Epidemiological Aspects and Trends of the Extensive Eradication Programs for Bovine Brucellosis Outbreaks-Associated Human Brucellosis in Korea, 2002~2013

  • Over the last decade, human brucellosis (HB) has become a principal zoonosis in Korea. The objective of this study was observing the epidemiological trends of the extensive eradication programs for bovine brucellosis (BB) outbreaks-associated HB in Korea from 2002 to 2013. The raw data analysis in this study was obtained from the website of brucellosis outbreaks by Korea Center for Disease Control and Prevention (KCDC) and Animal and Plant Quarantine Agency (QIA), Korea, 2002~2013. The number of cases of BB outbreaks-associated HB in Korea was increased after 2003 and peaked in 2006 before decreasing thereafter. There were a total of 89,240 infected BB in domestic cattle with a cumulative incidence rate (CIR) of 611.8 per 100,000 cattle; during the same period there were a total of 703 cases of HB with a CIR of 0.12 per 100,000 persons. Moreover, the correlation coefficient of brucellosis outbreaks between cattle and human was highly significant (r=+0.985). The attack ratio of HB was 7.88 per 1,000 BB cases. All of the control measures of HB were applied in the extensive eradication programs for brucellosis outbreaks of infection source in domestic animals and contributed significantly to the reduction in the outbreaks of HB in Korea.

Investigation of Ingredients and Hazardous Substances in Disinfectants Used against COVID-19 and Some Livestock Diseases (코로나바이러스감염증-19와 일부 가축전염병 방역소독제품의 함유성분 및 유해물질 조사)

  • Objectives: The Coronavirus Disease 2019 (COVID-19) pandemic has caused the death of 740,000 people around the world as of August 12, 2020. Foot-and-Mouth Disease, Avian Influenza, and African Swine Fever are serious livestock diseases. Government agencies in Korea have provided ingredient information and usage instructions for disinfectants used to counter those infectious diseases. The purpose of this study was to provide information on the chemical ingredients in disinfectant products used against COVID-19 and certain livestock diseases. Methods: We collected information from the Korean government. The Central Disaster Management Headquarters and Central Disease Control Headquarters provided information on disinfectant products used against COVID-19. The Animal and Plant Quarantine Agency of Korea provided information on efficacy-certified disinfectant products for use against selected livestock diseases. Health hazard and environmental hazard information on the ingredients in the disinfectants was collected from the Korea Occupational Safety & Health Agency's Material Safety Data Sheets, and toxicity value information was collected from United States Environmental Protection Agency's CompTox Chemicals Dashboard. Results: There were 76 COVID-19 disinfectant products in use, and the most common ingredients were benzalkonium chloride (51%), alkylbenzyl dimethyl ammonium (30%), and ethanol (3%). There were 216 livestock disease disinfectant products comprised of 89 acidic, 88 oxidic, 30 aldehydic, three alkaline, and six other products. Among the 49 active ingredients used in the disinfectants that were investigated, health and environmental hazard information was provided for many of them, but only 20 chemicals had official toxicological information. Conclusion: Since the disinfectants included numerous chemicals, an understanding of their chemical characteristics could be critical to prevent unintended human or environmental exposure.

Serologic survey of the ruminant bacterial infectious diseases in farmed deer and wild water deer in Jeonbuk province (사육사슴 및 야생고라니에서 소 세균성 전염병에 대한 혈청학적 연구)

  • Deer can be one of the susceptible animals to bovine infectious diseases, and thus, may play a role either as a reservoir or amplifier host for spreading the diseases to other species such as cattle and goat. This study was conducted to determine the serum antibodies to bacterial infectious diseases for brucellosis, tuberculosis (TB), paratuberculosis (Johne's disease) in deer. Serum samples were randomly collected from 78 deer from 31 farms at Jeonbuk province, and 7 wild water deer from Jeonbuk wild animal treatment center during 2005 to 2007, respectively. Four farm deer (5.1%) showed antibodies to tuberculosis using Antigen Rapid Bovine TB Ab Test Kit. One elk (1.3%) and one wild water deer had antibodies for paratuberculosis. Antibody against Brucellosis was not detected in tube agglutination test (TAT) and enzyme-linked immunosorbent assay (ELISA). These data suggest that caution should be applied to inspection of velvet, deer blood and meat for human consumption from deer because of zoonotic bacterial diseases in deer. In addition, farmed deer can be a transmissible host for zoonotic disease to diary or raising farm.

  • Farmed deer could be susceptible carrier to bovine viral infectious disease. But unfortunately, there has not been an overall study over this subject in Korea so far. Therefore, a study was conducted to see serum antibodies to bovine leukosis, food and mouth disease, bovine viral diarrhea and infectious bovine rhinotracheitis in deer using the sera of farmed deer. As a result, two deer in a farms showed positive in bovine leukosis antibodies, using ELISA. For wild water deer, no antibodies were found for those diseases. As a result, it can be assumed that deer were relatively low rate of exposure to highly contagious disease such as viral bovine infectious disease in Korea. As this study was conducted over limited in number of subject and regions, continued study should be carried out in order to prevent and control the interspecies transmission in the future.

Prevalence of major legal communicable diseases in bovine and swine in Jeonbuk province (2004~2008) (전북지역에서 2004~2008년에 소와 돼지에서 법정전염병 발생 동향 분석)

  • Prevalence of major legal communicable diseases in bovine and swine had been monitored in Jeonbuk province from year 2004 to 2008. At least 1 communicable disease had been reported in 687 heads from 68 bovine farms and 17 farms (25.0%) of the 68 positive farms had 1~2 additional outbreaks during the surveillance. By disease, enzootic bovine leukosis, Johne's disease and Akabane disease were occurred in 53 farms (582 heads), 14 farms (100 heads) and 1 farm (5 heads), respectively. Swine communicable diseases were occurred in 4,466 heads from 63 swine farms and 18 farms (28.6%) of the 63 positive farms had 1~2 additional outbreaks during the surveillance. By disease, Aujeszky's disease (AD), porcine epidemic diarrhea (PED), classical swine fever (CSF), porcine reproductive and respiratory syndrome (PRRS), porcine transmissible gastroenteritis (TGE), atrophic rhinitis (AR) and Japanese encephalitis in swine (JE) were occurred in 20 farms (70 heads), 20 farms (2,817 heads), 12 farms (258 heads), 6 farms (1,257 heads), 1 farm (50 heads), 1 farm (2 heads) and 1 farm (10 heads), respectively. In total, 10 communicable diseases (1 species of first-class, 3 species of second-class, and 6 species of third-class) were reported. The first-class diseases were CSF. Johne's disease, and Aujeszky's disease. JE was the second-class and Akabane disease, enzootic bovine leukosis (EBL), PED, PRRS, TGE and AR were third-class diseases.

Traceability Management Model Supporting Safety Critical Transaction of Livestock Products (축산물 거래의 안전성을 보장하는 이력추적관리모델)

  • A set of definitions for traceability and quick recall service function were urgently needed to facilitate handling of events threatening food safety caused by livestock diseases or germ contaminations. However, the research on the unified management system for both international and domestic livestock products distribution was scarce. The livestock products traceability evaluation model proposed in the paper was composed of four modules: the Forward Transaction Trace (FTT) module, the Backward Record Trace (BRT) module, the Forward Product Trace (FPT) module, and the Origin Pedigree (OP) module. The evaluation indexes for each module was included and finally a pilot system evaluating the proposed management model was suggested and displayed. The result of the paper was expected to be a solution to the distributed traceability system and the proposed traceability management model could be expected to any food traceability.

  • Simulation using the virtual reality has been applied in various fields such as exercise, education, disaster simulation training, but there is a little research on disease caused by virus transmission. In this study, we conducted simulation studies and analysis of avian influenza disaster response training. The annual avian influenza virus is recurring every year in Korea, but there are still few solutions and preventive measures for the preventing the avian influenza. The avian influenza can cause a great deal of societal harm and enormous economic damage. Prophylaxis is important because livestock epidemics, such as avian influenza and foot-and-mouth disease, have a large impact on farm households. Therefore, we proposed and analyzed contents that can be avoided through simulation of avian influenza disaster response presented in this study.

Application of the CRISPR/Cas System for Point-of-care Diagnosis of Cattle Disease (현장에서 가축질병을 진단하기 위한 CRISPR/Cas 시스템의 활용)

  • Recently, cattle epidemic diseases are caused by a pathogen such as a virus or bacterium. Such diseases can spread through various pathways, such as feed intake, respiration, and contact between livestock. Diagnosis based on the ELISA (Enzyme-linked immunosorbent assay) and PCR (Polymerase chain reaction) methods has limitations because these traditional diagnostic methods are time consuming assays that require multiple steps and dedicated equipment. In this review, we propose the use of the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) Cas system based on DNA and RNA levels for early point-of-care diagnosis in cattle. In the CRISPR/Cas system, Cas effectors are classified into two classes and six subtypes. The Cas effectors included in class 2 are typically Cas9 in type II, Cas12 in type V (Cas12a and Cas12b) and Cas13 in type VI (Cas13a and Cas13b). The CRISPR/Cas system uses reporter molecules that are attached to the ssDNA strands. When the Cas enzyme cuts the ssDNA, these reporters either fluoresce or change color, indicating the presence of a specific disease marker. There are several steps in the development of a CRISPR/Cas system. The first is to select the Cas enzyme depending on DNA or RNA from pathogens (viruses or bacteria). Based on that, the next step is to integrate the optimal amplification, transducing method, and signal reporter. The CRISPR/Cas system is a powerful diagnostic tool using a gene-editing method, which is faster, better, and cheaper than traditional methods. This system could be used for early diagnosis of epidemic cattle diseases and help to control their spread.

동물 결핵

  • 동물의 결핵은 Mycobacterium bovis의 감염에 의한 만성 소모성 질병이며 인수공통전염병이다. 동물로부터 사람으로의 결핵 전염은 생유 섭취하던 시대에 상당히 많이 보고되었다. 우유의 살균처리와 소에서 피내진단에 의한 양성우 살처분 및 보상금 지급 정책을 전개하면서 M. bovis의 사람전염은 급격히 감소하였다. 소 결핵은 우리나라에서 연간 0.15% 내외의 발생을 보이고 있으며, 발생의 주원인으로는 외부입식소, 인근발생농장, 과거발생농장의 사후관리소홀 등이다. 사람 결핵의 주원인균인 M. tuberculosis와 M. bovis는 유전체가 99.9% 유사하며, M. bovis를 M. tuberculosis의 아종으로 분류하기도 한다. 두 세균은 M. tuberculosis complex에 속하며, M. tuberculosis와 M. bovis이외에도 M. africanum, M. canettii, M. microti, M. pinnipedii 등이 있다. M. bovis는 M. tuberculosis complex중에서 가장 넓은 숙주범위를 가진다. M. bovis의 대표적인 숙주는 종이름에도 나타나 있듯이 소이다. 소결핵 전파원으로서는 M. bovis에 감염된 소가 가장 중요하다. 소 이외에도 면양, 산양, 말, 돼지, 사슴, 엘크, 영양 (antelope, kudus, elands, sitatungas, oryxes, addaxes), 개, 고양이, 흰족제비 (ferrets), 낙타, 여우, 밍크, 오소리, 쥐, 영장류, 라마, 맥 (tapirs), 코끼리, 코뿔소 (rhinoceroses), 주머니쥐, 땅다람쥐 (ground squirrels), 수달 (otters), 물개, 산토끼 (hares), 두더쥐 (moles), 너구리 (raccoons), 코요테, 사자, 호랑이, 표범, 살쾡이 (lynx) 등에 감염될 수 있으나, 대부분 종결숙주 (spillover host)로 가축의 결핵방제가 유지되고 있는 국가에서는 야생동물 결핵의 가축 전염이 문제시되고 있다. M. bovis는 주로 호흡기와 소화기를 통하여 감염되며, 결핵결절이 형성되는 부위를 관찰하면 감염경로를 추정할 수 있다. 결핵에 감염되면, 초기에는 뚜렷한 임상증상을 보이지 않으나, 아침, 추운 날씨, 또는 운동 중에 심한 기침을 하며, 호흡곤란을 일으킬 수 있다. 결핵은 감염되어도 대부분 무증상이기 때문에 피내진단, 결핵결절 병리소견, 원인균 분리 등에 의해 진단하여야 한다. 감염된 결핵균은 탐식세포에 탐식되어 특징적인 육아종성 결절 병변으로 진행된다. 현재 결핵은 피내진단과 결핵결절 병리소견 등에 의해 판정하고 있다. 최신 진단법으로는 피내진단을 대체할 수 있는 인터페론 감마 검사법과 우군의 결핵 스크리닝과 말기 결핵 검사에 우수한 항체진단법이 개발되어 있다. 그러나, 소 결핵 근절을 위해서는 일관성있는 진단법과 진단기준을 적용하는 것이 중요한 성공요인중 하나이다. 소결핵 청정국인 호주와 캐나다에서는 피내진단과 도축장 결절검사를 결핵 양성우 색출방법의 근간으로 삼고 있으며, 소결핵 근절의 최종단계에 이르러서는 특이적인 검사법을 적용하였지만, 근절목적상 민감성이 높은 피내진단법을 사용하였다. 이와 더불어, 피내진단 양성우의 부검소견과 원인균 분리를 통해 결핵을 확진하여 출처농장의 역추적 검사를 통하여 결핵 양성소를 제거하였다. 한편, 결핵의 농장간 및 지역간 전파방지를 위해 결핵 청정농장과 결핵 오염농장, 결핵 청정지역과 결핵 오염지역 구분을 통하여 결핵 오염농장과 결핵 오염지역으로부터 결핵 청정농장과 결핵 청정지역으로의 이동전 결핵 검진을 통해 개체 이동에 따른 결핵 전파를 근본적으로 차단하는 시스템을 엄격히 적용한 것이 주요한 성공 요인중 하나였다. 호주 결핵 근절정책 성공요인을 요약하면, 일관성 있는 결핵진단법 적용, 양성우 출처농장의 철저한 역추적 검사, 개체 이동전 결핵 음성증명 확인, 농가단체의 경제적 및 방역상 적극적인 지원 및 협조 결핵의 지속적인 모니터 링과 현장요구에 부응하는 방제신기술의 지속적인 연구개발 등을 들 수 있다. 최근 들어 국내 동물 결핵은 소, 특히, 한우의 결핵발생이 증가하고 있으며, 사슴 결핵발생도 증가하고 있다. 농장간 및 지역간에 결핵 감수성 가축, 특히, 소와 사슴의 거래가 아주 복잡하게 이루어지고 있는 현실을 고려할 때, 결핵전파의 주원인인 결핵감염 소나 사슴의 농장내 반입을 철저히 차단해야 할 것이다. 이때, 개체 검사는 물론이고, 출처농장에 대한 결핵 음성을 확인한 후 입식하여야 할 것이며, 입식 후에도 60일정도 격리사육하면서 피내진단등 결핵검진 후 음성인 경우에만 합사하여야 할 것이다. M. bovis는 사람을 비롯한 거의 모든 온혈동물에서 결핵을 일으킬 수 있기 때문에, 결핵 감염소로 판정된 농장 종사자는 각 시도 보건소의 협조를 받아 결핵검진을 받도록 해야 한다. 농장 가축에 접촉할 수 있는 야생동물의 접촉을 차단하여야 하며, 특히, 농장 사료의 야생동물에 의한 오염을 방지할 수 있는 사료창고관리를 철저히 해야 한다. 결핵 감염소를 다룰 때는 분비물 또는 가검물에 의해 감염될 수 있기 때문에 개인방역장비 - 방역복, 마스크, 비닐장갑, 비닐장화 - 를 착용한 상태에서 다루어야 한다. 특히, 결핵 감염소를 매몰 또는 소각하는 과정에서 결핵 감염소의 배설물 및 분비물 처리를 철저히 하여야 한다. 모든 작업을 마친 후에는 개인방역장비, 매몰 또는 소각에 사용하였던 장비 등을 청소 및 소독하고 필요시 소각 또는 매몰하여야 하며, 개인감염위험과 타인 감염위험을 방지하기 위해 노출부위를 세척하여야 한다.

Organic Matter Analysis and Physicochemical Properties of Leachate from a Foot-and-Mouth Disease Landfill Site (구제역 가축 매몰지 침출수의 물리 화학적특성과 유기물질 성상분석)

  • Foot and mouth disease (FMD) is one of the most notorious and contagious viral diseases afflicting cloven-hoofed animals. In this study, the physicochemical properties of leachate from a FMD landfill site at 773-1, Waryong, Andong, Korea and the ground water from 777, Waryong, Andong, Korea, were analyzed for 1 year from December $10^{th}$ 2010 to November $17^{th}$ 2011. The leachate was collected from the FMD landfill site during March, May, July, September and November, 2011 and changes in pH, brix, water content, insoluble solids, crude proteins, crude lipids, total and reducing sugars and ash content were determined. Considering the annual profiles of temperature and rainfall at the FMD landfill site, the dramatic changes in the physicochemical properties of the leachate from March to July, and especially from May to July, such as increases in pH, and a rapid reduction of brix and organic matter, may be closely linked to the growth of microorganisms in the leachate. The sharp decreases in the concentration of biominerals, such as Mg, Ca, and Fe from 1073, 4311 and 56.2 ppm in March to 151, 78, and 0.1 ppm in November, further suggest that decreases in organic matter in the leachate result from degradation by microorganisms originating from the intestines of the livestock. Analysis of the profiles of the organic materials in the leachate revealed that the properties of the leachate were similar to those of excremental matter-derived water. These results could be applied to a number of fields for the analysis of organic matter behavior, the development of the degradation process, and risk analysis in the environment for hygiene and food industries, of leachate from FMD landfill sites.
