Enhancement of Bt-Plus Toxicity by Unidentified Biological Response Modifiers Derived from the Bacterial Culture Broth of Xenornabdus nematiphila (Xenorhabuds nematophila 세균 배양액 유래 미확인 생리활성 물질의 비티플러스 살충력 상승효과)

  • 'Bt-Plus' has been developed by mixing spores of Bacillus thuringiensis (Bt) and culture broth of Xenorhabdus nematophila (Xn). Despite its high toxicity, it has some imitation to broaden its efficacy against diverse insect pest spectrum. This study focuses on enhancement of Bt-Plus toxicity against semi-susceptible insect, Spodoptera exitgua, by addition of Xn metabolites. Two main Xn metabolites, oxindole (OI) and benzylideneacetone (BZA), are known to enhance the Bt insecticidal activities. The addition of OI or BZA significantly increased Bt-Plus pathogenicity. However, when the freeze-dried Xn culture broth was added to Bt-Plus, much less amount was enough to enhance the toxicity compared to the amount of OI or BZA. An HPLC analysis indicated that there were more than 12 unidentifed bacterial metabolites in Xn culture broth. These suggest that there are potent biological response modifiers in Xn metabolites other than OI and BZA.

  • Mycoplasma genitalium has 367 conserved genes and the smallest genome among mono-culturable prokaryotes. Conservative metabolic pathways were examined among M. genitalium and 14 prokaryotes, one hyperthermophilic exosymbiotic archaeon Nanoarchaeum equitans and 13 intracellular eubacteria of plants or insects, with fewer conserved genes than M. genitalium. They have 11 to 71 metabolic pathways, however complete metabolic pathways ranged from 1 to 24. Totally, metabolic pathway hole is very high due to the lack of 45.8% of the enzymes required for the whole metabolic pathways and it could be suggested that the shared metabolic pathway with the host's enzyme would work or the essential substances are host dependent. The number of genes necessary for mass transfer through the cell membrane is also very low, and it may be considered that the simple diffusion or the protein of the host will function in the cell membrane of these prokaryotes. Although the tRNA charging pathway was distributed in all 15 prokaryotes, each has 5-20 tRNA charging genes. This study would give clues to the understanding of the metabolic pathways of intracellular parasitic bacteria of plant and endosymbiotic bacteria of insects, and could provide basic data for prevention of crop damage, development of insect pests and human medicines.

  • The effects of foliar spray of Kinetin (6-Furfurylaminopurine) on the growth of mulberry tree and the metabolic activities in the leaves were measured. The results of this experimented are summarized as follows: 1. Foliar application of Kinetin was effective in increasing of leaf area, the fresh and dry weight of the leaves, the length and diameter of shoots. 2. Chlorophyll content was generally increased in all treated leaves. 3. All the treated leaves were higher in GOT activity than the control leaves but were lower in GPT activities. 4. The activities of the carbohydrate metabolism in the mulberry leaves were higher than that of the control in all treated leaves.

Study on Development of Novel Biopesticides Using Entomopathogenic Bacterial Culture Broth of Xenorhabdus and Photorhabdus (Xenorhabdus 및 Photorhabdus 세균 배양액을 이용한 생물농약 개발에 관한 연구)

  • Two groups of entomopathogenic bacteria, Xenorhabdus and Photorhabdus, are known to suppress insect immune responses by inhibiting eicosanoid biosynthesis. This study used these bacterial culture broths to develop novel biochemical insecticides against the diamondback moth, Plutella xylostella. Though the bacterial culture broths alone showed little insecticidal activity, they significantly enhanced pathogenicity of Bacillus thuringiensis against the fourth instar larvae of P. xylostella. Sterilization of the bacterial culture broth by autoclaving or $0.2\;{\mu}m$ membrane filtering did not influence the synergistic effect on the pathogenicity of B. thuringiensis. Three metablites identified in the culture broth of X. nematophila also showed similar synergistic effects. In field test, both entomopathogenic bacterial culture broth also enhanced the control efficacy of B. thuringiensis against P. xylostella.

  • 벼멸구의 카보후란에 대한 저항성 기작을 구명하기 위해 실내에서 카보후란으로 30세대 도태하여 얻은 저항성계통($LD_{50};\;20.3{\mu}g/g$)과 약제를 12년 동안 처리하지 않은 벼멸구 감수성 계통($LD_{50};\;0.3{\mu}g/g$)을 완충용액과 마쇄하여, 105,000g에서 2시간 원심분리하여 얻은 상등액(에스테라제층)과 침전물(P450-산화효소층)을 효소액으로 하여 $^{14}C$-카보후란을 반응시켜 계통 간 대사물 량의 차이를 조사한 바 저해제(piperonyl butoxide; 산화효소저해제, diethylmalate; 글루타치온 전이효소 저해제, iprobenfos; 에스테라제 저해제)와 보조인자 (NADPH; P-450 산화효소, 글루타치온; 글루타치온전이효소)에 상관없이 카보후란의 대사물과 그 양이 계통간 차이가 없었다. 이상의 결과로부터 저항성 벼멸구에서 일반적으로 곤충에서 생화학적 저항성 기구로 잘 알려진 가수 분해 효소의 일종인 에스테라제와 p-450 산화효소, 글루타치온 전이효소의 활성 증가가 저항성 발달에 관여하지 않음을 알 수 있었다.

Effects of Grasshopper (Oxya chinensis sinuosa Mistshenko) Powder and Aerobic Exercise on Energy Metabolism in ICR Mice (벼메뚜기(Oxya chinensis sinuosa Mistshenko) 분말 섭취와 유산소성 운동훈련에 의한 마우스의 에너지 대사 변화)

  • The purpose of this study was to analyze the effect of grasshopper (Oxya chinensis sinuosa) powder ingestion with/without aerobic exercise (treadmill running) on energy metabolism. To achieve this purpose, 28 Institute of Cancer Research (ICR) mice were divided into four groups: normal diet control group (CON), a normal diet with exercise control group (COEX), a grasshopper powder-supplemented diet group (GH), and a grasshopper powder-supplemented diet with exercise group (GHEX). Duration of the powder ingestion and aerobic exercise training were 6 weeks. Body weight gain ratio was not significant. Fat mass significantly decreased in GH and GHEX. There were no changes in blood glutamic oxaloacetic transaminase and glutamic pyruvic transaminase levels between groups. Glucose transporter type 2 and glucose transporter type 4 protein levels were not significantly different between groups. Fibronectin type III domain-containing protein 5 level was the highest in GHEX. AMP-activated protein kinase level significantly increased in GHEX compared to the levels in the other groups. Glycogen synthase kinase 3 beta protein level was reduced in GHEX compared to that in CON. These results suggest that grasshopper powder ingestion and endurance exercise training influence energy metabolism.

  • Like vertebrates, insects synthesize various eicosanoids after the committed catalytic step of phospholipase A2 (PLA2). However, the subsequent biosynthetic steps exhibit some deviation from those of vertebrates. Due to little composition of arachidonic acid in insect phospholipids, PLA2 releases linoleic acid, which is another polyunsaturated fatty acid and relatively rich in insect phospholipids, to synthesize arachidonic acid via chain extension and desaturation. Resulting arachidonic acid is then oxygenated into a prostaglandin (PG), PGH2, by a specific peroxidase called peroxynectin, but not by cyclooxygenase. PGH2 is then isomerized to various PGs such as PGA2, PGD2, PGE2, PGI2, and a thromboxane (TXB2). All four epoxyeicosatrienoic acids such as 5,6-EET, 8,9-EET, 11,12-EET, and 14,15-EET are also synthesized from arachidonic acid by oxygenation of vertebrate types of monooxygenases. However, the other type of eicosanoids called leukotrienes are found in insect tissues but their synthetic pathway is unclear. Eicosanoids mediate various insect physiological processes such as metabolism, excretion, immunity, and reproduction. Thus, identification of novel compounds interrupting eicosanoid biosynthesis would be a novel approach to develop insecticides. This review focuses on PGs and their immune mediation.

Inheritance of Fenvalerate Resistance in the Diamondback Moth, Plutella xylostella Linne(Lepidoptera: Yponomeutidae) (배추좀나방의 Fenvalerate에 대한 저항성 유전)

  • The mode of inheritance of resistance to fenvalerate in the diamondback moth (Plutella xylostella L.) was studied using insecticide susceptibility and mixed function oxidase tests. Tehre were no differences in the concentration-mortality relationships between $F_1$ progenies $(S_{female}\timesR_{male},\;R_{female}\timesS_{male})$ obtained from reciprocal crosses with the susceptible and fenvalerate-selected strains(R), indicating the absence of sex-linked inheritance. Degree of dominance of the $F_1$ progenies $(S_{female}\timesR_{male},\;R_{female}\timesS_{male})$ were -0.50 and -0.46, respectively, in the insecticide susceptibility test and -0.85 and -0.81, respectively, in the mixed function oxidase test. These results suggest that inheritance of fenvalerate resistance is controlled by and incompletely recessive autosomal gene.

Complete genome sequencing of Pseudomonas fluorescens NBC275, a biocontrol agent against fungal pathogens of plants and insects (식물 및 곤충의 곰팡이 병원균에 항균력을 가진 Pseudomonas fluorescens NBC275 균주의 유전체 염기서열)

  • Pseudomonas fluorescens NBC275 (Pf275) isolated from soil sample collected at riverside of Nakdonggang showed antagonistic activity against fungal pathogens of plants and insects. Here we present complete genome sequence of Pf275. The genome comprises of 6,610,362 bp with GC content of 60.9%, which includes 5,869 predicted protein-coding genes, 16 rRNAs, and 65 tRNAs. Genome analysis revealed gene clusters encoding antimicrobial secondary metabolites such as pyoverdine, 2, 4-diacetylphloroglucinol, and phenazine, which are known to play essential roles in biocontrol of diseases.

  • Glucose, trehalose, and glycogen content were measured in haemolymph and the body during larval development of the oriental corn borer, Ostrinia furnacalis ($Guen\'{e}e$). Glucose content varied in haemolymph, but in the body it gradually decreased at all larval stages. As the larva proceeds growth, the initial high level of trehalose increased further in haemolymph whereas the low level trehalose gradually decreased in the body. Glycogen levels in haemolymph rose gradually during larval growth whereas those in the body increased sharply at the two latter instar larval stages. It is apparent that the existence of a homeostasis in carbohydrate levels between haemolymph ana the body.
