Methyl Methanesulfonate Stimulated $^3H$-Thymidine Uptake in Synchronized HeLa $S_3$ Suspension Cells Pretreated with Thymidine Analogs

  • The rate of $^{3}H$-thymidine uptake induced by MMS in asynchronous HeLa $S_3$ suspension cells was decreased in direct proprotion to dose increase. The combined treatment of BUdR or IUdR with MMS was more effective in reducing the rate of $^{3}H$-thymidine uptake. MMS-stimulated $^{3}H$-thymidine uptake was detected in synchronized $G_2$ stage cells of HeLa $S_3$ suspension cultures following treatment with thymidine double shock. BUdR and IUdR greatly enhanced MMS-stimulated $^{3}H$-thymidine uptake. IUdR was found to be a more effective sensitizer than BUdR in this respect.

Comparison of Methods for the Measurement of Bacterial Production in a Lake Ecosystem (호수 생태계에서 세균 생산량 측정 방법의 비교)

  • The bacterial secondary production was measured at 6 sites of Lake Soyang in October, 1989 by $^{3}$H-thymidine incorporation rate. Verfication for the method of bacterial secondary production measurement showed that $^{3}$H-thymidine incorporated into DNA, RNA and protein by average percentage of 38.45, 42.27 and 20.07%, respectively. THe more increased incoporated $^{3}$H-thymidine, the more increasde DNA fraction, but protein fraction was generally low. Incorporation of rate of /usp 3/H-thymidine. $^{3}$H-leucine into protein correlated with protein fraction of incorporated $^{3}$H-thymidine. Conversion factors were calculated as follows; $1.83*10 ^{20}$ cells/moles of thymidine incorporated/hr and 1.69*10$^{22}$ cells/moles of leucine incorporated/hr.

Comparison of Ag-NORs stain and [3H]thymidine incorporation in antiproliferative effect of caffeine on NIH3T3 cells (Ag-NORs 염색법과 [3H]thymidine incorporation법에 의한 caffeine의 NIH3T3 세포증식 억제효과 비교측정)

  • Inhibitory effect of caffeine on NIH3T3 cell proliferation was studied by using [$^3H$]thymidine incorporation and a modified one-step silver staining technique. The latter technique shows argyrophilic nucleolar organizer region-associated proteins (Ag-NORs), which are seen in nuclei as black dots. The result was compared with the counts of [$^3H$] thymidine incorporation. The results were summarized as follows; 1. The Ag-NORs numbers of NIH3T3 cells were $6.81{\pm}1.38$ at 24 hrs, $7.13{\pm}1.26$ at 48 hrs, $8.50{\pm}2.04$ at 72 hrs after incubation. 2. The numbers of Ag-NORs were significantly decrease in caffeine treated groups (p<0.01). 3. The counts of [$^3H$] thymidine incorporation were similar to the result of using Ag-NORs staining technique. It is concluded that Ag-NOR is a rapid, simple and compatible method to quantitate cell proliferation as compared with [$^3H$]thymidine incorporation.

  • Dose response for the nuscheduled DNA synthesis induced by various concentration of MMS was dose dependent and directly proportional to dose increased. Time dependence of unscheduled DNA synthesis was continued up to 4 hours, with the peak appearing 2$\\sim$3 hours after treatment with MMS and $^3 H$-thymidine labeling. Single treatment with BUdR or IUdR does not induce unscheduled DNA synthesis. BUdR and IUdR greatly enhanced MMS-induced unscheduled DNA synthesis, but the dose responses were different from that of single treatment with MMS. IUdR was found to be more effective sensitizer on MMS-induced unscheduled DNA synthesis.

Effects of Polyunsaturated Fatty Acids on Intestinal Cell Proliferation

  • The effect of the polyunsaterated fatty acids, linoleic acid(LA), arachidonic acid(AA) and conjugated dienoic linoleic acid(CLA) on IEC-6 cells (rat intestinal cell)proliferation and cell transduction have been determined in vitro. IEC-6 cells proliferation was assessed by cell growth and [3H]-thymidine incroporation analysis. At 10 μM concentration , the proliferationof cells supplemented with AA or LA was significantly higher than that of CLA. [3H]-thymidine uptake showed the same results. LA and AA increased [3H]-thymidine uptake more than CLA. The stimulatory effect of LA or AA was even more pronounced in the presence of IGF. Both cell number analysis and [3H]-thymidine incorporation revealed that IEC-6 cell proliferation was influenced differently by exogenous free fatty acids, in which AA or LA stimulated IEC-6 cell proliferation and CLA inhibited it. Tyorosine phosphorylation provides a key switch to regulate celluar acitivity in response to extracellular stimuli. At 20 μM and 10μM, AA with IGF-1 stimulated protein tyrosine phophorylation in IEC-6 cells, but LA's impact was less than that of AA. CLA and CLA with IGF-1 inhibited protein tyrosine phosphorylation in IEC-6 cells. These results suggest there is a possible correlation between cell proliferation and IGF receptor tyrosine knase activity driven by AA.

Influence of Panax Ginseng on Hepatic DNA Synthesis in Mice Receiving ACTH ( II ) (고려인삼이 ACTH를 받은 마우스의 간 조직 DNA 합성능에 미치는 영향(II))

  • It was planned to investigate, by observing incorporation of $[^3H]$ thymidine into liver cells, the influence of Panax Ginseng upon hepatic DNA synthesis in mice that received ACTH. Thirty male mice $(body\;weight:\;18{\sim}20\;g)$ were divided equally into the ginseng-ACTH and the saline-ACTH groups. Each animal of the ginseng-ACTH and the saline-ACTH groups received every day (subcutaneously) 0.05 m1/10 g body weight of ginseng extract (4 mg of ginseng alcohol extract in 1 ml of saline) and the same amount of saline, respectively, for 5 days. On the 5th experimental day, all animals received 0.01 unit of ACTH intraperitoneally one hour. after the last medication, and $1{\mu}Ci/g$ body weight of $[^3H]$ thymidine after one more hour. Five animals, at a time, of each group were sacrificed 1, 10, and 24 hours after thymidine administration, and their hepatic radioactivity was measured autoradiographically in terms of the % number of radioactive cells in 1,000 cell counts (Radioactive Index, R.1.). Following results were obtained: 1. The hepatic radioactive indices obtained from the saline-ACTH group 1, 10, and 24 hr after $[^3H]$ thymidine administration were $1.50{\pm}0.32,\;2.16{\pm}0.33\;and\;2.79{\pm}0.31\;(mean{\pm}S.D.)$, respectively. 2. The corresponding values obtained from the ginseng-ACTH group $(2.71{\pm}0.22,\;3.85{\pm}0.29,\;and\;5.06{\pm}0.31)$ were significantly higher than the values of the saline-ACTH group. It is inferred from the above results that the ginseng tends to prevent reduction in hepatic DNA synthesis caused by ACTH administration.

  • In present study, radioautography and electron-microscopy were applied by the author to the elucidation of the mechanism of diapause in Bombyx mori L. (1). Patterns of nucleic acid and protein syntheses during embryonic development of silkworms, incubated at high and at low temperatures, were demonstrated by means of radioautography with labelled precursors of nucleic acid and protein. On the third day after blastokinesis the embryo incubated at high temperature generally incorporated much of the $^3$H-glycine into the brain and the suboesophageal ganglion, and some into other regions. When incubated at low temperature, no difference in the pattern between the brain, the suboesophageal ganglion and other parts was observed. Radioautography with $^3$H-thymidine revealed no significant difference in DNA synthesis in embryos incubated at high and low temperatures. In diapausing eggs twenty days after deposition, only a few cells of the mesoderm incorporated the labelled material into their nuclei, but in the hibernated eggs all the nuclei of the mesoderm had thymidine incorporation. After blastokinesis only the anterior portion of the embryo around the brain and the suboesophageal ganglion had thymidine incorporation; this was not observed in the posterior portion.

  • Single crystals of deoxy-thymidine diphosphate-D-gluxose-4,6-dehydratase(abbreviated as dTDP-D-glucose dehydratase) from Escherichia coli Strain BL21 clone which harbors the gene of dTDP-D-glucose dehydratase in Salmonella typhimurium LT2 have been grown with and whithout substrates by sitting drop vapor diffusion at room temperature. The precipitating agent was 1.6 to 2.0 M Na, K phosphate buffer(pH 8.0). The crystals diffract to at least 2.5Å and belong to the hexagonal space group P61 with cell dimensions a=b=168.54Å, c=81.08Å. The asymmetric unit contains one dimer with a crystal volume per protein mass(VM) of 2.4Å3/Da and solvent content (Vsol) of 64% by volume.

The inference from the above result was that the ginseng suppressed DNA synthesis of cells in the mandibular gland.
