Studies on the isloation and action mechanism from natural folk herbs (천연 민간 생약으로부터 항암물질의 분리동정 및 그작용기전에 관한 연구)

  • 배풍등, 등혹 및 ginsenoside Rh$_1$의 간암세포에 대한 세포독성작용, 인삼 Rh$_1$의 세포보호작용, 비파의 ursolic acid 생체방어기전 활성화 및 산화억제작용, ononin의 radical 제거능을 검토하였다. 그 결과 배풍등 CHCl$_3$분획 및 등혹 CHCl$_3$분획의 간암세포에 대한 강한 세포독성작용을 나타내었으며 배풍등 CHCl$_3$은 sarcoma 180 이식 종양조직의 성장을 유의성있게 억제하였다. Ursolic acid는 지질과산화, 단백질 산화억제와 catalase, GSH S-transferase를 활성화시켰다. 인삼 saponin은 SOD 및 nonprotein-SH를 증가시키고, 지질과산화를 억제시켰다. Ginsenoside Rh$_1$ 및 Rh$_2$는 각각 radical에 대한 세포보호작용과 간암세포 세포독작용을 나타내었다.

Effect of Acer tegmentosum M. Extracts on Hepatocarcinoma Cell (산겨릅나무 추출물의 간암세포의 증식억제 효과)

  • The objective of this study was to investigate the anticancer effects of Acer tegmentosum M. extracts. HepG2 hepatocarcinoma cells were treated with ethanol, chloroform, ethylacetate, butanol, aqueous fraction and hot water extract. The antiproliferative effect was evaluated by trypan blue exclusion, MTT-based viability assay and morphology. The trypan blue test showed that anticancer effect of the A. tegmentosum M. extracts on HepG2 cells increased gradually in proportion to the increasing concentration of the fractions. The butanol fraction showed the highest anticancer activity against HepG2 cells (p<0.05). The MTT assay indicated that the growth inhibition by the butanol fraction was dose-dependent. These results suggest that A. tegmentosum M. has the potential to inhibit the growth of hepatocarcinoma cells.

The Interaction between HCV-Infected huh7.5 Cells and HCV-Specific T Cells (C형 간염 바이러스 감염 간암 세포주와 T 림프구의 상호작용에 대한 연구)

  • Recently, Hepatitis C virus (HCV) replication system has been established using human hepatoma cells (huh cell) and a variety of HCV clones. In this study, we established an infectious HCV replication system using huh7.5 cells and J6/JFH1 clone (genotype 2a). In addition, we investigated the antigen presentation capability of HCV-infected huh7.5 cells to HCV-specific T cells. Interestingly, HCV-infected huh7.5 cells were not capable of activating HCV-specific T cells. However, huh7.5 cells stimulated by exogenous HCV peptide were able to activate HCV-specific T cells, which was shown to produce TNF-${\alpha}$ and IFN-${\gamma}$. We further examined if HCV infection has an inhibitory effect on the expression of MHC class I molecule of huh7.5 cells. We found that HCV infection did not change the expression level of MHC class I molecule on huh7.5 cells.

Quinone Reductase Inductive Activity and Growth Inhibitory Effect against Hepatoma Cell of Oriental Melon Extract (참외 추출물의 Quinone Reductase 유도활성 및 간암세포 증식 억제효과)

  • This study was performed to elucidate anticancer activities of various parts, such as peel, flesh, placenta, seed, stalk and stem leaf of oriental melon. Chemopreventive and anticancer effects of oriental melon extract were evaluated by detoxifying enzyme, quinone reductase (QR) inductive activity, cytotoxicity and growth inhibitory effect against hepatoma cell. Stalk and stem leaf extracts of oriental melon showed the increment of QR inductive activity with dose-dependent manner and induced quinone reductase 3.9, 1.5-fold at $200{\mu}g/mL$ respectively compared to control. The growth inhibitory effect of oriental melon extract against mouse hepatoma cell (Hepa1c1c7) was investigated by crystal violet (CV) assay. Stalk and stem leaf of oriental melon showed potent growth inhibitory effect. Based on these result, the growth inhibitory effects of stalk, stem leaf at various concentration were examined in detail by MTT assay using human hepatoma cancer cell (HepG2). All of two parts showed growth inhibitory effects and expecially stalk exhibited inhibitory effect of 60.3% at maximum concentration. The above results suggest that stalk of oriental melon has a possibility as a source of natural cancer chemopreventive materials.

The Anti-cancer Effects of Bigihwan, Daechilgithang, and Mokwhyangbinranghwan Ethanol Extracts in Human Hepatocellular Carcinoma Cells (인체 간암세포에서 비기환(肥氣丸), 대칠기탕(大七氣湯) 및 목향빈랑환(木香檳榔丸)의 항암 활성 비교)

  • Hepatocellular carcinoma (HCC) is one of the most commonly diagnosed cancers in the word. Although radiation and chemotherapy are generally effective, there are various side effects that greatly limit the effectiveness of these treatments. Therefore, traditional herbs may have potential as important resources for the discovery of liver cancer therapeutics. In this study, we selected three Korean herbal medicine formulas from the Donguibogam, namely Bigihwan (BGH), Daechilgithang (DCGT), and Mokwhyangbinranghwan (MHBRH), and evaluated their anti-cancer effects on HCC cells. According to our results of three ethanol extracts, BGH was more effective at suppressing HCC growth than DCGT or MHBRH. Furthermore, flow cytometry analysis showed that inhibition of HCC proliferation by the three extracts was associated with the induction of apoptosis and autophagy. In particular, BGH significantly increased mitochondrial impairment and showed the possibility of inducing mitophagy in comparison with the other two extracts. BGH prominently upregulated the levels of microtubule-associated protein light chain-3 which was accompanied by a decrease in the expression of anti-apoptotic Bcl-2 without altering the expression of pro-apoptotic Bax. In addition, the levels of PTEN-induced kinase 1 were also markedly increased in BGH-treated HCC cells. Moreover, autophagy blocking improved cell viability and reduced apoptosis after the three treatments, indicating that autophagy by these extracts enhances HCC cells against cytotoxicity. In conclusion, our findings show that BGH demonstrates the highest anti-cancer activity among the three formulas and inhibits the proliferation of HCC cells through autophagy induction.

천연 민간 생약으로부터 항암물질의 분리동정 및 그 작용기전에 관한 연구

  • 간암세포에 대한 세포독성효과를 민간생약을 대상으로 screening한 결과, 덩굴차, 해당화, 보리수나무, 인진호가 강한 활성을 나타내었다. 그 외 죽역, 엉겅퀴, 등나무흑, 비파등도 중정도의 세포독성을 나타내었다. 활성성분을 규명할 목적으로 각 분획별로 활성을 검토해 보았다. 인진호는 CHCl$_3$ Fr. 및 EtoAc Fr.이 강한 활성을 나타내었고, 그들은 모두 용량의존적으로 세포독작용을 나타내었으며, CHCl$_3$ Fr.이 더 강한 활성을 나타내었다. 등나무혹은 CHCl$_3$ Fr 및 EtoAc Fr.이 강한 활성을 나타내었으며, EtoAc Fr.이 용량의존적으로 세포독작용을 나타내었다. 비파는 Hexane Fr 및 CHCl$_3$ Fr.에서 강한 활성이 있었고, 그들은 용량의존적으로 세포독성을 나타내었으며, CHCl$_3$ Fr.에서 더 강한 활성이 인정되었다. 그리고 비파에서 분리된 ursolic acid는 6-100$\mu\textrm{g}$/m1에서 용량의존적으로 간암세포 독성작용을 나타내었다.

Antiproliferation effects of ethanol extract of garlic peels on human cancer cell lines (마늘껍질 70% 에탄올 추출물의 인간 암세포 증식억제 활성)

  • Ethanol extract of garlic peels (GPE) was investigated for its antiproliferative effects on human cancer cell lines. Human lung cancer cell line A549 treated with $500{\mu}g/mL$ GPE resulted in the growth inhibition of A549 by 90%. In stomach cancer cell AGS proliferation inhibition activity, GPE showed 45% and 71% inhibition of AGS growth at $1,000{\mu}g/mL$ and $2,000{\mu}g/mL$, respectively. GPE inhibited the growth of the breast cancer cells MCF-7 effectively at low concentration and showed 78% and 90% inhibitions of MCF-7 growth at $200{\mu}g/mL$ and $500{\mu}g/mL$, respectively. GPE showed very significant antiproliferation effect on liver cancer cell line Hep3B and inhibited Hep3B cell growth by 57% at $100{\mu}g/mL$, and the inhibition's rate increased up to 87% at $500{\mu}g/mL$. Antiproliferation effect of GPE on colorectal cancer cell HT-29 showed 15% reduction of HT-29 cell growth at $200{\mu}g/mL$ and the growth rate was reduced in a dose dependent manner up to $1,000{\mu}g/mL$. These results indicated that GPE had high antiproliferation effects on breast and liver cancer cell lines at low concentrations ($200{\mu}g/mL$), and by higher concentrations over $500{\mu}g/mL$, GPE inhibited the growth of A549 and HT-29. The results of our study suggested the potential use of garlic peels for use as an excellent antiproliferative substance for human cancer cells.

  • Euonymus alatus(Thunb.) Sieb has been used for cancer theraphy such as Liver cancer, esophagus cancer, stomach cancer, and uterine cancer in folk. Eupolyphaga simemsis(ES) extracted from Euonymus alatus(Thunb.) Sieb increases HDL-C/TC ratio and LCAT activity. TGF-${\beta}$ and Insulin has been Knoen as factors to induce the supression of proliferation of Hep3B, Human hepatoblastoma cell. Plasma HDL3-C involved in carbohydrate methabolism in the liver was maintained in low level by Euonymus alatus(Thunb.) Sieb. The low level of plasma HDL3-C suppresses the transition from normal liver to Fat liver. Finally, this resuly in suppression of cancer in liver. In this study, Euonymus alatus(Thunb.) Sieb showed higher cell toxicity in hepatoblastoma cell(Hep3B) compared to the uterine cancer cell(HeLa). This means Euonymus alatus(Thunb.) Sieb seems not to affect in the all kinds of cancer. In future, we will study whether Euonymus alatus(Thunb.) Sleb have an effect on the liver cancer induced by virus and hepatocellular carcinoma. Also, the mechanims of suppression of cell proliferation in HCC need to investigated.

Induction of Apoptosis by Methanol Extract of Endlicheria anomala in Human Lung and Liver Cancer Cells (Endlicheria anomala 메탄올 추출물에 의한 인체 폐암세포주와 간암세포주의 자가사멸 유도)

  • Endlicheria anomala, a neotropical plant, is found in northern South America and the Amazon region. It is traditionally used to remove poisons and cure gangrene. According to recent data, this plant has diverse biological properties such as anti-oxidative, anti-inflammatory and anti-melanogenic properties. However, the anti-cancer effect of E. anomala and its molecular mechanisms remain unclear. In this study, we examined the anti-cancer effect and the active mechanism of methanol extract of E. anomala (MEEA) in human lung adenocarcinoma cells (A549) and human liver cancer cells (HepG2). Our data revealed that MEEA showed cytotoxic activity in a dose-dependent manner and induced apoptosis both in A549 and HepG2 cells. We verified evidences of apoptosis via formation of chromatin condensation, apoptotic body and accumulation of cells in the subG1 phase. Following observed apoptosis-related phenomena, we found that the induction of apoptosis by MEEA was associated with the increase of tumor suppressor p53 and cyclin-dependent kinase inhibitor p21 (WAF1/CIP1) expression. Furthermore, MEEA-induced apoptosis was characterized with proteolytic activation of caspase-3, degradation of poly ADP ribose polymerase (PARP), and up-regulation of pro-apoptotic Bax expression. Taken together, these findings indicate that MEEA may have potential cancer therapeutic utility in A549 and HepG2 cells.

Inhibitory Effect of Celeriac Extract on Cancer Cell Proliferation (셀러리악 추출물의 암세포 증식 억제 효과)

  • This study was carried out examine the effect of Celeriac Extract, which contains various anticancer ingredients, on the proliferation inhibition of human-derived cancer cells and the degree of inhibition. The five cell lines used in the experiment were lung cancer cells A549, prostate cancer cells DU-145, uterine cancer cells HeLa, breast cancer cells MCF-7, and liver cancer cells SNU-182. All cancer cells derived from the human body were used, and the inhibition of cancer cell proliferation with Celeriac Extract 10ug/mL, 100ug/mL, and 1000ug/mL was measured using the CCK-8 method. As a result of examining the inhibition of cancer cell proliferation, Celeriac Extract 1000ug/mL showed significant proliferation inhibition in lung cancer cells A549, prostate cancer cells DU-145, uterine cancer cells HeLa, and liver cancer cells SNU-182, and showed a concentration dependence. However, only a concentration-dependent decrease was observed in breast cancer cells MCF-7.In conclusion, it can be seen that the cell proliferation inhibition mechanisms of Celeriac Extract using various human-derived cancer cell lines provide the potential for cancer prevention and therapeutic development.
