Freezing Hardiness of Several Pear Cultivars According to Degree and Duration of Low Temperatures (저온 처리 온도 및 지속 시간에 따른 배의 품종별 내동성 비교)

  • Freezing hardiness of winter bud and branch of several pear (Pyrus pyrifolia) cultivars according to degree and duration of low temperatures was investigated by sprouting, electrolyte leaching rate and triphenyltetrazolium chloride (TTC). Sprouting rate as infected by degree and duration of low temperature were different between cultivars. The lower temperature, the longer duration, sprouting rate was decreased. Electrolyte leaching rate was showed above 30% at below $-30^{\circ}C$ treatment regardless of cultivars and duration. The lower temperature and the longer duration, Electrolyte leaching rate was increased. Electrolyte leaching rates of Manpungbae, Niitaka and Chuwhangbae at $-30^{\circ}C$ for 9 hours treatment which were observed high sprouting rate, were lower than those of other varieties. Absorbance rates by TTC test at $-21^{\circ}C$ treatment were 66.0 to 96.5% for 6 hours, 49.4 to 91.9% for 9 hours, and 37.3 to 89.4% for 12 hours. Freezing hardiness of pear cultivars at ecodormancy was different according to degree and duration of low temperature treatments.

Advanced Water Treatment of High Turbidity Source by Hybrid Process of Photocatalyst and Ceramic Microfiltration: Effect of Organic Materials in Water-back-flushing (광촉매 및 세라믹 정밀여과 혼성공정에 의한 고탁도 원수의 고도정수처리: 물 역세척시 유기물의 영향)

  • For advanced drinking water treatment of high turbidity water, we used the hybrid module that was composed of photocatalyst packing between outside of tubular ceramic microfiltration membrane and membrane module inside. Photocatalyst was PP (polypropylene) bead coated $TiO_2$ powder by CVD (chemical vapor deposition) process. Instead of natural organic matters (NOM) and fine inorganic particles in natural water source, modified solution was prepared with humic acid and kaolin. Water-back-flushing of 10 sec was performed per every period of 10 min to minimize membrane fouling. Resistance of membrane fouling ($R_f$) decreased and J increased as concentration of humic acid changed from 10 mg/L to 2 mg/L, and finally the highest total permeate volume ($V_T$) could be obtained at 2 mg/L. Then, treatment efficiencies of turbidity and $UV_{254}$ absorbance were above 98.5% and 85.7%, respectively. As results of treatment portions by membrane filtration, photocatalyst adsorption, and photo-oxidation in MF, MF + $TiO_2$, and MF + $TiO_2$ + UV processes, turbidity was treated little by photocatalyst adsorption, and photo-oxidation. However, treatment portions of humic acid by adsorption and photo-oxidation were above 10.7 and 8.6%, respectively.

Physicochemical and Functional Properties of Kochujang with Broccoli Leaf Powder (브로콜리잎 분말 첨가 재래식 고추장의 이화학적 및 기능적 특성)

  • Home-made broccoli Kochujang (HMBK) was prepared with the addition of 5% broccoli leaf powder. Its physicochemical and functional properties were measured in extracts (80% methanol, 80% ethanol, and distilled water) and compared with home-made Kochujang (HMK) and factory-produced Kochujang (FPK). Total phenolic content (TPC) was 22% higher in methanol extract from HMBK (524.2 GAE/100 g) compared to HMK (431.0 GAE/100 g). TPC was 8% higher in ethanol extract from HMBK (541.9 GAE/100 g) compared to HMK (499.9 GAE/100 g). DPPH radical scavenging activity was 1.6 times higher in the methanol extracts from HMBK than HMK. In contrast there was no difference in DPPH radical scavenging activity between HMBK and HMK. Oxygen radical absorbance capacities in methanol and ethanol extracts from HMBK were similar to HMK, but both were higher than extracts from FPK (55% and 23% higher, respectively). Inhibition of angiotensin I converting enzyme activity in methanol extracts from HMBK was similar to HMK, but it was 2.6 times higher than inhibition activities from FPK. Interestingly, only ethanol extract from HMBK showed a 10.7% and 18.3% inhibition on cell growth of the human colon adenocarcinoma grade II cell line (HT-29) and human lung carcinoma cell line (NCI-H1229), respectively. These results indicate home-made Kochujang with broccoli leaf powder contains high total phenolics, antioxidant activities, and cancer cell growth inhibition activities.

Photoeletrochemical Properties of α-Fe2O3 Film Deposited on ITO Prepared by Cathodic Electrodeposition (음극전착법을 이용한 α-Fe2O3 막의 광전기화학적특성)

  • Semiconducting $\alpha$-Fe$_2$O$_3$ film was prepared by the cathodic electrodeposition method on Indium Tin Oxide (ITO) substrate for photoelectrochemical cell application. After heat treatment at 50$0^{\circ}C$, the phase was changed from Fe to $\alpha$-Fe$_2$O$_3$. The phase, morphology, absorbance, and photocurrent density (A/$\textrm{cm}^2$) of the film depended on the preparation conditions: deposition time, applied voltage, and the duration of heat treatment. The $\alpha$-Fe$_2$O$_3$ film was characterized by X-Ray Diffractometer (XRD), Scanning Electron Microscope (SEM), and UV -Visible Spectrophotometer. The stability of the $\alpha$-Fe$_2$O$_3$ film in aqueous solution was tested at zero bias potential under the white-light source of 100 mW/$\textrm{cm}^2$. The apparent grain size of the films formed at -2.0 V was larger than that grown at -2.5 V. The $\alpha$-Fe$_2$O$_3$ film deposited at -2.0 V for 180 s and heat-treated at 50$0^{\circ}C$ for 1 h showed the predominant photocurrent of 834$\mu$A/$\textrm{cm}^2$.

  • Since many process parameters of FHD(Flame Hydrolysis Deposition) are involved in forming multi-component amorphous silica film ($SiO_2-B_2O_3-P_2O_5-GeO_2$), it has not been easy to predict the optical, mechanical and thermal properties of deposited film from the simple process parameters, such as source flow rate. Furthermore, the prediction of final composition of film becomes even more difficult after sintering at high temperature due to the evaporation of volatile dopants. The motivation of the study was to clarify the quantitative relationship between simple process parameters such as the flow rate of source gases and resulting chemical composition of sintered film. Hence, the compositional analysis of silica soot by FTIR(Fourier Transformation Infrared Spectroscopy) and ICP-AES(Inductively Coupled Plasma-Atomic Emission Spectrometry) under the control of the amount of dopant was carried out to obtain the quantitative composition. By measuring spectrum of absorbance from FTIR, the compositional change of B-O, Si-O, OH($H_2O$) in silica film was measured. The concentrations of these dopants were also measured by ICP-AES, which were compared with the FTIR result. The final quantitative relationship between simple process parameters and composition was deduced from the comparison between two results.

Changes in the Characteristics of Dissolved Organic Matter by Microbial Transformation and the Subsequent Effects on Copper Binding (생분해에 따른 용존 유기물질 성상 및 중금속 구리와의 결합특성 변화)

  • Microbial changes in the characteristics of dissolved organic matter (DOM) and the subsequent effects on the conditional stability constants of copper were investigated using 14 day-incubations of Pony Lake fulvic acid (PLFA), Suwannee River fulvic acid (SRFA) and the mixtures of the humic substances and glucose. After incubation, dissolved organic carbon (DOC) concentrations were diminished, and specific UV absorbance values and DOC-normalized fluorescence intensities increased. The microbial changes were minimal for the samples contaning humic substances only whereas they were much pronounced for the mixtures with glucose. The extent of the changes increased with a higher content of glucose in the mixtures. The same trend was observed even for glucose solution. Our results suggest that labile organic moieties may be transformed into more chromophoric and humidified components by biodegradation. For the mixture samples, the copper binding stability constants did not change or even decreased after incubation. Therefore, microbially induced enrichment of the fulvic- and humic-like carbon structures in DOM appears to result in little change or the decrease of the copper binding coefficients.

Determination of Complex Formation Constant of Sodium-Selective Ionophores in Solvent Polymeric Membranes (용매 고분자막 상에 고정된 나트륨 이온선택성 물질의 착물형성상수 결정)

  • The complex formation constants (${\beta}_{MLn}$) of potassium and various sodium-selective neutral carriers in solvent polymeric membranes have been determined using solvent polymeric membrane-based optodes and ion-selective electrodes (ISEs). Two different types of PVC-based membranes containing the H^+selective chromoionophore (ETH 5294) with and without a sodium ionophore (4-tert-bntylcalix[4]arenetetraacetic acid tetraethyl ester, ETH 2120, bis[(12-crown-4)methyl] dodecylmethylmalonate or monensin methyl ester) were prepared and their optical responses to either the changes in alkali metal cation (e.g., sodium and potassium) concentrations at a fixed pH (0.05 M Tris-HCl, pH 7.2) or varying pH at a fixed alkali metal cation concentration (0.1 M) were measured. The same type of membranes were also mounted in conventional electrode body and their potentiometric responses to varying pH at a fixed alkali metal cation concentration (0.1 M) were measured. The complex formation constants of the ligand could be calculated from the calibration plots of the relative absorbance vs. the activity ratios of cation and proton ($a_{M^+}/a_{H^+}$) and of the emf vs. pH. It was confirmed that the ratio values of the complex formation constants for the primary and interfering ions are closely related to the experimental selectivity coefficients of ISEs.

Comparison of antioxidant activity and prevention of lymphocyte DNA damage by fruit and vegetable juices marketed in Korea (시판 천연 과일주스와 채소주스의 항산화능과 임파구 DNA 손상 방지 효능 비교)

  • Purpose: Fruit and vegetable juices are known to be rich sources of antioxidants, which have beneficial effects on diseases caused by oxidative stress. The purpose of this study was to directly compare the antioxidant activities of fruit and vegetable juices marketed in Korea. Methods: We analyzed four fruit juices, two vegetable juices, two yellow-green juices, and six mixed vegetable juices. Antioxidant activities were analyzed using 2,2-diphenyl-1-picrylhydrazyl (DPPH) test, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonate) (ABTS) test, and oxygen radical absorbance capacity (ORAC) assay. Protective effects against DNA damage were determined using an ex vivo comet assay with human lymphocytes. Results: DPPH radical scavenging activities were in the following order: blueberry juice > mixed vegetable C juice > kale juice > mixed vegetable P juice > grape juice. ABTS radical scavenging activities were in the following order: blueberry juice > mixed vegetable C juice > grape juice > mixed vegetable P juice > kale juice. Peroxyl radical scavenging activities as assessed by ORAC assay were in the following order: blueberry juice > kale juice > mixed vegetable C juice > grape juice. Grape or blueberry juice showed strong abilities to prevent DNA damage in lymphocytes, and the difference between them was not significant according to the GSTM1/GSTT1 genotype. Conclusion: Antioxidant activities of fruit and vegetable juices and ex vivo DNA protective activity increased in the order of blueberry juice, grape juice, and kale juice, although the rankings were slightly different. Therefore, these juices rich in polyphenols and flavonoids deserve more attention for their high antioxidant capacity.

Improvement in Storage Stability of Export Peeled-Chestnuts Using Electrolyzed Acid-water (전해산화수를 이용한 수출용 깐밤의 저장성 향상)

  • This study was carried out to improve the storage stability of export peeled-chestnuts using electrolyzed acid-water, because ice-cold tap water generally used to store export peeled-chestnuts grew turbid during the cold chain, decreasing the quality. The changes of pH and turbidity in the electrolyzed water used for storing peeled-chestnuts were investigated. The pH value rapidly changed when stored at $30^{\circ}C$, whereas it very slowly changed at $4^{\circ}C$. This was especially evident by the change in pH from 2.5 to 3.4 of the electrolyzed acid-water after 8 days. In the case of turbidity, closely correlated with the pH change, the electrolyzed water used for storing peeled-chestnuts was very turbid when stored at $30^{\circ}C$, but it was not even until 14 days at $4^{\circ}C$, as evident by the absorbance $(OD_{660})$ of electrolyzed acid-water at pH 2.5 and $4^{\circ}C$ being only 0.01 after 14 days and 0.11 after 35 days. Sensory evaluation, tested by expert panel, of the chestnuts stored in electrolyzed water was not significantly different compared to the control.

Hemolytic Properties of Tolaasin Causing the Brown Blotch Disease on Oyster Mushroom (느타리버섯 갈반병 원인독소 Tolaasin의 용혈특성)

  • Tolaasin is a peptide toxin produced by Pseudomonas tolaasii and causes a brown blotch disease forming brown, slightly sunken spots and blotches on the cultivated mushrooms. It is a lipodepsipeptide consisting of 18 amino acids and its molecular mass is 1,985 Da. It forms a pore in plasma membranes, resulting in the disruption of membranes of fungal, bacterial, plant, and animal cells as well as mushroom tissue. In order to measure the toxicity of tolaasin, erythrocytes of blood were used to evaluate the tolaasin-induced hemolysis. Hemolytic activity of tolaasin was measured by observing the absorbance change either at 420 nm, representing the release of hemoglobins from red blood cells(RBCs), or at 600 nm, representing the density of residual cells. The hemolytic activity of culture-extract of P. tolaasii increased at early-stationary phase of growth and was maximal at late stationary phase. The hemolytic activity of tolaasin appeared high in the RBCs of dog and rat. The RBCs of rabbit and hen were less susceptible to tolaasin. The effects of various cations were also measured. $Cd^{2+}$ and $La^{3+}$. as well as $Zn^{2+}$ appeared inhibitory to the tolaasin-induced hemolysis. The effects of various anions on tolaasin-induced hemolysis were measured and carbonate showed the greatest inhibition to the hemolysis. However, phosphate stimulated the tolaasin-induced hemolysis and no effects were observed by chloride and nitrate.
