Protective effect of Mori Fructus in HaCaT human keratinocytes exposed to UVB (UVB에 노출된 HaCaT 인간 각질세포에서 상심자의 보호효과)

  • Objective : Mori Fructus has been used to relieve thirst, and treat dizziness, tinnitus, and insomnia caused by poor constitution. This study was performed to investigate the protective effect of the ethanol extract of Mori Fructus (MF) in ultraviolet B (UVB)-induced apoptotic cell death in human keratinocyte cells. Methods : MF was prepared by extracting 100 g of Mori Fructus in 1 L of 100% ethanol for 48 h. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to measure cell viability. Apoptosis was determined by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay, and the expression of apoptosis-related proteins was observed by Western blot. Tyrosinase activity was measured with a colorimetric commercial kit. Results : MF promoted cell vitality and inhibited apoptosis of UVB-induced HaCaT cells. MF pretreatment reduced TUNEL-positive cells and increased the expression of caspase-3 and -9. MF also displayed antioxidant effect with high radical scavenging ability. At 2 ㎎/㎖ concentration, the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical inhibition rates were 55.3 ± 4.6% and 48.5 ± 1.3%, respectively. Furthermore, MF showed a concentration-dependent inhibitory effect on tyrosinase. Conclusion : These results suggest that MF functions as a protective regulator in UVB-induced HaCaT cells by regulating apoptosis and partially exerting antioxidant effects. In addition, the tyrosinase inhibitory effect of MF shows the potential for MF to be used for skin pigmentation.

Anti-oxidative Effects of Taraxaci Herba and Protective Effects on Human HaCaT Keratinocyte (포공영 추출물의 항산화 효과 및 피부 각질세포 보호효과)

  • Objectives : This study was carried out to investigate anti-oxidative effects of Taraxaci Herba (TH) and protective effects on Human HaCaT keratinocyte. Methods : Anti-oxidative effects were measured by estimating the amount of total phenolics and flavonoids. In addition, DPPH free radical scavenging activities were estimated. Protective effects of TH on HaCaT keratinocytes against oxidative stress induced by hydrogen peroxide were also measured. Results : In our results, treatment with TH did not show cytotoxicity on HaCaT keratinocyte beneath the concentration of 200 $\mu$g/ml. 42.64$\pm$1.90 $\mu$g/ml of total phenolics and 28.09$\pm$1.84 $\mu$g/ml of flavonoids was detected from TH ethanol extract. In addition, DPPH free radical scavenging activities of TH were elevated in dose-dependent manner. In addition, The value of half maximal inhibitory concentration (IC$_{50}$) was 165.5 $\mu$g/ml. Finally, TH showed protective effect against cell death of HaCaT cell induced by hydrogen peroxide significantly. Conclusions : In conclusion, these results suggest that TH may have anti-oxidantic action in human skin and also suggest the possibility as cosmetic material.

Anti-inflammatory Effects of Myrrh Ethanol Extract on Particulate Matter-induced Skin Injury (미세먼지로 인한 피부 각질 세포 손상에서 몰약 에탄올 추출물의 항염증 효과)

  • Objectives: Myrrh have been used as a traditional remedy to treat infectious and inflammatory diseases. However, it is largely unknown whether myrrh ethanol extract could exhibit the inhibitory activities against particulate matter (PM)-induced skin injury on human keratinocytes, HaCaT cells. Therefore, this study was aimed to investigate the inhibitory activity of myrrh ethanol extract on PM-induced skin injury in HaCaT cells. Methods: To investigate the inhibitory effects of myrrh ethanol extract in HaCaT cells, the skin injury model of HaCaT cells was established under PM treatment. HaCaT keratinocyte cells were pre-treated with myrrh ethanol extract for 1 h, and then stimulated with PM. Then, the cells were harvested to measure the cell viability, reactive oxygen species (ROS), pro-inflammatory cytokines including interleukin (IL) 1-beta, IL-6, and tumor necrosis factor (TNF)-𝛼, hyaluronidase, collagen, MMPs. In addition, we examined the mitogen activated protein kinases (MAPKs) and inhibitory kappa B alpha (I𝜅-B𝛼) as inhibitory mechanisms of myrrh ethanol extract. Results: The treatment of myrrh ethanol extract inhibited the PM-induced cell death and ROS production in HaCaT cells. In addition, myrrh ethanol extract treatment inhibited the PM-induced elevation of IL-1beta, IL-6, and TNF-𝛼. Also, myrrh ethanol extract treatment inhibited the increase of hyaluronidase, MMP and decrease of collagen. Furthermore, myrrh ethanol extract treatment inhibited the activation of MAPKs and the degradation of I𝜅-B𝛼. Conclusions: Our result suggest that treatment of myrrh ethanol extract could inhibit the PM-induced skin injury via deactivation of MAPKs and nuclear factor (NF)-𝜅B in HaCaT cells. This study could suggest that myrrh ethanol extract could be a beneficial agent to prevent skin damage or inflammation.

Anti-aging effect of Codium fragile extract on keratinocytes damaged by fine dust PM10 (미세먼지 PM10으로 손상을 유도한 각질형성세포에서 청각 (Codium fragile) 추출물의 항노화 효과)

  • Objectives : Fine dust caused by environmental pollution cause oxidative damage and skin aging. In this study, The possibility of using the Codium fragile extract (CFE) as an anti-aging product for skin improvement was evaluated by confirming the protective effect of skin cells from PM10 (particulate matter 10) through inhibition of ROS and MMPs. Methods : In this study, elastase and collagenase inhibitory activities were evaluated. Cell viability was evaluated by treating keratinocytes (HaCaT cell line) with CFE at various concentrations. The cytoprotective effect from PM10 in keratinocyteswas evaluated using the 3-[4,5-dimethylthiazol]-2-yl]-2,5-diphenyl-tetrazoliumbromide (MTT) assay. ROS (reactive oxygen species) was measured in keratinocytes damaged by PM10 using DCF-DA (2′,7′-dichlorofluorescin diacetate) fluorescence staining. As an anti-aging effect of CFE, MMP-1 (matrix metalloproteinase-1) and MMP-1 (matrix metalloproteinase-9) inhibitory activities were evaluated. Results : As a result, CFE decreased the activity of elastase and collagenase. As a result of evaluating the toxicity of CFE, it is non-toxic at a concentration of 10 to 80 ㎍/㎖. Although cell viability of HaCaT cells treated with PM10 decreased, cell viability increased by 38% when treated with CFE 80 ㎍/㎖. Also, ROS decreased by 8.4%, and MMP-1 and MMP-9 decreased at CFE 80 ㎍/㎖. Conclusions : CFE showed excellent cell protection effect, and it is considered that it can be used in anti-aging products for skin improvement by effectively inhibiting ROS and MMPs from keratinocyte damage caused by fine dust.

  • The ultrastructures of the granular glands in the asiatic land salamander (Hynobius leechi) skin were observed by means of electron microsope The results were as follows; 1. The granular gland of the asiatic land salamander skin consisted of a body of gland and a duct. The body of the granular gland consisted of the glandular epithelial cells and the myoepithelial cells. The duct of the granular gland consisted of the keratinocytes. 2. The glandular epithelial cells in the asiatic land salamander skin were divided into the dark cell and the light cells in accordance with the electron density of the cytoplasm. 3. The secretory granules of the granular glands were round or oval in form and were divided into the various granules, showing the secretory granules showing weak electron density had the parts showing strong electron density near the granular membrane. 4. It is supposed that showing the different electron densities of the granules in a glandular epithelial cell is due to different mature stages and to different level of water absorption, and the chemical components of the granules showing different electron densities are identical.

Study on Effect of Skin Soothing by Helianthus annuus (Sunflower) Seed Oil (Helianthus annuus (Sunflower) Seed Oil 의 항염 및 피부 개선 효과 연구)

  • In this study, in vitro and clinical studies were conducted to assess the anti-inflammatory effects and skin improvement effects, including moisturizing, sebum secretion-regulating, skin barrier function enhancing, and soothing of Helianthus annuus (Sunflower) seed oil. In in vitro study using cultured human epidermal keratinocytes induced with inflammation by lipopolysaccharide, significant decreases in inflammatory cytokines interleukin-6, interleukin-8, and tumor necrosis factor alpha was revealed, indicating the anti-inflammatory effects of H. annuus (Sunflower) seed oil. Additionally, the results of clinical study on subjects with sensitive skin demonstrated improved skin hydration, regulation of sebum secretion, enhanced skin barrier function, as well as amelioration of skin redness and acne, indicating positive effects on overall skin conditions after application of H. annuus (Sunflower) seed oil containing test product for 4 weeks. Results of this study demonstrated the potential of H. annuus (Sunflower) seed oil as an ingredient for cosmetic, targeting consumers with sensitive skin.

Effect of Pulsed Electromagnetic Field Stimulation on TNF-α/IFN-γ induced inflammatory response in human skin keratinocytes HaCaT Cell to reduce inflammatory factors (인간 피부각질세포 HaCaT Cell에서 TNF-α/IFN-γ로 유도된 염증 반응에 대한 펄스형 전자기장(PEMF) 자극의 염증 인자 완화 효과)

  • The purpose of this study was to observe cell death in human keratinocytes stimulated against the infectious cytokines TNF-α and IFN-γ, and to observe the expression of Phospho-NF-κB due to phosphorylation of IkB to confirm the mechanism of inhibiting the expression of inflammatory cytokines. As a result of cell viability analysis, differences in PEMF stimulation time were observed little by little after 1 hour, 3 hours, 6 hours, 12 hours, 24 hours, and 48 hours, but there was no statistical significance according to PEMF stimulation time for each time (p>0.05). No significant difference was observed in the total amount of NF-κB present in the cytoplasm and nucleus, but a significant decrease in the expression of phosphorylated NF-κB was observed in the group exposed to PEMF stimulation for 24 hours (*p<0.05). The expression of IL-1β was observed in all inflammation-induced groups, and the concentration of IL-1β compared to α-Tubulin expression was reduced by about 54% in the PEMF-stimulated group for 24 hours compared to the control group (***p<0.001). As a result of the study, it is shown that PEMF stimulation does not negatively affect HaCaT cells from 0 to 48 hours and can inhibit the expression of inflammatory cytokines by inhibiting the pathway of NF-κB.

Study on the Propagation of Acanthopanax Plants II. Characteristics of Seed and Growth of Embryo in Stratifying Treatment (오갈피나무속(屬)(Acanthopanax) 식물의 번식(繁殖)에 관한 연구(硏究) II. 종자형질(種子形質)과 배(胚) 발육(發育)특성)

  • Seeds of Acanthopanx plants need about 2 years to germinate after sowing even if full matured fruits were harvested. I leave a room for doubt that this phenomenon could be brought by the immatured embryo in seed. If it is true, stratification method for the seed propagation of Acanthopanax plants would be used more effectively to promote the growth of embryo in relatively short time. Before stratification, seeds were devided into two parts. One of them was treated for 24hrs in the concentration of $GA_3$ 100 ppm. After stratification of non-treated and treated seeds, seeds for microtechnique were taken on interval of one month for three months, and fixed in Farmer's solution. The seed sizes of Acanthopanax plants were in biggest order A. sessiliflorus, A. seoulense, A. chiisanensis, A. koreanum and A. sieboldianum. The dehiscence phenomenon of seed coat didn't show in most of the seeds that stratified, but A. koreanum only dehisced in seeds treated in $GA_3$ 100ppm. The embryos of the stratified seeds that treated in $GA_3$ showed nomal growth, complete cotyledons and procambium in hypocotyl in any species, but the non-treated seeds could not expect the germination after sowing due to poor growing of the embryo in all species. In view of the results so far achieved, it has taken long times from sowing to germinate because the seed of Acanthopanax plants has immatured embryo.

Medicinal Herbal Complex Extract with Potential for Hair Growth-Promoting Activity (발모효과를 가지는 한방복합처방단)

  • To develop new therapeutic materials to prevent hair loss and enhance hair growth, we developed a medicinal herbal complex extract (MHCE) using 23 herbs traditionally used in oriental medicine. Medicinal Herbal complex extract was consist of Angelica gigas Nakai, Psoralea corylifolia Linne, Biota orientalis Endlicher, and Eclipta prostrata Linne, Rehmannia glutinosa Liboschitz var. purpurea Makino, Ligustrum lucidum Aiton, Polygonum multiflorum Thunberg, and Sesamum indicum Linne, Sophora angustifolia Sieboldet Zuccarini, Angelica dahurica Benthamet Hooker, and Leonurus sibiricus Linne, Salvia miltiorrhiza Bunge, Prunus persica Batsch, Commiphora molmol Engler, Chrysanthemum indicum Linne, Boswellia carterii Birdwood, Panax ginseng C. A. Meyer, Cnidium officinale Makino, Albizia julibrissin Durazzini, and Corydalis ternata Nakai that have traditionally been used for treating hair loss, preventing gray hair, anti-inflammation, and blood circulation in oriental medicine. In addition, we examined the hair growth effect of MHCE in vitro and in vivo. In vitro, we evaluated the effects of MHCE on cultured HFDPC, HaCaT cells, and murine embryonal fibroblasts (NIH3T3 cells). Also, we evaluated the ability of MHCE to prevent gray hair on murine melanoma cells (B16F1 cells). The hair growth-promoting effect of MHCE in vitro was also observed in vivo using C57BL/6 mice. Our results showed that MHCE significantly increased the proliferation of HFDPC (175 % proliferation at $50{\mu}g/mL$), HaCaT cells (133 % proliferation at $20{\mu}g/mL$), and NIH3T3 cells (120 % proliferation at $50{\mu}g/mL$). MHCE also showed consistent melanogenesis in B16F1 cells (154 % melanin synthesis at $50{\mu}g/mL$). Moreover, MHCE showed potential for hair growth stimulation in C57BL/6 mice experiments (98 % hair growth area on 4 weeks). These results indicate that MHCE may be a good candidate for promotion of hair growth.

New Whitening Agent From Pimpinella brachycarpa (참나물추출물의 멜라닌 생성저해 효과)

  • To develop a new whitening agent for cosmetics from natural products, Pimpinella brachcarpa was selected for its inhibitory effect on melanogenesis in B16 melanoma cells. Crude ethanolic extract of P. brachycarpa and its four fractions-hexane, ethyl acetate(EtOAc), butanol and aqueous were evaluated for antioxidative effects and tyrosinase inhibitory activity. To elucidate the mechanism of active compounds of P. brachycarpa, we investigated the changes in protein level of tyrosinase, TRP-1 and TRP-2 using Western blotting and the changes in mRNA level of tyrosinase using RT-PCR technique. Following UV irradiation, expression of ET-1 in HaCaT keratinocytes was measured by quantitative enzyme immunoassay(EIA) using human ET-1 antibody. Crude ethanolic extract of P. brachycarpa and its four fractions-hexane, EtOAc, butanol and aqueous had free radical scavenging effect by 87.2, 2.5, 97.2, 80.5, 49.8% at 100 ${\mu}g/mL$ and tyrosinase inhibitory effect by 18.3, 15.1, 55.4, 13.1, 0 % at 100 ${\mu}g/mL$. P. brachycarpa EtOAc fraction significantly inhibited melanin production in B16 melanoma cells. Treatment with P. brachycarpa extract for 72 h suppressed the biosynthesis of melanin up to 58 % at 100 ${\mu}g/mL$. Especially, the EtOAc fraction of P. brachycarpa reduced the tyrosinase activity and tyrosinase expression in B16 melanoma cells in a dose-dependent manner. mRNA levels of tyrosinase and TRP-1 were markedly reduced by the EtOAc fraction of P. brachycarpa. Moreover, at the concentrations of $12.5{\sim}50{\mu}g/mL$ of the fraction, the production of UV-induced ET-1 in HaCaT keratinocytes(24 h after 8 $mJ/cm^2$ UVB irradiation) was reduced about 40%(p<0.05). P. brachycarpa could be used as a new natural skin-whitening agent due to the inhibitory effect of on melanin biosynthesis and endothelin-1 expression.
