Identification of Orchid Mycorrhizal Fungi Isolated from Terrestrial Orchids in Mt. Hambaek, Korea (함백산의 난초과 식물의 뿌리에서 난균근균의 분리 및 동정)

  • In this study, orchid mycorrhizal fungi (OMF) were isolated from four terrestrial orchids on Mt. Hambeak, Platnathera chlorantha, Platnathera mandarinorum, Cephalanthera falcate, and Cephalanthera longibracteata. OMF were identified using morphological and sequences analysis of fungal internal transcribed spacer (ITS) regions by specific primer of basidiomycetous orchid mycorrhizas; ITS1-OF and ITS4-OF. Four species of orchid mycorrhizal fungi were identified as Ceratobasidium sp, Epulorhiza anaticula, Tulasnella calospora and Tulasnella sp.

Growth Stimulation of Alnus firma and Robinia pseudoacacia by Dual Inoculation with VA Mycorrhizal Fungi and Nitrogen-Fixing Bacteria and Their Synergistic Effect (VA 내생균근균(內生菌根菌)과 질소고정균(窒素固定菌)으로 이중접종(二重接種)한 사방오리나무와 아까시나무의 생장촉진(生長促進)과 접종(接種)의 상승효과(相乘效果))

  • To evaluate potential of VA mycorrhizal fungi for promoting growth of nitrogen-fixing trees and efficiency of fungal inoculation in relation to soil fertility and dual inoculation with bacteria, Alnus firma was grown for six months in pots with steam-sterilized soil after inoculation with Glomus mosseae, and Robinia pseudoacacia was grown in the fumigated field after inoculation with native Glomus sp. and Gigaspora sp. In unfertilized Alnus plants, 27% increase in dry weight(or 18% in height) was observed by Glomus inoculation, while plants inoculated with both VA mycorrhiza and actinomycete(crushed nodule inoculum) showed synergistic effect of 83% dry weight increase over uninoculated plants. In fertilized Alnus plants, mycorrhizal inoculation alone or dual inoculation with actinomycete resulted in depression of height and dry weight of plants. In case of Robinia, dual inoculation stimulated height growth by 23% (or dry weight by 25%) over the control in unfertilized field, while 13% more height growth(or 21% more dry weight) was observed in fertilized field. It is concluded that VA mycorrhizae, especially Glomus mosseae, have a potential for growth enhancement in Alnus, that synergistic effect of dual inoculation(mycorrhiza + actinomycete) exists in both Alnus and Robinia, and that responses of these plants to VA mycorrhiza are more pronounced in unfertilized soil.

Korean Tricholoma matsutake Strains that Promote Mycorrhization and Growth of Pinus densiflora Seedlings (균근 형성과 소나무 유묘 생장이 우수한 송이 균주의 선발)

  • Domestic and international production of Tricholoma matsutake has decreased owing to matsutake forests being left alone, host plant disease, forest fires, climate change, and so on. In order to identify strains that are suitable for the production of T. matsutake-inoculated seedlings, Pinus densiflora seedlings were inoculated with T. matsutake after in vitro rooting and mycorrhization was examined in the roots of T. matsutake-inoculated seedlings after 6 months. The mycorrhization rate was greater than 80% for 5 strains (NIFoS 421, 434, 1681, 1984, and 2001) out of 19 total strains. Seven strains (NIFoS 434, 441, 561, 562, 1016, 1807, and 1812) showed shoot/root ratios of less than 3.0 and had a seedling shoot biomass of 2.0 to 4.8 times higher than that of the root. Eight strains (NIFoS 441, 561, 562, 1016, 1807, 1812, 1984, and 2001) stimulated increases in shoot volume and three stains (NIFoS 441, 562, and 1812) promoted the growth of root biomass by mycorrhizal formation. In conclusion, 4 strains (NIFoS 434, 561, 1984, and 2001) out of 19 total strains tested showed higher mycorrhization rates and seedling growth than those of the other strains. We expect that the use of these four strains may contribute to T. matsutake-inoculated seedling production.

Effects of Orchid Mycorrhizal Fungi on the Growth of Cymbidium kanran and Phalaenopsis (난 균근균 처리가 한란과 호접란의 생육에 미치는 영향)

  • The effects of the orchid mycorrhizal fungi isolated from Cymbidium goeringii were investigated on the growth of orchid plants. The plants, a hybrid of Cymbidium kanran Jeju${\times}$C. kanran Nangoku and Phalaenopsis were inoculated with the fungal isolates of Rhizoctonia repens (P1), R. endophytica (P2) and R. repens (P3; different from P1) on the oatmeal agars for two months. Then the orchid plants were cultivated in the greenhouse for eighteen months. The difference of plant growth after cocultivation for two months was not found among the treatments. After cultivation for four months, the growth of hybrid plants of Cymbidium kanran Jeju${\times}$C. kanran Nangoku was observed to be distinguished in the Rhizoctonia repens (P1). After cultivation in the greenhouse for eighteen months, the plant height, the number of shoots, the number of leaves, the number of roots and plant fresh weight of the hybrid of Cymbidium kanran Jeju${\times}$C. kanran Nangoku, inoculated with Rhizoctonia repens (P1), were significantly increased compared to those of the control. The plants inoculated with Rhizoctonia repens (P3) were not different and the plants inoculated with R. endophytica were poor compared to the control. The plant height and fresh weight of the Phalaenopsis, inoculated with Rhizoctonia repens (P1), increased significantly compared to those of the control. Overall, Rhizoctonia repens (P1), was the most effective for the growth of a hybrid of Cymbidium kanran Jeju${\times}$C. kanran Nangoku.

  • The presence of endomycorrhizal fungi was examined on the soils collected from the followings; Cryptomeria japonica dominant forest (Wan San Park, Jun Ju city) and two coast areas (Digitaria sanginalis dominant; Sin Chang Ri, Young Il Kun and Pragmited communis dominant; Sap Kyo Cheon, A San). Six species in Endogonales were identified; Glomus intraradices, G. occulum, G. clarum, Acaulospora bireticulata, Scutellospora aurigloba, and Sc. gilmorei.

  • Vesicular-arbuscular mycorrhizal fungi were isolated from the soils of three different plant communities (Cassia mimsoides var. nomame, Themeda triandra var. japonica, Miscanthus sinensis var. purpurascens). Eight azygospores or chlamydospores of VA-mycorrhizal fungi were identified (eight species of four genera), and six among them newly described in Korea (six species of two genera); Glomus aggregatum, Gl. ambisporum, Gl. geosporum, Gl. mosseae, Scutellospora coralloidea, Sc. heterogama.

  • Truffles are cultivated by growing seedlings of compatible tree species in pasteurized substrate amended with truffle spores, maintaining the inoculated seedlings in the greenhouse until the mycorrhizal relationship is established and certified, planting the infected seedlings onto appropriate and properly prepared sites, and managing the resulting truffieres (truffle orchards) to maintain the truffle infections and encourage truffle fruiting. Questions remain to be addressed at each step of the process in developing best management practices for truffle cultivation.
