Quantification of Icariin Contents in Epimedium koreanum N. by Using a Near Infrared Reflectance Spectroscopy (NIRS를 이용한 삼지구엽초의 이카린 함량 분석)

  • Near infrared reflectance spectroscopy (NIRS) has become widely accepted for rapid quantitative analysis of components in some crops. Our object was to determine icariin contents in whole plant of Epimedium koreanum by using an NIRS system. Total 150 plant samples previously analyzed by HPLC were scanned by NIRS and 68 samples were selected for calibration and validation equation. A calibration equation calculated by MPLS(modified partial least squares) regression technique was developed and a coefficient of determination in calibration and validation sets were 0.95 and 0.82, respectively. A comparison between NIRS estimation and HPLC value was performed with the remaining samples not included in the calibration and validation sets. Most of samples also showed a positive correlation like a validation set. Our results demonstrate that this developed NIRS equation can be practically used as a mass screening method for rapid quantification of icarin contents in Epimedium koreanum N.

Development of On-line Sorting System for Detection of Infected Seed Potatoes Using Visible Near-Infrared Transmittance Spectral Technique (가시광 및 근적외선 투과분광법을 이용한 감염 씨감자 온라인 선별시스템 개발)

  • In this study, an online seed potato sorting system using a visible and near infrared (40 1100 nm) transmittance spectral technique and statistical model was evaluated for the nondestructive determination of infected and sound seed potatoes. Seed potatoes that had been artificially infected with Pectobacterium atrosepticum, which is known to cause a soil borne disease infection, were prepared for the experiments. After acquiring transmittance spectra from sound and infected seed potatoes, a determination algorithm for detecting infected seed potatoes was developed using the partial least square discriminant analysis method. The coefficient of determination($R^2_p$) of the prediction model was 0.943, and the classification accuracy was above 99% (n = 80) for discriminating diseased seed potatoes from sound ones. This online sorting system has good potential for developing a technique to detect agricultural products that are infected and contaminated by pathogens.

Evaluation of Chemical Composition in Reconstituted Tobacco Leaf using Near Infrared Spectroscopy (근적외선 분광분석법을 이용한 판상엽 화학성분 평가)

  • Near InfraRed Spectroscopy(NIRS) is a quick and accurate analytical method to measure multiple components in tobacco manufacturing process. This study was carried out to develop calibration equation of near infrared spectroscopy for the prediction of the amount of chemical components and hot water solubles(HWS) of reconstituted tobacco leaf. Calibration samples of reconstituted tobacco leaf were collected from every lot produced during one year. The calibration equation was formulated as modified partial least square regression method (MPLS) by analyzing laboratory actual values and mathematically pre-treated spectra. The accuracy of the acquired equation was confirmed with the standard error of prediction(SEP) of chemical components in reconstituted tobacco leaf samples, indicated as coefficient of determination($R^2$) and prediction error of sample unacquainted, followed by the verification of model equation of laboratory actual values and these predicted results. As a result of monitoring, the standard error of prediction(SEP) were 0.25 % for total sugar, 0.03 % for nicotine, 0.03 % for chlorine, 0.16 % for nitrate, and 0.38 % for hot water solubles. The coefficient of determination($R^2$) were 0.98 for total sugar, 0.97 for nicotine, 0.96 for chlorine, 0.98 for nitrate and 0.92 for hot water solubles. Therefore, the NIRS calibration equation can be applicable and reliable for determination of chemical components of reconstituted tobacco leaf, and NIRS analytical method could be used as a rapid and accurate quality control method.

Discrimination of Geographical Origin and Seed Content in Red Pepper Powder by Near Infrared Reflectance Spectroscopic Analysis (근적외선 분광분석법에 의한 고춧가루의 원산지 및 고추씨 혼입 판별)

  • Red pepper powder (Capsicum annum L.) is an important seasoning as a kimchi ingredient in korea and most korean consumer tend to eat the korean red pepper powder as the better than other oriental country such as China. Near infrared reflectance spectroscopy (NIRS) was applied for discrimination according to geographical origin (Korea, China) of red pepper powder. The objective of this study is to determine if NIR technique could be used to discriminate between the korean red pepper powder and non-korean red pepper powder according to seed content and maxing ratio in red pepper powder by using the new method. Rapid, precise and nondestructive analysis method for determination of the geographical origin of red pepper powder by near infrared spectroscopy and chemometrics were performed. It has been observed discriminant analysis with PLS is adequate to determinate the geographical origin of red pepper powder. It tend to difficult the discrimination of geographical origin according to increase the seed content of red pepper powder. The accuracy of discrimination in mixed red pepper powder was range from 95.2% to 100%.

Discriminant Analysis of Cigarette Brands by Nearinfrared Spectroscopy (근적외선 분광법을 이용한 제품담배 판별 연구)

  • This experiment was conducted to investigate the discrimination of cigarette brands and the similarity between Korea and America cigarette brands by near infrared spectra. Statistical tools such as principal component analysis (PCA) and mahalanobis distance(M.D) were used. The discrimination rate of the Korea and the America cigarette brands, determined by position number which was calculated with PCA and M.D, was 94% and 87%, respectively. The spectra of the 10 America cigarette brands were selected by averaging 5 sample spectra for each brand and another 5 spectra for each brand were investigated to use as the sample spectra. Comparing the sample spectra with the selected spectra by M.D using 410-1090 nm, 1110-1850 nm and 1970-2490 nm wavelength, the discrimination rate which was determined by the closest M.D between the sample and the selected spectra was 100% when each spectra was investigated on the same time. But the discrimination rate decreased 50% when the sample and the selected spectra were investigated on the different time. Excluding 1970-2490 nm wavelength, the discrimination rate increased up to 90% when the sample and the selected spectra were investigated on the different time. Comparing the spectra of Korea cigarette brands with those of America cigarette brands by M.D using only 410-1090 nm and 1110-1850 nm wavelength, the spectra of Expo(G) was similar to Winston, Vantage(U.L) and Benson & hedges(M.), the spectra of Hanaro(D) was similar to Carrel, Winston(L), Vantage(U.L), \Vantage and Carrel(L), the spectra of Hanaro(L) was similar to Winston(L) , Carton, Vantage and Carmel(L) and the spectra of Pinetree was similar to Kent, Kool, Kent(G.L), Merit and Benson & hedges(L), respectively. Rapid Near Infrared Transmittance Analysis of Ingredients on the Casing Materials (근적외선 투과 분광분석법을 이용한 가향액 중 가향제 분석)

  • It is very important to add uniformly casing materials on tobacco for taste and flavor. However, analysis of casing materials was spent much time, effort and money. The object of this study was the development of a rapid method for the determination of glycerine, propylene glycol(PG), sucrose, glucose, fructose and water in the casing materials using the NIR transmittance method. Hundreds of calibration samples, with extended ranges (50%, 75%, 100%, 125%, and 150% of standard addition) in each constituent, were prepared in the casing materials at the various temperatures $(25^{\circ}C\;and\;30^{\circ}C)$. Calibration equation was developed by modified partial least square (MPLS) method using second derivative. The standard error of calibration and $R^2$ between added value and NIR estimated value results were $0.007{\sim}0.034\;and\;0.996{\sim}1.000$ for the casing sample set, respectively. The standard error of prediction and R2 between added value and NIR estimated value results were $0.010{\sim}0.034\;and\;0.997{\sim}1.000$ for the casing sample set, respectively. The analysis result was not different significantly between the NIR and added value. These results show that the NIR measurement system is an effective tool to ensure quality on the casing materials.

Preparation of ZnO Thin Films with UV Emission by Spin Coating and Low-temperature Heat-treatment (스핀코팅 및 저온열처리에 의한 자외선 발광특성을 갖는 산화아연 박막의 제조)

  • Purpose: This research is that prepare amorphous or crystalline ZnO thin films with pure strong UV emission on soda-lime-silica glass (SLSG) substrates by low-temperature annealing. Methods: Growth characteristic and optical properties of the amorphous or nano-crystalline ZnO thin films prepared on soda - lime - silica glass substrates by chemical solution deposition at 100, 150, 200, 250 and $300^{\circ}C$ were investigated using X-ray diffraction analysis, ultraviolet - visible - near infrared spectrophotometer, and photoluminescence. Results: The films exhibited an amorphous pattern even when finally annealed at $100^{\circ}C{\sim}200^{\circ}C$ for 60 min, while crystalline ZnO was obtained by prefiring at 250 and $300^{\circ}C$. The photoluminescence spectrum of amorphous ZnO films shows a strong NBE emission, while the visible emission is nearly quenched. Conclusions: These results indicate it should be possible to cheaply and easily fabricate ZnO-based optoelectronic devices at low temperature, below $200^{\circ}C$, in the future.

Performance evaluation of Terrestrial Laser Scanner over Calibration Baseline (표준거리측정 시설을 이용한 지상라이다 성능 평가)

  • This study deals with the measurement of reflectivity as well as the distance accuracy with Terrestrial Laser Scanner(TLS) using time of flight methods and near infrared wave length, for a variety of user-made targets. Especially, point clouds' reflection to several targets was measured with Gretag Macbeth il spectrophotometer in the office. And the distance accuracy in comparison to reference distance for TLS performance evaluation, was tested after scanning the user-made targets and measuring the inter-pillars distances over the precise EDM calibration baseline. The results of test was shown that except white resin objects, with approx. 10m and 170m inter-pillar distances, other targets achieved the distance accuracy of several millimeters(mm) with respect to standard distances. Future work should be concentrate on a few parameters influencing on the distance accuracy such as atmospheric correction, instrument correction, the additive constant or zero/index correction, etc.

The Effect of Representative Dataset Selection on Prediction of Chemical Composition for Corn kernel by Near-Infrared Reflectance Spectroscopy (예측알고리즘 적용을 위한 데이터세트 구성이 근적외선 분광광도계를 이용한 옥수수 품질평가에 미치는 영향)

  • The objectives were to assess the use of near-infrared reflectance spectroscopy (NIRS) as a tool for estimating nutrient compositions of corn kernel, and to apply an NIRS-based indium gallium arsenide array detector to the system for collecting spectra and analyzing calibration equations using equipments designed for field application. Partial Least Squares Regression (PLSR) was employed to develop calibration equations based on representative data sets. The kennard-stone algorithm was applied to induce a calibration set and a validation set. As a result, the method for structuring a calibration set significantly affected prediction accuracy. The prediction of chemical composition of corn kernel resulted in the following (kennard-stone algorithm: relative) moisture ($R^2=0.82$, RMSEP=0.183), crude protein ($R^2=0.80$, RMSEP=0.142), crude fat ($R^2=0.84$, RMSEP=0.098), crude fiber ($R^2=0.74$, RMSEP=0.098), and crude ash ($R^2=0.81$, RMSEP=0.048). Result of this experiment showed the potential of NIRS to predict the chemical composition of corn kernel.

Study on Development of Non-Destructive Measurement Technique for Viability of Lettuce Seed (Lactuca sativa L) Using Hyperspectral Reflectance Imaging (초분광 반사광 영상을 이용한 상추(Lactuca sativa L) 종자의 활력 비파괴측정기술 개발에 관한 연구)

  • In this study, the feasibility of hyperspectral reflectance imaging technique was investigated for the discrimination of viable and non-viable lettuce seeds. The spectral data of hyperspectral reflectance images with the spectral range between 750 nm and 1000 nm were used to develop PLS-DA model for the classification of viable and non-viable lettuce seeds. The discrimination accuracy of the calibration set was 81.6% and that of the test set was 81.2%. The image analysis method was developed to construct the discriminant images of non-viable seeds with the developed PLS-DA model. The discrimination accuracy obtained from the resultant image were 91%, which showed the feasibility of hyperspectral reflectance imaging technique for the mass discrimination of non-viable lettuce seeds from viable ones.
