This research was conducted to determine the effects of chitosanoligosaccharide on liver poisoning induced by cadmium (Cd). Three groups of mice were used in this research. The group was only injected with cadmium (5.0 mg/kg; i.p.) (group Cd) and the other group was injected with cadmium and chitosanoligosaccharide (0.5% solution) at the same time (group Cd+Chi). In order to investigate the inhibitory action of chitosanoligosaccharide on liver damage, cadmium concentration in liver tissues and metallothionein (MT) concentration were relatively measured. In addition, histological observations were made to determine the morphologic injury of liver tissues. Cadmium concentration in liver tissues was drastically lower in groups Cd+Chi than in group Cd. MT concentration in liver tissues was lower in group Cd than in groups Cd+Chi. As the result of electron microscopic observation, mitochondria in group Cd showed a severe swelling phenomenon, RER fragment and ribosome dropout. However, in groups Cd+Chi, mitochondria with high electron density were distributed and RER forming a typical lamellae with ribosome was observed. From these results, cadmium toxicity on rat liver tissues could be lessened by chitosanoligosaccharide.
To investigate the effect of the squalene against the lead toxicity. A healthy male of ICR mice were used for experiment. The SOD was observed after the intraperitoneal injection in mice. The ultrastructural changes of the liver were observed after the intraperitoneal injection in mice. The experimental groups were divided into two groups. Group A was control group that squalene was not treated after intraperitoneal injection of lead acetate. Group B was squalene treatment group that squalene solution was injected after intraperitoneal injection of lead acetate. All groups were used to 10 mice. The results were as follow: SOD activity in the liver, Group A was lower than in normal. But, Group B was higher than in Group A (P<0.05). In the histological observation, Group A were showed that the inner cavity of mitochondria swellen and development of cristae weakened. Swelling of lamellae of rough endoplasmic reticulum (rER) was showed. It was concluded that the SQ will be effective for the recovery of hepatic cell at lead intoxication.
Sodium arsenite (As) aqueous solution containing 4 ppm wss given to male rats for 15 days as drinking water. Electrophoretic pattern of liver cytosol from As-fed rats appeared to be significantly different from that of the control rats. Although the normal protein content of the cytosol fraction of As-fed rat liver was decreased, 8 stress proteins were increased. In liver cytosol fractions of As-treated rat, one kind of arsenic-binding substance (AsBS) was observed. Molecular weight of AsBS was identified to be 500 D and composition of amino acid was glycine, glutamic acid and cysteine. Glutathione (GSH) appeared to bind to arsenic and GSH-As complex showed the same mobility as AsBS on gel filtration chromatography. GSH conjugated As prevented As from inhibiting respiration, conformational change and swelling-contration of mitochondria. According to the above results. it is concluded that in vfuo treated arsenic stimulated synthesis of stress protein, and arsenic-binding substance might be glutathione and have a protective role against arsenic toxicity.
This paper reports on Pb and Cd concentrations in the tissues of 76 individuals of 16 rare bird species found dead across Korea. In most of species, Pb concentration was higher in kidney and bone than in liver. The range of the mean liver Pb was 0.47 (Cygnus cygnus)∼3.74 ㎍/wet g (Asio flammeus). Based on hepatic Pb concentrations, 39 (51.3%) of the 76 individuals were classifed as background level (〈2㎍/wet g), 35 (46.1%) were elevated level (〉2㎍/wet g) and 2 (2.6%) were poisoned level (〉10㎍/wet g). These results were indicated many rare birds were suffered from chronic Pb exposure. Cd concentration was generally higher in kidney than in liver and bone. The range of the mean kidney Cd was 0.06 (Bubo bubo)∼l.75㎍/wet g (falco subbuteo). Only one of the 76 individuals had elevated Cd concentration (〉2.7㎍/wet g) in kidney. These results indicated most of rare birds had low Cd accumulations.
The flavin-containing monooxygenases(FMOs) (EC1.14.13.8) are NADPH0dependent flavoenzymes that catalyze oxidation of soft nucleophilic heteroatom centers in a range of structurally diverse compounds, including foods, drugs, pesticides, and other xenobiotics. In humans, FMO3 is quantitatively a major human liver monooxygenase. In the present study, the baculovirus expression vector system was used to overexpress human FMO3 in sect cells for stalytic studies. Microsomes isolated from Spodoptera frugiperda(Sf)9 cells infected with human FMO3 recombinant baculovirus catalyzed the NADPH-and O$_2$-dependent oxidation of methimazole, thiourea, and phenylthiourea. However there was no detectable activity with 1, 3-diphenylthiourea or larger thiocarbamides. Microsomes from control Sf9 cells were devoid of methimazole or thiourea S-oxygenase activity. 1, 3-diphenylthiourea is apparently completely excluded from the catalytic site, these amines drugs are probably approaching the upper size limits of xenobiotics accepted by human FMO3. The substrate specificity of this iosform in humans appears considerably more restriceted than that of pig, guinea pig, rat or rabbit FMO3.

The hepatoprotection by the methanol extract of Oenanthe javanica DC (water dropwort) (OJME) was investigated in Sprague Dawley rats with inducing liver damage by acetaminophen. After OJME administration for 1 week, the increase of hepatic lipid peroxide level by acetaminophen-induced hepatotoxicity was significantly reduced. In case of phase I microsomal enzyme systems including cytochrome P-450, aminopyrine N-demethylase and aniline hydroxylase, any significant differences between in control and in OJME-pretreated group was observed after acetaminophen treatment. However, the pretreatment of OJME maintained the hepatic glutathione level and the activity of liver cytosolic glutathione S-transferase, which was significantly decreased by the acetaminophen intoxication. Among the glutathione-generating system, glutathione reductase was more responsible for its biosynthesis rather than ${\gamma}-glutamylcystein$ synthetase. OJME itself showed the strong inhibition activity on DPPH radical generation. In conclusion, OJME administration maintains the liver glutathione pool and hepatic glutathione S-transferase activity, in addition with its high anti-oxidative capability, to show hepatoprotective effect from acetaminophen intoxication.
BACKGROUND: The purpose of this study was that serum metabolic and hematological variables were measured to investigate the detoxication effect of Taraxacum mongolicum extract on male albino rat exposed with lead and mercury. METHODS AND RESULTS: For this study, Pb and Hg-exposed( 50 ppm) albino rats was used, and the treatments were carried out in three doses of 100 mg/kg, 200 mg/kg and 500 mg/kg as dry weight of T. mongolicum extract. T. mongolicum extract could improved the body weight gain and feed efficiency ratio, except to food intake. The levels of biochemical factors elevated by Pb-Hg mix exposure, which are Bilirubin, Alkaline phosphatase(ALP), glutamic oxaloacetate transaminase(GOT), glutamic pyruvate transaminase (GPT), lactate dehydrogenase(LDH), ceatinine and blood urea nitrogen(BUN), were significantly reduced in all treated groups as compared to Pb-Hg mix exposure alone. T. mongolicum extract was shown to suppress the accumulation of Hg and Pb in serum by dose dependent manner. CONCLUSION: Therefore, this study suggest that T. mongolicum extract might have the potential effect to minimize the toxic effects of Pb and Hg.
Recently, cases of Aloe vera induced-toxic hepatitis have been reported. However, the precise inflammatory effects of Aloe vera extract have not been clearly elucidated yet. In this study, the inflammatory effects and the mechanism of 70% ethanolic Aloe vera extract on liver were evaluated by in vitro assays using human hepatoma HepG2 cells. Cell viability was investigated using MTT assay at $0.001{\sim}100{\mu}g/mL$ of Aloe vera extract. To evaluate inflammatory effect of the Aloe vera extract, the secretion of pro-inflammatory cytokine Interleukin 8 (IL-8) and Macrophage colony-stimulating factor (M-CSF) were detected. Aloe vera extract did not induce cell death at concentrations of $0.001{\sim}100{\mu}g/mL$. However, Aloe vera extract significantly increased the IL-8 secretion by 15.7~25.8% and the M-CSF secretion by 36.6~61.5% at the same concentrations. These results indicate that Aloe vera extract shows an inflammation-related mild hepatotoxicity than a severe toxicity such as cell death and this hepatitis is mediated by the secretion of inflammatory cytokine IL-8 and M-CSF.
본 연구는, 그 동안 간과되어 왔던 의류 상품의 판촉에 초점을 맞추어, 판촉의 여러 방법 별 소비자들의 반응 정도를 알아보고, 그 반응 경향이 소비자의 인구통계적 특성, 판촉에 대한 태도, 의복 쇼핑 성향, 기타 의복 구매 행동과 연관성을 가지는지를 규명함을 목적으로 한다. 수도권 거주 여성 444명을 대상으로 수집한 자료를 이용해 분석이 이루어졌다. 실증적 연구를 통해 다음과 같은 결과를 얻었다. 첫째, 판촉의 각 방법에 대한 소비자들의 반응 정도는 가격 할인이 가장 높았고 그 다음은 디스플레이, 쿠폰, 사은품, 백화점 패션쇼, 경품의 순서로 나타났다. 가격 할인, 디스플레이, 사은품, 쿠폰, 백화점 패션쇼 등 5가지 판촉 방법에 대한 소비자 반응 정도를 기준으로 군집분석한 결과 판촉에 대한 반응 유형에 있어 차이를 보이는 5개 집단을 규명할 수 있었다. 각 집단의 인구통계적 특성들 중에서는 소비자 자신의 학력과 자녀의 연령만이 집단간에 유의한 차이를 보였으며, 각 집단의 판촉 태도는 집단 간에 유의한 차이를 보였다. 둘째, 각 집단은 의복 쇼핑 성향 및 의복 구매 관련 정보원 이용에서 차이를 보였다. 결론에서는, 실증적 연구의 결과 판촉에 대한 소비자 반응 유형별로 인구통계적 특성, 판촉 태도, 의복 쇼핑 성향, 기타 의복 구매 행동에 차이가 있음을 알 수 있었으므로 의류 상품의 판촉 전략 수립시 표적 집단의 특성에 따라 판촉 방법 선택이나 판촉 정보 전달 경로 등을 조절하는 것이 필요함을 제언하였다. 예를 들어, 신중/경제적 쇼핑 성향을 가지며 판촉에 대한 태도가 호의적인 소비자들에 대해서는 다양한 정보 전달 경로를 통해 가격 할인이나 쿠폰을 제공하는 것이 좋은 판촉 방법이 될 것이며, 쾌락/유행 지향적이고 상표/품질을 중시하는 소비자들에 대해서는 잡지, 인적 정보, 카탈로그 등을 정보 전달 경로로 하여 디스플레이나 패션쇼를 통해 판매 촉진을 할 수 있을 것이다.0.10 $h^{-1}$과 0.09$h^{-1}$h-1이었고, 반감기는 각각 6.93 및 7.70시간으로 측정되었다. 각각의 농도에서 12시간 및 8시간 이후에는 g당 0.19$\mu\textrm{g}$ 및 0.18$\mu\textrm{g}$이하로 떨어짐을 알 수 있었다. Phosphamidon과 profenofos의 급성어독성은 profenofos가 높았고, BCF profenofos가 phosphamidon보다 약 100배 정도 높게 나타났으며, 배설속도는 phosphamidon이 profenofos보다 약 2배 정도 빨랐다.
Studies on the food-safety of pheasant and mallard, which belong to wild fowl as new meat resources. were carried out. The results were summarized as follows : 1. Food poisoning bacteria including Salmonella spp, was not detected from the inspections of small intestine, cecum, and rectum. 2. Parasite inspection tests on blood, feces, digestive organ, and thoracic organs were negative. 3. Antibiotic residues from the carcass muscle by simplified disk methods were not detected. 4. Seven different pesticide residue tests, such as DDT and BHT, on the muscle and liver were negative. 5. Four different kinds of toxic heavy metals such as Cd were much lower than the permissible concentration. Studies on the food safety tests and inspections from the pheasant and mallard were revealed that from taking this new food resources, the toxicities would be very low for the human health by the direct influences.
