Detection of Hepatitis B Virus by LAMP and DHPLC (등온증폭반응법과 변성 고성능 액체 크로마토그래피를 이용한 B형 간염 바이러스의 검출)

  • The denaturing high performance liquid chromatography(DHPLC) with fluorescence detector assay is very useful tool for detecting nucleic acids. Furthermore, loop-mediated isothermal amplification(LAMP) constitutes a potentially valuable tool for rapid diagnosis of pathogenic microorganisms. In this study, we evaluated the specificity, detection limit, and sensitivity of a LAMP method and DHPLC method for rapid detection of the hepatitis b virus(HBV). As a result, the LAMP assay reported here has the advantage of rapid detection whereas, DHPLC assay has more sensitivity than other assays. These findings suggest that LAMP and DHPLC assay may be good tool for rapid diagnosis of clinical HBV infection.

The Improved Method for Precise Determination of Pu Isotope Ratio using MC-ICP-MS (다중검출기유도결합플라즈마질량분석기를 이용한 Pu 동위원소비 정밀 분석법)

  • Plutonium is by far the most important of the transuranic elements which have been released into the environment due to radio-toxicity and long term radiation effects on humans. And Pu isotope ratio ($^{240}Pu/^{239}Pu$) is of great interest because this ratio is used as a fingerprint for different sources. Mass spectrometry has been used as an useful atom counting technique with several advantages over decay counting techniques for the determination of Pu isotopes. It enables a determination of Pu isotope ratio in the environmental samples with a low detection limit and a short determination time. An ICP-MS is the representative mass spectrometry for Pu determination. In this study, the precision of $^{240}Pu/^{239}Pu$ isotope ratio was improved by using 4 multiple ion counters of MC-ICP-MS. The detection limit of $^{239}Pu$ and $^{240}Pu$ were $0.10\;fg\;ml^{-1}$ ($0.24\;{\mu}Bq\;ml^{-1}$), $0.12\;fg\;ml^{-1}$ ($0.97\;{\mu}Bq\;ml^{-1}$), respectively. The relative standard deviation of $^{240}Pu/^{239}Pu$ isotope ratio was less than 1 % in trace level. The various reference materials (seawater, soil and sediment) were analyzed to verify this method and their analytical results were in good agreement with the certified (or recommended value) value.

  • Concentrations of polychlorinated biphenyl congeners (PCBs) in samples collected from 43 kindss of water, 35 kinds of soil and 11 kinds of sediment in various place of Korea have been monitored using GC/MS/SIM analytical method. In our investigation, mean recoveries were $83.8{\pm}10.4%$ for water, $94.9{\pm}12.2%$ for soil and sediment samples and $80.2{\pm}8.7%$ for the spiked $^{13}C{_{12}}$-PCBs. Detection limits of PCBs for water, soil and sediment samples are 0.01 ng/ml and 0.05 ng/g. PCB congeners in water samples were not detected within quantitation limit, but trace amounts were detected in most of soil and sediment samples. The contamination level of PCBs in Korea was evaluated to be lower than in advanced countries like USA, Europe and Japan.

Analysis of Glyphosate and Glufosinate in Animal Feeds using LC-MS/MS (LC-MS/MS를 이용한 동물 사료 내 글라이포세이트 및 글루포시네이트 분석)

  • The standards for the contents of glyphosate and glufosinate in foods are specific and well categorized. However, the standard of content in animal feeds is relatively inadequate and the classification is insufficient. There is also constant debate about the risk of glyphosate and glufosinate to human health, but the risk to animals has not been well studied. In this study, we established an analytical method in feeds that is estimated to be the path for animals to ingest glyphosate. The solvent extraction was carried out using 25% methanol. After centrifugation, samples were purified using solid phase extraction (SPE) and quantitatively analysed using LC-MS/MS after concentrated. Assessment of validation was conducted through detection limits, accuracy, and precision tests. The detection limits for the established method were 1.8 of ${\mu}g/kg$ of glufosinate and $2.4{\mu}g/kg$ of glyphosate. Accuracy was ranged from 94.4% to 103.4% and precision was range from 1.5% to 7.2%. Glufosinate was detected in one sample ($ND{\sim}8.8{\mu}g/kg$) and glyphosate was detected in all but one sample ($ND{\sim}337.0{\mu}g/kg$) by applying the analytical method to animal feeds (n=13).

Microbiological Hazard Analysis of Foodservice Facilities and Equipment at Kindergarten Foodservice Establishments (유치원 급식시설.설비에 대한 미생물학적 위해분석)

  • This study was conducted to analyze the food safety management practices and to provide data that could be used to improve food safety management at kindergarten foodservice establishments in Daegu and Gyeongbuk province. Microbiological hazard analysis was conducted from May to July, 2008 at the four foodservice establishments. A total (20 items) of foodservice facilities, cooking utensils, and equipment were tested about total plate counts, coliforms, E. coli, Staphylococcus aureus, Salmonella spp., and Listeria monocytogenes. The microbiological detected levels were higher than the critical limits for prepreparing and cooking knives, chopping boards, working tables (both prepreparing and cooking), refrigerator (inside wall), bart, basket, and tray. E. coli, Salmonella spp., and Listeria monocytogens were not detected in any samples. However, Staphylococcus aureus were detected in rubber gloves for cooking and trench in the bottom at one foodservice establishment. In conclusion, these results suggest that a prerequisite programs guideline should be provided to improve the food safety levels at kindergarten foodservice establishments and the foodservice manager must maintain proper food safety technique for foodservice facilities, cooking utensils, and equipment at kindergarten foodservice establishments to prevent cross-contamination and spread of foodborne pathogens.

Occurrence of Synthetic Musk Compounds in Surface and Waste Waters in Korea (국내 하천수 및 하수처리장 유입.방류수의 합성머스크화합물 오염실태 조사)

  • Synthetic musk compounds are widely used as fragrances in a variety of personal care products like soaps, sampoos, cosmetics and perfumes. The occurrence of synthetic musk compounds in municipal sewage effluent and other environmental samples could negatively impact the health of the ecosystem and humans, due to persistent and long-term chronic exposure of aquatic organisms. Fifteen synthetic musk compounds (musk ketone, musk xylene, musk ambrette, musk moskene, musk tibetene, HHCB, AHTN, ADBI, AHDI, DPMI, ATII, ethylenebrasssylate, ambretettolide, cyclopentadecanolide, OTNE) were analysed in surface waters and sewage treatment plants (STP) influents/effluents in Korea by GC/MS. Method detection limits were $0.005{\sim}0.398{\mu}g/L$. HHCB and AHTN were most frequently observed in both surface waters and STP influents/effluents.

Analysis and Monitoring of Residues of Aminoglycoside Antibiotics in Livestock Products (축산식품 중 아미노글리코사이드계 항생제 잔류량 분석 및 실태조사)

  • It is possible that veterinary medicines remain in livestock food products, according to the use of many and various veterinary medicines to protect against disease when livestock animals are breed in limited space. Concentrated and continuous monitoring of residues is needed due to increases in resistance to antibiotics and side effects by eating livestock food products. We developed an analysis method for detecting streptomycin, dihydrostreptomycin, neomycin, gentamicin and spectinomycin in meat using LC/MS/MS and measured sensitivity, precision, accuracy, linearity and recovery according to CODEX guidelines to acquire confidence in the analysis method. Based on the results, we acquired good sensitivity compared to the maximum residue limit (MRL) as limits of detection (LOD) were 0.002-0.016 mg/kg and limits of quantification (LOQ) were 0.006-0.050 mg/kg. The analysis method satisfied the CODEX guidelines. The linearity ($r^2$) values of aminoglycoside antibiotics were 0.9936-0.9980, recoveries were 60-110% and relative standard deviations (RSD) were within 15%. As a result of monitoring for residues in a total 250 samples of livestock foods such as pork, chicken, and beef by the confirmed method, dihydrostreptomycin and gentamicin were detected in 5 pork samples. The residues of these antibiotics were within the MRLs. Thus, the detection ratio was 2% as 5 samples were identified from 250 samples.

Determination of Mycotoxins in Agricultural Products Used for Food and Medicine Using Liquid Chromatography Triple Quadrupole Mass Spectrometry and Their Risk Assessment (LC-MS/MS를 이용한 식·약 공용 농산물의 곰팡이독소 분석 및 위해평가)

  • For this study, we surveyed concentrations of 8 mycotoxins (aflatoxin B1, B2, G1, G2, ochratoxin A, fumonisin B1, B2 and zearalenone) in agricultural products used for food and medicine by liquid chromatography-tandem mass spectrometry and conducted a risk assessment. Samples were collected at the Yangnyeong Market in Seoul, Korea, between January and November 2019. Mycotoxins were extracted from these samples by adding 0.1% formic acid in 50% acetonitrile and cleaned up by using an ISOLUTE Myco cartridge. The method was validated by assessing its matrix effects, linearity, limit of detection (LOD), limit of quantification (LOQ), recovery and precision using four representative matrices. Matrix-matched standard calibration was used for quantification and the calibration curves of all analytes showed good linearity (r2>0.9999). LODs and LOQs were in the range of 0.02-0.11 ㎍/kg and 0.06-0.26 ㎍/kg, respectively. Sample recoveries were from 81.2 to 118.7% and relative standard deviations lower than 8.90%. The method developed in this study was applied to analyze a total of 187 samples, and aflatoxin B1 was detected at the range of 1.18-7.29 ㎍/kg (below the maximum allowable limit set by the Ministry of Food and Drug Safety, MFDS), whereas aflatoxin B2, G1 and G2 were not detected. Mycotoxins that are not regulated presently in Korea were also detected: fumonisin (0.84-14.25 ㎍/kg), ochratoxin A (0.76-17.42 ㎍/kg), and zearalenone (1.73-15.96 ㎍/kg). Risk assessment was evaluated by using estimated daily intake (EDI) and specific guideline values. These results indicate that the overall exposure level of Koreans to mycotoxins due to the intake of agricultural products used for food and medicine is unlikely to be a major risk factor for their health.

Nested PCR Detection of Pseudomonas syringae pv. actinidiae, the Causal Bacterium (Nested PCR을 통한 참다래 궤양병균 (Pseudomonas syringae pv. actinidiae)의 검출)

  • A PCR method that combines biological and enzymatic amplification of PCR targets was developed for the detection of Pseudomonas syringae pv. actinidiae on kiwifruit leaves. A nested PCR was performed with primers designes from the coding sequence of the cfl gene, which is involved in production of the phytotoxin coronatine. The first and second primer sets efficiently amplified expected 665 and 310-bp fragments, respectively. With two successive amplifications, as few as 20 CFU/ml of P. syringae pv. actinidiae could be detected on ethidium bromide-stained agarose gel. Leaf samples were collected from 4 kiwifruit trees showing yellow halo spots on leaves and incubated in pepton-sucrose broth for 12 h at $16^{\circ}$C before PCR amplification. Positive detection was obtained with one sample, which was proved as a diseased plant in the next spring.

Test Generation for Partial Scanned Sequential Circuits Based on Boolean Function Manipulation (논리함수처리에 의한 부분스캔순차회로의 테스트생성)

  • This paper describes a test generation method for sequential circuits which improves the application limits of the IPMT method by applying the partial scan design to the IPMT method. To solve the problem that the IPMT method requires enormous computation time in image computation, and generates test patterns after the partialscan design is introduced to reduce test complexity. Scan flip-flops are selected for the partial scan design according to the node size of the state functions of a sequential circuit in their binary decision diagram representations. Experimental results on ISCAS'95 benchmark circuits show that a test generator based on our method has achieved 100% fault coverage by use of either 20% scan FFs for s344, s349, and s420 or 80% scan FFs for sl423. However, test gener-ators based on the previous IPM method have not achieved 100% fault coverage for those circuits.
