Isolation of Entomopathogenic Fungi for Infection to the Pine Gall Midges, Thecodiplosis japonensis from the Forest Soil in Korea (삼림 토양으로부터 솔잎혹파리 감염 사상균의 분리)

  • To develop a microbial pesticide for the control of pine gall midges. Thecodiplosis japonensis, entomopathogenc fungi were isolated from 233 soil samples in the damaged region of Thecodiplosis japonenesis, and identified with Beauveria spp. 29 strains and Paecilomyces spp. 2 strains. The morphology of entomopathogenic fungi was observed by scanning electron miroscope. In addition, the toxicity of entomopathogenic fungi was observed by scanning electron microscope. In addition, the toxicity of entomopathogenic fungi isolated from soil samples was determined by bioassay against Thecodiplosis japonensis larvae. The result showed that toxicity of relatively pathogenic strains, Beauveria spp. SFB-168-2 was 82.9%, suggesting that Beauveria spp. SFB-168-2 is effective entomopathogenic fungi for the control of pine gall midges.

Identification of Beauveria spp. Isolated from Mulberry Longicorn Beetle (Apriona germari Hope) using Polymerase Chain Reaction (뽕나무 하늘소(Apriona germari Hope)로부터 Beauveria속 사상균의 분리 및 PCR에 의한 동정)

  • To develope a microbial pesticide for the control of mulberry longicorn beetle, Apriona germari, Beauveria spp. were isolated from the infected Apriona germari larvae. The morphology of Beauveria spp. was observed by phase contrast and scanning electron microscope. In addition, the Beauveria spp. isolated from Apriona germari were identified by the random amplification of polymorphic DNA using polymerase chain reaction. The results showed that the Beauveria spp., SFB-1A and SFB-3A, isolated from Apriona germari were identified with B. bassiana and B. brongniartii, respectively, suggesting that the random amplification of polymorphic DNA is effective for the identification of Beauveria spp.

Identification of Metarhizium sp. Isolated from Protaetia brevitarsis seulensis (Kolbe) Using Ribosomal DNA Sequence (흰점박이꽃무지로부터 Metarhizium속 사상균의 분리 및 ribosomal DNA 염기서열에 의한 동정)

  • For the purpose of the protection of beneficial insects from pathogens and the development of control agent against pests, a strain of Metarhizium sp. was isolated from the infected Protaetia brevitarsis seulensis larvae in Korea. Under the scanning electron microscope, the isolate, Metarhizium sp. KMA-1, showed distinct formation of conidia on the palisade-like masse which were comprised of elongate chains and this shape is a typical feature of Metarhizium species. PCR techniques were used to identify the isolate and the primers used were designed on the basis of two kinds of rRNAs sequences, 28S rRNA and internal transcribed spacer(ITS). The specific PCR products from each primer set were amplified and the DNA sequences were determined for the similarity comparison. Sequence alignment of these fragments using GenBank database resulted in the highest homology similarity between the isolate Metarhizium sp. KMA-1 and M. anisopliae. From these results, the isolate Metarhizium sp. KMA-1 in this study was identified as M. anisopliae.

  • Insects are commonly infected by fungal diseases and are mostly susceptible to them. Increasing levels of insecticide resistance has recently become an issue for control programs; thus, research has focused on mosquito control using entomopathogenic fungi, including fungal pathogens such as Beauveria bassiana, Metarhizium anisopliae, and Lagenidium giganteum. Review discusses entomopathogenic fungi related to control programs for mosquito transmitted vector-borne diseases such as dengue, filariasis, malaria, and yellow fever, and how to use entomopathogenic fungi for mosquito control.

First Report on Hirsutella subulata, a Pathogen of Rice Stem Borer, Chilo suppressalis in Korea (국내 미기록 이화명충 병원사상균(Hirsutella subulata)에 관한 보고)

  • Hirsutella subulata (Hyphomycetes) was observed for the first time on diseased larvae of Chilo suppressalis in Korea. This fungus formed slender and slightly attenuated synnemata consisting of closely bound, longitudinal hyphae on the insect larvae. Typical characteristics of this species are ellipsoid or broadly obovoid phialides with slender, needle-like necks and asymmetric conidia. It is very similar to H. barberi but can be distinguished by its asymmetrical conidia.
