In vitro Anti-proliferative Characteristics of Flavonoids and Diazepam on MDA-MB-231 Breast Cancer Cells (Flavonoid류와 diazepam의 시험관 내 MDA-MB-231 유방암세포 증식 억제 효과)

  • The beneficial use of sedatives is often required for medically ill patients. This study examined the effect of plant flavonoids and diazepam peripheral-type benzodiazepine receptor (PBR) activation and glucose utilization in breast cancer cells, along with their interactions. In estrogen receptor negative MDA-MB-231 cells, the anti-proliferative activity of fisetin (3,7,3',4'-tetrahydroxyflavone) and diazepam was more prominent than in estrogen receptor positive MCF-7 cells. Unlike PBR ligands, treatment with $10^{-6}$ M concentration of diazepam for 3 days exhibited anti-proliferative effects, while similar to apigenin (4',5,7-Trihydroxyflavone) and fisetin, diazepam hardly affected the PBR mRNA expression by MDA-MB-231 cells. Treatment with $10^{-6}$ M concentration of flavonoids and diazepam for 3 days inhibited the glucose utilization of MDA-MB-231 cells. Treatment with $10^{-6}$ M concentration of flavonoids and diazepam for 6 days showed increased cytotoxicity and reduced the PBR mRNA expression of the MDA-MB-231 cells. Apigenin enhanced diazepam-induced anti-proliferative effects on the MDA-MB-231 cells as well. All together, this study showed the in vitro anti-proliferative activity of flavonoids and diazepam on MDA-MB-231 breast cancer cells, plus additive enhancements. In conclusion, this study provides experimental basis for advanced trials in the future.

  • Marker genes are defined as genes in which the expression level characterizes a specific experimental condition. Such genes in which the expression levels differ significantly between different groups are highly informative relevant to the studied phenomenon. In this paper, first the system can detect marker genes that are selected by ranking genes according to statistics after normalizing data with methods that are the most widely used among several normalization methods proposed the while, And it compare and analyze a performance of each of normalization methods with mult-perceptron neural network layer. The Result that apply Multi-Layer perceptron algorithm at Microarray data set including eight of marker gene that are selected using ANOVA method after Lowess normalization represent the highest classification accuracy of 99.32% and the lowest prediction error estimate.

A Study on the Reduction of Reservoir Capacity by the Sedimentation (퇴사로 인한 저수지내용적 감소에 관한 조사연구)

  • With 31 existing reservoirs selected in the Yong San River Basin, the sedimentation of the reservoirs has been calulated by comparing the precent capacity with original value, which revealed its reduced reservoir capacity. The reservoirs have a total catchment area of 13,871 ha. with a total capacity of 17.6 million cubic meter, and are short of water supply due to reduction of reservoir capacity. Annual sedimentation in the reservoir ranged from 120 to 3,770 cubic meter per square kilometer with great difference in its distribution, and the average value was 877 cubic meter. This wide disparity is analysed to come principally from the topography, geology, vegetation and hydraulics. The reservoir basin had a large portion of devastated land but has become green more and more in the last 10 years. It can be summarized that in the reservoirs with an average period of sedimentation of 26 years, the reduction rate of reservoir capacity amounts 12.5%, and 0.48% is shown for annual capacity reduction rate.

Material Decomposition through Weighted Image Subtraction in Dual-energy Spectral Mammography with an Energy-resolved Photon-counting Detector using Monte Carlo Simulation (몬테카를로 시뮬레이션을 이용한 광자계수검출기 기반 이중에너지 스펙트럼 유방촬영에서 가중 영상 감산법을 통한 물질분리)

  • Mammography is commonly used for screening early breast cancer. However, mammographic images, which depend on the physical properties of breast components, are limited to provide information about whether a lesion is malignant or benign. Although a dual-energy subtraction technique decomposes a certain material from a mixture, it increases radiation dose and degrades the accuracy of material decomposition. In this study, we simulated a breast phantom using attenuation characteristics, and we proposed a technique to enable the accurate material decomposition by applying weighting factors for the dual-energy mammography based on a photon-counting detector using a Monte Carlo simulation tool. We also evaluated the contrast and noise of simulated breast images for validating the proposed technique. As a result, the contrast for a malignant tumor in the dual-energy weighted subtraction technique was 0.98 and 1.06 times similar than those in the general mammography and dual-energy subtraction techniques, respectively. However the contrast between malignant and benign tumors dramatically increased 13.54 times due to the low contrast of a benign tumor. Therefore, the proposed technique can increase the material decomposition accuracy for malignant tumor and improve the diagnostic accuracy of mammography.

Investigation of Subsurface Structure of Cheju Island by Gravity and Magnetic Methods (중력 및 자력 탐사에 의한 제주도 지질구조 연구)

  • The geologic structure of the Cheju volcanic island has been investigated by analyzing the gravity and magnetic data. Bouguer gravity map shows apparent circular low anomalies at the central volacanic edifice, and the maximum difference of the anomaly values on the island appears to be 30 mgal. The subsurface structure of the island is modeled by three-dimensional depth inversion of gravity data by assuming the model consists of a stacked grid of rectangular prisms of volcanic rocks bounded below by basement rocks. The gravity modeling reveals that the interface between upper volvanic rocks and underlying basement warps downward under Mt. Halla with the maximum depth of 5 km. Magnetic data involve aeromagnetic and surface magnetic survey data. Both magnetic anomaly maps show characteristic features which resemble the typical pattern of total magnetic anomalies caused by a magnetic body magnetized in the direction of the geomagnetic field in the middle latitude region, though details of two maps are somewhat different. The reduced-to-pole magnetic anomaly maps reveal that main magnetic sources in the island are rift zones and the Halla volcanic edifice. The apparent magnetic boundaries inferred by the method of Cordell and Grauch (1985) are relatively well matched with known geologic boundaries such as that of Pyosunri basalt and Sihungri basalt which form the latest erupted masses. Inversion of aeromagnetic data was conducted with two variables: depth and susceptibility. The inversion results show high susceptibility bodies in rift zones along the long axis of the island, and at the central volcano. Depths to the basement are 1.5~3 km under the major axis, 1~1.5 km under the lava plateau and culminates at about 5 km under Mt. Halla. The prominent anomalies showing N-S trending appear in the eastern part of both gravity and magnetic maps. It is speculated that this trend may be associated with an undefined fault developed across the rift zones.

Physical and Mechanical Characteristics of Basalts in Northwestern and Southeastern Jeju Island (제주도 북서부 및 남동부 현무암의 물리적 & 역학적 특성)

  • Volcanic rocks in Jeju Island have vesicular structure caused by various environmental factors, and indicate the differences in geological and mechanical characteristics from region to region. In addition, the bedrock of Jeju Island shows stratified structure, that is, soft layers composed of pyroclastic rocks or cavities are irregularly developed between the basalt layers by several times of volcanic activity. In this study, various physical tests and unconfined compressive strength test were conducted for intact rocks sampled in northwestern onshore and offshore of Jeju Island. The results obtained in the tests were compared with the physical and mechanical characteristics of intact rocks sampled in southeastern offshore of Jeju Island. As a results, it was confirmed that the physical and mechanical characteristics of basalts sampled in northwestern Jeju Island were similar to those of basalts sampled in southeastern offshore of Jeju Island. In addition, it was possible to estimate approximate design parameters from the correlation of mechanical properties with physical properties of basalts in Jeju Island.

  • Site coefficient and amplification factor of current domestic Seismic Design Code (KBC-2009) have no consideration for the domestic ground condition in which the base rock is normally placed within 30m form the surface. Accordingly, in this study dynamic centrifugal test and analysis for pile foundation into sand and upper structure were achieved. and the response spectrums of free surface and basement were compared with each other. Within the period 1 sec., the measured spectral acceleration of free surface and basement was bigger than the design spectral acceleration of SC and SD site. However the measured spectral acceleration of free surface and basement for the period over 1.5 sec. was smaller than the design spectral acceleration of SC site. There was no severe difference of spectral acceleration according to the upper structure, embedded depth of foundation and free surface conditions. Consequently, normal domestic apartment housing for the period range over 1.5sec. could be design more economically applying these test result.

Ultrasonic Phantom Based on Plastic Material for Elastography (초음파 탄성 영상 평가를 위한 플라스틱 기반의 팬텀 개발)

  • A human tissue mimicking phantom is constructed to assess the performance of a medical ultrasound elasticity imaging system. In a human body, the tumor or cancer is stiffer than its surrounding normal tissue. A technique fur imaging the elasticity of such a tissue is referred to as elastography. Homogeneous elasticity phantoms with differing Young's moduli are constructed using a plastic hardener and softener to simulate the mechanical characteristics of a diseased human tissue. The Young's modulus of the fabricated homogeneous phantom materials were measured from 11.1 to 79.6 kPa depending on the mixing ratio of the amount of the hardener to that of the softener. An ultrasound lesion mimicking phantom was made of these materials, and ultrasound elasticity imaging was performed on it. It is confirmed in this paper that the fabricated plastic-based elasticity phantom is useful in representing the elastic characteristics of a human tissue.

  • To build tetrahedra mesh for FEM numerical analysis, Boundary Representation (B-Rep) model is required, which provides the efficient volume description of an object. In engineering, the parametric solid modeling method is used for building B-Rep model. However, a geological modeling generally adopts discrete modeling based on the triangulated surface, called a Sealed Geological Model, which defines geological domain by using geological interfaces such as horizons, faults, intrusives and modeling boundaries. Discrete B-Rep model is incompatible with mesh generation softwares in engineering because of discrepancies between discrete and parametric technique. In this research we have developed a converting program from Sealed Geological Model to Gmsh and COMSOL software. The developed program can convert complex geological model built by geomodeling software to user-friendly FEM software and it can be applied to geoscience simulation such as geothermal, mechanical rock simulation etc.

Weathering Characteristics of Granitic Grus in Naesung Stream Drainage, Yeongju-Bonghwa Basin, Korean Peninsula (내성천 유역분지인 영주-봉화 분지 화강암 구릉대의 풍화 특색)

  • Naesung stream famous for 'sandy river', a tributary to the Nakdong River, flows through Yeongju-Bonghwa Basin, its drainage. If the dismantlement of granitic hills in basin is in final stage, weathering materials from hills into stream are finer materials like silty or sandy loam than coarse sand, because sand as weathering mantles is provided from granitic hills, in general. So the granitic hills in Yeongju-Bonghwa basin is dissecting present. As a results of the CIA analysis(A-CN-K and A-CNK-FM ternary diagram), chemical weathering of granitic grus in Yeongju-Bonghwa basin is too very weak for calcium and sodium to be dissolved and go as far as to be more weak than that of Jeongeup, Nonsan and Namwon, common granitic grus in Korean Peninsula. Therefore, the chemical characteristics of granitic hills in Yeongju-Bonghwa basin show that the alteration of weathering mantles just finished disintegration and is dissected at a standstill. Plenty of sands provided from granitic hills is filling the channel of Naesung stream.
