Distribution of Anthocyanin Contents According to Growth Stages in Black-seeded Soybean Germplasms (검정콩 유전자원의 생육기별 안토시아닌 함량 분포)

  • There has been known C3G (Cyanidin-3-Glucoside), D3G (Delphinidin-3-Glucoside), and Pt3G (Petunidin-3-Glucoside) were main anthocyanin pigments in black-seeded soybean. Anthocyanin contents of total 1,032 black-seeded soybean germplasms were analyzed by HPLC. Average of total anthocyanin content was 11.67 mg/g on the all materials ranged from 0.54 to 23.45 mg/g. Mean value of C3G, D3G, and Pt3G contents in all black-seeded soybeans were 8.81, 1.78 and 0.79 mg/g, respectively. Environmental conditions influenced anthocyanin contents during seed development. Delayed flowering, especially of later maturing germplasms, has been showed to result in increased anthocyanin content. So, prolonged maturation period germplasm is generally higher than that of shorten genotypes. It may be concluded that the higher levels of anthocyanin content was associated with the late dates of harvest maturity. Also larger seeds showed high anthocyanin contents than smaller. That inclination is similar in C3G's occasion because C3G content contribute highly to total anthocyanin content than other pigments.

Protective Effects of Black Soybean Seed Coat Extracts against Oxidative Stress-induced Neurotoxicity (산화적 손상에 의해 유도된 신경세포독성에 대한 검정콩 껍질 추출물의 보호효과)

  • Rat pheochromocytoma cells (PC12) and mice were utilized as in vitro and in vivo models to determine the neuroprotective effects of a 70% acetone extract of black soybean seed coat (BSSCE). BSSCE showed higher total phenolic contents than other extracts. Intracellular reactive oxygen species accumulation from $H_2O_2$ treatment of PC12 cells was significantly reduced when BSSCE was present in the media compared to PC12 cells treated with $H_2O_2$ only. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium-bromide (MTT) reduction assay and lactate dehydrogenase assay also showed significantly increased protective effects in PC12 cells. In addition, BSSCE improved the in vivo cognitive ability against amyloid beta peptide-induced neuronal deficits.

Effects of Acidic Treatments for Anthocyanin and Proanthocyanidin Extraction on Black Bean (Glycine max Merrill.) (검정콩 안토시아닌 및 프로안토시아니딘 추출에 대한 산 처리의 영향)

  • This study was performed to investigate the effects of various acidic treatment for anthocyanin and proanthocyanidin extraction from black bean. Extracts were prepared with 80% methanol solutions containing HCl, acetic, formic, phosphoric, and citric acids of different concentrations (0.1, 0.2, 0.3, 0.5, and 0.7%). Total anthocyanin content ranged from 0.74 mg/g in 0.7% citric acid to 1.74 mg/g in 0.3% HCl, depending on acid type and concentration. The major anthocyanins were delphinidin-3-glucoside (D3G), cyanidin-3-glucoside (C3G), and petunidin-3-glucoside (Pt3G). The highest C3G content was 1.12 mg/g in 0.3% HCl. Proanthocyanidin content ranged from 2.01 mg/g in 0.5% acetic acid to 5.29 mg/g in 0.3% HCl. Thus, acidic condition is a significant factor affecting anthocyanin and proanthocyanidin extraction from black bean, and the optimum extraction conditions were determined as 80% methanol containing 0.3% HCl.

In vitro Biological Activities of Anthocyanin Crude Extracts from Black Soybean (In vitro 실험에서 검정콩 안토시아닌 조추출물의 효능 분석)

  • This study was carried out to investigate the antioxidative and anti-inflammatory activity of crude anthocyanin compounds extracted from black soybean. The crude anthocyanin compounds were extracted with 80% methanol and concentrated to powder. The most abundant compound isolated from the extract was C3G(cyanidin-3-glucoside). The superoxide dismutase (SOD) assay was conducted to assess the antioxidative activity of the crude extract. SOD, which catalyzes the dismutation of the superoxide anion into hydrogen peroxide and molecular oxygen, is one of the most important antioxidative enzymes. The black soybean anthocyanin extracts inhibited more than 90% of the superoxide radical at a concentration of 0.1% and 100% at a concentration of 0.5%, indicating that this extract displayed excellent antioxidative activity. To assess the anti-inflammatory activity of the extract, a NO(Nitric oxide) production assay in RAW 264.7 cells was performed. NO is an important physiological messenger and effector molecule in many biological systems, including immunological, neuronal and cardiovascular tissues. In this assay, the anthocyanin extracts showed a high anti-inflammatory potential, where the inhibitory potency for NO production was similar to the positive control, particularly for EGCG(epigallocatechin-3-gallate), which is known to have excellent anti-inflammatory activity. Thus, it can be concluded that the anthocyanin extracts from black soybean have distinctive pharmaceutical activities and may be used as an excellent source materials to supplement the health benefits of various food products.

Development of Prediction Model by NIRS for Anthocyanin Contents in Black Colored Soybean (근적외분광분석기를 이용한 검정콩 안토시아닌의 함량 분석)

  • Near infrared reflectance spectroscopy (NIRS) is a rapid and accurate analytical method for determining the composition of agricultural products and feeds. This study was conducted to measure anthocyanin contents in black colored soybean by using NIRS system. Total 300 seed coat of black colored soybean samples previously analyzed by HPLC were scanned by NIRS and over 250 samples were selected for calibration and validation equation. A calibration equation calculated by MPLS(modified partial least squares) regression technique was developed in which the coefficient of determination for anthocyanin pigment C3G, D3G and Pt3G content was 0.952, 0.936, and 0.833, respectively. Each calibration equation was applied to validation set that was performed with the remaining samples not included in the calibration set, which showed high positive correlation both in C3G and D3G content file. In case Pt3G, the prediction model was needed more accuracy because of low $R^2$ value in validation set. This results demonstrate that the developed NIRS equation can be practically used as a rapid screening method for quantification of C3G and D3G contents in black colored soybean.

Inhibitory Effect of Various Cereal and Bean Extracts on Carcinogenicity in vitro (곡류 및 두류 에탄올 추출물의 in vitro 발암 억제 효과 비교)

  • To investigated the anticarcinogenic activity of 70% ethanol extracts from various cereal in vitro, antimutagenic activity, inhibitory effect of DNA strand scission and tumor promotion were examined. The antimutagenic activity of the beans such as black bean and small red bean was generally higher than that of cereals examined. However inhibitory activity of 70% ethanolic extracts against DNA strand scission induced mitomycin C showed that millet, job's tear, black bean and soy bean among cereals and beans tested in this study inhibited effectively the DNA strand scission. Antioxidative activity of some cereal extracts determined by using linoleic acid model system showed that Job's tear, millet and black bean were higher antioxidative activity than other cereals and beans. Conventional short-term antipromoter assay system using activation of Epstein Barr virus (EBV) clearly demonstrated that sorghum, buckwheat, black bean and small red bean have inhibitory effects on promotion in cellular carcinogenesis.

Changes of Isoflavone Contents During Maturation under Different Planting Dates in Black Soybean (파종기 차이에 따른 등숙기간 중 검정콩의 아이소플라본 함량 변화)

  • This study was carried out to investigate the influence of different planting time on the synthesis of isoflavone in black soybean, Three varieties used in this experiment were lpumgeomjeongkong, Cheongjakong and Heugcheongkong which had different ecotypes, repectively. Seeds were sown at different time, May 15th, May 30th and June 15th with planting density of $60{\times}15cm$. In order to analyze the content of isoflavone, we collected sample every 5 days from 30 days after flowering to harvest and analyzed them with UPLC. As sowing was delayed, the content of isoflavone increased in all of three varieties. The content of genistein was greater than daidzein and glycitein. Increase of Glycitein was not distinct from 55 days after flowering(DAF) and it was stable against temperature change during the seed developing period. Although the content of genistein in Ilpumgeomjeongkong from 50 to 55 DAF, in Cheongjakong from 40 to 55 DAF and in Heugcheongkong from 60 to 65 DAF was lower than the content of daidzein, it was higher than that of daidzein afterward. In the statistical analysis on the relationship between average temperature and the content of aglycone isoflavone at 5-day intervals from 30 DAF during the grain filling period, genistein in Ilpumgeomjeongkong showed meaningful correlation as y=-15.28x+407.9 ($R^2=0.505^*$), diadzein in Cheongjakong showed meaningful correlation as y=-6.188x-164.5($R^2=0.454^*$), and genistein showed significantly high correlation as y=-11.59x+297.6 ($R^2=0.545^{**}$). Taking all the above results into consideration, it was suggested that the regions suitable for high content of isoflavone in black soybean be the northern area of Gyeonggi-do and Gangwon-do, Chungcheongbuk-do and inland area of Gyeongsangbuk-do, where are relatively low average temperature from flowering stage($R_2$) during the grain filling period.

Growth and Yield Responses of Soybean under Night Illumination at Different Growth Stages (콩의 생육단계별 야간조명에 따른 생육 및 수량 반응)

  • This experiment conducted to know physiological characteristics and stress effect on different growth stage of soybean by night illumination. Soybean variety, Shinpaldalkong 2, Keumjungkong and Muhankong were treated by night illumination with 20~30 Lux (0.05~0.08W m$^{-2}$ , 0.24~0.36 $\mu$㏖ S$^{-1}$ m$^{-2}$ ) for 15 days at six different growth stage, seedling, pre-floral initiation, post-floral initiation, pod filling and seed ripening stage. Night illumination delayed flowering to 2~8 days compared to control. Delay of flowering by night illumination severely effected at the pre-floral initiation stage. Stem length was increased all the night illumination treatments except the pod filling stage. Number of nodes in Shinpaldalkong 2 and Keumjungkong 1 were increased until before post-floral initiation stage but in Muhankong were increased until after post-floral initiation stage by night illumination treatments. Number of pods were decreased all the night illumination treatments except seedling stage compared with control. Yield decreased all the treatments and severe the loss rate degree showed the order of prefloral initiation, post-floral initiation, seedling and flowering stage.

Yields and Isoflavone Contents of Soybean Cultivar in Highland Area (국내 육성 콩 품종의 고랭지 재배 시 수랑 및 Isoflavone 함량)

  • This study was performed to obtain fundamental information on growth adaptability and isoflavone contents of soybean cultivars in highland area of Korea. For this study, we cultivated nineteen cultivars and investigated yield and isoflavone contents. In Jinbu-myeon located at altitude of 600 m, most of cultivars showed more than 2.5 t/ha seed yield except three cultivars, and the range of isoflavone contents among nineteen cultivars was 715 to $2,545\;{\mu}g/g$. Cultivars with both high seed yield and isoflavone contents were Manrikong, Sinpaldalkong2, Cheongjakong, and Cheongjakong2 in this area. In Daegwallyeong-myeon located at altitude of 800 m, the range of seed yield and isoflavone contents was 1.4 to 2.4 t/ha and 437 to $2,370\;{\mu}g/g$, respectively. Cultivars with both high seed yield and isoflavone contents were Ilpumgeomjeongkong and Dooyookong in this area. Cultivars selected on the basis of seed yield and isoflavone contents in 2004 and 2005 showed also similar results for yield and isoflavone contents at farmer's field in highland area in 2006. In conclusion, it could be expected to produce soybeans having both higher yield and better quality if the selected soybean cultivars are cultivated in highland area.

Variation in Pod Shattering in a RIL Population and Selection for Pod Shattering Tolerance in Soybean [Glycine max (L.) Merr] (콩 RIL 집단의 내탈립성 변이 탐색 및 유망계통 선발)

  • Pod shattering during the maturing stage causes a serious yield loss in soybean. It is the main limiting factor of soybean cultivation and mechanization. It is important to develop varieties suitable for mechanical harvesting and to develop energy-efficient agricultural machinery to save labor and costs. 'Daewonkong,' developed by the National Institute of Crop Science (NICS) in 1997, is an elite cultivar that occupies more than 80% of the soybean cultivation area in Korea because of its strong tolerance to pod shattering. The objectives of this study were to investigate the variation in pod shattering degree in a RIL population developed from a 'Daewonkong' parent and to select promising lines with pod shattering tolerance. 'Daewonkong' demonstrated a high level of tolerance to pod shattering compared to the 'Tawonkong' and 'Saeolkong' varieties, with no shattered pods after 72 hours of drying. Screening of pod shattering showed a clear distinction between the tolerant and susceptible varieties. Also, the distribution of shattering pod ratio in the two populations showed a similar pattern for three years. The promising lines with pod shattering tolerance included 27 lines in the 'Daewonkong'×'Tawonkong' population and 21 lines in the 'Daewonkong'×'Saeolkong' population. The promising lines are expected to be widely used as breeding parents for creating soybean cultivars with pod shattering tolerance.
