  • 온주밀감은 제주도에서 재배면적 비율이 아주 높아 매년 많은 량이 생산되며 많은 과피 들이 폐기 처리되고 있다. 본 연구는 제주도의 상징이며 변색방법이 간편하고 폐자원을 활용할 수 있는 천연염재로서 온주밀감 과피에서 염료를 추출하여 염 액의 농도, 염색온도, 염색시간, 염색횟수를 변인으로하여 면, 모, 견, 나일론직물에 염색 한 후 표면색과 염색성을 검토하였다. (중략)

Effects of Night Temperature at Veraison on Berry Skin Coloration of 'Kyoho' Grapevines (포도 '거봉' 품종의 변색기 야간 온도 처리가 과피 착색에 미치는 영향)

  • We analyzed berry skin coloration, anthocyanin accumulation, and plant hormone contents in berry skins to determine the effect of night temperature at veraison on berry skin coloration in 'Kyoho' grapevines (Vitis labruscana L.). Vines were grown under 21, 24, and 27℃ at night for 20 days at veraison, from 40 to 60 days after full bloom (DAFB). Berry skin coloration of 'Kyoho' grapes was more suppressed in 27℃ treated vines, followed by that in 24℃ treated vines, than that in 21℃ treated vines. Cluster and berry weight and soluble solids content was lower in 24 and 27℃ treated vines than in 21℃ treated vines. Anthocyanin started to accumulate from 60 DAFB in berry skin of 21℃ treated vines, and malvidin and total anthocyanin content increased until 100 DAFB. The total and most of the individual anthocyanins decreased in 24 and 27℃ treated vines; however, peonidin did not decrease in 24℃ treated vines compared to that in 21℃ treated vines. Abscisic acid (ABA) peaked at veraison in berry skins of 21℃ treated vines and decreased thereafter until 100 DAFB. The increase in ABA content was inhibited in berry skins of 24 and 27℃ treated vines. Gibberellin (GA) content in berry skins decreased rapidly at veraison, with the decrease being slower under 27℃ than under 21℃. ABA/GA in berry skins of 21℃ treated vines peaked at 60 DAFB and decreased thereafter until 100 DAFB. However, ABA/GA decreased in berry skins of 24 and 27℃ treated vines, with reduced anthocyanin accumulation. Therefore, high night temperature (above 24℃) at veraison suppressed the berry skin coloration of 'Kyoho' grapes with changes in anthocyanin contents and composition due to the decrease in ABA/GA ratio and fruit soluble solids contents.

Effects of Bagging Periods on Pericarp Characteristics and Berry Cracking in 'Kyoho' Grape (Vitis sp.) (괘대시기가 '거봉' 포도의 과피 특성과 열과에 미치는 영향)

  • The berry growth and pericarp characteristics were characterized to confirm the effects of bagging periods on berry cracking during berry development in 'Kyoho' grape. The berry weight was the highest at 13.4 g in late period of bagging treated at 7 to 9 weeks after full bloom (WAFB) as compared with the lowest of 12.3 g in total period bagging. The berry cracking rate under critical turgor pressure in the non-bagging control was 53.3%, while those of bagging treatments were decreased in the order of 42.7%, 37.3%, 33.3%, and 18.7% in bagged during 3 to 9, 3 to 5, 5 to 7, and 7 to 9 WAFB, respectively. In the results of observation on histological characteristics of pericarp, berry lenticels of whole bagging treatments had smaller and normal shape compared with non-bagging control treatment. Especially on the pericarp of late period bagged during 7 to 9 WAFB, suberization around stomata and micro-cracking were not observed and structural strength of pericarp was increased with thicker sub-epidermal layer and cell wall. Therefore, the results indicate that bagging treatment for two weeks just before the veraison when the day length and daylight is relatively longer and stronger can effectively reduce berry cracking by strengthening structure of pericarp in 'Kyoho' grape.

Major Metabolites Involved in Skin Blackening of 'Niitaka' Pear Stored under Cold Temperature (신고 배 저온 저장 중 발생하는 과피 흑변에 관여하는 주요 대사체)

  • Oriental pear (Pyrus communis L. cv 'Niitaka') was stored at $0^{\circ}C$ for 5 months and major metabolites involved in blackening of the peel were analyzed by untargeted GC-MS and targeted HPLC methods. In this study, peels of sound and skin-blackened pears were analyzed and compared. Skin-blackened fruit was clearly characterized by a distinctive pattern in changes which included a decrease of malic acid, succinic acid, and ascorbic acid, while an increase of fumaric acid, threonine, and gluconic acid, which indicated both reduced metabolic activity and anti-oxidative capacity of the cells. Chlorogenic acid was a major phenolic compound and the peel of sound fruit showed high levels of free phenolic compounds compared than the peel of skin-blackened fruit which are believed to be related to oxidation of phenolics in skin-blackened tissue. The changes or profiling of major metabolites by targeted or untargeted analysis method could become a useful tool for understanding physiology, disorder mechanism, and identifying metabolic networks connecting primary and secondary metabolism in postharvest research.

단감의 저장 중 갈변발생에 영향을 미치는 인자에 관한 연구

  • 저장 중 단감은 과피 및 과육의 변색 발생으로 인한 품질저하가 심각하게 발생하고 있는데, 아직까지 이를 방지하기 위한 원인 규명은 매우 미흡한 실정이다. 이에 본 연구에서는 단감의 재배지, 수확시기, 크기, 예조기간, 저장온도, 저장기간, 포장재의 두께 등을 달리하여 저장하면서 처리조건에 따른 단감 과육의 갈변 발생 관련성을 조사하였다. 단감의 변색발생 증상은 저장 1개월 이후부터 일부 나타나기 시작하여 저장기간이 경과할수록 심하였다. 단감의 갈변 발생률 및 갈변 정도를 실험 처리구별로 살펴보면 과수원, 수확시기, 예조기간, 저장온도, 포장재의 두께, 포장 내 가스조성에 따라 차이를 보였는데 단감의 수확시기가 늦을수록, 수확후 예조기간이 짧을수록, 저장을 위해 사용한 포장필름의 두께가 얇을수록 갈변발생정도가 심하였다. 또한 단감의 과수원에 따라서도 갈변의 발생률이 차이를 보여 동일지역이더라도 재배조건에 의하여서도 영향을 받는 것으로 나타났다. 이와 같은 결과를 종합하여 보면 단감 과육의 갈변발생은 수확 전 및 수확 후 인자들의 단독적인 영향이기보다는 복합적인 영향에 의한 것으로 추정되었다.

Comparison of Fruit Characteristics and Quality in 'Heukgoosul' Grapes Set on Primary and Secondary Shoot (포도 '흑구슬'에서 1차지 및 2차지 착생 과실의 특성 및 품질 비교)

  • This experiment was performed in order to define the characteristic and quality of the fruits set on primary and secondary shoots in 'Heukgoosul' grape. The numbers of flower clusters per shoot were 1.1 and 0.6 in primary shoots and secondary shoots, respectively. The fruits of primary shoot showed 12.1 g of higher berry weight than 8.9 g of the fruits set on secondary shoots. Fructose and glucose contents were significantly higher in the fruits of secondary shoots during veraison, but the levels gradually decreased, reaching a similar level with those of primary shoot at the harvest stage. The acid content decreased rapidly from day 50 after full bloom in both primary and secondary shoots, but the rate of decrease slowed down from day 70 after full bloom in secondary shoots. Berries of secondary shoots showed rapid increase of skin coloration during veraison and reached a high degree of coloration within a short period when compared with those of primary shoots.

Skin Coloration and Endogenous Hormonal Changes of 'Kyoho' Grape by High Temperature at Veraison (변색기 고온에 의한 포도 '거봉'의 과피 착색 및 내생 호르몬 변화)

  • We analyzed the anthocyanin accumulation, abscisic acid (ABA), gibberellic acid (GA) contents and metabolic genes expression in berry skins under high temperature (High T) at veraison, in order to investigate the cause of bad coloration of 'Kyoho' grape due to High T in summer season. The coloration of 'Kyoho' grapes was stopped by High T for 10 days from veraison, and the fruit quality was not affected except skin color. Total anthocyanin of skins was decreased by High T treatment and malvidin and peonidin were decreased compared to control. In berry skins, ABA content did not decrease by High T treatment, but it was rather higher than that of control. GA content was increased about two times compared to the control after 10 days of High T treatment, which caused decreased ratio of ABA/GA. Analysis of expression of anthocyanin biosynthetic genes showed that the early biosynthetic genes were not affected by High T and the expression of UFGT was decreased by temperature treatment. ABA biosynthetic gene expressions were not affected by High T and the expression of GA20ox1 and GA2ox1/2, which are known to regulate the biosynthesis and inactivation of GA, were increased and decreased by High T, respectively. Therefore, the bad coloration of 'Kyoho' grapes under the High T at veraison was due to inhibition of anthocyanin biosynthesis of skin, and it was suggested that the anthocyanin biosynthesis was controlled by the ratio of ABA and GA rather than ABA content.

Effects of Calcium Concentrations of Coating Bag on Pericarp Structure and Berry Cracking in 'Kyoho' Grape (Vitis sp.) (괘대봉지의 칼슘 코팅농도가 '거봉'포도의 과피구조와 열과에 미치는 영향)

  • The effects of calcium concentrations of coating bag treatment to reduce berry cracking were investigated through the changes of pericarp structure and berry cracking rate in 'Kyoho' grape. The soluble solids and anthocyanin contents in harvested grapes were highest at $18.1^{\circ}Brix$ and $2.56{\mu}g{\cdot}cm^{-2}$ in non bagging group compared with those of calcium coating bag treatments. The firmness of pericarp was lowest in non bagging group ($1.18kg{\cdot}5mm^{-1}{\O}$) compared with bagging treatments (1.23, 1.24, 1.27, $1.35kg{\cdot}5mm^{-1}{\O}$) which increased effectively in proportion to calcium concentration. As a result of histological observation of the fruit skin, the bagging with higher calcium concentration developed thicker epidermal and sub-epidermal layer of cell wall than that of non bagging. Moreover, the strengthened berry skin of calcium treatments effectively decreased berry cracking rate under critical turgor pressure. However, the 9% calcium coating bag treatment which was the most effective for cracking reduction seriously decreased marketability of harvested grape with white color staining on berry skin caused by eluted calcium from the coated paper bag. Based on our results, we recommend that 6% calcium coating bag be available for berry cracking reduction and higher quality production.

Effect of High $CO_2$ Pre-treatment on Quality of 'Hikawa Hakuho' Peach Fruits (고 이산화탄소 전처리가 '일천백봉' 복숭아의 품질 변화에 미치는 영향)

  • This study was conducted to evaluate the efficacy of high a)z pre-treatment (60, $100\%$ for 3, 6, 12 hr) on quality of 'Hikawa Hakuho' peach fruits. High a)z treatment inhibited the respiration and ethylene production, especially with long tenn treatment for more than 6 hr. Discoloration of peel, softening and decay were reduced by high $CO_2$ treatment $CO_2$ concentration of $60\~100\%$ did not affect soluble solids content and titratable acidity of fruits. $CO_2$ treated fruits, however, showed higher sweetness score compared with control. As the $CO_2$ concentration and treatment duration increased from 60 to $100\%$ and from 3 to 12 hr, respectively, the efficacy on quality increased. Fruits treated with $100\%\;CO_2$ for 3 hr and $60\%\;CO_2$ for 12 hr, however, showed very low flavor score due to the residual $CO_2$ in flesh. High $CO_2$ injury was not noted at any $CO_2$ levels tested in this study. Above results suggest that $60\%\;CO_2$ treatment for 6 hr resulted in as good control of respiration, ethylene production, decay, discoloration and softening without off-order.

Expression of Genes Affecting Skin Coloration and Sugar Accumulation in 'Hongro' Apple Fruits at Ripening Stages in High Temperatures (고온에 의한 변색단계별 '홍로' 사과의 착색 및 당 축적 관련 유전자 발현 분석)

  • High temperature is one of the important environmental factors limiting cultivation of apple (Malus domestica Borkh). The expression of genes related with anthocyanin synthesis and sugar accumulation in response to high temperature was studied in the 'Hongro' apple fruits at different developmental stages in different temperature conditions through real-time PCR. Expression of ${\hat{a}}$-amylase (BMY) and polygalacturonase (PG) genes related with sugar synthesis was higher in late ripening stages than in initial ripening stages. Expression of four genes such as phenylalanine ammonia-lyase (PAL), chalcone synthase (CHS), flavanone 3-hydroxylase (F3H), and malate dehydrogenase (MDH), which were related with fruit skin coloration, increased gradually in apple fruits of the middle and late ripening stages. Interestingly, the expressions of all genes were highly inhibited expressed at $30-35^{\circ}C$ compared to $25^{\circ}C$ in all ripening stages. In the further work, investigation of expression levels of various genes could be conducted in the level of transcriptomics in fruits at the middle ripening stages to get meaningful information of ripening metabolism in apple in high temperatures.
