갓(Brassica juncea)의 주 항균물질의 구조 분석 (Structural Analysis of Major Antimicrobial Substance Obtained from Leaf Mustard(Brassica juncea))

  • A major component(compound A) in the ethylacetate fraction exhibited a strong antimicrobial activity was identified by UV, IR, FABMS and NMR. The compound A showed strong absorbance at 209, 259 and 359nm, indicating a flavonoid ring structure. IR spectrum possessed absorbance of OH at 3400∼3300cm-1, ketone at around 1650cm-1, and aromatic C=C at around 1660cm-1. Molecular weight of the compound A calculated as 478 from the information of m/z 479(M+H)+ and m/z 477(M-H)+ in the FABMS spectrum. Molecular formula of this compound was found to be C22H22O12 from m/z 479.1220(+3.1mmu for C22H23O12) of HRFABMS spectrum and from 13C-NMR spectrum. 1H-NMR and 13C-NMR spectra of the compound A revealed aromatic proton and benzene rings. Distortionless enhancement by polarization transfer(DEPT) exhibited that the compound A possessed 10 quaternary carbons and 3 substituted benzene rings including a methoxy group substitution. The compound A was identified as isorhamnetin 3-O-β-glucopyranoside by spectrophotometric methods in conjunction with 1H-1H COSY, 1H-13C COSY and HMBC, which revealed a flavone with OH group at 3, 5, 7, and 4' and methoxy group at 3' positions esterified to glucose.

Electrophoretic and Immunological Evaluation of Secalin in Rye, Triticale, and Wheat-Rye Translocation Wheat

  • Seed storage proteins have been used for studying biochemical genetics and end-use quality aspects. We conducted enzyme-linked immunosorbent assay (ELISA) and one-dimensional SDS-PAGE (1D SDS-PAGE) to evaluate different cereal crop species and Korean wheat lines for rye secalin proteins. The antisecalin antibody showed consistent specificity for rye secalin with little cross-reactivity to gliadins. Immunological cross-reactivities measured by the ELISA technique using competition assay showed significant differences of absorbance among rye, triticale, wheat-rye translocated wheat and non-translocated wheat. The absorbance values were lowest in rye followed by triticale, translocated wheat and non-translocated wheat. The ELISA for discrimination of wheat-rye translocation on the basis of antigen-antibody reactivity showed that none of the Korean wheat lines possessed 1RS and secalin proteins. The competitive ELISA experiment demonstrated specific determination for secalin that was originated from rye chromosomal parts. The result of 1D SDS-PAGE for identifying rye secalin subunits showed all three rye specific secalin protein subunits (75 KDa, 45 KDa, and 40 KDa) for rye and triticale, and 1RS specific secalins (45 KDa and 40 KDa) for 1AL/1RS and 1BL/1RS translocated wheats. All Korean wheats were lacking 1RS of rye chromosome and secalin.

  • This study was conducted to develop a method to change the color of traditional Dongchimi using natural dyes to meet changing consumer demands. It was intended to improve the manufacturing process ability for color Dongchimi, and the best optimum extraction solvent and method from the natural dyes were proven to be confirmed and valid. The extraction process was evaluated using different solvents including distilled water, 20% ethyl alcohol, and 1% citric acid, and the quantity of the extracted anthocyanin was then measured based on the absorbance. The greatest absorbance was observed when 1% citric acid was used. Based on these findings, it would be most efficient to allow the traditional Dongchimi to produce citric acid naturally instead of using artificial additives during its production. Celosia cristata L. was extracted in its largest quantity when reacted with 20% ethyl alcohol. Therefore, instead of applying Celosia cristata L. to Dongchimi as the sub-ingredient during the preparatory stage, it will be more efficient to use Celosia cristata L. as the fractional ingredient in order to first extraction.

  • 토양세균의 혼합배양에 의한 MCPP [2-(2-Methyl-4-Chlorophenoxy) Propionic Acid]의 미생물학적 분해를 조사하였다. 혼합배양은 Pseudomonas 종, Flavobacterium 종, 그리고 Achromobacter종을 포함하였다. 세균들은 유일한 탄소 및 에너지원으로서 MCPP를 이용하였으나, 부분적인 분해만을 부여주었다. MCPP의 분해는 2-methyl-4-chlorophenol의 형성에 의하여 진행되었으며, 이는 HPLC 에 의하여 발견되었고, GC-MS에 의하여 확인되었다. 이 중간대사물질은 일시적으로 생성되었으며, reverse phase HPLC와 Uv absorbance에 의하여 다른 대사산물의 존재는 관찰되지 않았다.

Comparison of Flocculati on-Spectrophotometry and Streaming Current Detector Method to the Control of Flocculants for the Removal of Humic Acid

  • Flocculation-spectrophotometry and streaming current detector( SCD ) method were investigated and compared in order to determine the optimum dosages of synthetic cationic polymers of different charge density and molecular mass for the removal of humic acid. The optimum dosage for each of the polymers was determined with the dosage at which the lowest absorbance of humic acid was shown for the formal. and was determined with the dosage required during charge neutralization of humic acid for the latter It was in good agreement between both methods and there is a strong inverse correlation between the optimum dosage and charge density of the polymers, with highly charged polymer giving the lowest optimum dosage, pointing out the importance the charge neutralization. By flocculation-spectrophotometry, it was found that the absorbance of humid acid with the amount of each of the polymers dosed, changes sharply for polymers of high charge density, but changes rather broadly for polymers of low and middle charge density, Both methods showed that a stoichiometric correlation exists between the optimum dosage of each of the cationic polymers and the negatively charged humic acid.

Effect of Organic Photosensitizers on the Antimicrobial Property of Polyurethane coated Leather

  • Cow leather coated with polyurethane film that contains various organic photosensitizers was investigated to demonstrate the antimicrobial properties in the application of the material to protective clothing and home appliances. To prepare the antimicrobial coating on leather surfaces with high potency against microbes, photoactive agents, such as benzophenone (BP), 4,4'-bis(dimethylamino) benzophenone (MK), 4,4'-dihydroxybenzophenone (DHBP) and methylene blue (MB), were incorporated into polyurethane-based coating solutions. The photoactive antimicrobial agent treated leather samples were characterized by SEM, color appearance, color fastness against abrasion, and antimicrobial tests. The optical properties of organic photosensitizers indicated that active UV absorbance ranges were different: BP (around 250 nm), MK (around 360 nm), DHBP (around 305 nm) and MB (around 295 nm &570 nm-685 nm). The intensity of the UV absorbance curve at the UVA light wavelength for the antimicrobial test showed the highest value with MK; subsequently, this was followed by MB, DHBP and BP in decreasing order. The treated-leather samples demonstrated excellent antibacterial activity under UVA light. The antimicrobial effects for the Staphylococcus aureus were superior to Escherichia coli. Moreover, the polyurethane finishing showed an effective durability to abrasion. The overall results indicated that DHBP is the most suitable PU coating additive to provide antimicrobial properties to leather as well as color and surface appearance than MK, MB, and BP.

Early Detection of Epiphytic Anthracnose Inoculum on Phyllosphere of Diospyros kaki var. domestica

  • We developed a polyclonal antibody (PAh) based- ELISA system to accurately and rapidly monitor inocula on plant surface before onset of anthracnose. Titer of mouse antisera against conidia of Colletotrichum gloeosporioides was determined by using indirect ELISA. It was high enough to be detectable up to ${\times}$ 12,800 dilutions. Absorbance readings exceeded (1.5even at a 10$^{-5}$ dilution. Sensitivity of PAb was precise enough to detect spore concentration as low as 50 conidia/well by indirect ELISA. PAb1 and PAb2 proved to be very sensitive and highly specific to the target pathogen, C. gloeosporioides, apparently discriminating other unrelated pathogens, or epiphytes. Absorbance values for original isolate exceeded 1.0, but no reaction was detected with other isolates, except three other anthracnose fungi: C. gloeosporioides (pepper strain), Glomerella cingulata (apple strain) and C. lagenarium. Our data suggest that PAb1 and PAb2 bind with the protein epitope that partially contains residues of amino acid, arginine, and Iysine. This kit fulfills the require-ments for detecting inoculums before infection and during onset of anthracnose on sweet persimmon.

직물에 흡착된 양이온계면활성제 정량방법에 관한 연구 (Analytical Methods for Cationic Surfactants Sorbed by Fabrics)

  • At present, analytical methods available for determining the amount of cationic surfactants(DSDMAC) on the fabrics are spectrophotometric method based on formation and extraction of the colored ion pair with anionic dye, radiochemical analysis and HPLC analysis. This study presents new analytical methods based on the formation of cationic surfactant(DSDMAC)-disulfine blue complex on the cotton fabric. Test methods for determining the amount of DSDMAC-disulfine blue complex on the cotton fabrics are measuring the reflectance of the colored fabric and the absorbance of the methanol solution of DSDMAC-disulfine blue complex extracted from dyed cotton fabric. Linear relationships between the K/S value of the fabric treated with DSDMAC followed by disulfine blue and the amount of DSDMAC sorbed by cotton fabric were obtained. Thus, the amount of DSDMAC sorbed by cotton fabric can be determined by K/S value of the fabric. DSDMAC-disulfine blue complex on the cotton fabric was extracted with methanol. The amount of DSDMAC sorbed by cotton fabric was estimated by measuring the absorbance of the methanol solution extracted from dyed cotton fabric.

키틴의 염료 흡착에 의한 염액의 색소제거에 관한 연구(제1보) (Decoloration in Dyebath by Dye Absorption of Chitin(Part I))

  • The adsorption ability of dyes on chitin, a natural polymer was investigated for decolorization of dye wastewater. Chitin was manufactured in lab by decalcification in dilute aqueous HCI solution and deproteination in dilute aqueous NaOH solution with shrimp shells. Absorbance of residue solution of dyebaths after dye adsorptions of chitin were measured in varieties of dye concentration and dipping periods. Four kinds of dyestuffs were used, C.I.Acid Blue 29. C.I.Direct Blue 6, C.I.Reactive Orange 12 and C.I.Basic Red 18. When chtin 1g was dipped in 0.05% of dyebath with stirring, maximum adsorption ratio of each kind of dyes was exhibited as 91.6% for C.I.Acid Blue 29, 95% for C.I.Direct Blue 6, 58.2% for C.I.Reactive Orange 13 and 75.8% for C.I.Basic Red 19. It shows that chitin has better adsorption abilities of ionic dyes of acid, direct and basic dye than non-ionic reactive dye. And chitin has better adsorption abilities of anionic acid direct dyes than cationic basic dye because of the presence of nitrogen atoms. All kinds of dyestuffs used showed speedy absorption effects by chitin, so chitin can absorb much amount of dyes in 5 mimutes reach to equilibrium of adsorption in 2 hours after dipping. Basic dye was absorbed the most speedily in 5 minutes, although maximum adsorption ratio is not high. That reason can be thought that chitin surface is essentially negatively charged due to polar funtional groups.

메칠오렌지에 의한 말레인산클로르페니라민의 분광광도 정량 (Spectrophotometric Determination of Chlorpheniramine Maleate with Methyl Orange)

  • A singly charged methyl orange(MO) anion was found to be extracted with chlorpheniramine maleate(CPM) as a 1 : 1 complex in chloroform. Of various solvents, MO-chloroform system gave a yellow color for CPM, while in the absence of CPM, an organic phase showed almost no color. In this way, the spectrophotometric method was investigated for the determination of CPM by solvent extraction. The addition of alcoholic bolic acid solution to the solvent extract gave a higher color stability and transparency at least 5 days, but the extract alone lost its color intensity significantly. CPM is determined by measuring the absorbance of the extracts over a range of $1{\sim}7{\times}10^{-4}M\;(39{\sim}273\;{\mu}g/ml)$ in aqueous solution at 423 nm. The molar absorptivity was $2.26{\times}10^3\;l,\;mol^{-1},\;cm^{-1}$. The absorbance of the extract was constant in the range of pH $3.7{\sim}4.6$. This novel method was applied for the determination of CPM in artificial and commercial preparations in comparison with the analytical method of CPM tablets in K.P.IV. The results obtained showed that the former was better in accuracy and time consumption than the latter.
