Development of an Efficient Bioassay Method to Evaluate Resistance of Chili Pepper Cultivars to Ralstonia solanacearum (고추 풋마름병에 대한 효율적인 저항성 검정법 확립)

  • Bacterial wilt caused by Ralstonia solanacearum is an important disease in cultivation of chili pepper, causing plant death and significant yield losses. Cultivation of disease-resistant varieties is the most suitable measure to control bacterial wilt of chili pepper. To establish an efficient screening method for resistant chili pepper to R. solanacearum, six resistant or susceptible cultivars to the R. solanacearum were selected and the development of bacterial wilt on the cultivars according to several conditions was investigated. Drenching bacterial suspension into the cut roots using a scalpel was more simple and effective to distinguish resistant and susceptible cultivars than inoculation methods of root-dipping or soil-drenching without wounding. A resistant pepper, 'MC4' to R. solanacearum showed high resistance under the developed conditions which were 21- to 28-day-old pepper inoculated with $1{\times}10^8cfu/ml$ of bacterial suspension. On the other hands, the susceptible cultivars represented high disease severity under the conditions. These results indicated that we developed an efficient method to evaluate resistance of chili pepper cultivars against bacterial wilt. In addition, we successfully evaluated resistance degree of 140 commercial chili pepper cultivars to R. solanacearum using the developed method.

Occurrence and Biovar Classification of Bacterial Wilt Caused by Ralstonia solanacearum in Eggplant (Solanum melongena) (가지의 Ralstonia solanacearum에 의한 풋마름병 발생과 생리형의 분화)

  • Batcterial wilt caused by Ralstonia solanacearum is one of important and widespread diseases worldwide as well as in Korea. Bacterial wilt disease caused by R. solanacearum has been reported mainly in solanaceous crops including eggplant (Solanum melongena), tomato (Solanum lycopersicum), potato (S. tuberosum), and pepper (Capsicum annuum). A total of 48 strains of R. solanacearum from eggplant were collected during 2005 and 2006. They were confirmed as R. solanacearum by PCR amplification with primer pair flipcF/flipcR resulting in production of 470-bp DNA fragment. The 15 isolates exhibited pathogenicity on eggplant and tomato, but less virulent on pepper than other species. The biovar of collected isolates, which have been reported of five types worldwide, were classified as biovars 3 and 4 by physiological test. Biovar 4 was the dormant type without pathogenicity on eggplant rootstock, whereas biovar 3 had pathogenicity on eggplant rootstocks that is resistant to R. solanacearum, indicating necessity of breeding new rootstock with resistance to R. solanacearum biovar 3

Breeding Lines with Multiple Resistance to both Bacterial Writ and Phytophthora Blight in Pepper(Capsicum annuum L.) (고추 풋마름병.역병 복합 저항성 계통육성)

  • In an effort to breed lines resistant to both bacterial wilt and Phytophthora blight, F5 and BC1F4, and F6 and BC1F5 generations of the crosses between the lines previously bred for resistance to Phytophthora blight ((16-2-2-3-2 = Phytophthora-resistant 'Kalmi', 19-1-3-7-1-1, 19-2-4-5-3-2 = Phytophthora-resistant 'Subi', Kim et al., 1996) and sources of resistance to bacterial wilt (KC350 = MC-4 or KC353 = PBC631, Kim et al., 1998) were evaluated for resistance to both bacterial wilt and to Phytophthora blight in 1999 and in 2000, respectively. Plants which were highly resistant to both bacterial wilt and Phytophthora blight were selected for development of the next generations.

Development of a Forecasting Model for Bacterial Wilt in Hot Pepper (고추 풋마름병 예찰 모형 개발)

  • A population density model for bacterial wilt, which is caused by Ralstonia solanacearum, in hot pepper was developed to estimate the primary infection date after overwintering in the field. We developed the model mechansitically to predict reproduction of the pathogen and pathogensis on seedlings of the host. The model estimates the pathogen's populations both in the soil and in the host. In order to quantify environmental infection factors, various temperatures and initial population densities were determined for wilt symptoms on the seedlings of hot pepper in a chamber. Once, the pathogens living in soil multiply up to 400 cells/g of soil, they can infect successfully in the host. Primary infection in a host was supposed to be started when the population of the pathogen were over $10^9$ cells/g of root tissue. The estimated primary infection dates of bacterial wilt in 2011 in Korea were mostly mid-July or late-July which were 10-15 days earlier than those in 2010. Two kinds of meterological data, synoptic observation and field measurements from paddy field and orchard in Kyunggi, were operated the model for comparing the result dates. About 1-3 days were earlier from field data than from synoptic observation.

Physiological, Biochemical and Genetic Characteristics of Ralstonia solanacearum Strains Isolated from Pepper Plants in Korea (고추에서 분리된 Ralstonia solanacearum 계통의 생리, 생화학 및 유전적 특성)

  • Totally sixty three bacteria were isolated from lower stems showing symptoms of bacterial wilt on pepper plants in 14 counties of 7 provinces, Korea. The isolates showed strong pathogenicity on red pepper (cv. Daewang) and tomato (cv. Seogwang) seedlings. All virulent bacteria were identified as Ralstonia solanacearum based on colony types, physiological and biochemical tests and polymerase chain reaction (PCR). All R. solanacearum isolates from peppers were race 1. The bacterial isolates consisted of biovar 3 (27%) and biovar 4 (73%). Based on polymorphic PCR bands generated by repetitive sequence (rep-PCR), the 63 R. solanacearum isolates were divided into 12 groups at 70% similarity level. These results will be used as basic materials for resistant breeding program and efficient control against bacterial wilt disease of pepper.

  • 고추$\cdot$참깨등 경제작물에 발생하는 병해는 수십여종이 있으나 그중 국내에서 흔히 발생하며 피해가 큰 병해는 곰팡이병으로 노균병, 잿빛곰팡이병, 탄저병, 균핵병, 시들음병, 역병 등 6종, 세균병으로는 무름병, 풋마름병, 세균성점무늬병등 3종, 그리고 바이러스에 의한 병을 들수 있다. 이 병해들은 그 종류별로 병징, 병발생 양상, 전염방법 등에 특징이 있기 때문에 기생하고 있는 작물이 다를지라도 병의 구별이 가능하고 동일종류에서는 병의 방제방법도 비슷하다. 최근 문제되고 있는 병해를 중심으로 진단법과 방제법을 알아본다.

  • Above 130 accessions of pepper consisting of 50 introductions from Mexico and Nepal, and resistant and susceptible controls were tested for resistance to bacterial wilt and to Phytophthora root rot at seedling stage by artificial inoculation. Aa the results, KC897, KC939, KC936 were newly found resistant to bacterial wilt in addition to already known resistance sources such as KC126, KC350, KC351, KC353. No new sources of resistance to Phytophthora root rot were found among the introductions from Mexico and Nepal.

  • Selection procedures for breeding lines resistant to both bacterial wilt caused by Ralstonia solanacearum and Phytophthora blight caused by P. capsici were executed to generations from $F_2$ and $BC_1F_1$ to $F_4$ and $BC_1F_3$ of crosses between a Phytophthora resistant breed, 'Chilbok No. 1' and bacterial wilt resistant accessions introduced from Vietnam during 2009 and 2010. The breeding populations or lines were tested for resistance to P. capsici and resistant plants were selected. The resistant selections were inoculated with R. solanacearum to discard susceptible plants. Resistance to P. capsici was conspicuously improved by selection from $F_2$ and $BC_1F_1$ and the final selections showed a similar level of resistance to P. capsici as a commercial Phytophthora resistant cultivar, Muhanjilju. A few $BC_1F_2$ selections were crossed to a cytoplasmic male sterile line, Chilbok-A, to identify their nuclear genotype interacting with male sterile cytoplasm. Majority of them was fixed to maintainer (Nrfrf) and only two resulted segregating into male sterile and male fertile plants indicating that the pollen parents were heterozygous in the fertility-restoring gene.
