Antidepressant Effect of Acer tegmentosum Maxim on Forced Swimming Test in the Rat (강제수영실험을 통한 산청목의 항우울효과)

  • Acer tegmentosum Maxim (AT) is a species of the maple genus, which is native to North-Eastern China and Korea. Traditionally, AT has been already used for pain relief in Korea. On the other hand, its antidepressant-like activity and related molecular mechanisms is not completely understood. Using the Forced Swimming Test (FST), the effects of a subacute treatment with AT(100, 200, and 400 mg/kg, p.o.) on the immobility and FST-induced changes to the immune parameters, cortisol, ACTH, and cytokine, in rats were investigated. The tendency of immobility showed a dose-dependent decrease in FST. The levels of cortisol, IL-6 and $IL-1{\beta}$ in the peripheral blood were increased significantly after FST exposure. Overall, these results suggest that AT treatment can decrease the immobility time and the release of pro-inflammatory cytokines in the FST, suggesting that the anti-inflammatory effects of AT might be involved in the antidepressant-like effect.

Antifatigue Effect of Chlorella vulgaris in Mice (클로렐라의 항 피로 효과 연구)

  • A unicellular algae, Chlorella vulgaris(CV), was used as a biological response modifier. The effect of CV on forced swimming test and blood biochemical parameters related to fatigue was investigated. Blood urea nitrogen(BUN); creatine kinase(CK); lactic dehydrogenase(LDH); glucose(Glc); total protein(TP); and albumin were determined. CV was orally administered to mice in the range of 0.05 to 0.15 g/kg/day. A forced swimming test results on 3 and 7 day after administration of CV, showed that immobility time was decreased in the CV-administered group(0.15 g/kg). In addition, the contents of BUN in the blood serum were decreased in CV-fed group. The contents of CK and LDH were tended to decrease, but not statistically significant. The plasma Glc level was increased in CV-fed groups(0.05 and 0.1 g/kg) compared to control group. It had no effect on the elevation of TP and albumin level. The results indicate that CV could improve physical stamina.

Effects of Lycii Radicis Cortex in HPA-Axis System on the Forced Swimming Test (지골피(地骨皮)가 강제수영부하실험에서 HPA-Axis System에 미치는 영향)

  • In order to investigate the antidepressant effects of Lycii Radicis Cortex on the change of HPA-Axis system, the forced swimming test was performed. The expressions of CRF and c-Fos in the PVN were measured with immunohistochemical method and the concentration of ACTH in Serum was evaluated with ELISA method. And the results obtained were as follows. Results : 1. The duration of immobility in the forced swimming test was significantly decreased in the LRC100 group and the LRC400 group(P<0.001). 2. The expressions of CRF and c-Fos were significantly reduced in the LRC100 group and the LRC400 group(P<0.001). And the concentration of ACTH in Serum were significantly reduced in the LRC 100 group(P<0.05). According to the results above mentioned, it can be considered that Lycii Radicis Cortex has antidepressant effects.

Effects of Mixture of Lycii Radicis Cortex and Moutan Cortex on Corticotropin-Releasing Factor, c-Fos, and Tyrosine Hydroxylase in Forced Swimming Test (지골피(地骨皮)와 목단피(牧丹皮) 복합제(複合劑)가 강제수영부하실험에서 CRF, c-Fos와 Tyrosine Hydroxylase에 미치는 영향)

  • Objectives : The goal of this study was to investigate the antidepressant effects of Mixture of Lycii Radicis Cortex and Moutan Cortex on the change of HPA-Axis and Catecholamic system. Methods : We were performed the Forced Swimming Test. The expressions of Corticotropin-Releasing Factor (CRF) and c-Fos at paraventricular nucleus (PVN), and tyrosine hydroxylase (TH) at ventral tegmental area (VTA) and locus coeruleus (LC) were evaluated by immunohistochemical method. Results : The duration of immobility in the Forced Swimming Test was significantly decreased in A100, A400(p<0.05~p<0.01). CRF expressions at PVN was significantly decreased in A400(p<0.05).No other group showed significant difference in expression of c-Fos at PVN comparing with control group. TH expressions at VTA was significantly decreased in A100 and A400, respectively(p<0.001). TH expressions at LC was significantly decreased in A100(p<0.01). Conclusions : According to the above results, Mixture of Lycii Radicis Cortex and Moutan Cortex has antidepressant effects via the reduction of CRF expression and the Catecholamine System activity.

The Regulatory Effect of Angelica gigas Nakai on Immune Enhancement and Cytokine Production in vivo and in vitro (참당귀(Angelica gigas Nakai)의 체력증진 및 면역조절효과)

  • Natural products are important sources for drug development because they have a wide variety of useful biological properties. Angelica gigas Nakai (AGN) has been used as an herbal medicine for treatment of colds, pain, and anemia. The present study was designed to evaluate the regulatory effect of AGN on immune enhancement in vivo and in vitro. To investigate the immune-enhancing effect of AGN, we used forced swimming test (FST) experimental model. Mice were orally administered by AGN or distilled water for 14 days and then immobility time and biological parameters in serum were measured. The results showed that immobility time in AGN treatment group was significantly reduced in comparison with the control group. Plasma levels of blood urea nitrogen and lactate dehydrogenase in AGN group was significantly decreased compared with control group. Additionally, we showed that AGN treatment significantly increased immune-related cytokines such as interleukin (IL)-4, IL-2, and interferon (IFN)-𝛾 levels in Molt-4 cells. Collectively, the findings provide experimental evidence that AGN may be effective in improving immune function.

And the experimental groups were divided into the Extraction after Mix group(A) and Mix after Extraction group(B). The effects of both group were compared. Results : The duration of immobility in the FST was decreased significantly in three groups except B100(P<0.01). B100 was shown the significant increase(P<0.05). The expression of CRF in PVN was decreased significantly in all group(P<0.05~P<0.01). The expression of c-Fos in PVN was decreased in A100, A400 and was increased significantly in B100, B400(P<0.05). The expression of TH in VTA and LC were decreased significantly in all group(P<0.001). Conclusions : Effects of Extraction after Mix group(A) and Mix after Extraction group(B) on the HPA-Axis system and Catecholamine system were validated.

Effects of Mixture of Bupleuri Radix and Lycii Radicis Cortex on the Change of HPA-Axis and Catecholamic System in the Forced Swimming Test (시호(柴胡)와 지골피(地骨皮) 합제(合劑)가 강제수영부하실험에서 HPA Axis와 카테콜라민 시스템에 미치는 영향)

  • Objectives : The effects of Extraction after Mixture of Bupleuri Radix and Lycii Radicis Cortex on the change of the HPA-Axis system and the Catecholamic system was investigated. Methods : After performing the Forced Swimming Test(FST), the expressions of corticotropin releasing factor(CRF), c-Fos in the paraventricular nucleus(PVN), and tyrosine hydroxylase(TH) in the ventral tegmental area(VTA) and locus coeruleus(LC) were measured by immunohistochemical method. Results : The duration of immobility in FST was significantly decreased in A 100(Extraction after Mixture of Bupleuri Radix and Lycii Radicis Cortex, 100mg/kg) and A 400(Extraction after Mixture of Bupleuri Radix and Lycii Radicis Cortex, 400mg/kg)(p<0.001). The expression of CRF in the PVN was shown the tendency to reduce in A100 and A400. The expression of c-Fos in the PVN was shown the tendency to reduce in A100 and A400. The expression of TH in the VTA was shown the tendency to reduce mildly in A100 and A400. The expression of TH in the LC was significantly reduced in A400(p<0.001). And the dose dependent reduction tendency was shown, respectively. Conclusions : According to the results above mentioned, the immobility, c-Fos and CRF expression was reduced at lower dose and was increased at higher dose. Therefore there is contradictory effects on the HPA Axis system in accordance with the dose. But in the effects on the catecholaminergic system, it significantly reduced the expression of TH in the LC. It was validated that the effect on the catecholaminergic system was ruled by Bupleuri Radix rather than Lycii Radicis Cortex via mainly the noradernergic system.

The expression of corticotropin-releasing factor(CRF), c-Fos in the paraventricular nucleus(PVN), and tyrosine hydroxylase(TH) in the ventral tegmental area(VTA) and locus coeruleus(LC) was measured with immunohistochemical method and the concentration of seum adrenocorticotropic hormone(ACTH) was measured with ELISA method. And the experimental groups were divided into the extraction after mixing(A) and mixture after extraction(B). The effects of both group were compared. Results : The duration of immobility in the forced swimming test was significantly decreased in the A400 group(P<0.01). The expression of CRF in PVN were significantly reduced in the A100, A400, B100, B400groups(P<0.001). but the expression of c-fos in PVN weren't reduced in all groups. And the concentration of ACTH in Plasma were significantly reduced in the A 100 group(P<0.01). The expression of TH in LC were significantly reduced in the A 400, B 100 and B400 groups(P<0.05~P<0.01). Conclusion : Mixture of Citri Peticulatae Viride Pericarpium and Lycii Radicis Cortex has antidepressant effects. But the difference between mixing and extracting methods was not shown.

The Effects of Different Type of Triglyceride Supplements on Exercise Performance Time, Energy Substrates, Insulin Hormone and Lipase Activity in the Trained Rats (서로 다른 형태의 지방산 투여가 훈련된 흰쥐의 지구성 운동수행력, 안정시기와 운동스트레스 시기의 에너지 기질, Insulin 호르몬과 Lipase 활성에 미치는 영향)

  • The purpose of this study was to investigate the effects of different type of triglycerides (MCT & LCT) on weight, survival time, energy substrate (FFA, TG, pyruvate, lactate), insulin and lipase in the trained rats. Fifty-four Sprague-Dawley rats were divided into 3 groups: control group (CG, n=18), MCT supplement group (MG, n=18), and LCT supplement group (LG, n=18). They also were divided into 3 periods: trained resting (R, n=6) and trained & exercise load (E, n=6), and survival time test was performed to know the supplemented effects. Body weight of all animals was checked every week, MCT group and LCT group received supplementary MCT and LCT orally and preliminary swimming training for 6 days before the start of main experiment. All animals received 15-minute swimming training 5 times during first week of experiment, and swimming training time was increased 15 minutes every week until it reached 90 minutes at last 9th week. After last swimming training, animals were fasted for 12 hours and blood samples were taken from abdominal aorta in the Department of Animal Medicine at the D university Animal Center. Among the CGE, MGE, and LGE groups, the MGE had the greatest increase in physical performance time. In the FFA levels, there was significant differences(p<.05) in CG, MG and LG groups, and also there was major difference of FFA levels in the MG and LG. In the lipase levels, there was signifi.ant differences (p<.05) in CG, MG and LG groups. MG was the greatest than the other groups. In the insulin hormone levels, there was the great differences (p<.05) in LG compare to CG groups, whereas there was no significant difference in the CG and MG. In conclusion, these results suggest that regular prolonged physical training with MCT supplementation, improves exercise performance time through the increase of energy substrate utilization, lipase activity and FFA levels, irrespective of insulin hormone responses.
