감자(Solanum tuberosum L.)는 식물학적 분류상으로 가지과에 속하는 작물로, 재배기간이 짧고 단위면적당 생산량이 많아 세계적으로 벼, 밀, 옥수수에 이어 네 번째로 재배되고 있다. 감자의 원산지는 남아메리카 안데스 산맥의 고원지대로 유럽을 거쳐 여러 나라들로 퍼져나갔으며, 1824년경 우리나라로 감자가 들어왔다는 기록이 남아있다. 국내 감자 재배작형 비율을 살펴보면, 봄재배가 63%, 여름재배 16%, 가을감자 14%, 그리고 겨울시설재배가 7% 정도를 차지하고 있으며 소비자들에게 연중 햇감자를 공급하고 있다. 1960년대부터 국내에서 감자 교배육종을 시작하면서 육종기반을 구축하였으며 현재까지 40여 품종을 육성하였다. 과거에는 부족한 주식을 대체하기 위한 구황작물로 감자를 주로 소비하였기 때문에, 식용감자 품종 위주로 선발하기 위하여 조숙성과 수량성에 중점을 두어 '조풍', '추백' 등의 품종을 육성하여 보급하였다. 최근 국민 소득의 향상과 식습관 변화 등으로 식품 소비구조가 다양화되면서 감자를 식용뿐만 아니라 감자칩, 프렌치프라이 등 가공제품이나 더 나아가 화장품 원료로 이용되면서 육종 방향과 목표를 다양하게 설정하여 품종을 육성하고 있다. 그 결과로, 전분함량이 높고 칩가공성이 우수한 '다미', '골든볼', '은선' 등 칩가공용 품종, 감자모양이 길고 건물함량이 높아 가공적성이 우수하고 프렌치 프라이로 적합한 '골든에그'와 '얼리프라이', 기형, 열개 등 생리장해 발생이 적고 일반 부식용인 '수선', '추원' 등 다양한 용도를 가진 품종을 육성하여 보급하고 있다. 이와 같이 현재까지 육성된 감자 용도별 주요 품종들의 현황과 육종 방향에 대해서 소개하고자한다.

The study was performed to investigate the effect of a plug culture method in the early stage of a potato breeding program. The 105, 200 and 288-cell plug trays were used for raising potato seedlings. The % germination in plug trays ranged between 94 and 96%. Percent tuberization measured at 50 and 60, and 70 days after sowing was the greatest in 105-cell and 200-cell trays, respectively. However, tuberization was most delayed in 288-cell trays. Thus, it is considered that 105-cell tray is the most effective for the selection of early maturing plants. Plant maturity was determined by the morphological characteristics at the seedling stage of stolons cultured in plug trays. Most of seedlings with shorter stolons at seedling stage (1st generation) were the early-maturing, while most of seedlings with longer stolons were the late-maturing at the clonal stage (2nd generation). These results mean that the plant maturity in the early potato breeding program can be effectively grouped by observing the morphological characteristics of stolons after plug culture.
The tetraploid Solanum acaule is a wild potato species from Bolivia widely used for potato breeding because of its diverse attractive traits, including resistance to frost, late blight, potato virus X, potato virus Y, potato leafroll virus, potato spindle tuber viroid, and cyst nematode. However, the introgression of useful traits into cultivated potatoes via crossing has been limited by differences in endosperm balance number between species. Somatic fusion could be used to overcome sexual reproduction barriers and the development of molecular markers is essential to select proper fusion products. The chloroplast genome of S. acaule was sequenced using next-generation sequencing technology and specific markers for S. acaule were developed by comparing the obtained sequence with those of seven other Solanum species. The total length of the chloroplast genome is 155,570 bp, and 158 genes were annotated. Structure and gene content were very similar to other Solanum species and maximum likelihood phylogenetic analysis with 12 other species belonging to the Solanaceae family revealed that S. acaule is very closely related to other Solanum species. Sequence alignment with the chloroplast genome of seven other Solanum species revealed four InDels and 79 SNPs specific to S. acaule. Based on these InDel and SNP regions, one SCAR marker and one CAPS marker were developed to discriminate S. acaule from other Solanum species. These results will aid in exploring evolutionary aspects of Solanum species and accelerating potato breeding using S. acaule.

"Hongyoung" was bred within the potato breeding program of National Institute of Highland Agriculture (NIHA). It was originated from a cross made between Atlantic as a female and AG34314 as a male parent in 2003. "Hongyoung" was evaluated as a clone number Daegwan 1-102 for the growth and tuber characteristics, yielding ability, and resistance to major diseases in the regional yield trials from 2006 to 2007. Finally, this clone was renamed as "Hongyoung" and was presented to Korean Seed & Variety Service in 2007 for registration as a new potato cultivar. "Hongyoung" has a round shape, shallow eye depth, red skinned and fleshed tuber characteristics. Its mean tuber yields from the regional yield trials was 37.4 MT/ha, and it showed the highest yield in spring cropping. "Hongyoung" showed high resistance to common scab (Streptomyces scabies) and potato leaf roll virus (PLRV), however it was susceptible to both potato virus Y (PVY) and late blight (Phytophthora infestans). The dry matter content of "Hongyoung" was low to medium 17.7%, therefore it is suitable for table use. Its anthocyanin content was the highest in autumn cropping with 31.8 mg/100g fresh weight and it contained 3.6 times higher anthocyanin than the control, Jasim.

It was originated from a cross made between Atlantic as a female and AG34314 as a male parent in 2003. Jayoung was evaluated as a clone number Daegwan 1-104 for the growth and tuber characteristics, yielding ability, and resistance to major potato diseases in the regional yield trials from 2006 to 2007. Finally, this clone was renamed as Jayoung and was presented to Korean Seed & Variety Service in 2007 for registration as a new potato cultivar. Jayoung has a oval shape, shallow eye depth, dark purple skinned and fleshed tuber characteristics. Its mean tuber yields from the regional yield trials was 37.3 MT/ha, and it showed the highest yield in spring cropping. Jayoung showed high resistance to both common scab (Streptomyces scabies) and potato leaf roll virus (PLRV), however it was susceptible to late blight (Phytophthora infestans). The dry matter content of Jayoung was fairy high 20.3%, therefore it is suitable for both chip processing and table usage. Its anthocyanin content was the highest in autumn cropping as 43.7 mg/100 g fresh weight and its mean anthocyanin content was 4.7 times higher than that of Jasim.
Solanum demissum is one of the wild Solanum species originating from Mexico. It has wildly been used for potato breeding due to its resistance to Phytophthora infestans. S. demissum has an EBN value of four, which is same as that of S. tuberosum, so that it is directly crossable for breeding purposes with the cultivated tetraploid potato (S. tuberosum). In this study, the chloroplast genome sequence of S. demissum obtained by next-generation sequencing technology was described and compared with those of seven other Solanum species to develop S. demissum-specific markers. Thetotal sequence length of the chloroplast genome is 155,558 bp, and its structural organization is similar to those of other Solanum species. Phylogenetic analysis with ten other Solanaceae species revealed that S. demissum is most closely grouped with S. hougasii and S. stoloniferum followed by S. berthaultii and S. tuberosum. Additional comparison of the chloroplast genome sequence with those of seven other Solanum species revealed two InDels specific to S. demissum. Based on these InDels, two PCR-based markers for discriminating S. demissum from other Solanum species were developed. The results obtained in this study will provide an opportunity to investigate more detailed evolutionary and breeding aspects in Solanum species.
Solanum stoloniferum, one of the wild tetraploid Solanum species belonging to the Solanaceae family, is an excellent resource for potato breeding owing to its resistance to several important pathogens. However, the sexual hybridization of S. stoloniferum with S. tuberosum (potato) is hampered due to the sexual incompatibility between the two species. To overcome this and introgress the various novel traits of S. stoloniferum in cultivated potatoes, cell fusion can be performed. The identification of the fusion products is crucial and can be achieved with the aid of molecular markers. In this study, the chloroplast genome sequence of S. stoloniferum was obtained by next-generation sequencing technology, and compared with that of six other Solanum species to identify S. stoloniferum-specific molecular markers. The length of the complete chloroplast genome of S. stoloniferum was found to be 155,567 bp. The structural organization of the chloroplast genome of S. stoloniferum was similar to that of the six other Solanum species studied. Phylogenetic analysis of S. stoloniferum with nine other Solanaceae family members revealed that S. stoloniferum was most closely related to S. berthaultii. Additional comparison of the complete chloroplast genome sequence of S. stoloniferum with that of five Solanum species revealed the presence of six InDels and 39 SNPs specific to S. stoloniferum. Based on these InDels and SNPs, four PCR-based markers were developed to differentiate S. stoloniferum from other Solanum species. These markers will facilitate the selection of fusion products and accelerate potato breeding using S. stoloniferum.

This study was carried out to discriminate potato cultivars and breeding lines by specific molecular markers using random amplified polymorphic DNA (RAPD) analysis. The genotypes of potatoes used for analysis were eight cultivars and five breeding lines. Some of those show much phenotypic resemblances among them because 'Jopung', 'Daekwan70', 'Gawon', and 'Daekwan72' have immediate parental relationship with 'Superior', 'Irish Cobbler', 'Namsuh', and 'Atlantic', respectively. So, there are many difficulties to distinguish the varieties by the morphological characteristics. Three URP primers, URP2, URP4, and URP8 were selected for promising primers to discriminate potato genotypes or cultivars. The three URP primers were shown very high reproducibility because of the relatively high annealing temperature and long primer size. Although the results of similarity analyses did not always reflect the genetic relationship between potato varieties, the reproducible pattern of amplified DNA bands by URP primers showed possibility for molecular markers for discrimination of potato genotype or cultivar.

The experiment was conducted to investigate the potential for environmentally friendly cultivation of cv. Haryeong, which was bred by the National Institute of Highland Agriculture in 2006, through evaluation of the late blight field resistance at the farmer's practices level. cv. Haryeong showed higher field resistance to late blight as 590.3 in mean Area Under the Disease Progress Curve (AUDPC) during the cultivation period than cv. Superior as 1377.7 in AUDPC. However there was no observed different trend among the experimental treatments. Mean of tuber yield of cvs. Haryeong and Superior were 2,870 kg/10a and 2,301 kg/10a, respectively, and in all experimental treatments Haryeong was 25% higher than those of Superior. Mean of dry matter content of Haryeong and Superior were 18.1% and 16.1%, respectively, and there were no significant differences among the experimental treatments. cv. Haryeong was confirmed as a suitable cultivar for environmentally friendly cultivation in this experiment. In the future, additional research on fertilization and disease management will be required.

Potato late blight caused by the fungus Phytophthora infestans is one of the most vital diseases damaging the potato plant. It is for this reason that breeding potato cultivars resistant to late blight is now becoming a major concern around the world. The B3C1 clones has been introduced by the Highland Agriculture Research Center, RDA. The clones which came from International Potato Center in 2005 have a durable resistance to late blight. The clones were bred under a short-day condition in Peru. However, there was still no report on the adaptability of these clones to the long-day condition in Korea. Therefore, this study was conducted to evaluate the late blight resistance and major agronomic characteristics of B3C1 clones under Korea's long-day condition. This study was also done to generate genetic resources for developing new varieties resistant to late blight. In this study it was found out that in naturally infested field with P. infestans, AUDPC (area under disease progress curve) values of all B3C1 clones were significantly lower than those of the control varieties, 'Superior', 'Atlantic', and 'Haryeong'. It was found out that B3C1 clones had a high level of resistance to late blight and that they could be used as genetic resources to breed potato varieties with late blight resistance. However, several undesirable characteristics such as extremely late maturity, excessive growth of stems and stolons, and production of tubers that cannot easily be removed from the stolons were also observed. Among the twenty B3C1 clones, two clones, LB-8 (CIP393077.159) and LB-11 (CIP393371.159), were selected for cultivating at the highland area of Korea. Two B3C1 clones were crossed with Korean breeding lines and clonal selection for the progenies is still in progress.
