Accumulation of oxyresveratrol in Ramulus mori upon postharvest storage (숙성에 의한 뽕나무 상지 내 옥시레스베라트롤 축적)

  • Oxyresveratrol (trans-2, 3', 4, 5'-tetrahydroxystilbene), found in many plants including grape, peanut and mulberry, is a phytoalexin, an antimicrobial and antioxidative substance that rapidly accumulates in areas infected by the pathogen. We examined the accumulation of oxyresveratrol in nine Morus alba L. cultivars with respect to storage time and temperature postharvest and infection with GRAS microorganisms. Among the nine cultivars, the Suwon cultivar showed the highest oxyresveratrol content (9.6-fold increase) postharvest, when stored at $30^{\circ}C$ for 7 days. The optimal temperature and postharvest storage time for oxyresveratrol accumulation was $30^{\circ}C$ and 6 days. When Ramulus mori was infected with five microorganisms, the accumulation of oxyresveratrol increased over 4-fold in response to B. coagulans infection. These results suggest that oxyresveratrol accumulation is influenced by storage temperature, storage time, Ramulus mori cultivars, and microbial attack. Therefore, postharvest storage for an appropriate time period at a suitable temperature might be a useful way to industrially produce Ramulus mori cultivars with high oxyresveratrol content.

Production and physicochemical properties of black bean yogurt made with lactic acid bacteria isolated from vinegar and kimchi (식초와 김치 유래 젖산균으로 제조한 서리태 발효우유 제조 및 물리화학적 품질 특성 연구)

  • This study evaluated the physicochemical properties of a novel black bean milk yogurt made by lactic acid producing bacteria isolated from vinegar (Bacillus acidiproducens, BA) and kimchi (Lactobacillus plantarum, LP). Commercially available Lactobacilli mixture for yogurt (YF) were regarded as control bacteria. To investigate optimal conditions to make black bean yogurt, different types of bacteria (BA and LP) and dilution ratios of black bean milk and water (1:1, 1:1.5, and 1:2) were used. Titratable acidity and pH were affected by these dilution ratios, but not by the different types of Lactobacilli used. Titratable acidity and pH were lowest in the 1:1 diluted sample. The visible cell count, sugar content, and viscosity were significantly higher in the sample containing the black bean milk: water ratio of 1:1 than the rest of the samples. In the sensory test, black bean yogurt made with BA showed a higher overall acceptability score, similar to yogurt made with YF. In conclusion, black bean yogurt made by fermentation with BA and with 1:1 black bean milk:water dilution ratio was better than ones made with LP and at other dilution ratios.

Optimization of sterilization conditions for the production of retorted steamed egg using response surface methodology (반응표면분석을 이용한 레토르트 계란찜의 살균조건 최적화)

  • The purpose of this study was to determine the optimum sterilization conditions for the production of retorted steamed egg using response surface methodology. Sterilization processes for eighteen conditions using varying sterilization temperature ($X_1$), time ($X_2$), and method ($X_3$) as the independent variables were carried out through a $3^2{\times}2$ experimental factorial design. Quality evaluations after sterilization included measurements of $F_0$ value ($Y_1$), peak stress ($Y_2$), pH ($Y_3$), color value ($Y_{4-6}$), and organoleptic test [preference for appearance ($Y_7$), overall acceptability ($Y_8$), and preference for texture ($Y_9$) and egg taste ($Y_{10}$)]. Dependent variables ($Y_{1-10}$) of eighteen conditions were more affected by temperature and time than by the sterilization method. Eight factors were selected among the dependent variables as significant factors related to the quality of the steamed egg. Finally, by establishing an optimum range of each dependent variable and contour analysis, the optimum sterilization conditions for the production of steamed egg were determined to be $120^{\circ}C$ for 25 min using a 2-step sterilization process.

Determination of Shelf-life of Black Mini Tomato Based on Maturity and Storage Temperature (흑색 방울토마토의 숙기 및 저장온도에 따른 상품성 유지기간 구명)

  • Black mini tomato 'Hei-G' fruits were harvested at different stages of maturity (immature-mature green and mature-black red) and stored at different temperatures (8, 12, and $20^{\circ}C$) to investigate the quality and lycopene content during storage. Weight loss increased dramatically at higher temperature for both harvesting stages without significant differences. Firmness of immature fruits decreased below the initial level of mature fruit (8.1N) after 5, 8, and 19 days storage, when they were stored at 20, 12, and $8^{\circ}C$, respectively. Soluble solid contents of mature fruit increased at initial storage, and were higher as compared to immature fruits before deterioration at each storage temperature. Decrease in titratable acid of mature fruits depended on storage time and temperature. However, titratable acid of immature fruits showed little change during storage, and so it did not affect flavor. Hunter a value changed greatly in immature fruit stored at high temperature. Unlike ripe tomatoes, there was no significant difference in black tomato Hunter b values of immature and mature fruit at initial and 12 days storage. However, immature fruits stored at $8^{\circ}C$ did not reach full maturity and color development and ripening. High storage temperature increased lycopene production while low storage temperature blocked lycopene development. Shelf life of the immature fruits, which was evaluated by elapsed days to conventional mature stage, was 12 and 15 days when they were stored at 20 and $12^{\circ}C$, respectively. The optimum storage temperature to maintain the quality and lycopene content of mature fruits was $12^{\circ}C$. Moreover, the shelf life of mature fruits stored at $20^{\circ}C$ could reach up to 5 days.

Vitamin B5 and B6 Contents in Fresh Materials and after Parboiling Treatment in Harvested Vegetables (채소류의 수확 후 원재료 및 데침 처리에 의한 비타민 B5 및 B6 함량 변화)

  • This study was aimed to determine the changes in vitamin $B_5$ and $B_6$ contents compared to fresh materials after parboiling treatment of the main vegetables consumed in Korea. The specificity of accuracy and precision for vitamin $B_5$ and $B_6$ analysis method were validated using high-performance liquid chromatography (HPLC). The recovery rate of standard reference material (SRM) was excellent, and all analysis was under the control line based on the quality control chart for vitamin $B_5$ and $B_6$. The Z-score for vitamin $B_6$ in food analysis performance assessment scheme (FAPAS) proficiency test was -1.0, confirming reliability of analytical performance. The vitamin $B_5$ and $B_6$ contents in a total of 39 fresh materials and parboiled samples were analyzed. The contents of vitamin $B_5$ and $B_6$ ranged from 0.000 to 2.462 and from 0.000 to $0.127mg{\cdot}100g^{-1}$, respectively. The highest contents of vitamin $B_5$ and $B_6$ were $2.462mg{\cdot}100g^{-1}$ in fresh fatsia shoots (stem vegetables), and $0.127mg{\cdot}100g^{-1}$ in fresh spinach beet (leafy vegetables), respectively. Moreover, the vitamin $B_5$ and $B_6$ contents for parboiling treatment in most vegetables were reduced or not detected. In particular, the contents of vitamin $B_5$ in parboiled fatsia shoots and vitamin $B_6$ in parboiled yellow potato and spinach beet were decreased 20- and 4-fold compared with fresh material, respectively. These results can be used as important basic data for utilization and processing of various vegetable crops, information for dietary life, management of school meals, and national health for Koreans.

Genotype Frequency of Human Papillomavirus Determined by PCR and DNA Sequencing in Korean Women

  • Human Papilloma viruses (HPVs) are etiological agents for cervical cancer and are classified into low- and high-risk categories. The aim of this study was to determine the frequency of the HPV genotype in the HPV screening test of Korean women using PCR-direct sequencing. Consensus primers of L1 legion were used for the amplification of HPV DNA and the PCR products (450 bps) obtained were analyzed by automatic sequencing. Sequences were compared with those in GenBank by using the BLAST program. Cervical swab samples of 3,978 women (20-73 years) were tested and the average age was 37.6 years. In this study, 1,174 samples were HPV positive out of 3,978 cervical swab samples screened (29.5%) and 136 samples (11.6%) showed a double infection. A total of 1,310 HPV genotypes were analyzed. The HPV positive rate was the lowest in the 20 years group (69.5%) and most of the samples of the > 60 years group were found HPV positive. Among thirty seven different HPV types identified by sequencing, 21 were HPV high risk types and 16 HPV low risk types were 69.8% (914/1,310) and 26.0% (340/1,310), respectively. In HPV high-risk types, 16 (13.21%), was the most frequently found. HPV 53 (9.62%) and 58 (9.24%) were also frequently found. This group was followed by HPV types 70 (5.50%), 33 (4.73%), 66 (4.20%), 18 (4.05%), 52 (4.05%), 31 (3.97%) and 56 (3.51%) in descending order of frequency. Among HPV low-risk types, 62 (4.20%), 6 (3.59%), 81 (3.59%), 84 (3.51%), and 11 (2.6%) were frequently found. In conclusion, PCR-direct sequencing could be used for quick and reliable typing of known and novel HPVs from clinical specimens. This data could be useful for epidemiological study of HPV and it also allows type-specific follow-up of women who have been treated for cervical intraepithelial neoplasia.

Functional Properties of Medicinal Plant Extracts (한방식재료 추출물의 기능성)

  • The purpose of this study was to investigate the functional properties of medicinal plant extracts. Four kinds of medicinal plants, Dioscorea batatas(DB), Armeniacae Semen(AS) Crataegus pinnatifida Bunge(CP) and Ponciri Fractus(PF), were extracted with water and 70% ethanol and the extracts were tested for their electron donating ability(EDA), nitrite scavenging ability(NSA) and inhibitory effects on cancer cells(MDA cell and A549 cell)growth. EDA at 100-1,000 ppm of water extract ranged from 3% to 14%, 14% to 36%, 29% to 72% and 14% to 43%, and that of ethanol extract ranged from 9% to 62%, 27% to 59%, 33% to 89% and 14% to 44%, in DB, AS, CP and PF, respectively. NSA of extracts measured at various pH(1.2, 3.0, 4.2, 6.0) showed the highest ability in all extracts at pH 1.2 and decreased with increasing pH. The highest NSA of water extracts of 1,000 ppm at pH 1.2 was 6%, 31%, 55% and 44% and that of ethanol extract was15%, 32%, 69% and 52%, in DB, AS, CP and PF, respectively Inhibition ratio of water and ethanol extracts on MDA cell growth was 24% and 17%, 51% and 93%, 46% and 69%, and 48% and 47%, while that on A549 cell was 18% and 9%, 6% and 3%, 7% and 3%, and 43% and 11%, at 1,000 ppm, in DB, AS, CP and PF, respectively.

A Study on the Trace Metals in Potatoes and Root Vegetables (서류 및 구근류의 중금속 함량)

  • This study was conducted to estimate the contents of 6 trace metals in potatoes and root vegetables purchased in wholesale market in the Seoul region, specifically, Cu. As, Zn, Pb, Cd and Hg. The minimum and maximum results obtained for each metal are summarized as follows for potatoes; Cu : 0.094∼0.344, As : N.D∼0.070, Zn : 0.247∼1.547, Pb : N.D∼0.268, Cd : N.D∼0.005, Hg : 0.001∼0.027, and for root vegetables; Cu : 0.021∼0.510, As : 0.004∼0.045, Zn : 0.143∼3.137, Pb : N.D∼0.372, Cd : N.D∼0.009, Hg : 0.001∼0.195. The daily intakes of trace metals from potatoes and root vegetables were estimated on the basis of the daily intake of these foods by Koreans, which were 0.073∼22.253 and 0.567∼156.219$\mu\textrm{g}$ from potatoes and root vegetables, respectively. The weekly intakes of trace metals were 0.009∼2.596 and 0.066∼18.224$\mu\textrm{g}$ for potatoes and root vegetables, respectively. The weekly intakes of trace metals for potatoes and root vegetables were very much lower than the PTWI(provisional tolerable weekly intake), enough to be within a safe range.

Effects of Addition Baekbokryung(White Poria cocos Wolf) Powder on the Quality Characteristics of Sulgidduk (백복령 가루 첨가가 설기떡의 품질 특성에 미치는 영향)

  • In an attempt to improve the taste and storage characteristics of Sulgidduk, a Korean traditional rice cake product, baekbokryung(White Poria cocos Wolf) powder was supplemented as one of the ingredients and Product Quality waw assessed. The baekbokryung Powder was added in rations of 0, 5, 7, 10, and $15\%$(w/w) per rice powder during the raw material preparations. The sensory, objective quality, and microbiological characteristics of the products were examined at 4-hr intervals during storage for 36 hours at $20^{\circ}C$ The sensory evaluation was conducted based on the acceptability and intensity characteristics of the product. In the acceptability test, a $5\%$ treatment wae renerally ranked with the highest score, among all the treatments while the preference differed according to ages of the panels ; namely, those in their 20's to 30's liked a $5\%$ supplementation while those in their 40's to 60's Preferred a $7\%$ one. The addition of baekbokryung Powder proportionally decreased the moisture content of the products. The colorimetric redness(a) and yellowness(b) of the Products increased with increasing added baekbokryung Powder. The redness values began to decrease slowly from the beginning while the rates were accelerated after 8 to 12 hours of the storage. In the textural characteristics, hardness, gumminess and adhesiveness values increased with increasing added baekbokryung powder. The rapid increase in adhesiveness during the initial 4 hour of storage and the following stabilization was noted. Cohesiveness and springiness values tended to decrease gradually with increasing added baekbokryung Powder. Except for the control sample, no significant differences in the total microbial counts among the treatments were noted. After 20 hours of storage, the control sample exhibited a rapid increase in total microbial count, while the Proliferations of microorganisms were rather suppressed in baekbokryung Powder treatments according to the added amounts The results of the study support the benefits of baekbokryung powder supplementation in Sulgidduk in the aspects of taste and functionality. The degree of further increase of baekbokryung Powder in Sulgidduk without causing the adverse qualify effects remains for future study.

Effect of Jehotang Extract on the Growth of Intestinal Bacteria and Immunostimulation (제호탕의 장내 세균 및 면역 활성에 미치는 연구)

  • Water extracts of Jehotang were evaluated for their growth-promoting effects on Bifidobacterium longum, Lactobacillus sp., L. acidophilus, and Clostridium perfringens. Addition of Jehotang water extract to modified EG media at 0.1 mg/mL increased growths of B. longum, Lactobacillus sp., and L. acidophilus, with 1.8-fold increase in growth of L. acidophilus compared to that of control. Studies on these strains by agar diffusion method showed Lactobacillus sp. and L. acidophilus were activated by addition of Jehotang extract at 10 mg/disc. Proliferation responses of mice splenocytes and Peyer's patch cells to ConA by LPS-stimulation at 500 mg/kg B.W./day Jehotang extract were investigated in vitro. Upon treatment of 1 mg/mL Jehotang water extract to mice, proliferations of splenocytes and Peyer's patch cells increased 1.4- and 1.6-fold compared to control, respectively. In mice administered Jehotang extract, production of intestinal secretory IgA (sIgA) increased 2.4-fold compared to control. These results indicate water extract of Jehotang stimulated intestinal immune system of mice. In mice treated with Jehotang extract, production of lymphocytes was 4% lower, whereas those of granulocytes and platelets were 4% and slightly higher than control, respectively.
