홍삼 및 기능성 소재를 이용한 주류첨가용 액상제제 개발 (Development of Liquid Phase Product from Red Ginseng and Medicinal Herbs for Alcoholic Beverage)

  • 본 연구에서는 홍삼 농축액의 crude saponin과 생약재의 물추출물이 ADH와 ALDH의 활성에 미치는 영향 및 이들 생약재를 이용한 주류첨가용 액상제제 개발을 위한 모델을 제시하였다. 재료자체의 비중이 커서 용매비율을 낮게 한 갈근의 경우, 추출에 따른 고형분의 함량이 매우 낮았고, 3시간이후 고형분의 함량에는 변화가 없었다. 갈화, 지구자, 상엽의 경우는 4시간이후 고형분 함량의 변화가 없었다. 홍삼의 crude saponin, 상엽, 갈화, 갈근, 지구자의 경우 적정농도에서 ADH의 상대활성을 104-114%까지 증가함을 보였고, 상엽, 지구자는 ALDH의 상대활성을 102∼106%까지 증가시켰다. 알코올 분해에 영향을 미치는 생약재를 원료로 액상제제를 제조하여 소주에 혼합한 후 1차 소비자 기호도 조사 결과, 경옥고향, 쌍화향, 칡향을 첨가한 세가지 시료 상호간에 유의적인 차이를 나타냈고, 쌍화향을 첨가한 시료가 가장 양호한 것으로 나타났다. 1차 기호도 조사결과 가장 양호한 것으로 나타난 쌍화향을 첨가한 시료와 향 무첨가 시료, 시판중인 K사제품간의 2차 기호도 조사 결과, 세가지 시료 상호간에 유의적인 차이를 보이지 않았다.

각종 약용 식물 첨가가 전통 인삼주의 품질 특성과 생리기능성에 미치는 영향 (Effects of Medicinal Plants on the Quality and Physiological Functionalities of Traditional Ginseng Wine)

  • 고부가가치를 가진 전통 인삼주를 개발하기 위하여 각종 약용식물들의 첨가가 인삼주의 품질과 생리기능성에 미치는 영향을 조사하였다. 먼저 전통 인삼주 제조용 최적효모를 선발하기 위하여 2종류의 시판 알코올발효효모와 청주제조용 Saccharomyces cerevisiae를 이용하여 인삼주를 제조한 결과 시판효모인 Fermivin으로 발효시킨 전통 인삼주가 에탄올 함량이 17.0%로 비교적 높았고 기호도도 제일 우수하였다. 또한 각종 약용식물첨가가 전통 인삼주의 품질에 미치는 영향을 조사한 결과 시판효모인 Fermivin을 0.4%, 시판곡자와 정제효소를 각각 g당 36sp씩 2:1의 비율로 혼합하여 담금한 주모에 1% 인삼과 0.5%의 하수오와 갈근을 첨가하여 $25^{\circ}C$에서 20일간 발효시킨 인삼주가 에탄올함량이 16.8%로 제일 높았고 기호도도 제일 우수하였다. 최적 발효일수를 결정하기 위하여 발효 중 생리기능성의 변화와 기호도를 조사한 결과 발효 20일에 항고혈압활성을 나타내는 안지오텐신전환 효소 (ACE)저해활성이 78.9%로 높았고 기호도도 제일 우수하였다. 그러나 HMG-CoA reductase 저해활성은 발효 15일에 8.2%, 20일에 9.2%를 보였을 뿐 여타의 생리기능성은 없거나 매우 미약하였다. 이상의 결과를 종합하여 볼 때 항고혈압활성이 우수한 고급 전통 인삼주를 제조하기 위해서는 시판곡자와 정제효소를 각각 g당 36sp씩 2:1로 혼합하여 증자미와 물과 시판효모인 Fermivin 0.4%와 인삼 1%로 1차 담금하여 $25^{\circ}C$에서 4일간 발효시킨 후 갈근과 하수오를 각각 0.5%첨가하여 다시 $25^{\circ}C$에서 16일간 발효시키는 것이 제일 좋은 것으로 사료된다.

갈근 추출물이 난소 적출로 비만이 유발된 Rat의 체중 변화와 유전자 발현에 미치는 영향 (Effects of Pueraria lobata on Body Weight and Gene Expression in Obese Rats Muscle with Estrogen Deficiency)

  • Objectives: GG is the EtOH fraction of extract of Pueraria lobata. In this study, we aimed to elucidate a possibility that GG reduce obesity and obesity-derived complications such as cardiovascular and metabolic disease. Methods: The effects of GG on the estrogen-deficient obese rats and the level of gene expression in muscle of rats were investigated. Results: GG decreased body weight in obese rats with estrogen deficiency. GG increased leptin gene expression in obese rats with estrogen deficiency. GG decreased TNFa gene expression in obese rats with estrogen deficiency. And GG increased PPAR-gamma, PGC-1a, Prdx6, FDFT1, and ACC gene expression of those in obese rats. Conclusions: We conclude GG might reduce body weight and regulate gene expression of muscle in obese rats.

양릉천 갈근 약침이 흰쥐의 고지혈증에 미치는 영향 (The Effect of Puerariae Radix Herbal-acupuncture at $GB_{34}$(Yangleungcheon) on Hyperlipidemia in Rats)

  • Objectives & Methods : The purpose of this study is to observe the effects of Puerariae Radix Herbal-acupuncture at $GB_{34}$(Yangleungcheon) on hyperlipidemia in rats. The author made several items to compare body-weight of the rats, weight of its liver, serum, enzymes in liver as well as the histological changes in each group. Results : 1. In the Puerariae Radix herbal-acupuncture group, there was a significant improvement of the levels of total cholesterol, HDL-cholesterol, atherosclerosis index and phospholipid in serum as compared with the control group but there was no significant difference as compared with the NP or saline group. 2. There was a significant improvement of HMG-CoA reductase activity and catalase level in Puerariae Radix herbal-acupuncture group as compare with the control group, the NP group and the saline group. 3. The pathohistological changes of liver and aorta was suppressed in the Puerariae Radix herbalacupuncture group than the control group. Conclusions : From the above results, it is suggested that Puerariae Radix herbal-acupuncture at GB34 has a therapeutic effect on hyperlipidemia.

곽향과 갈근 복합제제의 대장암 세포주 HT-29 증식 저해효과 및 $PGE_2$ 생성 억제효과 (Inhibitory Effect of Mixture of Ethanol Extracts in Agastachis Herba and Pueraria Radix on the Proliferation and $PGE_2$ Production of HT-29 Human Colon Cancer Cell Line)

  • Ethanol extracts of the whole herb of Agastachis Herba (A) and of Pueraria Radix (P) alone and of their mixture (A+P) downregulated the cell growth, cyclooxygenase-2 (COX-2) expression, prostaglandin $E_2\;(PGE_2)$, and cGMP production. A, P, and A + P inhibited the cell growth of HT-29 colon cancer cells in a concentration- and time-dependent manner but not the growth of normal colon cell, CCD-112CoN. In addition, they markedly inhibited the productions of $PGE_2$ and cGMP as well as the mRNA expression of COX-2. These data suggest that non-toxic concentration of A, P, and A + P have a significant effect on the in vitro growth of HT-29 cells, specifically through the inhibition of the $PGE_2$ production via COX-2.

갈근해주탕(葛根解酒湯) 합(合) 단주환(斷酒丸)과 해주지갈탕(解酒枳葛湯)이 실험적(實驗的) Alcoholism 생쥐의 행동(行動)에 미치는 영향(影響) (The Effects of Galgunhaejutang + Danjuhwan and Haejujigaltang on the Experimental Behaviors of Alcohol-Intoxicated Mice)

  • This experiment was designed to investigate the effects of Galgunhaeiutang + Danjuhwan and Haejujigaltang on the experimental behaviors of alcohol-inticxicated mice. ICR mice were used as subjects for alcohol intoxication in this experiment and sample group received Herb Medicine. The foot-step and rearing score were measured in mice. Also Rota-Rod treadmill & number of successful trials swimming time in the T-maze were done. And the following results were observed; 1. The food-step score was decreased in Galgunhaejutang + Danjuhwan group at the 8th hour, the difference was statistically significant. But Haejujigaltang group showed no significant difference. 2. The number of rearing was decreased in Galgunhaejutang + Danjuhwan group at the 3rd and 8th hour, the difference was statistical significant, while Haejujigaltang group showed no significant differnce. 3. The motor incoordination measered by the Rota-Rod treadmill was decreased in Galgunhaejutang + Danjuhwan group at the 8th hour, but Haejujigaltang group showed no significant difference. 4. The number of successful trials in T-maze showed no significant difference in both groups. 5. The passing time in T-maze was increased in Galgunhaejutang +Danjuhwan group at the 8th hour, but Haejujigaltang group showed no significant difference.

갈근추출액이 알레르기 염증반응에 미치는 영향 (The Effects of Puerariae Radix on Allergic Inflammation)

  • Objectives To evaluate that Okwada affected which factors for treatment of lung fibrosis. Methods Bleomycin induced lung fibrosis model made in mice. After Okwada lyophilized, power sample obtained and melt in distilled water. Okwada solution administered mice through oral route on 21 days after bleomycin instillation and this procedure performed once a day for 7 days. We divided by three groups; normal (control), bleomycin induced lung fibrosis without treatment (experimental), bleomycin induced lung fibrosis with treatment (treatment). On six weeks after bleomycin instillation, mice sacrificed and removed lung. We performed Western blot analysis for TGF-beta, phosphodiesterase 5A, interleukin (4,5,13) and compared therapeutic effects of Okwada. Results On western blot analysis, all normal and experimental mice detected TGF-beta, phosphodiesterase 5A, interleukin 4,5,13. The amount of band of TGF-beta, phosphodiesterase 5A, interleukin 5 in experimental and treatment group was similar. However, interleukin 4,13 of treatment group decreased compared with experimental group. Conclusions Okwada would be effected the lung fibrosis through suppression of interleukin 4,13.

갈근해기탕(葛根解肌湯)의 항 Allergy 및 항염증 효과 (Inhibitory Effect of Galgeunhaegi-Tang on Compound48/80 Stimulated Allergic Reaction)

  • The present study was conducted to investigate the anti-allergic activity of Galgeunhaegi-Tang(GHT). We investigated the anti-allergic effects of GHT in RBL-2H3 basophilic leukemia cells by compound48/80, a mast cell degranulator and compound 48/80 induced anaphylactic shock in mice. GHT significantly inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. In addition, GHT effectively inhibited anaphylactic shock in mice by 40% at a dose 100 mg/mouse versus PBS treated control after the l.p injection(8 mg/kg) of compound 48/80. The in vitro anti-inflammatory activities of GHT in LPS-stimulated RAW 264.7 cells were investigated. GHT inhibited NO production in LPS-stimulated RAW 264.7 cells and effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. These result provide evidences that GHT may be beneficial in the treatment of allergic inflammtory disease.

인체 모유두세포의 증식에 미치는 갈근 에탄올추출물의 효과 (Effect of Puerariae Radix Ethanol Extract on the Proliferation of Human Dermal Papilla Cells)

  • In this study, we investigated the effect of Puerariae Radix ethanol extracts (EPR). The effect of the EPR on proliferation of human hair dermal papilla cells(HHDPCs) by MTT assay and observed Expression of mechanisms that regulate cell proliferation extracellular signal-regulated kinase(ERK) and Akt by western blot. The results showed EPR increased the proliferation of HHDPCs and up-regulation phosphorylation of ERK and Akt. ERK and Akt increased by EPR inhibited phosphorylation by PD98059 (ERK inhibitor) and LY294002 (Akt inhibitor), and cell proliferation was also inhibited. These results suggested EPR increases the proliferation of HHDPCs through phosphorylation of ERK and Akt, and therefore is a beneficial effect for the alopecia treatment.

HPLC에 의한 한국산 칡차의 분석 (Studies on the Content of Pueraria Radix in the Tea by HPLC)

  • 시판 갈근으로부터 Isoflavanoid 성분인 Daidzein과 Daidzin을 분리 정제하고 이를 표준물질로 하여 시판 칡차 8종에 대한 칡성분 분석을 HPLC 방법으로 시도하여 다음과 같은 결론은 얻었다. 1. 칡차의 성분 분석법은 종래의 TLC법에 비하여 HPLC를 이용, 신속하고 정확하게 분석할 수 있었다. 2. 추출방법에 따라 성분함량이 크게 영향을 받아 Daidzein을 분석할 경우는 냉침법이, Daidzin의 분석시는 가열 추출법이 양호하였다. 3. 한국산 갈근중 Daidzein과 Daidzin은 0.08% 및 0.66% 함유되었다. 4. 시판 칡차중 고형차류는 칡의 성분이 표기량에 비하여 매우 적게 함유되거나 전혀 함유되지 않은 예도 있으며 액상차의 경우는 엑기스 고형분이 4∼5%가 함유되었다.
