갈근(葛根)과 숙지황(熟地黃) 추출물의 cisplatin에 의한 rat mesangial cell의 apoptosis에 대한 보호효과 (Preventive Effect of Puerariae Radix and Rehmanniae Radix Preparata on Cisplatin-induced Rat Mesangial Cell Apoptosis)

  • One of the major side effects of cisplatin is nephrotoxicity, leading to acute renal failure. Recent study has suggested a role of hydroxyl radicals and p53 in renal cell injury by cisplatin. This study determined the possible involvement of oxidative stress in p53 activation. In rat mesangial cells, cisplatin treatment induced apoptosis and p53 activation. Pifithrin-$\alpha$, a pharmacological inhibitor of p53, suppressed cisplatin-induced apoptosis. Cisplatin also induced reactive oxidative species (ROS) generation. Of interest, cisplatin-induced apoptosis was prevented by N-acetyl-cysteine (NAC), a general antioxidant. NAC diminished p53 activation during cisplatin treatment. Puerariae Radix and Rehmanniae Radix Preparata with antioxidative activity were reduced the cisplatin-induced ROS generation, caspase-3 activity and p53 activation. In conclusion, ROS may contribute to p53 activation to initiate cisplatin-induced apoptosis in rat mesangial cells. In result, antioxidative effect of Puerariae Radix and Rehmanniae Radix Preparata prevented cisplatin-induced apoptosis through inhibition of p53 activation.

갈근으로부터 분리한 암세포 증식저해물질 (Anti-proliferative Components in the Roots Extract from Pueraria thunbergiana)

  • The antitumor activity of the roots extract of Pueraria thunbergiana was investigated on the basis of cytotoxicity upon the cultured human tumor cell lines, in vitro. The purification of methylene chloride (MC) soluble part and ethylacetate (EA) soluble part of extract by column chromatography furnished seven isoflavonoids, two triterpenoids, one but-2-enolide. The structures of them were established by chemical and spectroscopic means to be lupeol (1), $\beta$-sitosterol (2), biochanin A (3), (-)-tuberosin (4), calycosin (5), daidzein (6), puerarin (7), daidzin (8), (+)-puerol-B 2-O-$\beta$-glucopyranoside (9), formononetin-7-O-$\beta$-glucopyranoside (10). Each isolates ($1{\sim}10$) were evaluated for inhibitory activities on the proliferation of cultured human tumor cell lines such as A549, SK-OV-3, HCT-15 and SK-MEL-2, respectively.

갈근탕의 사염화탄소에 의한 간세포 독성 억제효과 (Protective Effect of Galgeun-Tang Against $CCl_4$ Induced Hepatotoxicity)

  • Galgeun-tang (GGT) has been a great source for treating cold diseases in the folk medicine recipe. Carbon tetrachloride ($CCl_4$) is one type of hepatotoxin that can eventually cause liver injury. During the experiment, we first studied the protective effects of GGT against $CC_4$-induced hepatotoxicity. GGT was pretreated for 3 h, and 1% $CCl_4$ was added to mouse primary liver cells. After 4 h, ROS generation and expression of antioxidant enzymes (catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx)) were analyzed by FACS and real time PCR. Also, the activities of ALT and LDH were measured using cultured medium. The hepatic levels of TNF-alpha and iNOS, which are related to inflammation and stress response gene, HSP72 and HO-1 were analyzed by PCR or real time PCR. Liver tissues were analyzed by HE stain. From the observation, we discovered that GGT treatment protects $CCl_4$-induced hepatotoxicity, and that GGT pretreatment decreases ROS generation, TNF-alpha and iNOS expression. However, gene expression of CAT, SOD, GPx, HSP72 and HO-1 were increased by GGT. These results lead to the conclusion that GGT has protective effects against $CCl_4$-induced hepatotoxicity.

  • Puerariae radix (PR) is a popular natural herb and a traditional food in Asia, which has antithrombotic and anti-allergic properties and stimulates estrogenic activity. One of the major side effects of cisplatin is nephrotoxicity, leading to acute renal failure. Recent study has suggested a role of ROS and p53 in renal cell injury by cisplatin. We studied that protective effects of PR on cisplatin-induced apoptosis in rat mesangial cell. Rat mesangial cell was preincubated with PR (50, 100, 150 and 200 ${\mu}g/m{\ell}$) for 12 hr and then treated with 30 ${\mu}M$ cisplatin for 24 hr. Protective effect of PR on cisplatin-induced apoptosis in ECV304 cells was determined using MTT assay, FDA-PI staining, flow cytometric analysis, caspase-3 activity assay, ROS assay and western blot. Our results showed that PR inhibited in cisplatin-induced apoptosis and ROS production in ECV304 cells. Moreover, PR reduced ERK, p38 and JNK activation that increased in cisplatin-treated rat mesangial cell. Furthermore, activation of p53 by cisplatin in rat mesangial cell was inhibited by PR treatment. These results suggest that protective effect of PR on cisplatin-induced apoptosis in rat mesangial cell may be associated with reduction of ERK, p38, JNK, p53 activation.

  • The radix of Pueraria thunbergiana BENTHAM (Leguminosae) is traditionally prescribed to attenuate the clinical manifestations of inner ear dysfunction and various clinical situations including fever, gastrointestinal disorders, skin problems, migraine headaches, lowering cholesterol and treating chronic alcoholism in Oriental Medicine. In the present study, we examined the effect of ethanol extract of P. thunbergiana radix (EPR) on cisplatin-mediated HEI-OC1 auditory cell death. In addition, to investingate the protection mechanism of EPR on free radicals. Treatment of EPR protected cells from cisplatin and reduced lipid peroxidation in a dose-dependent manner. Furthermore, EPR demonstrated significant scavenging activity against various free radicals, including superoxide radical, hydroxyl radical, hydrogen peroxide, and DPPH radical. These results indicate that EPR protects cisplatin-induced damages of HEI-OC1 cells through inhibition of lipid peroxidation and augmenting scavenging activities against free radials.

유기산을 이용한 한약재의 효소적 갈변방지 (Inhibition of Enzymatic Browning in Medical Herbs (Crude Drug Materials) by Organic Acid)

  • 한약재의 효소적 갈변방지를 위하여 기존의 방법(연탄건조, 유황훈증, 아황산염류 처리 등의 건조가공법)을 대체하여 한약재의 안전성을 확보할 수 있는 새로운 갈변방지 기술의 개발이 필요하였다. 먼저 유기산 및 그 유도체를 대상(구연산 외 12종)으로 한약재의 갈변방지 효과를 실험한 결과, 1% 수준에서 구연산, 아스코르빈산, 이소아스코르빈산, 초산 순으로 효과가 있었다. 그 중에서도 구연산과 아황산염 ($Na_2S_2O_5$)에 대하여 독활, 갈근, 작약을 대상으로 비교 실험했을 때, 구연산은 아황산염 ($Na_2S_2O_5$)과 유사한 갈변방지 효과를 보였다. 앞으로 한약재의 갈변방지를 위하여 구연산의 사용은 기존의 방법을 대체할 수 있는 새로운 기술로 기대된다.

인진(茵蔯)과 갈근(葛根)이 d-galactosamine, 급성(急性) alcohol중독(中毒) 및 $CCl_4$중독(中毒) 백서(白鼠)의 간손상(肝損傷)에 미치는 영향(影響) (Studies on the effects of PUERARIAE RADIX(葛根) and ARTEMISIAE HERBA(茵蔯) on experimental liver damages induced by alcohol, d-galactosamine and $CCl_4$)

  • Recent survey shows that chronic liver disease such as chronic hepatitis, liver cirrhosis and hepatoma is the third leading causes for death in Korea. In oriental medicine, viral hepatitis is related to Hwangdal(黃疸) and alcoholic liver disease is related to Joosang(酒傷). ARTEMISIAE HERBA and PUERARIAE RADIX have long been used in treating those symptoms. This study was done to evaluate the effect of AR1EMISIAE HERBA and PUERARIAE RADIX on viral and alcoholic hepatitis. ARTEMISIAE HERBA and PUERARIAE RADIX were decocted respectively with water and followed by vaccum evaporation. The solution was diluted to adequate concentration. Sprague-Dawley rats were used in this experiment. Each group was given PUERARIAE RADIX or ARTEMISIAE HERBA solution orally and CCl4, d-galactosamine or alcohol was given orally 30 minutes later. After 24 hours of starvation, blood samples were taken to check serum GOT, GPT, LDH and ALP activities, TC, TG, glucose and BUN levels. These results show that ARTEMISIAE HERBA has better effect on liver injury induced by d-Galactosamine than PUERARIAE RADIX and that both ARTEMISIAE HERBA and PUERARlAE RADIX have good effect on acute alcoholic liver disease while in the liver injury induced by $CCl_{4}$, PUERARIAE RADIX has better inhibitory effect on serum AST, ALT and ALP levels and ARTEMISIAE HERBA has better inhibitory effect on serum total cholesterol and triglyceride. And the result that high concentration group has better effect shows these effects are concentration-dependent. Further study on the mechanism of these herbs is still required.

원인 불명열 환자를 갈근해기탕(葛根解肌湯)으로 치험한 1례 (A Clinical Study of the Treatment Prescribed Kalkunhaeki-tang for FUO (Fever of Unknown Origin) Patient)

  • 1. Objectives FUO is defined as a temperature higher than $38.3^{\circ}C$ on several occasions and lasting longer than 3 weeks, with a diagnosis that remain uncertain after 1 week of investigation. Diagnosing the cause of FUO is particularly difficult. 2. Methods We have tried researching effect of herbal medicines(Kalkunhaeki - tang treatments) and Acupoint Injection to Taeumin FUO patient. 3. Results A Taeumin patient diagnosed to FUO was treated with Taeumin herbal medicines(kalkunhaeki - tang treatments) and change of patient's fever and general condition were improved. 4. Conclusions The constitutional treatment with Kalkunhaeki - tang may have an effect on management of FUO. So we report the healing process and results of this patient in this study.

갈근해기탕(葛根解肌湯)의 기원(基源), 변천(變遷), 구성(構成)에 대한 고찰(考察) (The Study on The Origin, Changes, and Composition of Galgeulhaegi-tang)

  • 1. Objectives : This paper was written in order to understand the origin and changes of Galgeunhaegi-tang. 2. Methods : We analysis Galgeunhaegi-tang through pathology and new prescription in "Donguibogam(東醫寶鑑)", "Chobongwon(草本卷)", "Gabobon(甲午本)" and "Sinchukbon(辛丑本)" of "Dongyi Suse Bowon". 3. Results and Conclusions : We can find the first origin of Galgeunhaegi-tang in "Sinchukbon(辛丑本)" in "Donguibogam(東醫寶鑑)" which invelves Galgeunhaegi-tang in the categories of pestilence(瘟疫) and Yangmyeonggyeongbyeong(陽明經病). Also, we can find the origin in Jugoeng(朱肱)' Jojung-tang. Seungeumgalgeun-tang in the "Chobongwon(草本卷)" developed by a Galgeunhaegi-tang of "Gabobon(甲午本)", and Galgeunnabokja-tang in the "Chobongwon(草本卷)" developed by a Galgeunhaegi-tang of "Sinchukbon(辛丑本)". It is considered that Galgeunhaegi-tang of "Gabobon(甲午本)" solves heat depression(鬱熱), due to not throwing off, by using Galgeun(葛根) and Daehwang(大黃), and accompanied dryness(燥) by using Hangin(杏仁) and Sanjoin(酸棗仁) which moisten dryness(燥). It is considered that Galgeunhaegi-tang of "Sinchukbon(辛丑本)" solves heat depression(鬱熱) by using Galgeun(葛根) and accompanied dryness(燥) by using Gobon(藁本) which sets free dryness(燥).

갈근분말 첨가 냉동쿠키의 제조조건 최적화 (Optimization of Iced Cookie with Arrowroot Powder Using Response Surface Methology)

  • The aim of this study was to use arrowroot powder to develop a recipe for a nutritional cookie having the optimal composition of ingredients and texture to elict a strong preference response in all age groups. Wheat flour was partially substituted with arrowroot powder to reduce the wheat flour content of the cookie. Measurements were made and analyzed according to the Response Surface Methodology technique, which showed 16 experimental points including 2 replicates for arrowroot powder, yellow sugar and butter. The compositional and functional properties of the test were measured, and the values obtained were applied to a mathematical model. A canonical form and perturbation plot showed the influence of each ingredient on the final mixture product. The result of sensory evaluation showed very significant values for color (p<0.01), appearance (p<0.001), texture (p<0.05), overall quality (p<0.05), and flavor (p<0.05). Instrumental analysis showed significant values for lightness (p<0.001), redness (p<0.01), yellowness (p<0.01) and spread ratio (p<0.001). The optimal sensory ratios were determined to be 15g for the arrowroot powder, 70 g for sugar and 80 g for butter.
