Gene Expression Analysis of Phenylbutazone-induced Liver Damage in Mice (페닐부타존에 의해 간손상이 유발된 생쥐의 유전자 발현 분석)

  • The KFDA (Korea Food & Drug Administration) has performed a collaborative toxico-genomics project since 2003. Its aim is to construct a toxicologenomic database of 12 hepatotoxic compounds from mice livers. Phenylbutazone which is non-steroidal anti-inflammatory drug was assigned. It was administered at low (0.0238 mg/kg) and at high (0.238 mg/kg) dose (5 mice per group) orally to the postnatal 6 weeks ICR mice, then the serum and liver were collected at the indicated time (6, 24 and 72 h) after administration. Serum biochemical markers for liver toxicity were measured and histopathologic studies also were carried out. The gene expression profiling was carried out by using Applied Biosystems 1700 Full Genome Expression Mouse. The 2-way ANOVA was used to find genes that reflected phenylbutazone-induced acute toxicity or dose-dependant changes. By self-organization maps (SOM), we identified groups with unique gene expression patterns, some of them are supposed to be related to phenylbutazone induced toxicity, including lipid metabolism abnormality, oxidative stress, cell death and cytoskeleton destruction.

  • The Flavin-containing monooxygenase (FMOs) (EC1.14.13.8) are NADPH-dependent flavoenzymes that catalyze oxidation of soft nucleophilic heteroatom centers in a range of structurally diverse compounds, including foods, drugs, pesticides, and other xenobiotics. In humans, FMO3 is quantitatively a major human liver monooxygenase. In the present study, the baculovirus expression vector system was used to overexpress human FMO3 in insect cells for catalytic studies. Six commercially available organic selenium compounds were examined for substrate activity with microsomes isolated from Spodoptera frugiperda (Sf)9 cells infected with human FMO3 recombinant baculovirus. While none of the aromatic heterocyclic selenides tested showed detectable activity, all dialkyl- and alkylaryl-selenides free from ionic groups catalyzed the NADPH- and O$_2$-dependent oxidation. Kinetic constants demonstrate that (based on Km) dialkyl-and alkylaryl- selenides are better substrates for human FMO3 than analogous nitrogen or sulfur compounds .

Accumulation of Tributyltin (TBT) in Blood, Liver and Muscle of Olive Flounder (넙치의 혈청, 간 및 근육 중 Tributyltin 축적 경향)

  • Accumulation of tributyltin (TBT) in serum, liver and muscle of olive flounder (Paralichtys olivaceus) was investigated in a 60-d static renewal exposure at $0.1{\mu}g$ TBT/L. Tributyltin accumulated rapidly from 83 ng Sn/g to 2,227 ng Sn/g on a wet weight basis in the serum of the olive flounder and to a greater extent than in the other tissues. The accumulated TBT concentrations in tissues were in the order of serum>liver>muscle on wet or dry-weight basis. High concentrations of dibutyltin (DBT: 990 ng Sn/g dry wt) and monobutyltin (MBT: 141 ng Sn/g dry wt), degradation products of TBT were found in liver of olive flounder at the end of exposure. On the other hand, DBT and MBT was below the detection limits in muscle during the exposure, and only low concentration of DBT (56 ng Sn/g) were detectefd in serum. Butyltin compounds were also quantitatively determined in feral fine-spotted flounder (Pleuronichthys cornutus) collected from Gwangyang Bay as one of polluted area and Sori Island as a reference site. All three butyltin compounds were detected from the fine-spotted flounder from Gwangyang Bay up to 3,107 ng Sn/g of total butyltins in liver, while 120 ng Sn/g of total butyltin concentration was found in the liver of fish from Sori Island.

  • Hepatic antioxidant defense systems were examined in rats treated with tetrabromobisphenol-A (TBBPA), a brominated flame retardant, at the doses of 0, 250, 500 and 1,000 mg/kg for four weeks. Hepatic ratio of glutathione disulfide to glutathione (GSH) and levels of malondialdehyde, oxidative stress markers were not changed in rats treated with TBBPA. Hepatic expression of antioxidant enzymes including GSH peroxdiase-1 (GPX-1)/GSH reductase (GR), alpha-, mu- and pi-class glutathione-S-transferase (GST) and gamma-glutamylcysteine ligase catalytic subunit was determined using immunoblot analysis. Alpha-class GSTs, GPX-1 and GR levels were significantly decreased in rats treated with TBBPA at the dose of 500 or 1,000 mg/kg. These results show that TBBPA results in down-regulation of hepatic expression of antioxidant enzymes related with GSH, suggesting the liver in TBBPA-treated rats may be more sensitive to oxidants.

Preventive Effects of Chitosan on the Disorders of Hepatic Functions and Lipid Metabolism in Rats Treated with 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD) (다이옥신계 TCDD (2, 3, 7, 8-tetrachlorodibenzo-p-dioxin)에 노출된 흰쥐의 지질대사 및 간 독성물질대사에 관한 키토산의 예방효과)

  • This study was conducted to fine out the preventive effects of chitosan and chitosan oligomer on the disorders of hepatic functions and lipid metabolism induced by 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD) using adult male rats (SD) for four weeks. Rats were fed chitosan ($4\%$) or chitosan oligomer ($4\%$) diets respectively before 3weeks of TCDD treatment (50 ug/kg BW) by intraperitoneal injection and then continually supplied these diets for one week until being sacrificed. The elevation of serum total and LDL cholesterol levels induced by TCDD treatment was significantly reduced in the rats fed chitosan diets. The increment of liver triglyceride levels caused by TCDD treatment was tended to suppress in all rats fed chitosan and chitosan oligomer diets. Fecal total lipid and cholesterol excretion were high levels in the rats fed chitosan diets. The hepatic cytosolic catalase activities significantly decreased by TCDD treatment appeared recovering trend by chitosan diets. In hepatic microsomal cytochrome p-450, NADPH cytochrome p-450 reductase, ethoxycoumarin-o-deethylase (ECOD) and benzphetamin N-demethylase (BPND) chitosan than chitosan oligomer diets apparently decreased the increasing levels by TCDD treatment. In histochemical observation the fat droplets and apoptosis of hepatocytes by TCDD treatment were markedly alleviated by chitosan and chitosan oligomer diets. These results indicate that chitosan, more than chitosan oligomer can exert preventive effects on some disorders of hepatic functions and lipids accumulation by TCDD.

  • In recent years, we have been working on the improvement project of mercury 3-lecture type at the Geummundo lighthouse for more than 100 years. We have installed toxic mercury removal, LED light source, and dualization system for nonstop. It can be seen as an example. Circular restoration of the entire frame is performed, and the real-time status can be grasped through the installation of the double-ball bearing and the remote control system. After installation, optical measurement through Hanvit secured the optical performance of 2,000,000 cd of floating brightness of 300cd, proving the efficient possibility of introducing the domestic rotary type light source using LED light source.

  • In this study, we evaluated the effect of fibrin, a natural material, on the local inflammatory reaction of PLGA in vivo. PLGA degradation products can decrease the pH in the surrounding tissue, causing local inflammatory reaction. To solve this problem, fibrin/PLGA scaffolds were implanted in 5-week-old Wister rats. To evaluate the influence of fibrin content on inflammatory cytokine expression induced by PLGA, RT-PCR analysis was used. Fibrous wall thickness and macrophage infiltration were evaluated by H&E and ED-1 immunohistochemical staining, respectively. In this study, we showed that fibrin/PLGA scaffolds reduced inflammatory reaction as compared to PLGA scaffold. We concluded that fibrin could reduce inflammatory response of PLGA.

Behavioral and Physiological Responses of Juvenile Red Seabream Pagrus major exposed to Ethanol Seawater (에탄올에 대한 참돔 Pagrus major의 행동 및 생리학적 반응)

  • Behavioral and physiological responses of juvenile red seabream (Pagrus major) to different concentrations of ethanol were investigated. No swimming and no reaction to touching by a wooden stick was observed at 0.6% ethanol group in behavioral response, and survival rate was 100% after 5 hours of treatment. Red blood cell count, hematocrit and hemoglobin levels in plasma were not significantly different among all groups. AST activities in plasma significantly decreased as ethanol concentration increased. On the contrary, ALT activities in plasma significantly increased as ethanol concentration increased. Cortisol level in plasma was the lowest in 0.6% ethanol group. Glucose levels in plasma increased significantly when ethanol concentration increased more than 0.4%. Oxygen consumption of fish in 0.6% ethanol seawater was constantly lower than that of fish in control seawater from 2 hours after the exposure to ethanol seawater until the end of experiment.

  • Population density of the spiders (Micryphantidae and Lycosidae) in the two Paddy field at Suweon and Iri was studied following single root-zone application of carbofurans encapsulea and liquid-formulated and two to four paddy water applications of carbofuran to rice plants. In two field tests, the spider populations were reduced In all insecticide treatments. Among them, the reduction was greater in the capsule placement of carbofuran than in the liquid injection and broadcast of carbofuran. Their reduction was also enlarged with the increase of application rate of insecticide. The significant reduction of spider population in root-tone appliration of carbofuran was considered due to the food-chain toxicity on the spider-hopper system.

  • In this study, we investigated the anti-obesity activity of the herbal extract mixture (HEM). The inhibitory effect of HEM on triglyceride accumulation of 3T3-L1 preadipocyte was examined by Oil-Red O staining. HEM inhibited the triglyceride accumulation of 3T3-L1 preadipocyte cell and reduced glycerol-3-phosphate dehydrogenase (GPDH) activity. We further investigated the effect of HEM in prevention of obesity in male ICR mouse for 5 weeks. Experimental groups were divided into high fat diet group (HFD), HFD supplemented with 100 mg/kg HEM group (HEM1) and HFD supplemented with 200 mg/kg HEM group (HEM2). Body weight and food efficiency ration of HEM1 and HEM2 was decreased by 52% and 50% and by 45% and 50%, respectively. The amount of adipocyte in body weight was decreased. Blood triglyceride and total cholesterol of HEM1 was significantly decreased. These results indicate that HEM may be useful in preventing obesity.
