A New Soybean Cultivar, "Galchae" for Sprout with Brown Seed Coat, Small Seed Size and High Sprout Yielding (갈색종피.소립 고수율 나물용 콩 신품종 "갈채")

  • A new sprout-soybean cultivar, "Galchae" was developed at the National Institute of Crop Science (NICS) in 2008. Galchae was selected from a cross between YS1287 and Jinju#1. The preliminary, advanced, and regional yield trials to evaluate the performance of Iksan 64 were carried out from 2005 to 2008. This cultivar has a determinate growth habit with purple flower, brown pubescence, brown seed coat, brown hilum, rhomboid leaflet shape and small seed size (8.4 grams per 100 seeds). The maturity date of "Galchae" is 6 days later than the check variety, "Dawon". It has good seed quality for soybean-sprout and resistance to lodging. This cultivar has resistance to soybean mosaic virus (SMV) and necrotic symptom (SMV-N). The average yield of "Galchae" was 2.51 ton per hectare in the regional yield trials (RYT) for double cropping carried out for two years from 2007 to 2008.

Isolation of Monocytes with High Purity and Yield from Peripheral Blood Mononuclear Cells by Flotation Density Gradient Centrifugation (부유밀도구배 원심분리를 이용하여 말초혈액단핵구로부터 고순도 및 고수율의 단세포 분리방법)

  • In this work, a simple, inexpensive and reproducible technique of flotation density gradient centrifugation was developed to isolate monocytes with high purity and yield from peripheral blood mononuclear cells (PBMC) using Histopaque solution, density and osmolarity of which were modified to 1.072 g/ml and 335 mOsm with phosphate-buffered saline (PBS) and sodium chloride (NaCl) solution, respectively. The average purity of monocytes was 74.75${\pm}$3.84%, with the individual purity ranging from 71.44% to 82.38%. The average yield of monocytes was 32.62${\pm}$11.16%, with the individual yields ranging from 21.02 to 53.63%. The monocytes isolated by floatation density gradient centrifugation could be successfully cultured into morphologically, phenotypically and functionally dendritic cells in vitro. In conclusion, the entire procedure seemed to be faster and more convenient, simple and cost-effective than other monocyte isolation methods, including plastic adherence and density gradient methods, and has the potential to be developed as a closed system for clinical scale generation of dendritic cells.

Extraction of starch from frozen potato whole-tissues using cellulase and its physicochemical properties (셀룰로오스분해효소에 의한 동결감자로부터 전분의 추출 및 물리화학적 특성)

  • This study investigated the impact of cellulase treatment on the extraction yield of potato starch (PS), and compared the physicochemical properties of PS by conventional (CSE) and enzymatic (ESE) starch extraction. In ESE, the PS extraction yield was predominantly influenced by reaction temperature, time and their interaction, compared to the cellulase concentration. When potatoes were treated for 8 h at $40^{\circ}C$ with 1.5% cellulase, the PS extraction yield was about 3.4-fold higher than that by CSE. Compared to CSE-PS, ESE-PS showed lower total starch contents and higher amylose contents, resulting in lower swelling factors and distorted pasting viscosity profiles accompanied by absence of peak and breakdown viscosities. However, ESE did not affect the gelatinization characteristics of PS. Overall results suggested that ESE can provide the highest yield of PS, and ESE-PS can be a potential starch source for extending the utilization of PS in food industries.

Studies on Nutrio-physiology of Low Productive Rice Plants (수도저위생산력(水稻低位生産力)의 원인구명(原因究明)에 관(關)한 영양생리적연구(營養生理的硏究))

  • Present study was undertaken to elucidate the relationship between uptake of nutrients and photosynthetic activities, and the translocation of several mineral nutrients in rice plants which were grown under different cultural conditions, utilizing radioactive tracer technique. Particular emphasis was placed on the analysis of patterns of nutrient uptake, the relationship between nutritional conditions and yield components. For this, rice plants grown on either low or high yielding fields at different growth stage were subjected to this study. The results are summarized as follows; 1. Varietal difference was observed in the uptake of potassium and phosphorus. Kusabue and Jinheung had good capacity but Paldal had rather poor capacity for the uptake of the both nutrients. 2. For rice plants, a high positive correlation was found between the oxidation of alpha plaus-naphthylamine by root and uptake of phosphorus. 3. Carbon assimilation rate repended on rice varieties. It was high in Noindo, Gutaenajuok #3 Suweon #82 and Jinheung but low in Taegujo, Kwanok, Yugu #132 etc. 4. Heavy application of nitrogen increased carbon assimilation in rice plants but this also depressed translocation of certain carbohydrates to ears. 5. Carbon assimilation wan greatly hampered in rice plants deficient in magnesium, phosphorus or potassium. 6. Total dry matter after ear formation stage, was much higher in rice plants grown in high yielding fields than those grown in low yielding fields. 7. Leaf area index(LAI) reached maximum at heading stage and decreased thereafter in high yielding fields. But in low yielding fields, it reached maximum before heading and sharply decreased thereafter due to early senescence of lower leaves. 8. In general, light transmission ratio (LTR) of leaves was higher in the early growth stage and lower in later stages. Higher ratio of LTR to leaf area index, was found in the rice grown in high yielding fields than those in low yielding fields. 9. Net photosynthetic activity decreased with the increase in leaf area index but was higher in high yielding fields than in low yielding fields. 10. After the ear formation stage, nitrogen, potassium and silicon as weil as $K_2O/N$ in straw were higher in high yielding fields than those in low yielding fields. 11. Nitrogen, phosphorus, potassium and magnesium taken up by rice plants in low yielding fields before heading stage were readily translocated to ears than those in high yielding fields. This suggests greater redistribution of nutrients in straw occurs due to lower uptake, in later growth stages, by rice plants grown in low yielding fields and hence results in early senescence due to nutrient deprivation. 12. In the high yielding fields nitrogen uptake by rice was slow but continuous throughout the life of the plants resulting in a large uptake even after heading. But, in low yielding fields the uptake was fast before heading and slow after heading. 13. A high positive correlation was found between the contents of nitrogen and potassium in the straw at heading stage and grain yield. Positive correlation was also found to hold between the contents of potassium, silicon, $K_2O/N$, $SiO_2/N$ in the straw at harvesting stage, and grain yield. 14. Carbon assimilation was greately hampered in rice plants deficient in magensium, phosphorus or potassium. 15. Uptake of nitrogen, phosphorus, potassium, silicon and manganese by rice was considerably higher in high yielding fields and reached maximum at ear formation stage. 16. In rice, a high positive correlation was discovered between total uptake of nitrogen, phosphorus, potassium, calcium, magnesium, silicon, manganese at harvesting stage and grain yield. 17. In rice, a high positive correlation was found between the total uptake of nitrogen, phosphorus, potassium, calcium, magnesium, silicon at harvesting stage, and number of spikelets per $3.3\;m^2$. In addition, a correlation was found between the total uptake of nitrogen and potassium and number of panicles per hill.

Effects of Water Temperature and Salinity on Dietary Feeding and Body Composition of Juvenile Jicon scallop, Chlamys farreri Transplanted from China (중국에서 이식된 비단가리비, Chlamys farreri의 먹이섭취와 체조성에 미치는 수온과 염분의 영향)

  • The feeding, clearance rate, ingestion rate and body composition of Jicon scallop, Chlamys farreri was investigated under combination in water temperature (24, 27, 30$^{\circ}C$) with salinity (24, 27, 30 %o). The Maximum feeding, Clearance rate showed 8.88 L h$^{-1}$ g$^{-1}$ and 25.42 $\times$ 10$^7$ cells h$^{-1}$ g$^{-1}$ under the experiment condition of 24$^{\circ}C$ and 30%o. While, the minimum feeding, clearance rate and ingestion rate were 0.15 L h$^{-1}$ g$^{-1}$ and 0.73 $\times$ 10$^7$ cells h$^{-1}$ g$^{-1}$ under the experiment condition of 30$^{\circ}C$ and 24%o, respectively. It was indicated that body composition of C. farreri were affected by water temperature and salinity.

  • 이번호에는 충남 연기군의 유일한 육용종계장으로 40년간 오로지 양계산업에 종사한 조은농장을 소개하고자 한다. 이곳은 다양한 효모제와 첨가제를 사용하면서 일체 항생제를 쓰지 않는 농장이다. 최근 추세가 무항생제를 선호하기 때문에 따라간다? 그렇지 않다! 조은농장 사장인 박순종 씨는 30년 전부터 이런저런 첨가제를 사용하면서 최소한 몇 년간 사육해보고 생산성에 효과가 있는 것으로 결과를 얻어 오로지 무항생제 축산업만을 고수하고 있다. 특히나 3년 전부터는 장기능 발달로 생체 활성화 효과가 있는 비타민 미네랄 활성화제제를 첨가 급이하면서 배부율이 크게 향상되는 등 높은 부화성적으로 농가 수익창출을 보고 있는 농장이다. 조은농장 박순종 사장을 만나 인터뷰한 내용을 정리했다.

Effect of Vitamin E Supplementation in Diets on Pork Quality (사료내 비타민 E의 추가급여가 돈육질에 미치는 영향)

  • This study was carried out to examine the effect of vitamin E additive supplements on the meat quality of finishing pigs. Two treatments were provided as commercial diet and commercial diet +vitamin E 200 IU. Dressing percent and carcass length were not different between treatments. Longissimus muscle color and longissimus muscle firmness were a significant differences between two treatments(P<0.05). However, longissimus muscle marbling. longissimus muscle loin area and back fat thickness were not significantly differences. Water holding capacity was increased along the level supplement of vitamin E increased(P<0.05). However. there were not differences on shear force, thawing loss, and cooking loss between treatments. L*, a*, b* and TBARS value were not different between treatments. In conclusion. longissimus muscle color and water holding capacity were affected by vitamin E supplement.

  • The feeding, clearance rate, and ingestion rate of Manila clam (Ruditapes philippinarum) were investigated with small group (10.8$\pm$.58 mm in shell length) and large group (37.5$\pm$3.8 mm in shell length) under combination in water temperature (24, 27, 33, $36^{\circ}C$) With salinity (12, 15, 24, 27$\textperthousand$). The maximum feeding, clearance rate, and ingestion rate in small and large groups were $27^{\circ}C$ and 27$\textperthousand$, respectively. While, The minimum feeding, clearance rate, and ingestion rate in both groups were $36^{\circ}C$ and 12$\textperthousand$, respectively. There results were suggested that the feeding, clearance rate, and ingestion rate of Manila clam were significantly influenced by external factors such as water temperature and salinity. The reason for the mass mortality of Manila clam during the heavy rainfall in summer season can be explained by high temperature and low salinity.

  • Coal fly ash of thermal power plants converted into mesoporous materials MCM-41. The synthesized material was characterized by XRD, FT-IR, SEM, and EDS techniques. The catalytic activity of prepared material was studied for the synthesis of 5-arylindene malononitriles via Knoevenagel condensation of aromatic aldehydes and malonontrile is described. The features of present method are easy handling, stability, reusability, and eco-friendliness of catalyst, high yields, short reaction time, simple experimental and work up procedure.

에틸렌을 이용한 단일벽 탄소나노튜브의 저온합성

  • 단일벽 탄소나노튜브(Single-walled carbon nanotubes, SWNTs)는 나노 스케일의 크기와 우수한 물성을 갖고 있어, 전자, 에너지, 바이오 등 다양한 분야로의 응용이 기대되고 있다. 이러한 응용의 실현을 위해서는 경제적, 산업적인 면에서 보다 손쉬운 합성법이 요구된다. SWNTs의 합성에는 대면적의 균일한 CNTs를 합성할 수 있다는 장점이 있는 열화학기상증착법(Thermal chemical vapor deposition, TCVD)이 가장 일반적으로 사용되고 있다. 하지만 탄화수소가스를 효율적으로 분해하기 위하여 $900^{\circ}C$ 이상의 고온공정이 요구되며, 이는 경제적, 산업적인 면에서 사용이 제한적이다. 따라서 저결함, 고수율의 SWNTs를 저온합성 할 수 있는 공정의 개발이 지속적으로 필요하다. 본 연구에서는, TCVD법을 이용하여 에틸렌 원료가스로 SWNTs의 저온합성 가능성을 확인하였다. 합성을 위한 기판과 촉매로는 실리콘 산화막 기판(SiO2/Si wafer)에 철 나노입자를 지닌 ferritin을 스핀코팅 후 산화하여 이용하였다. 저온합성 공정의 변수로는 합성온도와 원료가스인 에틸렌의 분율을 설정하여, 변수가 SWNTs의 결정성과 수율에 미치는 영향을 고찰하였다. 합성된 SWNTs의 분석의 용이함과 손지기(Chirality)의 제어 가능성을 확인하기 위하여 나노 다공성 물질인 제올라이트(Zeolite)를 보조 기판으로 사용하였다. 실험결과 에틸렌 원료가스로 합성한 SWNTs는 메탄을 원료가스로 사용한 경우보다 낮은 $700^{\circ}C$ 부근에서도 합성이 가능함을 확인하였다. 또한 에틸렌의 분율과 합성 시간의 정밀한 제어를 통해 SWNTs의 합성온도를 더욱 감소시키는 것도 가능할 것으로 예상된다.
