방선균을 이용한 항생물질 발효

  • 방선균에서 항생물질을 발효 생산하기 위해서는 이와 관련된 방선균의 생리학적 및 생화학적 특성을 고찰해 볼 필요가 있다. 우선, 항생물질은 방선균의 2차 대사산물로써 이의 생산은 미생물의 성장과는 거의 연계되어 있지 않다. 즉, 방선균의 발효 과정을 살펴보면, 일반적으로 균체가 성장하는 증식 단계(trophophase)와 항생물질이 생산되는 발효 단계(idiophase)로 구분할 수 있다(2). 다시 말해서, 항생물질과 같은 2차 대사산물은 균체의 성장이 어느 정도 완료되어진 이후에 생합성되어지기 시작하며, 이는 방선균의 생활 주기상의 분화과정과도 밀접한 관계를 갖고 있는 것으로 알려져 있다. 또 하나의 특징은 한 종류의 방선균으로부터 유사한 화학적 골격을 지닌 여러 종의 항생물질들이 동시에 생산되어지는 경우가 많으며, 외부 환경에 따라 그 생산량이 크게 영향을 받는다는 사실이다. 따라서 방선균에서 목적하는 항생물질만을 과량생산하기 위해서는 배지의 조성을 비롯하여 pH, 발효온도, 통기, 점도 등 여러가지 발효 조건들을 잘 조절해 주어야 한다. 이러한 관점에서 방선균을 이용한 항생물질 발효에 있어서 그 생산량을 증대시키기 위해 고려해 주어야 할 사항들을 고찰해 보기로 한다.

Growth of an Obligatory Methanotroph Methylosinus trichosporium OB3b on Methanol (메탄자화균 Methylosinus trichosporium OB3b의 메탄올 기질에서의 성장)

  • An obligatory type II methanotroph Methylosinus trichosporium OB3b was cultivated on methanol as a sole carbon and energy source. The effects of methanol concentration, pH, temperature, nitrogen source and phosphate concentration on cell growth were investigated and the results were compared with the growth on methane, which had been studied previously. When $(NH_4)_2SO_4$ was used as a nitrogen source, the maximal specific growth rate (${\mu}max$) on methanol was $0.20hr^{-1}$ and the carbon conversion efficiency(CCE) was 43%. In comparison, on methane, ${\mu}max$ and CCE were $0.08hr^{-1}$ and 32%, respectively. Ammonia was found to be a better nitrogen source for methanol-growing cells. Cell yield on nitrogen (YX/N) was the same regardless of nitrogen source as 7.14g dry cells/g N, but the yield on methanol(YX/N) was higher with ammonia(0.8g dry cells/g MeOH) than with nitrate(0.64g dry cells/g MeOH). Optimal pH and temperature were 7.0 and $30^{\circ}C$, respectively. Methanol inhibition on cell growth was observed at above 0.5%(v/v). Inhibition by phosphate was observed at above 60mM, although the inhibition on methanol dehydrogenase activity started at a much lower level of 20mM. Based on the experimental findings, the cellular physiology of M. trichosporium OB3b growing on the two closely-related carbon sources were discussed extensively.

Screening of ${\gamma}-Linolenic$ Acid-Producing Fungi (감마-리놀렌산 생산 곰팡이의 탐색)

  • For a study on the production of ${\gamma}-linolenic$ acid(GLA) by microorganisms, fifteen strains of Mucorales obtained from culture stocks and ten isolate strains were compared in their cell growth, lipid content, fatty acid composition and pellet size formed in shake flask culture. Among the fungi examined, the isolated fungus, designated as FB-354, was found to be the most suitable one for the production of GLA mainly due to its high contents of lipid, 29.9% of dry cell weight and GLA, 16.8% of the total fatty acids. The strain FB-354 was tentatively identified as Mucor sp. on the basis of morphological characteristics. Fungal oil produced by Mucor sp. FB-354 was fractionated into 81.1% of neutral lipid, 7.2% of glycolipid, and 11.8% of phospholipid. Although the GLA content in the phospholipid fraction was as high as 21.4%, most of the GLA was found in the neutral lipid fraction.

  • To develope food preservative, antimicrobial activities of Pinus densiflora (PD) ethanol extract against Listeria monocytogenes Scott A. Listeria monocytogenes Brie I and Listeria monocytogenes ATCC 19111 were investigated. The ethanol extracts of PD showed strong antimicrobial activities on Listeria monocytogenes. The crude ethanol extracts of PD were further fractionated by ether, ethyl acetate and butanol. The ether fraction from ethanol extract showed the strongest antimicrobial effects on Listeria monocytogenes in tryptic soy broth containing 40 mg/mL ether fractions compared with other fractions. The effect of ethanol extract of pinus densiflora against Listeria monocytogenes culture for growth stage in tryptic soy broth at 35$^{\circ}C$ showed the strongest antimicrobial activites for lag phase. The morphological changes of the cells were observed with transmission electron microscope (TEM) and scanning electron microscope (SEM) and the cells were injured by treatment of 40 mg/mL ethanol extract of Pinus densiflora.

Isolation and Characterization of Bacteria Capable of Degrading Bisphenol A (Bisphenol A 분해세균의 분리 및 특성)

  • Eighty-seven microbial strains capable of growing on bisphenol A (BPA) as a sole carbon source were isolated from soils, waste waters and sludges. Among them, three bacterial strains were finally selected as potential decomposers through measuring BPA-degradation efficiency by HPLC analysis. Two of these bacterial strains were identified as Serratia marcescens 1901 and S. marcescens 1902, and another was Pseudomonas putida 1401 by 16S rDNA partial sequences and based on morphological and physiological properties. They showed higher cell growth and BPA degradation in PAV (PAS medium containing vitamin mixtures) than in PAS medium. The degradation efficiencies of these bacterial strains were within a range of 20-40% in the PAV containing 500 mg/1 or 100 mg/l of BPA fur 3 days. S. marcescens 1901 showed higher degradation efficiency at 100 mg/1 of BPA than those of other selected strains, while S. marcescens 1902 and P. putida 1401 degraded a high concentration of BPA (500 mg/l) with a degradation efficiency of 40% for 3 days. The BPA degradation using a mixed culture of three selected strains showed the similar level of dog-radation efficiency with that using a pure culture.

  • An extracellular polysaccharide, methylan, was produced under the specific conditions by Methylobacterium organophilum from methanol. The specific growth rate of cells was approximately constant regardless of C/N ratio and the specific product yield was maximum at a C/N ratio of 30. Methylan production was suppressed by the deficiency of mineral ions such as Mn$^{++}$ or Fe$^{++}$ ion. The optimal pH for cell growth and methylan production was 7. Whereas the optimal temperature for cell growth was found to be 37$^{\circ}C$, that for methylan production was 3$0^{\circ}C$. The methanol concentration above 4% completely inhibited the cell growth. The initial methanol concentration for the maximal production of methylan was 0.5% (v/v) and above this concentration, methylan production was markedly inhibited. To overcome the substrate toxicity and inhibition for both cell growth and methylan production, a fed-bach culture of intermittent feeding within 5 g/l methanol was conducted under the optimal culture condition. Methylan production of was stimulated by nitrogen limitation and methylan was accumulated up to 8.7 g/1 and cell mass also increased up to 12.4 g/l.

Characteristics of Submerged and Solid-State Fermentations for Production of Arachidonic Acid Mortierella alpina (Arachidonic Acid 생산을 위한 Mortierella alpina 곰팡이의 심부 및 고체 발효 특성 연구)

  • The objective of this work was to evaluate a solid-state fermentation process for the practical production of arachidonic acid(AA) by Mortierella alpina ATCC 32222. In the present investigation, batch culture kinetics for both submerged- and solid-state fermentations was carried out at $25^{\circ}C$ to identify the relationship between growth and arachidonic acid (AA) production. Glucose and yeast extract were used in submerged fermentations by using flasks, while rice bran was used as a sole raw material in the other type of fermentations by using a series of Petri dishes. It was evident that a mixed-growth associated pattern existed between the two variables, irrespective of modes of fermentations. The effect of carbon to nitrogen (CfN) ratio on AA production in solid-state fermentation was studied in the range of 6.5 - 20. As a result, an optimum condition was found to be 6.5. Supplementary carbon source was not necessary to meet the optimum C/N ratio. Unlike the Previous results obtained by other researchers, a supplement of sodium glutamate up to $4\%$ (w/w) to the rice bran medium did not have a positive effect on the AA productivity. However, an increase in AA productivity was obtained with the rice bran medium supplemented with sesame oil.

Effect of the Environmental and Nutritional Conditions on the Growth of Marine Microalga Isochrysis Galbana Parke (해양 미세조류 Isochrysis galbana Parke 성장에 대한 환경 및 영양 조건의 영향)

  • The marine microalga Isochrysis galbana Parke was studied to optimize its growth conditions in flask culture. Important medium components studied include nitrogen source, buffer, trace elements and vitamins. Environmental conditions include pH, temperature, light intensity, mixing extent and working volume. The medium prepared from natural sea-waters gave a higher final cell density than the medium prepared from synthetic sea-water Nitrate was a better source than ammonium. In the range of 0.4∼2mM, the final cell density was proportional to the initial nitrate concentration and the cell yield was estimated to be 8.5g dry cell wt/g N. For phosphate, optimal growth was observed in 0.1∼1.0mM but a considerable variation in pH was resulted. The addition of Tris at 5mM or 7mM could stabilize the medium pH, but this significantly reduced both growth rate and final cell density, The effect of trace elements and vitamins was negligible. Optimal temperature and initial pH were $20^{\circ}C$ and 8. When the intensity of incident light was varied in the range of 400∼2100 lux, the growth rate increased from 10mL to 70mL, the final cell density decreased although the initial growth rate did not change. Optimal agitation speed was 100rpm when working volume was 30mL. With optimal conditions, the maximum specific growth rate obtained was 0.021hr-1 and the final cell density was 1.1g/L.

Study on the Cultural Conditions of Starch Utilizing Yeast Sporobolomyces holsaticus (전분이용성효모, Sporobolomyces holsaticus FRI Y-5의 배양조건에 관한 연구)

  • It was investigated under several cultural conditions to produce biomass directly from starch by an strictly aerobic and amylolytic yeast, Sporobolomyces holsaticus FRI Y-5. Its optimal temperature and initial pH of medium for growth were $23^{\circ}C$ and 6.9, respectively. Activation energy, Ea, for growth was calculated to be 17.33 Kcal/mole from the Arrhenius relationship. When each of 13 nitrogen sources was added to the basal medium, $(NH_2)_2CO$ had the best effect, on which concentration of cell after 3 day incubation was 10.6 g/l and cell yield was 0.451. The yeast growth was affected by $MgSO_4,\;K_2SO_4\;and\;ZnSO_4$ as a mineral source and was best on the medium containing all of them. The addition of yeast extract (5g/l) could enhance the production of biomass and cell yield to 77% and 32%, respectively.

Optimization of Tyrosinase Production using Neurospora crassa (Neurospora crassa를 이용한 Tyrosinase 생산의 최적화)

  • Neurospora crassa (KCTC 6079) produces tyrosinase (EC 1.14.18.1) during sexual differentiation under derepressed conditions in the presence of inducers such as amino acid analogues, antimetabolites or protein synthesis inhibitors. The selection of inducer concentration and induction time as well as inducer type are critical for the optimization of the enzyme production. The best inducer was found to be cycloheximide. Since cycloheximide was toxic to the cells, an optimal inducer concentration and an optimal induction time were determined to maximize the enzyme production from batch cultures. Mathematical models for the cell growth and the enzyme production were proposed and used for process optimization. By optimizing the induction conditions, maximum tyrosinase productivity was increased significantly.
