Studies on Microbial Utilization of Agricultural Wastes (Part 4) Effect of Acid in Neutralization after Alkali Treatment of the Wastes on Cellulosic Single Cell Protein Production (농산폐자원의 미생물학적 이용에 관한 연구 (제사보) 기질처리시의 알칼이ㆍ산중화 조건에 대하여)

  • Experiments were carried out to establish the effects of acids in neutralization after alkaline treatment of rice straw, with which cellulosic single cell protein can be produced by cellulose utilizing bacteria, Cellulomonas flavigena KIST 321, previously isolated by authors. Following results were obtained. 1. Rice straw as carbon source was pretreated with 10 volumes of 1 normality of NH$_4$OH or NaOH(NaOH/substrate:40%, and then washed with water or neutralized with H$_3$PO$_4$, H$_2$SO$_4$, HCl and CH$_3$COOH. Among the above mentioned methods, neutralization with H$_3$PO$_4$after alkaline treatment was proved to be the most effective on its digestibility and SCP production. Dry cell 12.28g/$\ell$ and 78% digestibility were obtained. 2. When rice straw was treated with NaOH solution, the result suggested that the productibity of cell-mass was attained on treatment of rice straw with 6% of NaOH (NaOH/substrate ratio) for 15~24hrs at room temperature. 3. When rice straw was treated with NaOH, a volume of water to substrate is adequate by two or three fold and the amount of NaOH can be economized up to 5% for the weight of rice straw. 4. The steaming of rice straw at 121$^{\circ}C$ for 30min. in alkaline treatment of rice straw gave the similiar effectiveness to that at room temperature for 15~24hrs. and accelerated the sterilization of the substrate. 5. Finally, the level of inorganic phosphate in a medium was investigated. 11.2g of dry cell was produced at the concentration of 0.2%, phosphate (phosphorous level 0.04%) in medium even though treated rice straw was neutralized with HCI instead of H$_3$PO$_4$, and 12.2g/$\ell$ at the concentration of 0.3% phosphate (phosphorous 0.04%) on neutralization with H$_2$SO$_4$.

  • The problem of temperament is not much of importance in Philosophical discussion compared to subjects such as liqi and Xingli. However, temperament is of great importance in practical study theory. "The reason people study is because they want to change their temper." the scholars said. The theory of change in temperament in neo-Confucianism is pursued in two directions. The first is to purify the temperament in a clear state. The second is to overcome the bias of temperament and lead it to a state of the middle path. The former is the study that purifies the murky water and turn it to a clear state, as the bead in it is brilliantly revealed. The latter is the study of maintaining without bias toward the path of nature itself. While the ultimate purpose of temperament change is to realize bright nature intact without obstruction of temperament, it goes through two processes specifically. It is to maximize intellectual and practical ability and overcome bias in inclination and character. The purification of temperament and the realization of Zhong are in sync with the orientation of the theory of temperament change.

  • Cultivation condition of yeast on the utilization of fermentable substrate from the cellulosic wastes such as rice hull, rice straw and corn starch cake was investigated. The results obtained were summarized as follows;1. Corn starch cake was respectively added to rice hull and rice straw in order to increase sugar concentration in the hydrolyzate, and then hydrolyzed. As the result, concentration of sugar in hydrolyzed solution of rice hull was 9.12%, in that of rice straw was 7.98%. 2. It was found that calcium carbonate as a neutralizer was the most effective to prepare the culture broth of yeast. 3. An optimal growth of Hansenula subpelliculosa GFY-2 was observed in the medium prepared by adding 0.3% of ammonium sulfate, 0.4% of potassium phosphate dibasic, 0.02% of magnesium sulfate, sodium chloride and calcium chloride to hydrolyaed sugar solution, respectively. 4. Hansenula subpellicuiosa GFY-2 cultured in the substrate solution which of rice hull and rice straw added to corn starch cake was assimilated more than 90% of sugar in the hydrolyzate within 48 hours. The yeast cells yielded in rice hull was 46.5%, and that of rice straw 45.4% to utilized sugars.

  • Various kinds of native herbage grasses like-Miscanthus sinensis, Arundinella hirta, Cymbopogen geirngii, Themeda japonica etc. are widely distributed in every Korean mountain. So we investigated the availability of native grass, Miscanthus sinensis as a substrate for the production of S. C. P. The results were obtained as follows. 1) At the alkali treatment, NaOH was the most effective with the exclusion of lignin, and pretreated Miscanthus with NaOH appeared to be a good substrate for the microbial growth. 2) When Miscanthus was treated with one to 10% NaOH, the microbial growth increased in proportion to the increased alkali concentration. Beyond 4% NaOH, a slight increase was observed. 3) Phosphoric acid, as a neutralizer, was the most effective in cell production after alkali treatment. 4) On the effect of incubation time, the productivity was best found at 60 hours, and the cell weight was 9.23 mg per 1 ml, and the microbial digestibility of substrate was 75.2%.

  • Present experiments were designed to estimate the effects of pretreatments by various kinds of alkalis to the agricultural wastes such as cereal straws as the substrate on the production of cellulosic single-cell protein. Among the various kinds of alkalis NaOH was proved to be the most effective on improving the digestibility of cellulose by the bacteria isolated. NH$_4$OH which is inferior to NaOH in the effectiveness of treatment might have more economic advantage in the price, and the ammonium salt resulted from the neutralization can be used as the nitrogen source by bacteria. The treatment with higher concentration than 1 normality of NH$_4$OH didn't increase the productivility of cell mass. About five per cent of (NH$_4$)$_2$SO$_4$ in medium resulted from the neutralization didn't have any influence in the cell mass productivility. Futhermore, the cell mass productibility was higher in the case of neutralization than alkali free washing. The digestibility of straws was increased from 7.9% to 46.0% by NH$_4$OH treatment, and 6.3∼6.45g of dry cell were obtained from 40g of NH$_4$OH treated straws. In the case of NaOH treatment, 8.6g of cell mass was obtained from 40g of substrate.

The Production of Sodium Gluconate by Aspergillus niger (Aspergillus niger를 이용한 글루콘산 나트륨의 생산)

  • Sodium gluconate was produced by neutralization of gluconic acid formed during the submerged culture fermentation of glucose with Aspergillus niger ACM 7. The fermentation characteristics of Aspergillus niger ACM 7 were investigated quantitatively according to the change of the initial glucose concentrations and the initial pHs of fermentation broth. The maximum specific growth rate was estimated to be $0.20hr^{-1}$ at 95g/$\ell$ of initial glucose concentration. The maximum fermentability of sodium gluconate was 95% at the initial glucose concentration of 26g/$\ell$. However, the maximum sodium gluconate productivity was 1.18g/$\ell$/hr when the initial glucose concentration was 110g/$\ell$. The optimum pH was found to be 5.5 for both the cell growth and the sodium gluconate production. With optimized culture conditions, the productivity of sodium gluconate in a fed-batch culture(production fermentor, 16,000$\ell$) increased up to 7.1g/$\ell$/hr.

  • Lactulose (4-O-${\beta}$-D-galactopyranosyl-D-fructose) is a non-digestible synthetic ketose disaccharide which can used in food and pharmaceutical fields due to its useful functions for encephalopathy, chronic constipation, hyperammonemia, etc. Therefore, the lactulose is regarded as one of the most important disaccharides and have been concentrated much interesting as an attractive functional material in the current industry. From this reason, the research related on the production of lactulose has been carried out various academic and industrial research groups. To produce lactulose, two main methods, chemical production and enzymatic production have been used. Commercially lactulose produced by alkaline isomerization of lactose as chemical production method but it has many disadvantages such as rapid lactulose degradation, purification, and waste management. From these reasons, lactulose produced by enzymatic method which solves these problems has been suggested as a proper method for lactulose production. Two different enzymatic methods have been reported as methods for lactulose production. Lactulose can be obtained through hydrolysis and transfer reaction catalyzed by a ${\beta}$-galactosidase which requires fructose as co-substrate and exhibits a low conversion. Alternatively, lactulose can be produced by direct isomerization of lactose to lactulose catalyzed by cellobiose 2-epimerase which requires lactose as a single substrate and achieves a high lactulose yield. This review summarizes the current state of lactulose production by chemical and biological methods.

Remediation of Acid Mine Drainage from an Abandoned Coal Mine Using Steel Mill Slag, Cow Manure and Limestone (제강슬래그, 우분 및 석회석을 활용한 폐 석탄광의 산성광산배수 처리)

  • In order to remediate acid mine drainage (AMD) from the Jeongam coal mine, steel mill slag, cow manure and limestone were used. As a result of batch test, the proper amounts for treating 1 L of acid mine water from the mine were determined as 15 g of steel mill slag, 15 g of cow manure and 500 g of limestone. After feasibility test, remediation system was arranged in the order of steel mill slag tank combination of cow manure and limestone, precipitation tank and oxidation tank. During 54 days' operations, the pH values of the treated waters increased from 3.0 to 8.3 and 61 % of sulfate concentration in an initial water was decreased. In addition, the removal efficiencies for metals in the water were nearly 99.9% for Al, Fe, Zn and 92.6% for Mn. Thus, the combination of steel mill slag, cow manure and limestone can be used as neutralization 때d metal removal for acid mine drainage.

An origin and development, the thought and understanding of actual world of Noron (노론의 연원과 전개, 철학사상과 현실인식)

  • Since Noron(老論) had organized in the period of Sookjong(肅宗), it constantly had led the political situation of Choson until Choson(朝鮮) perished as the grasping political power. Studies and thoughts development of Noron can be devided into four periods. First, the term of politics of faction of the period of Sookjong. Second, a period of Youngjo(英祖) and Joungjo(正祖). Third, a period of politics of power(勢道政治). Fourth, the latter term of 19century. We can look into an origin and development aspect in outline by dividing like this. The general character of Noron can be summarized by the respect of Song Si-yeol(宋時烈, 1607-1689), the theory of a party of a man of virtue(君子黨論) based on the theory of moral civilization of Choson(朝鮮中華論), the succession of Lee i(李珥; 1636-1684)'s neo-confucianism, rejecting all teaching that does not conform to neoconfucianism and protecting right studies, and oppression of Roman Catholic. The noticeable scholars of Noron were Kwon sang Ha(權尙夏; 1641~1721), Kim chang hyup(金昌協; 1651~1708), Lee jea(李縡; 1680~1746) etc. These scholars of Noron following Song Si-yeol had tried to raise "Learning of the Way"(正明道) by respecting Zushi and removing injustice(尊朱子攘夷狄), also believed people should embody moral values in their society and country. and possessed an will guiding to stabilize the country by rejecting uncivilization(尊王攘夷). Above all, they insisted, the King of Choson should rule with 'lighting heavenly reason'(明天理). Also they insisted the King and countrymen should together strive to recover civilization of moral humanity and destroy uncivilzation. But gradually they lost the motive and purpose of moral politics in the seventeenth century. Finally Noron Byeokpa(?派) take over the reins of government. It resulted in the bad effect of politics of autocrat(勢道政治) having their own way to use power of authority after death of Jungjo(正祖). The peculiar character of Noron politics can valued as the extreme aspect of 'according of politics and scholarship'(政學一致).

AN EVALUATION OF CHEMICAL DEGRADATION OF LIGHT-CURED RESTORATIVE COMPOSITES (광중합 복합레진의 화학적 분해 평가)

  • The aim of this study was to evaluate the resistance to degradation of four commercial composite resins in an alkaline solution. The brands studied were Charisma, Filtek P 60, Palpique Estelite, and Spectrum. Preweighed discs of each brand were exposed to 0.1N NaOH solution at $60^{\circ}C$. After 2 weeks they were removed, neutralized with HCl, washed with water and dried. Resistance to degradation was evaluated on the basis of following parameters: (a) mass loss(%) - determined from pre-and post-exposed specimen weights; (b) Si loss(ppm) - obtained from ICP-AE analysis of solution exposed to specimens; and (c) degradation depth(${\mu}m$) - measured SEM and CLSM from polished circular sections of exposed specimens. The results were as follows: 1. The sequence of mass loss was in descending order by Palpique Estelite, Filtek P 60, Charisma, and Spectrum. 2. The sequence of the degree of degradation layer depth was in descending order by Filtek P 60, Charisma, Palpique Estelita, and Spectrum. 3. The sequence of the Si loss was in descending order by Chrisma, Spectrum, Palpique Estelite, and Filtek P 60. 4. The correlation coefficient between mass loss and degradation layer depth was relatively high(r=0.704, p<0.05). 5. When observed with SEM, destruction of bonding was observed between resin matrix and filler. 6. When observed with CLSM, degradation layer depth of composite resin surface was observed.
