Effects of SDN® as a Microbial Culture on Milk Production, Milk Composition and Somatic Cell Counts of Lactating Cows (미생물 배양액 SDN®의 급여가 착유우의 유생산, 유성분 및 체세포수에 미치는 영향)

  • Holstein cows(n=12) were assigned to one of three diets containing control diet and diets containing a microbial culture, 50ml(T1) and 100ml(T2) SDN(R)(amicrobial culture), per day. The basal diet containing concentrate mixture, corn silage and timothy hay were fed midlactating cows for 12 weeks. Milk production tended to be higher for cows fed T2 diet (20.8kg/day) than fed T1(19.7kg/d) and control diet (19.2kg/day).  There was a tendency of an increase in 4% FCM for cows fed T2 diet(19.6kg/day) than T1(18.8kg/d) and control diet(18.4kg/day). Milk components were not found to be different between cows fed control diet and SDN(R) diets. There was a tendency an increase in milk protein for cows fed control diet(3.43%) compared with microbial diets, T1 and T2(3.08% and 3.20%). However, milk protein production was not significantly different between control diet(0.65kg/d) and T1(0.61kg/d) or T2(0.67kg/d). Somatic cell counts for cows fed T1(72,000) and T2(60,000/ml) were lower than cows fed control diet (108,000/ml) (P<0.05). In conclusion, the cows that were fed diets containing SDN(R) as a microbial culture resulted a tendency of an increase in milk production and a reduction of somatic cell counts which indicates improved milk quality and hygiene.

Effectiveness of Real-time Oxygen Control in Fresh Produce Container Equipped with Gas-diffusion Tube (기체확산 튜브 부착 신선 농산물 용기에서의 실시간 산소농도 제어의 효과)

  • Simplified control logic was devised to fabricate and operate the modified atmosphere (MA) container of fresh produce equipped with gas-diffusion tube whose opening/closing was controlled in response to real time $O_2$ concentration. This is a simplified ramification of the previously developed control logic using both $O_2$ and $CO_2$ concentrations ([$O_2$] & [$CO_2$]). The developed logic was applied to and tested by a container system filled with spinach at $10^{\circ}C$ having optimum MA window of [$O_2$] of 7~10% and [$CO_2$] of 5~10%. It was shown that setting the proper on-off limit (11%) for $O_2$ control based on the assumed relationship $[O_2]+[CO_2]$=21% could attain the desired $CO_2$ concentration just below the upper tolerance limit ($[CO_2]_H$, 10%). The $O_2$ control point can be the lower tolerance limit or adjusted one (21-$[CO_2]_H$) depending on the commodity's MA requirement. The developed logic using single $O_2$ sensor could attain the equilibrated [$O_2$] of 11% with [$CO_2$] of 8~9% which was desired and similar to that of its predecessor ([$O_2$] of 9~10% with [$CO_2$] of 10%) using both $O_2$ and $CO_2$ sensors. Both MA containers (one only with single $O_2$ sensor control and one with $O_2$ and $CO_2$ sensors) could also keep the spinach quality without significant difference between them, but significantly better than perforated control package of air.

Effects of Forced Molting and Storage Time on the Egg Quality (강제환우와 보존기간이 난질에 미치는 영향)

  • An experiment was conducted to investigate the effects of forced molting and egg storage time on the various egg qualities. A total of 240 ISA Brown layers (60 wk of age) were employed as the unmolted treatment (Control). Two hundred and forty ISA Brown layers, molted at the age of 55 wk, were used as a forced molting treatment (T1), and the same number and strain of layers, molted at the age of 70 wk, were also used as the another forced molting treatment (T2). A total of 120 eggs were sampled from each treatment, and divided into six sets, 20 eggs per set. These six sets were stored for 1, 3, 6, 9, 12, and 15 days at $18^{\circ}C$ temperature, respectively. Eggs from T1 were collected from laying hens at the age of 68 wk, which started molting at 60 wk of age and achieved 50% egg production at 63 wk of age. Eggs from T2 were collected from hens at 82 wk of age, which started molting at 70 wk of age and achieved 50% egg production at 78 wk of age. The eggshell strength of T1 was significantly (p<0.05) higher than the Control and T2, and the storing periods did not affect the eggshell strength at all. Neither the forced molting nor the storing periods did not exert any consistent effect on the egg weight, eggshell thickness, eggshell color and egg yolk color. The albumin heights of T1 and T2 were significantly (p<0.05) lower than the Control, and it was remarkably reduced gradually as the storage periods increased in all three treatments. The Haugh unit showed very similar trends as the albumin height, indicating that both albumin height and Haugh unit were very much related to each other. In conclusion, the forced molting improves the eggshell strength, but decreases the albumin height and Haugh unit. The storage of eggs also decreases the albumin height and Haugh unit regardless of molting.

Comparison of Exposure Dose by Using AEC Mode of Abdomen AP Study in Radiography (복부 전후 방향 검사의 자동노출제어 사용 시 선량 비교 연구)

  • We evaluated the effectiveness of TL (Time Limit) method by comparing with NTL (Non-time limit) method when it is used for examinations for abdomen Anterior Posterior (AP) in this paper. The evaluation was conducted based on the comparison of dose, and of signal to noise ratio (SNR) and contrast to ratio (CNR) on both methods. The experiments were conducted with XGEO GC 80 (Samsung, Korea), Unfors ThinX RAD (Unfors, Sweden) and Rando Phantom (Alderson research laboratories, USA) and shielding material with the size of $5.5{\times}9{\times}0.1cm^3$. It was set to activate only two upper ionization chambers in automatic exposure control(AEC) mode and the tube-voltage was set to 80kVp. When the exposure time was limited, it is limited to 51 msec. The images both by NTL AEC method and TL AEC method were acquired when with and without attachment of shielding material on the upper ionization chambers. The images were evaluated by SNR and CNR which are the image evaluation methods using 'Image J'. The NTL AEC method showed increases in dose as much as 130.7% at maximum and 80% at minimum than other methods. The TL AEC method showed decreases in mAs and exposure dose than the NTL AEC method as much as 43.8% and 44.4% respectively. There were no significant differences in SNR or CNR for the experiments (($p{\geq}0.05$). Therefore, it is suggested that the TLAEC mode is more effective when examining patients who have high BMI index or a patient with a metallic substance in the body after surgery.

Effects of Hagocho (Prunella vulgaris L.), Gamgook (Chrysanthemum indicum L.) and Galgeun (Pueraria Radix) on Plasma Lipid Compositin and Histological Consideration in Hyperlipidemic Rat (하고초, 감국 및 갈근 증류액이 비만쥐의 지질구성 및 각종 장기의 해부조직학적 소견에 미치는 영향)

  • Effects of prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix on plasma lipid composition and histological consideration were investigated in hyperlipidemic rats. Concentration of plasma ${\beta},-lipoprotein$ showed a tendence to decrease in prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix treatment groups. However these values were showed not significantly different from control group. Concentration of plasma FFA in prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix treatment groups showed a lower values compared to control group and concentration of plasma FFA of prunella vulgaris L. and pueraria Radix treatment group showed a lower values than prunella vulgaris L. and chrysanthemum indicum L. treatment group. Concentration of plasma glucose and triglyceride showed a tendence to decrease in prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix treatment groups. However plasma glucose values showed not significantly different from control group. Plasma total cholesterol concentration showed a low and HDL-cholesterol concentration showed a high in prunella vulgaris L. and pueraria Radix treatment group. However LDL-cholesterol concentrations were not significantly different in treatment groups. Histological consideration of heart, liver and kidney in prunella vulgaris L. and pueraria Radix treatment groups showed a good features in fat accumulation condition than those of other treatment groups. However in the condition of high fat accumulation in tissues, heart, liver and kidney were showed a slight congestion and a bashed cell nucleus.

Antioxidant Activity of Solvent Fractions from Cultivated and Wild Gromwell (재배와 야생 지치의 추출물과 용매별 분획물의 항산화효과)

  • In order to set up an accurate quality criteria for the Boraginaceae that have been traditionally used for medical purposes and food colorant, and to assess its viability as functional food ingredient, antioxidant tests were conducted on the wild and cultivated plants. Variety of indicators including total contents of phenol, DPPH, SOD-liked effect, hydroxy radical-scavenging effect, lecithin oxidation inhibitory effect, etc were analyzed. Wild and cultivated gromwell's total contents of phenol in their methanol extracts were 0.14% and 0.13%, while they were most active in ethyl acetate extracts and n-hexane extracts, respectively. $IC_{50}$ values of methanol extract of the wild and cultivated plants were 794.41 ${\mu}g$/mL and 971.86 ${\mu}g$/mL, indicating that the wild plant is more responsive (p<0.05) to low concentration. Also the wild and cultivated plants were most active in ethyl acetate fraction and n-hexane extracts when their $IC_{50}$ values were measured by each solvent extracts. SOD-liked effects of both plants were concentration dependent while methanol extracts were more active (p<0.05) in 500 ${\mu}g$/mL than other solvent extracts. Hydroxy radical-scavenging effect of both plants showed less than 50% activity in concentration of 1,000 ${\mu}g$/mL except in chloroform fraction and n-hexane fraction. Lecithin oxidation inhibitory effects of the wild and cultivated plants were active in methanol and solvent extracts of 200~1000 ${\mu}g$/mL. Especially it showed 90% of high inhibitory effect in 1,000 ${\mu}g$/mL of chloroform fraction. Hence, both wild and cultivated Boraginaceae were analyzed to be viable as functional food ingredient.

Effect of N-Application Level According to Seedling Density on Tiller Development, Yield and Quality in Direct-Seeded Rice on Flooded Paddy Surface (벼 담수표면산파 시 입모밀도에 따른 시비 조절이 분얼발생, 수량 및 품질에 미치는 영향)

  • This research was carried out to determine the effect of controlling nitrogen application on tiller development, yield, and quality of rice under scanty or excess seedling stands in direct-seeded rice on flooded soil surface. Seedling stand was set to 3 levels: scanty $(60/m^2)$, optimum $(120/m^2)$, and excess $(200/m^2)$. In the scanty plot, additional 4 kg/10a nitrogen was applied at the 3rd leaf stage to promote tiller development. On the other hand, applying 3.3 kg/10a nitrogen at 5th leaf stage in the excess plot was omitted to suppress tillering. Maximum numbers of tillers per plant were 15.2 and 8.6 in scanty and excess plots, respectively, as compared with 9.8 in optimum plot. Productive tiller rate decreased with the increase in number of seedling stands. Regardless of seedling density, the first tiller developed on the 3rd node of rice stem from the bottom. The primary tillers developed at 3, 4, 5, 6, 7 nodes in scanty plot, 3, 4, 5 nodes in optimum plot, and 3, 4 nodes in excess plot. The secondary tillers developed only in some portion of plants in scanty and optimum plots. The order of tiller emergence was negatively correlated to stem length, panicle length, non-productive tiller number, grain number per panicle, and fertility in scanty plot, and to perfect grain ratio in excess plot. In the optimum plot, however, the order of tiller emergence was not correlated to any of the mentioned characteristics. The perfect grain ratio of scanty plot was the highest because green-kerneled rice was a very small portion in the primary tillers as compared with those of optimum and excess plots. Yield indexes of scanty and excess plots were 99%, and 97%, respectively, of the yield (494 kg/10a) in optimum plot. In conclusion, when seedling stands are not at optimum level, rice yield and quality similar to optimum planting density level can be obtained by means of controlling nitrogen application.

Comparison of Green Color Retention of Zoysiagrass and Cool-season Grass under Multilayer System, USGA System, and Mono-layer System of Sports Field (스포츠용 다단구조, USGA구조 및 약식구조 지반에서 한국잔디 및 한지형 잔디의 녹색기간 비교)

  • This study was initiated to evaluate green color retention under three different soil systems. Several turfgrasses were evaluated in multi-layer, USGA, and mono-layer systems. Turfgrass entries were comprised of three cultivars of Korean lawngrass (Zoysia japonica Steud.) as warm-season grass (WSG) and three blends and three mixtures of Kentucky bluegrass (KB, Poa pratensis L.), perennial ryegrass (PR, Lolium perenne L.), and tall fescue (TF, Festuca arundinacea Schreb.) as cool-season grass (CSG). Significant differences were observed in visual turf color and green color retention among soil systems and turfgrasses. Both the multi-layer and USGA systems were highly associated with better color ratings and longer color retention, as compared with the mono-layer system. Seasonal variation of visual turf color greatly occurred from late December to early spring. CSG exhibited longer color retention than did WSG. The latter maintained green color for approximately 6 months, regardless of the soil system. Spring green-up of Korean lawngrass occurred from early to middle May, while it underwent discoloration from late October to early November. Among the CSGs green-up occurred between early March and early April and leaf color was maintained until middle December to early February. Therefore, the CSGs were green for 8.5 to 11 months, depending on turfgrass and soil system. The mean period of green color duration across all soil systems was approximately 10-11, 9-10 and 8.5-9.0 months for PR, KB and TF, respectively. As for the CSG mixtures, the greater the proportion of PR, the longer the green color retention, while the higher the proportion of TF, the shorter the color retention. There was greater variation in green color duration among the CSGs than the WSGs. Across soil systems, color retention differences of 2 to 6 days were observed for the Korean lawngrass, but 7 to 36 days for the CSGs. These results demonstrate that green color retention varied greatly according to soil systems and also among turfgrasses. Selections of turfgrass and soil system should be made using a concept-oriented approach, when establishing garden, park, soccer field, golf course and other sports field. Information obtained in this study can be used to select soil systems and turfgrasses based on the expected degree of leaf color retention.

Enhancement of Stem Firmness in Standard Chrysanthemum 'Baekma' by Foliar Spray of Liquid Calcium Compounds (액상 칼슘 화합물 엽면살포에 의한 스탠다드 국화 '백마'의 줄기 경도 강화)

  • This study was conducted to enhance the stem firmness of standard chrysanthemum 'Baekma' bred in Korea for commercial quality improvement and inhibition of stem breaking during transportation through foliar spray with calcium agents. Calcium agent screening 'Baekma' was examined using $CaCl_2{\cdot}2H_2O$, $Ca(NO_3)_2{\cdot}4H_2O$, and OS-Ca (natural liquid calcium compounds extracted from oyster shell) depending on each concentration (0, 0.001, 0.01, 0.1, and 1.0%, respectively). All calcium agents sprayed with 1.0% caused chemical injury such as stem bending or leaf burn. OS-Ca also showed more sensitive response to chemical injury than the other calcium agents because OS-Ca was absorbed very well by 'Baekma' leaves. Maximum stem firmness measured during the final harvest was greater in OS-Ca than in the other calcium agents. Especially, maximum stem firmness was greatest in 0.01% OS-Ca. However, elastic strength and maximum bending stress were greater in 0.001% OS-Ca than in the others. Thus, OS-Ca ranged from 0.005 to 0.05%, which did not show any chemical injury, was finally selected as the first candidate for hardening the stem of 'Baekma'. The next experiment using OS-Ca was conducted with the concentrations of 0, 0.005, 0.01, and 0.05%, respectively. From the results, 0.05% OS-Ca showed better plant growth and parameters such as plant height, stem diameter (upper and middle part), the number of leaves, and dry weights of each part than the other concentrations of OS-Ca and control. As for stem firmness depending on OS-Ca concentration, the Ca content within stem, maximum firmness, elastic strength, and maximum bending stress of stem in 'Baekma' sprayed with 0.05% OS-Ca showed the highest values among all the treatments and it turned out to be very high level of significance between control and OS-Ca treatments. However, the area and percentage of the inside cavity within horizontal stem section in 'Baekma' did not show any significance between any treatments including control. Thus, stem firmness of 'Baekma' did not show any correlation with the inside cavity area of stem. In conclusion, we recommend foliar sprays with 0.05% OS-Ca at vegetative growth stage to enhance stem firmness of 'Baekma' during transportation.

Yield and Quality of Forage Mixture as Affected by Maturity of Rye Cultivar and Oat-Rye Seeding Rate (호밀품종의 조만성과 연맥-호밀의 파종량이 혼파사초의 수량과 사료가치에 미치는 영향)

  • Evaluation works of oat in mixture with rye on forage yield have not been reported. The objective of this study was to determine yield and quality of forage mixture as affected by maturity of rye cultivar and oat-rye seeding rate at Suweon from 1999 to 2000. The experiment was arranged in a split plot design with three replications. Main plots consisted of maturity of rye. such as early(cv. Koolgrazer) and late(cv. Kodiak)maturing cultivars. Sub-plots consisted of seeding rate (T1: Oat 2000 and rye 0kg/ha, Ts: Oat 150 and rye 40kg/ha, T3: Oat100 and rye 80kg/ha, T4: Oat50 and rye 120kg/ha. and T5: Oat0 and rye 160kg/ha). Crude protein(CP) content of oat-rye mixture harvested in the fall was not influenced by maturity of rye cultivar, but that of oat-rye mixture was increased from 13.6 to 19.3% as the seeding rate of rye increased(P<0.05), however, maturity of rye cultivar significantly affected CP content of oat-rye mixture in the spring(P<0.01). Acid detergent fiber(ADF) content of oat-rye mixture harvested in the spring was not significantly affected by rye cultivar, but the ADF was decreased from 27.8 to 20.7% as the seeding rate of rye increased(P$<$0.01). When rye was harveste in the spring, ADF content of late maturing cultivar 'Kodiak' was shown as 28.0%. This was lower than that of early maturing cultivar 'Koolgrazer' which was shown as 35.8%(P$<$0.01). Among treatments, neutral detergent fiber(NDF) and in vitro dry matter digestibility(IVDMD) of oat-rye mixture showed a similar trend made on ADF. In this experiment, the highest forage yield (12.356kg/ha) was obtained from early maturing rye cultivar and seeding rates of 100kg/ha of oat and 80kg/ha of rye mixture. A significant interaction between maturity of rye cultivar and seeding rate was found(P$<$0.01). The above results indicate that an early maturing rye cultivar at the seeding rate of 100kd/ha in mixtures with 80kg/ha of oat could be recommended as a succeeding cropping system after corn for silage.
