Mycorrhizal Development and Growth Stimulation of Pinus thunbergii Seedlings Inoculated with Pisolithus tinctorius at Two Soil Mixtures Treated with Six Nitrogen Levels (배양토(培壤土) 및 질소(窒素) 시비(施肥) 수준(水準)이 모래밭 버섯균(菌) (Pisolithus tinctorius)을 접종(接種)한 해송(海松)(Pinus thunbergii) 묘(苗)의 생장(生長)과 균근(菌根) 형성(形成)에 미치는 영향(影響))

  • This study was carried out for observation of growth, mycorrhizal formation and nutrient absorption of Pinus thunbergii seedlings treated with two soil mixtures and various nitrogen levels after inoculation with mycorrhizal fungus, Pisolithus tinctorius. 1. Seedlings grown on vermiculite applied with $50-150{\mu}g/ml$ nitrogen levels were well developed with pinnate type and cluster-like mycorrhizae. But seedlings on sandy loam had monopodial type in addition to the above-mentioned two types. 2. Optimum fertilization level for mycorrhizal formation is 50 or $150{\mu}g/ml$ N that showed best mycorrhizal formation of $86.4({\pm}3.14)%$ or $73.0({\pm}7.21)%$, respectively, but increased nitrogen levels decreased formation of mycorrhizal short roots. Seedlings applied with $450{\mu}g/ml$ nitrogen level decreased in net assimilation rate (NAR) and crop growth rate(CGR) during early growth of the seedlings, and they were increased since Aug, when nutrient application was stopped. 9. Inorganic nutrient absorption was increased more in seedlings grown on vermiculite and inoculated growth medium than those grown on sandy loam and noninoculated one, and it was gradually increased with increasing nitrogen increasing nitrogen level until $350{\mu}g/ml$. But $450{\mu}g/ml$ nitro gen level rather reduced absorption of nutrient.

  • Quercus acutissima seedlings were grown for two years in fumigated or non-fumigated in nursery soil in a medium with and without vegetative inoculum of the ectomycorrhizal fungi, Pisolitlius tinctorius (Pt) and Rhizopogon rubescens (Rr). Mycorrhizal formation were 42% in fumigated Pt inoculation and 36% in fumigated Rr inoculation. $2.16{\ell}\;per\;m^2$ of Pt vegetative inoculum in fumigated soil stimulated the seedling height (98%), root collar diameter (132%) and weight (420%). And Rr inoculation in fumigated soil increased the seedling height (44%), root collar diameter (23%) and weight (157%) compared with non-treated plot. There was no effect of Pt and Rr inoculation on the growth in non-fumigated soil. Nitrogen and $P_2O_5$ contents in foliage were not different by the treatment but $K_2O$ and Ca in fumigated soil were higher than non-fumigated soil.

  • Populus alba ${\times}$ P. glandulosa cuttings in nursery bed were inoculated with mycelium of ectomycorrhizal fungus, Pisolithus tinctorius (Pt) to evaluate effectiveness of the fungus in growth stimulation of poplar. Pt was cultured in 1l glass bottles with vermiculite-peatmoss mixture moistened with modified Melin-Norkrans' solution. The nursery bed was arranged for microplots of $1{\times}2m$ in size and fumigated with methyl bromide before inoculation and cutting placement. Fifty cuttings were placed in each microplot and two treatments (fumigation only and fumigation plus Pt inoculation) were replicated three times. At the end of the first growing season, inoculated plants grew 19% faster in height and produced 49% more dry weight (above-ground portion) than uninoculated plants. Survival rate of inoculated cuttings was also improved by 20% over that of uninoculated cuttings. Inoculated cuttings developed abundant fine root system with golden brown zigzag tips. In the middle of September a sporocarp of Pt was produced from an inoculated plot, suggesting successful establishment of mycorrhiza between poplar and Pt fungus.

Spore Diversity of Arbuscular Mycorrhizal Fungi in Upo Wetland (우포 습지에 분포하는 수지상균근균 포자의 다양성)

  • We extracted arbuscular mycorrhizal fungal (AMF) spores from rhizospheres of three plants from Upo Wetland, Korea. We identified the isolated AMF spores based on morphological characteristics and phylogenetic analysis of partial 18S rDNA nucleotide sequences. The species diversity of AMF spores was calculated among the study sites and host plants. Consequently, nine species from six genera of AMF spores were identified. We confirmed the species diversity of the AMF spores in rhizospheres affected by host plants in the wetland. In the course of this study, we confirmed a previously unreported AMF species in Korea: Diversispora epigaea. We described the morphological features and molecular characteristics of this previously unreported AMF species.

  • The histological studies were conducted to investigate the symbiotic relationships between Gastrodia elata and Armillaria mellea by using light and electron microscopes. The fungus, A. mellea, penetrated into the cortex of G. elata, in which endomycorrhizal mycelia in the cortical cells appeared to be dissolved and digested, and seemed to be consequently used as nutritional sources for G. elata growth. Staining of infected tissues revealed that protein- and fat-like substances were localized in the cells. The nuclei of cells infected by the fungal mycelia were hypertrophied 1.5 to 2 times as those without the fungal infection.

Effects of Herbicides on the Growth, Nodulation and VA Mycorrhizal Infection in Soybean (Glycine max) (제초제(除草劑) 처리(處理)가 콩 생육(生育) 및 근류균(根瘤菌) 착생(着生)과 VA 균근균(菌根菌) 감염(感染)에 미치는 영향(影響))

  • A field experiment was done to evaluate the growth response, rhizobia nodulation and vesicular-arbuscular mycorrhizal fungi (VAMF) infection of soybean treated with the different concentration of three-herbicides, alachlor, simazine, linuron. The results obtained were summarized as follows ; Emergence rate of soybean was non-significantly decreased by increasing the herbicide concentration. In simazine treatment, low emergence rate showed even in recommended concentration, especially the lowest emergence rate in treble concentration of recommended dose was about 50%. The plant length of soybean lowered by increasing the herbicide concentration, but there were almost no differences in recommended dose. Particularly that in a high concentration of herbicides at three weeks after treatment(WAT) was decreased remarkably. In the plant diameter of soybean shoot, no differences in the recommended concentration of alachlor and linuron at 2 WAT, but those of recommended dose was decreased with the lapse of time, and the lowest treatment was simazine-treated plot. Leaf area and fresh weight of soybean tended to decrease over the control even in recommended concentration and it was decreased with the increment of the herbicide dose. Yield component, fresh weight of rhizobia nodule and chlorophyll content of soybean leaves were reduced with the increment of herbicide dose, espicially remarkable tendency was revealed in simazine-treated plot. Seasonal infection rate of VAM showed a slight decrease by increasing of herbicide dose, and the lowest infection was simazine treatment. Significant correlation was realized among the reduction of rhizobia nodulation, VAM colonization and the agronomic characters of soybean and it was considered that the reduction of rhizobia nodulation and VAM colonization by misapplication of the herbicides might be a causal factor for decrease in soybean yield.

  • Experiments on the GA production ability by ectomycorrhizal fungus, Pisolithus tinctorius was carried out to investigate specific physiological phenomena of growth increase in host plants by formation of mycorrhizae, The culture extract of P. tinctorius was purified by solvent fractionation, sephadex LH-20 chromatography, silica gel partition chromatography and TLC, successively. GA activities in the purified GA fractions were monitored by micro-drop bioassay using dwarf rice seedlings, 'Tan-ginbozu'. $30{\sim}60%$ EtOAc election fractions of silica gel pardon chromatography and the zone of Rf $0.1{\sim}0.4,\;0.6{\sim}0.8$ of TLC exhibited the GA-like activities. The GA activities were increased with the more treated amount of culture extracts. This activity in 100ml of culture solution was equivalent to 0.1ng of $GA_3$.

  • Red alder(Alnus rubra Bong.) seedlings inoculated with Frankia only or both Frankia and spores of Alpova diplophloeus(Zeller & Dodge) Trappe & Smith were grown in a greenhouse for ten weeks. The ten-week-old seedlings were fertilized with six nitrogen(N) and phosphorus(P) fertility regimes (no fertilization, 10mM $NH_4NO_3$, 50mM $NH_4NO_3$, 5mM $KH_2PO_4$, 10mM $NH_4NO_3+5mM$ $KH_2PO_4$, and 50mM $NH_4NO_3+5mM$ $KH_2PO_4$) three times a week for ten weeks. The higher N-fertilization significantly increased mycorrhiza formation by greenhouse contaminant mycorrhizal fungi, but decreased N-fixation and P concentration in nodule tissues. P-fertilization significantly increased nodule and shoot dry weight, and P concentration in plant tissues. When N was highly fertilized, however, the P-fertilization effect disappeared in nodule P concentration but doubled in leaf P concentration. A. diplophloeus inoculation significantly increased diameter growth and $CO_2$ exchange rate, but decreased leaf dry weight. Our results suggest that the higher N- or P-fertilization affect nitrogenase activity and mycorrhizal development but the effects are changed by their interactions.

Arbuscular mycorrhizal spores found from the soils of the leguminous plants in Korea (두과식물에서 발견된 내생균근 포자들)

  • Sixty five soil specimens were collected from the nineteen leguminous plant roots throughout 9 sites in four Provinces from Augast 12 to November 21, 1991. VA-mycorrhizal spores isolated from the collected soils were identified and classified into four genera 21 species. However, six spores were newly found and described here: Acaulospora elegans, A. undulata, Glomus laminated spores of. macrocarpus var. macrocarpus, Gl. WUMI%3, Gl. scintillans, Scutellospora verrucosa. Species of three spores that belong to 3 genera; Acaulospora sp., Glomus sp., Scutellospora sp. were unidentified.
