Comparisons of Decomposition Rates, Carbon and Nitrogen Dynamics of Branches in Pinus densiflora and Quercus variabilis Stands (소나무와 굴참나무 임분의 가지 분해율과 탄소 및 질소 동태 비교)

  • This study was carried out to understand how factors of decomposition such as quality of the substrate (tree species), forest types, and soil·meteorological conditions affect decomposition rates and dynamics of carbon (C) and nitrogen (N) of branches in adjacent Pinus densiflora and Quercus variabilis stands in Sancheong and J inju in Southern Korea. The branch bags (30 cm × 30 cm) with branch samples (a diameter of 2.0 - 4.0 cm and length of 10 cm) were placed on the forest floor at each stand in May 2018. The branch bags were collected in August and November 2018, February and May 2019, respectively. The decomposition rates of branches in P. densiflora stands were 4.49 % for Sancheong, and 5.75 % for Jinju. Whereas, the decomposition rates in Q. variabilis were 20.01 % for Sancheong, and 24.68 % for Jinju, respectively. The decomposition of branches was more rapid in Q. variabilis compared with P. densiflora in both regions. C and N in decomposed branches were more mineralized in Q. variabilis, whereas C and N were more accumulated in P. densiflora. These results indicated that the decomposition rates, C and N of decomposed branches may be affected by differences in substrate quality such as initial N concentration and C/N ratio rather than differences between both regions including different environmental factors.

  • 세계적인 자원고갈과 지구온난화와 같은 환경문제가 발생됨에 따라 대체에너지 개발에 대한 연구가 활발히 이루어지고 있다. 섬유소 기질을 이용한 바이오에탄올 생산은 세계적으로 막대한 자원이 있으며 광합성에 의해 재생산되는 무한한 재원으로서 환경적으로도 대기오염물질을 적게 배출하여 유용한 에너지원으로 각광받고 있다. 하지만 섬유소 기질은 cellulose, hemicellulose, lignin과 같은 고분자 화합물이 유기적으로 결합된 단단한 결정구조로 이루어져 있어 이를 분해하여 원하는 물질을 얻기 위해서는 전처리 과정이 필요하다. 전처리 공정은 바이오에탄올을 생산하는 당화 및 발효공정의 효율 및 반응시간 단축에 기여하며, 특히 섬유소 기질일 경우에는 필수불가결한 공정이다. 전처리 공정은 물리적, 화학적, 생물학적 방법으로 나누어지며, 이러한 방법들 중 기질의 특성과 처리효율에 따라 기술들을 병합하여 사용하기도 한다. 이에 본 연구에서는 산 처리, 암모니아 처리, 과산화수소 처리 및 효소를 이용한 생물학적 처리를 단독 또는 병행하여, 전환된 당 성분 및 함량을 조사함으로서 섬유소계 기질인 채소 음식물류 쓰레기를 대상으로 바이오에탄올을 경제적으로 생산하기 위한 적합한 전처리법을 검토하였다. 전처리 방법별 당화율을 살펴보면, 산 처리와 암모니아-과산화수소-계면활성제 처리가 각각 65.3 % 및 65.7 %로 가장 높았으며, 과산화수소 처리는 16.2 %로 가장 낮았다. 반면 전처리 공정 없이 효소를 이용한 당화만을 실시한 경우에는 4.3 %의 낮은 당화율을 나타내었다. 섬유소계 기질의 전처리 효율을 향상시키기 위해 첨가하는 계면활성제의 효과는 암모니아-과산화수소 및 암모니아-과산화수소-계면활성제 처리의 당화율을 비교한 결과, 뚜렷한 효과를 확인할 수 없었다. 전처리 방법별 당의 성분 및 함량을 비교한 결과 육탄당은 암모니아-과산화수소-계면활성제 전처리에서 가장 많이 검출되었다. 오탄당은 산 처리 후 그 함량이 현저히 높았으며, 오탄당 중 xylose의 함량이 60.49 mg/g로 가장 많이 차지하고 있었다. 이 결과로부터 전처리 방법에 관계없이 당화율은 유사한 수준을 보이지만, 당화된 당의 성분 및 함량에는 큰 차이가 있음을 알 수 있었다. 이당류의 경우 과산화수소 및 암모니아-과산화수소 처리를 제외한 나머지 전처리 방법에서 유사한 수준을 나타내었다. 암모니아 처리 및 과산화수소 처리를 순차적으로 병행한 암모니아-과산화수소 처리에서는 각각의 처리시보다 육탄당의 함량은 증가하였으나 암모니아 처리시보다 이당류의 함량은 감소한 것으로 나타났다.

  • Enzymatic hydrolysis of insoluble cellulose was performed in a bubble column with tangential flow ulrafiltration membrane unit. The reactor was operated in a batch mode as well as semi-continuous and continuous with continuous removal of products through the tangential flow ultrafiltration membrane. The optimum superficial gas velocity was 1-3cm / sec so as to avoid bubble coalescence and enzyme denaturation. In continuous and selni-cotinuous process, the conversion was gradually increased but the total reduced sugar concentration was drcastically dereased with the dilution rate. It was concluded that the bubble column attaching tangential flow ultrafiltration membrane unit was effective on continuous hydrolysis of cellulose and recovery of enzyme.

  • In order to prepare digested Protein source for the Weanling Food from soybean, an attempt was made to decompose steamed soybean protein to amino acids and peptides by protease and cellulase produced from Aspergillus niger and Aspergillus sojae. In this paper, the optimum condition for digestion of soybean protein were studied and also investigated the effects of decolorization of it. As the results, followings were obtained; 1. As steaming conditions, a treatment under 15 lb of pressure and 10 minutes of heating shows most effective. 2. The optimum pH of Asp, sojae enzyme for the digestion of soybean protein is 6.0, while that of Asp. niger enzyme is 4.4. In successievly-decomposing with Asp. sojae and Asp. niger, it shows the most effective on ratio of water-soluble-nitrogen to total nitrogen and amino-nitrogen to total nitrogen than any other separate treatments. 3. The suitable amount of the enzyme solution to that of the soybean substrate paste, in volume, is 1 : 2. 4. Digestion ratio of soybean protein indicates the gradual and steady effects of increasing time of digestion, but 8 hour-digestion regarding to putrefaction was suitable. 5. The most effective decolorization was successively passed on culumns of active carbon and anion exchanger (Dowex 2-x-8) at room temperature. In separate treatments, the effective order of decolorization was as follows; (Dowex 2-x-8)>Active carborn>Amberite IR-120 6. The powder type of the soy protein source obtained by concentration below $60^{\circ}C$ contains 12.51% of moisture, 66.31% of protein, 4.25% of fat, 12.75% of carbohydrate, 4.18% of ash.

  • These experiments were conducted to investigate the hydrolysis products on the various oligosaccharides of Bacillus cirulans F-2 $\alpha$-amylase, and the hydrolysis rate on the various raw starches of Bacillus circulans F-2 $\alpha$-amylase, Bacillus amylotiquefaciens $\alpha$-amylase and Rhizopus niveus glucoamylase. The results obtained were as follows : 1. Maltotetraose, maltopentaose, maltohexaose, maltoheptaose and maltooctaose were hydrolyzed, but maltose and maltotriose were not hydrolyzed by Bacillus circulans F-2 $\alpha$-amylase. Among maltotetraose, maltopentaose, maltohexaose, maltoheptaose and maltooctaose, especially maltotetraose was hydrolyzed weakly by Bacillus circulans F-2 $\alpha$-amylase. 2. The Hydrolysis rate of oyster glycogen was slightly lower than soluble starch, amylose and amylopectin. 3. The hydrolysis rate of com starch was higher in shaking incubation than in stationary incubation, but the hydrolysis rate of potato starch was not definite according to kinds of enzyme. 4. On com, rice, arrowroot, high amylose corn, banana, sago, yam and potato starch, Bacillus circulans F-2 $\alpha$-amylase exhibited a remarkably higher hydrolysis rate than Bacillus amyloquefaciems $\alpha$-amylase and Rhizopus niveus glucoamylase.

Effects of Non-ionic Surfactant Tween 80 on the in vitro Gas Production, Dry Matter Digestibility, Enzyme Activity and Microbial Growth Rate by Rumen Mixed Microorganisms (비이온성 계면활성제 Tween 80의 첨가가 반추위 혼합 미생물에 의한 in vitro 가스발생량, 건물소화율, 효소활력 및 미생물 성장율에 미치는 영향)

  • The non-ionic surfactant (NIS) Tween 80 was evaluated for its ability to influence invitro cumulative gas production, dry matter digestibility, cellulolytic enzyme activities, anaerobic microbial growth rates, and adhesion to substrates by mixed rumen microorganisms on rice straw, alfalfa hay, cellulose filter paper and tall fescue hay. The addition of NIS Tween 80 at a level of 0.05% increased significantly (P<0.05) in vitro DM digestibility, cumulative gas production, microbial growth rate and cellulolytic enzyme activity from all of substrates used in this study. In vitro cumulative gas production from the NIS-treated substrates; rice straw, alfalfa hay, filter paper and tall fescue hay was significantly (P<0.05) improved by 274.8, 235.2, 231.1 and 719.5% compared with the control, when substrates were incubated for 48 hr in vitro. The addition of 0.05% NIS Tween 80 to cultures growing on alfalfa hay resulted in a significant increase in CMCase (38.1%), xylanase (121.4%), Avicelase (not changed) and amylase (38.2%) activities after 36 h incubation. These results indicated that the addition of 0.05% Tween 80 could greatly stimulate the release of some kinds of cellulolytic enzymes without decreasing cell growth rate in contrast to trends reported with aerobic microorganism. Our SEM observation showed that NIS Tween. 80 did not influence the microbial adhesion to substrates used in the study. Present data clearly show that improved gas production, DM digestibility and cellulolytic enzyme activity by Tween 80 is not due to increased bacterial adhesion on the substrates.

Purification and Characterization of a Lipolytic Enzyme Produced by Aeromonas hydrophila PL43 (Aeromonas hydrophila PL43이 생산하는 지질분해 효소의 정제 및 특성)

  • A bacterial strain, producing an excellent lipolytic enzyme, was isolated from the intestinal tracts of an earthworm (Eisenia fetida). The strain was identified as Aeromonas hydrophila by phenotypic, chemotaxonomic characteristics and 16S ribosomal DNA analysis, and was designated as Aeromona hydrophila PL43. The lipolytic enzyme from A. hydrophila PL43 was purified via 35−45% ammonium sulfate precipitation, DEAE-sepharose fast flow ion-exchange, and sephacryl S-300HR gel filtration chromatography. The yield of the purified enzyme was 3.7% and 2.5% of the total activity of crude extracts with p-nitrophenyl butyrate (pNPB) and p-nitrophenyl palmitate (pNPP) as substrates, respectively. The molecular weight of the purified enzyme was approximately 74 kDa using gel filtration, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), and zymography. The optimal activity of purified enzyme was observed at 50℃ and pH 8.0 using pNPB, and 60℃ and pH 8.0 using pNPP. The purified enzyme was stable in the ranges 20− 60℃ and pH 7.0−10.0. The activity of purified enzyme was inhibited by PMSF, pepstatin A, Co2+, Cu2+, and Fe2+, but was recovered by metal chelating of EDTA. The Km and Vmax values of the purified enzyme were 1.07 mM and 7.27 mM/min using pNPB and 1.43 mM and 2.72 mM/min using pNPP, respectively.

Purification and Characteristics of Xylanases from Produced Thermophilic Alkalophilic Bacillus K17 (고온, 알칼리성 Bacillus K17이 생성하는 Xylanase의 정제 및 특성)

  • The culture filtrate of thermophilic alkalophilic Bacillus K17 strain contained two types of xylanases were purified by ammonium sulfate fractionation, DEAD-Sephadex A-50 column chromatography, CM-Sephadex C-50 column chromatography and Sephadex G-100 gel filtration. The purified enzymes were found to be homogeneous by sodium dodecyl sulfate and disc polyacrylamide gel electrophoresis. Xylanase I and II were characterized with respect to molecular weight, optimal temperature and pH, thermal and pH stability, and Michaelis constant. Xylanase II was more active and stable, and showed greater substrate affinity and molecular weight than xylanase I. The activities of xylanases I and II were inhibited by Cu$^{++}$, Ag$^+$, Hg$^{++}$ and Fe$^{++}$. Xylanase I hydrolyzed xylan to yield xylobiose and higher amount of xylooligosaccharides, but xylanase II produced xylose other than xylobiose and xylooligosacchrides.

Lignocellulose Biodegradation and Interaction between Cellulose and Lignin under Sulfate Reducing Conditions (황산염 환원 조건에서 리그노셀룰로오스의 분해 및 리그닌과 셀룰로오스의 상호작용)

  • In this study, the biodegradation test on lignocellulose under sulfate reducing conditions was carried out. In particular, the interaction between cellulose and lignin was investigated with various g-cellulose/g-lignin (C/L) ratios: 42.15, 4.59, 2.51, 1.14 and 0.7. It was shown that the rate of cellulose degradation decreased in proportion to the lignin content. Assuming first order degradation kinetics, the consequences of competitive inhibition were graphically shown for different C/L ratios. The relation between cellulose reduction rate and C/L ratio was expressed by logarithm function with a determination coefficient of 0.97. Lignocellulose reduction rate was also described as a logarithm function of C/L ratio showing a inhibition effect by lignin. In the mean time, the rate of lignin decomposition was higher at C/L ratio of 2.51 and 1.14 compared with C/L ratios of 4.59 and 0.7, indicating that excessive extra carbon source is not appropriate for lignin biodegradation.

  • Experiments were conducted to determine effects of whole and processed corns on in situ disappearance rates of nutrients in the rumen and in vitro degradability of dry matter by rumen microorganisms. Whole corn(WC) was processed into four different types; ground corn(GC), cracked corn(CC), flaked corn(FC), and soaked corn(SC). In the in situ experiment, the rate of ruminal DM disappearance after 48 hour incubation was highest in GC(76.1%) and lowest in WC and SC(12% ; P<0.01). The rate of ruminal CP disappearance after 48 hour incubation was highest in GC and CC(48 and 38%, respectively; P<0.01). The rate of ruminal OM disappearance after 48 hour incubation was highest in GC(76.14%) and lowest in WC and SC(11.82 and 12.26%, respectively; P<0.01). In the in vitro experiment, the two-stage incubation technique was used to measure digestibilities of whole and processed corns. The digestibility of DM was higher in GC, CC and FC(86.95, 85.84 and 82.29%, respectively) than in WC(15.36%; P<0.01).
