Behaviour of the soil residues of the herbicide quinclorac in the micro-ecosystem (pot) (Micro-ecosystem(pot)중 제초제 quinclorac 토양잔류물의 행적)

  • Rice plants were grown for 42 days in the specially made micro-ecosystem(pot) containing two different soils treated with fresh and 60-day-aged residues of [$^{14}C$]quinclorac, respectively, to elucidate the behaviour of the herbicide quinclorac residues in the soils. Amounts of $^{14}CO_{2}$ evolved from two soils treated with different residues with and without vegetation were all less than 2.2% of the total $^{14}C$, indicating that there was little microbial degradation of quinclorac in soil. $^{14}C$-Radioactivity absorbed and translocated into rice plants from soil A and B containing fresh quinclorac residues was 8.4 and 24.2%, respectively, of the originally applied $^{14}C$, while 5.5 and 17.7%, in aged residue soils. These results indicate that larger amounts of $^{14}C$ were absorbed by rice plants from soil B with less organic matter and clay than soil A, and the uptake of [$^{14}C$]quinclorac and its degradation products decreased with aging in soil. After 42 days of rice growing, 84.5 and 61.8% of the $^{14}C$ applied freshly to soil A and B, respectively, remained in soil, whereas, in the case of aged soils, 86.3 and 67.7% of the $^{14}C$ applied did. Meanwhile, without vegetation, more than 98.3% of the $^{14}C$ applied, in both fresh and aged residues, remained in soil, suggesting that quinclorac was relatively persistent chemically and microbiologically. Most of the non-extractable soil-bound residues of [$^{14}C$]quinclorac were incorporated into the organic matter and largely distributed in the fulvic acid portion.

Fungicidal Effect of Slightly Acidic Hypochlorous Water against Phytopathogenic Fungi (미산성 차아염소산수의 식물병원균류에 대한 살균 효과)

  • Slightly acidic hypochlorous water (SAHW) is well known for having a powerful and broad spectrum antimicrobial activity, and is harmless to the environment and humans. SAHW (pH 5~6.5, 20~30 ppm available chlorine concentration) was generated by electrolysis of dilute solution of HCl (4%) in a chamber of a non-membrane electrolytic cell. Our objective was to determine SAHW has a potential fungicidal activity on some phytopathogenic fungi. Spores of Botrytis cinerea, Colletotrichum acutatum and Phytophthora capsici were not culturable on agar media at approximately 10 seconds after treatment by SAHW. However, inactivation of Penicillium hirsutum was required over 3 min. Dilution of SAHW with sterilized distilled water (SDW) at the ratio of 1:1 (SAHW:SDW) against C. acutatum showed 100% inactivation but, the efficacy in 1:2 decreased until 63.2%. Control value of SAHW was 70.4% against C. acutatum on pepper fruits when applied upto 24 h postinoculation. SAHW has a powerful and wide spectrum antifungal activity and could be applied as a potential alternative to fungicidal agent for control of plant disease.

Biochemical Characteristics and Growth Control for fungi isolated from mural painting of Tomb No.6 at Songsan-ri, Gongju (공주 송산리 6호분에서 분리한 진균의 생화학적 특성 분석 및 생장제어 연구)

  • Fungi were isolated from mural painting in tomb no.6 at songsan-ri, Gong-ju. Antifungal susceptibility of essential oils extracted from natural medicine was tested and it confirmed applicability for mural painting in tombs. 26 species of fungi collected from air-borne and wall surfaces were identified to 15 species of Ascomycetes, 2 species of Zygomycetes, 1 of Basidiomycetes. Wheat starch and gelatin degradability were evaluated as isolated fungi. SY-18, SY-23, SY-25 showed high degradability of wheat starch. SY-18, SY-21, SY-23 were decomposed into gelatin. Biochemical characteristics of decomposing fungi to wheat starch glue and cowhide glue were analyzed by using ${\alpha}-amylase$ and gelatinase activity. An Antifungal test was conducted in Anethole and Eugenol. Anethole and Eugenol mixture(1:2) showed high antifungal susceptibility. Natural adhesives help microbial growth and can cause structural damage in mural painting. The expectation of this study is the possibility to control microbial growth in wall painting using natural essential oils. It can be used as a data for conservation method to control microbial damages.

  • The objective of this study was investigate the characteristic of biosurfactant produced from the iso-lated strain. The strain was isolated from soli samples of Duck-Yu Mountain and it was identified as Bacillus atrophaeus DYL-130 by 16S rDNA and gyrA gene nucleotide sequence analysis. The surface ten-sion of culture filtrate of Bacillus atrophaeus DYL-130 decreased to 28 mN/m and its biosurfactant con-centration was determined by diluting the culture filtrate until the critical micelle concentration (CMC). The emulsifying activity and stability of crude biosurfactant was measured by using water-immiscible hydrocarbons and oils as substrate. The biosurfactant was purified by affinity chromatography and the surface activity of purified biosurfactant was measured by drop-collapsing method and it could be effectively emulsify toluene.

Cyanidin-3-O-glucoside Ameliorates Postprandial Hyperglycemia in Diabetic Mice (당뇨 마우스에서 cyanidin-3-O-glucoside의 식후 고혈당 완화 효과)

  • Cyanidin-3-O-glucoside (C3G) shows anti-inflammatory and antioxidant effects; however, its effect on postprandial blood glucose levels remains unknown. Alpha-glucosidase inhibitors regulate post-prandial hyperglycemia by impeding carbohydrate digestion in the small intestine. Here, the effect of C3G on ${\alpha}-glucosidase$ and ${\alpha}-amylase$ inhibition and its ability to ameliorate postprandial hyperglycemia in streptozotocin (STZ)-induced diabetic mice were evaluated. ICR normal and STZ-induced diabetic mice were orally administered soluble starch alone or with C3G or acarbose. The half-maximal inhibitory concentrations of C3G for ${\alpha}-glucosidase$ and ${\alpha}-amylase$ were 13.72 and $7.5{\mu}M$, respectively, suggesting that C3G was more effective than acarbose. The increase in postprandial blood glucose levels was more significantly reduced in the C3G groups than in the control group for both diabetic and normal mice. The area under the curve for the diabetic mice was significantly reduced following C3G administration. C3G may be a potent ${\alpha}-glucosidase$ inhibitor and may delay dietary carbohydrate absorption.

Effects of Storage Temperature and Materials on Maintenance of Quality of Solidago virgaurea spp. gigantea in Modified Atmosphere Packaging (MAP 포장재의 종류와 온도가 울릉미역취의 품질에 미치는 영향)

  • Modified atmosphere packaging (MAP) was assessed in terms of extending the shelf life of Solidago virgaurea spp. gigantea. In June 2005 Solidago virgaurea spp. gigantea were harvested from Ulleung Island, packaged in $30\;{\mu}m$ PP, $30\;{\mu}m$ Antifogging-OPP, $30\;{\mu}m$ LDPE, $18\;{\mu}m$ Macroperforated-HDPE, $60\;{\mu}m$ nylon/PE, or $85\;{\mu}m$ PVC/PE film, then stored at 4, 10 or $20^{\circ}C$ for up to 14 days. The $O_2$ concentration decreased to $9{\sim}10%$ in the PP and A-OPP packaging, to $12{\sim}16%$ in LDPE packaging, and to 2% in nylon/PE and PVC/PE packaging at $10^{\circ}C$. The $CO_2$ concentration increased to 5% at $10^{\circ}C$ in PP, A-OPP and LDPE packaging, but increased consistently in the nylon/PE and PVC/PE packaging. Weight loss was markedly reduced by all MAP films except M-HDPE, and which was enhanced with decreasing storage temperature. The soluble solids and pH were not affected by the packaging material at $4^{\circ}C$ and $10^{\circ}C$, but were affected at $20^{\circ}C$. The highest score for sensory qualities (appearance, color and overall acceptability) was obtained for vegetables stored at $4^{\circ}C$ in PP films. Our results show that MAP using PP films and $4^{\circ}C$ storage can effectively maintain the quality of Solidago virgaurea spp. gigantea.

Determination of Emulsion Stability Index in W/O Emulsion (유중수적형 유화계내에서의 유화안정지수 산출법의 확립)

  • To evaluate the emulsion stability indices of W/O emulsion system, we developed the simple and sensitive "VOLUMETRIC METHOD". This technique involved the first step of homogenizing the milk fat-water system with Ultra-turrax T25, then the volume of the added water phase was measured immediately. After quiescent incubation in test tubes at room temperature for a desired storage time, the bottom volume of the separated water layer was measured. And then "emulsion stability index(ESI)" was calculated by the following equation : $ESI=(1-V_s/V_a){\times}100$, where $V_a$ means the volume of the added water in the W/O emulsion and $V_s$ represents the volume of the separated water in the W/O emulsion for a desired storage time. The emulsion stability indices of W/O emulsion system at sorbitan trioleate, span 60, and tween 20 were $95.4{\pm}1.8$, $56.1{\pm}2.8$, and $41.6{\pm}2.2$ respectively. Furthermore, the differences between "VOLUMETRIC METHOD" and "Titus et al method" were less than 5.0 of ESI Value.

Enzymatic Characteristics of ${\alpha}-Galactosidase$ for the Removal of Flatulence Factor in Soybean (대두(大豆)의 Flatulence Factor 제거(除去)를 위한 ${\alpha}-Galactosidase$ 효소제의 특성)

  • For the removal of raffinose and stachyose related to flatulence in soybean, ${\alpha}-Galactosidase$ activity of six commercial enzyme preparations was compared and their enzymatic characteristics were investigated. Among the tested enzymes, one product from Aspergillus niger was shown to be the most potent in ${\alpha}-Galactosidase$ activity. The enzyme characteristics of the selected preparation were shown to be pH 4.0-4.5 for optimum activity, pH 4-5 for optimum stability and $45^{\circ}C$ for optimum activity. Upon reaction on a synthetic substrate, $p-nitrophenyl-{\alpha}-D-galactoside$, Michaelis constant was 2.08 mM and maximum velocity was 435 micromoles of substrate/minute/g enzyme preparation. The enzyme was proved to be essential for SH group for its activity and capable of hydrolyzing raffinose, sucrose and $p-nitrophenyl-{\alpha}-D-galactoside$ almost completely. Thin-layer chromatographic analysis exhibited that the enzyme treatments of raffinose and stachyose were resulted to produce only monosaccharides in 2 hours of hydrolysis. It was, therefore, assumed that the flatulence factor in soybean foods can be easily removed by the use of enzymes showing ${\alpha}-Galactosidase$ activity.

Properties of Antimicrobial Substances Produced by Bacillus amyloliquefaciens CJW15 and Bacillus amyloliquefaciens SSD8 (Bacillus amyloliquefaciens CJW15와 SSD8이 만드는 항균물질들의 특성)

  • Two Bacillus strains, CJW15 and SSD8, with strong antibacterial activities were isolated from cheonggukjang. Both were identified as B. amyloliquefaciens strains after gene sequencing of rRNA and recA. CJW15 strongly inhibited the growth of B. cereus (ATCC14579), Listeria monocytogenes (ATCC19111), and Lactococcus lactis (ATCC11454). In comparison, SSD8 inhibited the growth of B. cereus (ATCC14579) and Enterococcus faecium (ATCC19953). The antibacterial activities of the two strains were not affected when exposed to a temperature of $100^{\circ}C$ for 15 min and were quite stable in acidic (pH 3) and alkaline (pH 12) pH conditions. Enzymatic treatments (trypsin, pepsin, proteinase K, and protease) had no effect on the activity of CJW15, but reduced the activity of SSD8 by half. Both isolates possess genes involved in the synthesis of lipopeptides (e.g. surfactin, fengycin, iturin, and iturin A), and genes encoding subtilin, a bacteriocin. Moreover, both isolates have fibrinolytic activities as well.

The Relationships between the Microorganisms and the Red-Colored Phenomena of Ginseng (Panax ginseng C.A. Meyer) (인삼뿌리의 적변현상과 근권미생물)

  • To clarify a significant difference between red-colored phenomena (RCP) and microbes isolated from rhizosphere soil of healthy ginseng (HES) and red-colored ginseng (RCS), we have examined growth and cellulase activities of the microbes according to pH variation and iron status. The soil microbes could not grow at pH 3.0 on the YEB medium. The growth of bacterium isolated from RCG at pH from 5.0 to 9.0 showed small differences and the growth of bacterium HES was lower than that of others. The growth of bacteria from RCS and surface soil (SUS) at pH 5.0 were also lower than that of pH 7.0 and pH 9.0. However, the bacteria isolated from red-colored ginseng (RCG) and RCS are able to grow on the medium contained 2 mM Fe$\^$3+/ at pH 3.0. Furthermore, the growth of bacterium from RCG increased about two times in the medium contained iron at pH 7.0 compared with minus iron. The cellulase activity of isolated bacteria increased two times in the medium contained 2 mM Fe$\^$3+/ compared with minus iron. The activity of extra-cellular cellulase was higher by one hundred times than that of intracellular level. The cellulase activity of the bacterium from RCS at pH 5.0 was higher by two times than that of pH 7.0. Especially, intracellular activity of the bacterium from RCS on the medium contained 2mM Fe$\^$3+/ increased about six to seven times compared with control (minus iron). Also, extra-cellular activity increased about eleven to twelve times compared with control. These results indicate that the soil microbes seem to be related iron redoxidation by proton extrusion and with cell wall digestion by secreted cellulase.
