The Experimental Study on the Antibiotic Effects of Fel Ursi Pharmacopuncture Solution on Bacterial Species which cause Keratitis (웅담약침액이 다종의 각막염 유발균에 미치는 항균력에 대한 실험)

  • Objectives : This experimental study was performed to investigate the effectiveness of Fel Ursi Pharmacopunture solution(FUPS) manufactured by using alcohol/water extraction method. To identify the use of it as eyedrops, antibacterial test on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans was performed. Methods : After treatment FUPS on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, we investigated anti-bacterial effects of FUPS on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans in terms of measuring MIC and size of inhibition zone respectively. Results : After FUPS was treated, significant changes of MIC on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans were not observed at all. Conclusions : The present study suggests that FUPS doesn't have anti-bacterial effects on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans which cause Keratitis. Perhap These results recommend that FUPS doesn't have anti-bacterial effects but have other mechanism which suppress inflammation.

  • Objectives : This experimental study was performed to investigate the effect of Fel Ursi & Bovis Calculus pharmacopuncture solution(FUBCPS) manufactured by using alcohol/water extraction method for identify ing the use of it as eyedrops. Methods: FUBCPS was manufactured by using alcohol/water extraction method. Measure the size of inhibition zone and MIC(Minimum Inhibition Concentration) after administering FUBCPS on bacterial species. Staphylococcus aureus. Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans, which cause keratitis. Administering cravit(Levonoxacin medicine) on bacterial species also performed to compare the anti-bacterial potency of this material, measured by using the size of inhibition zone Results : After administering FUBCPS on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans). there was no response to MIC and there was no anti -bacterial potency also. Conclusions : This study suggests that FUBCPS dose not have anti-bacterial effects on bacterial species which cause Keratitis. These study result recommends that we need to research more on herbal medicines of eye drop which have anti-bacterial effects on keratitis

The Experimental Studies on Anti-bacterial Effect of Tangpo-san on Three species of causative bacteria of Keratitis (탕포산(湯泡散)이 삼종(三種)의 각막염(角膜炎) 유발균들에 미치는 영향)

  • This experimental study was performed to investigate the effect of herbal eye drops, Tangpo-san and Coptidis rhizoma on Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa keratitis. The following results were obtained by using Minimum inhibition Concentration(MIC) and inhibition Zone. 1. MIC on Staphylococcus aureus in Tangpo-san was 100%, in Coptidis rhizoma was 100% and in Cravit was 0.1% 2. MIC on Staphylococcus epidermidis in Tangpo-san was 100%, in Coptidis rhizoma was 10% and in Cravit was 0.1%. 3. MIC on Pseudomonas aeruginosa in Tangpo-san, Coptidis rhizoma was not showing and in Cravit was 0.1%. 4. The size of inhibition zone on Staphylococcus aureus for Tangpo-san was 13.3mm in $50{\mu}{\ell}$, for Coptidis rhizoma was 26mm in $50{\mu}{\ell}$ and for Cravit was 31mm in $50{\mu}{\ell}$, showing the highest antibacterial effect. 5. The size of inhibition zone on Staphylococcus epidermidis for Tangpo-san was 16mm in $50{\mu}{\ell}$, for Coptidis rhizoma was 25mm in $40{\mu}{\ell}$ and for Cravit was 34mm in $50{\mu}{\ell}$, showing the highest antibacterial effect. 6. The size of inhibition zone on Pseudomonas aeruginosa for Tangpo-san, Coptidis rhizoma was not and for Cravit was 24.7mm in $50{\mu}l$, showing the antibacterial effect. In addition, the results shows that the herbal eye drops, Tangpo-san and Coptidis rhizoma can be used to cure Staphylococcus aureus, Staphylococcus epidermidis keratitis and if further study is performed, the use of herbal eye drops will be valuable and beneficial in the clinical medicines.

Effects of natural and chemical disinfectants on soft contact lens infected with Pseudomonas aeruginosa : Scanning Electron Microscopic study (Soft contact lens에 있어서 천연보존제와 합성보존제의 살균효과 : 주사 전자현미경적 관찰)

  • The effects of grapefruit seed extract and $H_2O_2$ disinfectants on Pseudomonas aeruginosa were investigated by antimicrobial assay and scanning electron microscopy. Pseudomonas aeruginosa is a Gram negative bacteria and a common causative microorganism of bacterial keratitis. After incubation of soft contact lens in Pseudomonas aeruginosa suspension for 120 minutes containing the disinfectants, the grapefruit seed extract showed antimicrobial activity as good as the chemical disinfectant. The living rates of Pseudomonas aeruginosa in grapefruit seed extract and $H_2O_2$ disinfectants were $1{\times10^3CFU/m{\ell}$ and $0.5{\times}10^3CFU/m{\ell}$, respectively showing no major difference in the killing activity. However, the mechanisms of the antimicrobial activity were quite different as indicated by scanning electron microscopy of the bacterial surface morphology. Considering chemically or other toxins induced keratitis, the natural disinfectant isolated from grapefruit seed extract shows promising results as a soft contact lens disinfectant.

A Study on the Cytotoxicity and Antimicrobial activity of Synthetic preservative(benzalkonium chloride) and Natural preservative(chitosan) (합성보존제(benzalkonium chloride)와 천연보존제(chitosan)의 세포독성 및 항균 활성에 관한 연구)

  • This study was performed to examine the cytotoxicity and antimicrobial activity of synthetic or natural preservative. Fibroblast cells L929 were used for cytotoxicity test and Pseudomonas aeruginosa ATCC27853, Staphylococcus aureus ATCC25923, Escherichia coli ATCC25922, Trichoderma reesei ATCC6967 were used for antibacterial and antifungal activities. Benzalkonium chloride(BAC) as a synthetic preservative and chitosan as a natural preservative were used for this study. Minimum inhibitory concentration(MIC) of BAC was 0.1~0.01% for P. aeruginosa and 0.001~0.0001% for S. aureus and 0.1~0.01% for T. reesei, MIC of chitosan was 2% for P. aeruginosa and 1I % for S. aureus. This study suggest that chitosan might be useful as an eyedrop.

Sclera Segmentation for the Measurement of Conjunctival Injection (결막 충혈도 측정을 위한 공막 영상 분할)

  • Conjunctival injection is the initial symptom of various eye diseases such as conjunctivitis, keratitis, or uveitis. The quantification of conjunctival injection may help the diagnosis and follow-up evaluation of various eye diseases. The size of the sclera is an important factor for the quantification of conjunctival injection. However, previous manual segmentation is time-consuming.Automatic segmentation is needed to extract the objective region of interest. This paper proposed a method based on the level set algorithm to segment the sclera from an anterior eye image. The initial model of the level set algorithm is calculated using the Lab color space, k-means algorithm and the geometric information. The level set algorithm was applied to the images in which the valley between the eyeball and skin was enhanced using the hessian analysis. This algorithm was tested with 52 images of the anterior eye segment. Results showed that the proposed method performs better than those with the level set algorithm using an arbitrary circle, or the region growing algorithm with color information. The proposed method for the segmentation of sclera may become an important component for the objective measurement of the conjunctival injection.

Clinical Bacteriological and Observations on Infectious Bovine Keratoconjunctivitis (IBK) in an Integrated Dairy Farm (대단위(大單位) 낙농목장(酪農牧場)에서의 소전염성결막각막염(傳染性結膜角膜炎)(IBK)의 임상적(臨床的) 및 세균학적(細菌學的) 관찰(觀察))

  • A group of 80 Holstein calves, many with clinical signs of infectious bovine keratoconjunctivitis in summer seasons, was assembled for bacteriologic and ophthalmologic studies at an integrated dairy farm. Observations were started from 1980 to 1984 but bacteriological study was only carried out from thor spring in 1981 and continued during the Autumn in 1981. Corneal lesions and drugs sensitivity were also observed and the results obtained were as follows: 1. Occurrences of IBK-affected cattle during the 5 years were predominent to calves in summer. 2. Among 142 eyes from 80 cattle, 53 isolates of Moraxella bovis(37.3%) were isolated from 37 cattle(46.3%). The prevalence of the infection for clinical eyes were higher than nonclinical eyes calves. 3. Miscellaneous organisms were found from age groups of 5 months to 9 months old. It were 8 hemolytic Neisseria spp., 3 Fungi, Mycoplasma sp. and 1 Pseudomonas sp., but 9 samples were unknown. 4. Prevalence of infection with M. bovis in bilateral clinical eyes were higher than unilateral eyes. 5. Incidence of corneal lesion was predominent in early stage as a watery tears(21.0%) and late stage as a leukomas of corneal opacity(22.8%). 6. The number of isolations of M. bovis and incidence of IBK varied from year to year; Higher incidence of IBK clinical signs were showed in calves from 5 months to 10 months old and the number of isolations of M. bovis was declined in order 8 months(100.0%), 4 months(75.0%), 5 months(66.6%) and 8 months(66.6%) old calves. 7. Chloramphenicol, Nitrofurantoin and Cephalothin were highly sensitive against M. bovis isolates and then Tetracycline, Neomycin, Erythromycin and Kanamycin were intermediate but low sensitive to Streptomycin, Colistin and Penicillin.

The Bacterial Contamination in Glasses for Vision Correction (시력 교정용 안경의 세균 오염)

  • Purpose: Recently, bacterial contamination of equipment and accessories required for vision correction has become a main causal factor in ophthalmic diseases. Thus, We investigated on both the actual condition of bacterial contamination from glasses of vision correction. Methods: Investigation of microorganisms was carried out with a group of 145 glasses wearers, composed of 36 elementary school students, 37 middle school students, 38 high school students, 10 college students, and 32 aged men. Results: Seventeen species of bacteria are detected from glasses of vision correction: B. cereus, B. licheniformis, Bacillus sp., CNS, Enterococcus sp., Escherichia coli, Proteus sp., Pseudomonas sp., Serretia sp., Streptococcus sp., Staphylococcus epidermidis, Staphylococcus aureus, Streptococcus hemolyticus,, Acinetobacter sp., Enterobacter cloacae, GNR, and Pseudomonas aeruginosa. Among 17 species of bacteria, there are some potential causative agents for keratitis, corneal ulcer, Acute dacryocystitis, Orbital cellulitis, Periphlebitis retinae, Marginal blepharitis, and Acute conjunctivitis. Enterobacter cloacae, Pseudomonas aeruginosa and Staphylococcus epidermidis cause keratitis. Pseudomonas sp., and Staphylococcus aureus cause corneal ulcer. Staphylococcus aureus causes acute dacryocystitis, orbital cellulitis, periphlebitis retinae, marginal belpharitis. Streptococcus hemolyticus causes acute conjunctivitis. Conclusions: In summation, it is verified that hazardous, opportunistic and infectious microorganisms exist in glasses for vision correction. Ophthalmic diseases are predicted. Therefore, supplementary research on the development of a cleaning solution to cleanse the infection and of an effective method to remove microorganisms is required.

Microbe Pollution Condition on Soft Contact Lenses (소프트 콘택트렌즈에서의 미생물 오염양상)

  • To know the pollution level concerning the microbes contamination condition in soft contact lenses, 2 experiments were operated with different methods. Firs experiment, we observed lenses surfaces through a scanning electronic microscope after pollution sort contact lenses with the suspensions of Staphylococcus aureus, Pseudomonas aeruginosa and Entrobacter aerogenes which can be major bacteria causing keratitis as wearing on soft contact lenses. As 1 hour after pollution with the soft contact lenses, we observed that many bacteria adhered to the lenses, and the pollution condition got worse with passing time increase(2, 4, 6, 12, and 24 hours, respectively). At 12 hours lapse of polluting soft contact lenses with P. aeruginosa, some bacteria adhered to the lenses. And as 24 hours lapsed after pollution soft lenses with E. aerogenes, bacteria adhered to them, and atypical substance was observed some of the surface of lenses. Second experiment, we observed the microbe pollution condition in soft contact lenses by let 21 people in twenties(men & women), who doesn't eye disorders, wear soft contact lenses during 4 hours. As the result, many kinds of bacterium and fungus were observed, and especially, the bacteria could be identified with their colors and the shapes of colony, so we found that single type, two types, and three types of bacteria adhered to their lenses.

The Experimental Study on Anti-bacterial Potency of Variou Herb-medicine used for eye disease on Staphylococcus epidermidis Keratitis (다종의 안질환 사용 처방이 각막염 유발균인 Staphylococcus epidermidis에 미치는 영향)

  • Objective : This experimental study was performed to investigate the effect of Herbe-medicine used for eye disease(Sean-tang, Jinpi-san, Tangpo-san, Copitdis Rhizoma) on Staphylococcus epidermidis keratitis. Methods : After administering various herbal eye drops on Staphylococcus epidermidis, I measured MIC and the size of inhibition zone. MIC was measured by dropping from 20 to $50{\mu}{\ell}$ according to density. Anti-bacterial potency was measured by the size of inhibition zone with change of volume under 2 days culture condition. Also continuous anti-bacterial potency of herbal eye drops was measured by the size of inhibition zone according to 2 days and 6 days culture each under the $50{\mu}{\ell}$ condition. Results : 1. MIC on Staphylococcus epidermidis in Sean-tang, Jinpi-san was 1%, $50{\mu}{\ell}$ 2. MIC on Staphylococcus epidermidis in Tangpo-san, Coptidis rhizoma was 10%, $50{\mu}{\ell}$ 3. MIC on Staphylococcus epidermidis in Cravit was 0.1%, $20{\mu}{\ell}$. 4. Under the 2 days culture condition, the size of inhibition zone on Staphylococcus epidermidis by volume for Sean-tang was 10.0mm in $50{\mu}{\ell}$, Jinpi-san was 16.0mm in $50{\mu}{\ell}$, Tangpo-san was 17.5mm in $50{\mu}{\ell}$, Coptidis rhizoma was 31.0mm in $50{\mu}{\ell}$ and Cravit was 34mm in $50{\mu}{\ell}$, showed the highest anti-bacterial potency. 5. Under the $50{\mu}{\ell}$ condition, the size of inhibition zone on Staphylococcus epidermidis by 2 and 6 culture days for Sean-tang was 47.5mm in 6days, Jinpi-san was 36.0mm in 6days, Tangpo-san was 45.0mm in 6 days and Cravit was 48.0mm in 6 days, which showed the highest anti-bacterial potency. 6. Under the $50{\mu}{\ell}$ culture condition, the size of inhibition zone on Staphylococcus epidermidis by 2 and 6 culture days for Coptidis rhizoma was 31.0mm in 2 days and 6 days, which showed the same anti-bacteria1 potency. Conclusions : The present author think that Herbe-medicine used for eye disease can be used to cure Staphylococcus epidermidis keratitis and if further study is performed, the use of the Herbe-medicine used for eye disease will be valuable and beneficial in the clinical medicines.
