin region on disc PAGE became heavier, as follicles became atretic. Two bands less than(M. W. 20,000) were detected only in the large follicular fluid. Another band(M. W. 43,000) was not detected in necrotic group, whereas all other groups showed it. It could be concluded that the component and composition of the proteins follicular fluids changes according to the follicular size during atresia. Therefore detection of the changing pattern of proteins in the follicular fluid can be used as a basic criterion for the identification of follicular atretic stage.

A Study on the Fine Structural Changes of Porcine Ovarian Follicles during Atresia (돼지 난소내 여포의 폐쇄에 따른 미세구조의 변화에 관한 연구)

  • This experiment has been done in order to study the correlation between the ultrastructure changes and the atresia phenomenon of the follicles in porcine ovary. The ovaries were assorted according to the presence or absence of corpus luteum. Thereafter, the follicles were classified into normal, pyknotic, necrotic and cystic groups by atretic characteristics on the histological observation, and then their ultrastructures were examined with an electron microscope. The results were as followings. 1. In normal group, granulosa cells represented the ultrastructural characteristics of protein-synthesizing cells. Since the initiation of atresia, the line structure of granulosa cells showed many of the characteristic features of steroid-secreting cells, followed by gradual pyknosis. 2. In necrotic group of the ovary without corpus luteum, the theca interns became hypertrophic and displayed the ultrastructural features of active steroidsecreting cells. But this phenomenon was not seen in the follicles of the ovary with corpus luteum. 3. Degenerative changes of cumulus cells were similar to those of granulosa cells, and the degenerating oocytes showed the degeneration of cellular organelles, cytoplasmic vacuolization and disappearance of microvilli on the surface. The degeneration of granulosa cells tended to procede that of oocytecumulus complex in the follicles of the ovary having no corpus luteum, but this tendency was reversed in the case of presence of corpus luteum.   • It is the purpose to investigate the physiological toxity and mutagenicity by heavy metal compounds such as potasium dichromate, sodium arsenite and selenium dioxide on the development of Drosophila melanogaster. These chemical compounds were highly toxic at every developmental stage of Drosophila. The relative values of fecundity, viability and developmental time obtained on food media containing 5 different levels of potasium dichromate (500, 600, 700, 800, and 900 ppm), also 5 different levels of sodium arsenite and selenium dioxide (10, 15, 20, 25 and 30 ppm). Resulting in the relative values of fecundity and viability generally decreased, on the contrary developmental time generally reduced as the concentration of chemicals in the food increased. A significant reduction in the fecundity was observed between 800 and 900 ppm (p<0. 01) on the potasium dichromate media, between 15 and 20 ppm (p<0. 05) on the sodium arsenite media, between 20 and 25 ppm (p<0. 02) on the selenium dioxide media. A significant reduction in the viability was observed between 700 and 800 ppm (p<0. 01) on the potasium dioxide media, between 25 and 30 ppm (p<0. 01) on the sodium arsenite media, between 20 and 25 ppm (p<0. 01) on the selenium dioxide media. The developmental time was significantly reduced between 800 and 900 ppm (p<0. 001) on the potasium dioxide media, between 15 and 20 ppm (p<0. 001) on the sodium arsenite media, and between 25 and 30 ppm (p<0. 01) on the selenium dioxide media.

  • This paper presents a LNG Liquefaction cycle configuration using two stages of methane expansion (i.e. spliting into two stages as warm & cold to generate an additional inflection point within a cold composite curve) and a single stage of nitrogen expansion to improve the efficiency of the conventional Methane & Nitrogen Expansion Cycle. In comparison with Double Nitrogen Expansion Cycel and Methane & Nitrogen Expansion Cycle, the cycle efficiency has increased approximately from 13.92 and 13.13 to 12.08 kW/ton/day (8~15% efficiency increase). A Life Cycle Cost (LCC) analysis based on Net Present Value (NPV) also show an improvement in therms of project NPV, against a minor increment of a CAPEX.

  • To analyze the influence of variables of roadheaders, the linear cutting testing data of pick cutter were collected from the former literatures. The input factors were set up as uniaxial compressive strength, cutting depth, cutting spacing, attack angle, skew angle, and output factors were determined as specific energy, average cutting force, maximum cutting force, average vertical force, and maximum vertical force. After composing a table of the design of experiment (DOE). The contribution level of each factor was calculated by analysis of variance (ANOVA). As a result, the factors having greatest influence on cutting force and specific energy were uniaxial compressive strength and cutting spacing.

  • Roadheaders have been operated in the construction of Korean tunneling projects. The note introduces operation manual and management case of transverse type roadhead in a Korean tunneling site. The cutting and reaction forces of axial and transverse type cutting heads were qualitatively analyzed. The shaping surfaces of tunnel faces were visualized in both cases of fixed and auto-controlled telescopic cylinder conditions. Excavating with fixed cylinder, concave surfaces were shaped on tunnel face. The total processes of sumping and shearing were illustrated for excavating hard rock tunneling. The supplementary graphical explanations for total tunneling procedures in Korea were provided.
