Simultaneous Analysis of Semi-Volatile Organic Base/Neutral Priority Pollutants in Soil (토양 중의 비휘발성 염기/중성 유기 Priority Pollutants 동시 분석에 관한 연구)

  • This study was aimed at the detection of 40 semi-volatile organic base/neutral compounds from soil among 129 priority pollutants listed by EPA. Two extraction procedures, sonication extraction and Soxhlet extraction, were studied as a extraction and concentration method for priority pollutants in soil. Extracts were analyted by GC/MS-SIM(selected ion monitoring). The analytical methods were tested by standard compounds spiked into blank soil. Accuracy and precision of the methods were measured by calculation of mean recovery and mean relative standard deviation. And the method detection limits were estimated.

Ethanol Concentration Sensor Using Microfluidic Metamaterial Absorber (에탄올의 농도를 검출하기 위한 미세유체 메타물질 흡수체)

  • In this paper, we proposed a novel ethanol concentration sensor using microfluidic metamaterial absorber. The metamaterial absorber comprises a split-ring-cross resonator(SRCR) and a microfluidic channel. The SRCR can generate LC resonance that is very sensitive to changes in the effective dielectric constant around the capacitive gap. In addition, microfluidic channels can change the effective dielectric constant of the dielectric substrate by using an infinitesimal quantity of a liquid on the order of microliters. The proposed absorber can detect the electrical properties of different concentration of ethanol. The performance of the proposed absorber is demonstrated using the absorption measurements of a fabricated prototype sample with waveguides. In addition, the simulated results and measurement results show good agreement.

Development of an Optical Range Finder for Surface Roughness Measurements (표면 요철 측정을 위한 광학적 거리 측정기 개발)

  • We develope a high repetition rate, short distance, optical range finder for surface roughness measurements of large structures, such as a highway road, etc. For range measurement based on a triangulation principle, we use a light emitting diode and an one dimensional Position sensitive photodetector for a light source and an angle detector of the reflected light at the object, respectively. The range finder has automatic power control and electrical background noise rejection capabilities which enable it to overcome variations of an object reflectance and to eliminate time-varying, as well as constant, background light noises. Our experimental results show less than ${\pm}1.5mm$ of measurement errors regardless of an object reflectance, for $22{\sim}38cm$ object ranges which are determined by considering the installation of the range finder and the depth of surface roughness.

Evaluation of a Diffusive Sampler for the Measurement of Formaldehyde using Colorimetric Method (흡광광도법을 이용한 포름알데히드 확산측정기의 평가)

  • The badge-type diffusive sampler for the measurement of formaldehyde in indoor air using three types of colorimetric methods such as chromotrophic acid(CTA), 3-methyl-2-benzothiazolinone hydrazone(MBTH), and 4-amino-3-hydrazino-5-mercapto-1,2,4-triazole(AHMT) method. The washing of the collection filter with several cleaning solution was effected with satisfactory results, regardless of the types of cleaning solutions. The concentrations of absorbance solution in each colorimetric method were experimentally determined by considering the sampling rates. The variation blank values in each colorimetric method was below 15%. As compared with CTA and AHMT methods, the reproducibility of MBTH method was excellent and was below 10% relative standard deviation. The collected formaldehyde mass and time-weighted concentration had a good correlation (correlation coefficient > 0.93). The limit of detection and limit of quantitation, and minimum sampling time were closely correlated to the sampling rates for the measurement of formaldehyde in each method.

Comparison of Colorimetry and HPLC Method for Quantitative Analysis of Chitooligosaccharide (키토올리고당의 측정법으로 비색법과 HPLC법의 비교)

  • The quantitative analysis of chitooligosaccharide was compared to using colorimetry and HPLC method. HPLC method required less than 10mins per sample in analytical time of glucosamine and its the recovery rate was 98.4% (10 mg/ml, w/v). Also there was no the effects of interfering substances(false positive response) by HPLC method. The content of chitooligosaccharide in processed chitooligosaccharide products obtained using HPLC showed lower levels compared to colorimetry. Thus, HPLC method was more sensitive, effective and precise than the colorimetry currently used to determine the glucosamine of chitooligosaccharide.

HPLC-based Analysis of Biogenic Amines in Aging-Cheese (HPLC를 이용한 숙성치즈로부터 바이오제닉 아민 분석법 개발)

  • Biogenic amines have been used as chemical indicators of fermented foods. So far, several chromatography methods have been developed to detect biogenic amines in foods. However, few methods have identified these compound in domestic cheese. We analyzed the biogenic amines (histamine dihydrochloride, tyramine hydrochloride, ${\beta}$-phenylethylamine hydrochloride, putrescine dihydrochloride, cadaverine, spermidine, tryptamine hydrochloride, ethanolamine hydrochloride and butylamine) in cheese by using HPLC. The calibration curves of the biogenic amines were found to be linear over the concentration range of 10-50 ppm with a correlation coefficient of above 0.99. The limit of detection (LOD) and limit of quantitation (LOQ) of the biogenic amines in the given order were 3.7 and 11.3 ppm, 3.4 and 10.4ppm, 3.4 and 10.3 ppm, 4.0 and 12.2 ppm, 3.4 and 10.4 ppm, 3.4 and 10.5 ppm, 3.5 and 10.7 ppm, 4.1 and 12.5 ppm, and 3.4 and 10.4 ppm, respectively. Recovery rates of the biogenic amines in the given order were 112, 104, 93, 108, 91, 102, 101, and 92%, respectively. The findings of this study suggest that HPLC is a suitable method for the determination of biogenic amines, thereby indicating its potential application in the quality control of aging cheese.

Determination of Pantothenic acid in Fortified Foods by HPLC (시판 영양강화식품중 판토텐산의 분석)

  • This study was conducted to develop an HPLC method for determining pantothenic acid in fortified foods which has typically been determined by microbiological assay (MBA) according to AOAC and Korean Food Code approved methods. Pantothenic acid was determined by reversed-phase ion-pair HPLC using UV absorption (200 nm) after extraction with 20 mM potassium phosphate solution by sonication. The recovery of spiked samples and detection limit (LOD) by HPLC were 83.5∼109.6% and 0.5 ppm (mg/kg), respectively. The LOD of the microbiological assay (MBA) was much lower than that of HPLC. The concentrations of pantothenic acid analyzed in all tested samples (n=13) confirmed compliance with declared label claims. The range of recovery ratio by the HPLC method when compared to the microbiological assay was 91.9∼117.6%. There was not significant difference (p<0.01) between the HPLC and MBA methods and the equation of the regression curve was y=1.1428x-0.2269 (r=0.9842). This proposed HPLC method for determining pantothenic acid appears to be suitable for determining pantothenic acid concentrations above 0.25 mg/100 g in fortified foods.

  • Environmental low level tritium analysis was studied using liquid scintillation counter(LSC) and electrolytic enrichment method. To obtain low level blank count, various counting vials were investigated. Among them, teflon coated PE vial had a lower blank count rate (1.86 cpm) and we obtained 2.01 Bq/L detection and tritium separation factor was above 20. LSC counting efficiency obtained $28.70{\pm}0.27%$ using the NIST tritium standard water sample.

  • A cylindrical object buried in the lossy half space is reconstructed from the measured scattered fields above the lossy half space. The position, the size and the medium parameters i.e. relative dielectric constants and conductivity of the buried object as well as the medium parameters of the background lossy half space are obtained from the scattered fields by using the iterative inversion method and the optimization hybrid algorithm combining the genetic algorithm and the Levenberg-Marquardt algorithm. Illposedness of the inversion due to the measurement errors in the scattered fields are regularized by filtering out the evanescent modes in the spatial frequency spectrum domain.

DIFFERENCES IN VIBRIO PARAHAEMOLYTICUS DETECTION RATIO BY ANALYTICAL METHODS (측정방법에 따른 장염비브리오균의 검출율의 비교)

  • The differences in Vibrio parahaemolyticus detection ratio were compared between the isolation methods, the Most Probable Number technique and single dilution tube method. During the period from February to October in 1976, 298 samples of sea water, 112 of bottom deposit, 169 of shellfish, and 80 of fish samples collected along the south coastal area of Korea were examined to determine the detection ratio of Vibrio parahaemolyticus. It was often observed that Vibrio parahaemolyticus was detected in higher diluted samples even through negative in lower dilution. Three hundred and forty three samples out of 659 samples submitted to the test by MPN procedure appeared positive for Vibrio parahaemolyticus showing $52\%$ detection ratio. Whereas only 149 samples, corresponding $22.5\%$, were positive for Vibrio parahaemolyticus in the lowest dilution grade. The positive result was $24.5\%$ in the lowest dilution grade and $50\%$ by MPN Procedure in sea water samples, $28.6\%$ and $65.2\%$ in bottom deposit, $22.5\%$ and $56.2\%$ in shellfish and $7.5\%$ and $32.5\%$ fish samples. When tested by triplicate tubes, $61.7\%$ of 149 Vibrio parahaemolyticus positive samples appeared positive in one tube, $28.9\%$ of them were positive in two tubes and $9.4\%$ of them were positive in all three tubes. The detection ratio determined by MPN procedure was more than two times higher than that of single dilution in triplicate tubes.
