Characteristics of Fermented Omija (Schizandra chinensis Baillon) Sugar Treatment Extracts by Lactobacillus sp. (Lactobacillus sp. 균주를 이용한 오미자 당침액의 발효특성)

  • This study was carried out to investigate the characteristics of FOST (fermented omija sugar treatment extracts) using Lactobacillus brevis HLJ59. Antioxidant activities of FOST were evaluated through viable cell number of L. brevis HLJ59, DPPH radical scavenging activity, reducing power and SOD-like activity, compared to non-FOST(non-fermented omija sugar treatment extracts). Also it was to evaluate Angiotensin Converting Enzyme (ACE) and Urease inhibitory activity of FOST. The viable cell number of L. brevis was about $2.05{\pm}0.21{\times}10^8$, $6.31{\pm}0.56{\times}10^{11}$, and $8.14{\pm}0.14{\times}10^9$ at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. DPPH radical scavenging activity of FOST was about 60.3%, 71.8%, and 44.5% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. The reducing power of FOST was about 0.92, 1.19, and 0.73 (OD at 700 nm) at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, and SOD-like activity of FOST was about 50.4%, 53.7%, and 33.4% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. ACE and Urease inhibitory activity by FOST was about 47.4%, 78.2%, 56.4% and 58.1%, 83.4%, 63.2% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. The results indicated that the fermentation of omija sugar treatment extracts using Lactobacillus brevis HLJ59 increased the antioxidant activities campared to the non-fermented omija sugar treatment extracts.

  • The biological magnification of Pb, Cu, Zn and Fe in vascular plant, Persicaria thunbergii, Trapa japonica and Nymphoides peltata natually grown at the river and riverside and relationship between the concentration of heavy metals in these vascular plants and in the soil were investigated in the Chonju river, Chonju city. The biological magnification values of these metals in P thunbergii were founded as follows ; Pb, Cu, Zn and Fe were 80.4~254.6$mu\textrm{g}$/g, 284.6~688.4$mu\textrm{g}$/g, 635.5~1979.4$mu\textrm{g}$/g and 1160.0~3590.9$mu\textrm{g}$/g respectively. In the case of T. japonica, Pb, Cu, Zn and Fe were 107.8~306.0$mu\textrm{g}$/g, 282.7~963.0p$mu\textrm{g}$/g, 1328.3~3546.$mu\textrm{g}$/g and 656.8~9944.0$mu\textrm{g}$/g and in N. peltata, 140.1~ 193.9$mu\textrm{g}$/g, 187.7~327.3$mu\textrm{g}$/g, 1126.6~1723.6$mu\textrm{g}$/g and 611.7~1914.6$mu\textrm{g}$/g respectively.   • 본 연구는 돼지 난포란의 동결보존 후 생존성과 난자의 활성화 처리에 따른 체외발생율과 이를 이용한 핵 이식배의 체외발생율을 조사하였다. 활성화 처리된 배는 $5\%$ FBS가 첨가된 NCSU 23 배양액으로 $38.5^{\circ}C$, $5\%\;CO_2$$95\%$ air의 조건으로 배양하였다. 1. 난포란을 EDS와 $5\%$ PVP로 동결 후 $10\%$ FBS가 첨가된 NCSU 23 배양액으로 $0{\~}10$시간 배양했을 때 체외발생율은 $36.0\%$로서 대조군인 비동결 난포란의 체외발생율 $46.0\%$에 비해 낮았다. 2. Ethanol과 cyclojexamide로 처리 후 42 및 46시간 배양한 배의 분할율은 각각 $33.3\%$, $36.0\%$$27.1\%$, $30.0\%$로서 대조군의 $8.8\%$, $11.4\%$에 비해 높게 나타났다. 3. 동결 및 비동결 난포란을 이용한 핵이식 배의 융합율과 발생율 간에는 유의한 차이가 없었다. 4. Ethanol과 cyclojexamide로 활성화 처리한 난자를 이용하여 재구축한 핵 이식배의 발생율은 $2.8\%$, $5.3\%$$1.5\%$, $2.9\%$로서 대조군의 $0.0\%$, $0.0\%$에 비해 높은 발생율을 나타냈다.

Cross-resistance and Inheritance of Resistance in Laboratory-selected Strains of the Brown Planthopper(Nilaparvata lugens Stal) (벼멸구의 저항성 유발, 교차저항성 및 저항성 유전에 관한 연구)

  • Cross-resistance and inheritance of resistance in laboratory-selected strains of the brown planthopper to various types of the insecticides were investigated. The fenobucarb-selected ($R_{f}$), carbofuran-selected($R_{c}$), and diazinon-selected($R_{d}$) strains were 50.3, 49.2 and 5.8 times less sensitive to the corresponding insecticides than th susceptible strain. both $R_{f}$ and $R_{c}$ strains were highly resistant to the other carbamate insecticides, and moderately resistant to cypermethrin and deltamethrin, but nearly not resistant to fenvalerate and the organophosphorus insecticides except malathion and phenthate. Moderate resistance to malathion and phenthoate in the $R_{f}$ and $R_{c}$ strains was obtained at the rate of 13.0-12.0 and 8.5-7.5 times, respectively. The $R_{d}$ strain showed low levels of resistance to the carbamate, organophosphorus and pyrethroid insecticides, but negatively correlated cross-resistance to fenvalerate. Resistance of the brown planthopper to all the test insecticides was inherited by partially dominant autosomal factor(s).

Association between systemic disease activity restriction and oral health

  • The purpose of this study was to analyze the responses of 5,824 adults(2,574 males and 3,250 females over the age of 19 years) using raw data from the 7th period of the National Health and Nutrition Examination Survey to investigate the relationship between systemic disease activity restriction and oral health. There were many systemic disease activity restrictions in adults with oral chewing and speaking problems, and it was statistically significant(p<.001). Factors influencing activity restriction due to systemic disease include age(odds ratio 1.03), Male(odds ratio 0.84), education level(odds ratio 0.57, 0.45, 0.31), drinking(odds ratio 1.38), chewing(odds ratio 1.86) and speaking(odds ratio 1.84) problems. There was a higher probability of activity restriction due to systemic disease when they received treatment for periodontal disease(odds ratio 1.27) and broken teeth(odds ratio 2.1). Also, it was statistically significant that the quality of life decreased when there was chewing and speaking problems.

Plant Production from Desiccated Somatic Embryos of Acanthopanax chiisanensis (지리오가피 (Acanthopanax chiisanensis) 체세포배의 건조처리를 통한 식물체 증식)

  • An efficient method of plant regeneration from Acanthopanax chiisanensis somatic embryos was developed. Cotyledonary somatic embryos were obtained in liquid Murashige and Skoog (MS) medium from embryogenic cell suspension cultures. They were desiccated for 0 to 72 hr and then cultured on MS medium containing NAA, BA, GA$_3$, (0-0.5mg/L). The highest multiple shoots formation (100%) was obtained from 72 hr desiccated somatic embryos on ifs medium with 0.5mg/L NAA+0.5mg/L BA or 0.5 mg/L NAA+0.5mg/L BA+0.5mg/L GA$_3$ after 6 weeks culture. Plant conversion from multiple shoots was not high. The highest plant conversion from multiple shoots was obtained on 1/3MS medium with 1.0mg/L GA$_3$. Converted plantlets were transferred to ex vitro condition and the highest survival rate (70%) of the plantlets was obtained on plastic pots containing vermiculite and sand. These results indicate that micropropagation procedure can be applied for an efficient mass propagation of Acanthopanax chiisanensis.

Effects of Energy Substrates in Culture Media on Developmental Capacity of Mouse Embryos (배양액에 첨가하는 에너지원이 생쥐 배 발생 능력에 미치는 영향)

  • This study was conducted to examine the effects of energy substrates in different conoentration of carbohydrates in the human oviduct and uterus on the in vitro development of mouse 2-cell embryos. Two-cell embryos were collected from ICR female mice at $46{\sim}50\;hr$ after 5 IU hCG injection and cultured in three different media [control: 0 mM, Guoup A: glucose (G) 0.5 mM + pyruvate (P) 0.32 mM + lactate (L) 10.5 mM, Group B: G 3.15 mM + P 0.1 mM + L 5.83 mM] for 72 hr. Rates of morula formation of group A (72.3%) and B (56.6%) were significantly higher higher (p<0.05) than that of control (34.9%) at 24 hr. However, blastocyst rate was significantly higher (p<0.05) in control (51.8%) than group A (39.8%) and B (28.9%) at hr. At 72 hr, no differences were found in the number of zona-intact, zona-escape and total blastocysts among groups. Mean and ICM cell numbers were significantly higher (p<0.05) in group A (78.0, 13.4) and B (64.4, 11.8) than control (53.1, 5.7), respectively, The percent of ICM were significantly higher (p<0.05) in group A (22.9%) and B (23.7%) than control (14.2%). No differences were round in the TE cell numbers ($34.1{\sim}45.1$). The ICM:TE ratio was significantly higher $34.1{\sim}45.1$ in control (1:6.0) than group A (1:3.4) and B (1:3.4). This study shows that energy substrates added to culture media especially, the oviductal level of carbohydrates increase the developmental capacity of 2-cell mouse embryos.

Somatic Embryogenesis in Immature Zygotic Embryo Cultures of Korean Soybean (Glycine max L.) Culitivars and Effect of 2,4-Dichlorophenoxyacetic Acid on Somatic Embryo Morphology (한국 품종 대두(Glycine max L) 미숙배로부터 체세포배발생과 배지의 2,4-Dichlorophenoxyacetic Acid 농도가 체세포배의 형태에 미치는 영향)

  • Immature zygotic embryos of five Korean soybean cultivars cultured on Murashige and Skoog's (MS) medium supplemented with various concentrations of 2,4-Dichlorophenoxyacetic acid (2,4-D) produced somatic embryos without forming an intervening callus. The highest frequency (up to 90%) of somatic embryo formation was obtained when zygotic embryos were cultlued on medium containing 1 to 2 mga 2, 0-D in four cultivars. However the frequency was highly variable to the cultivars. Transversely sliced primary somatic embryo halves were also capable of forming secondary embryos at frequencies of up to 70% when cultured on medium containing 0.1 to 1 mg/L 2,4-D. Somatic embryos formed on zygotic embryos cultured on medium containing 0.1 to 0.2 mg/L 2,4-D had two cotyledons more frequently than one horn-type cotyledon and those on medium containing 0.5 to 4mg/L 2,4-LD had a horm-type cotyledon at a prominently higher freequency. However somatic embryos on medium containing 10mg/L or higher concentrations of 2,4-D were usually shunted at the globular stage even after transfer to medium containing lower concentrations of 2,4-D or other growth regulators. non somatic embryos with one or two cotyledons or a hem-type cotyledon were transferred to medium containing $GA_3$, those with two cotyledons converted to plantlets at a higher frequency (25%) than the others.

The Anti-Sticking Effect of Mixture of Trisodium Phosphate and Citric Acid on Oral Streptococcus species (구강 내 사슬알균 종들에 대한 제3인산나트륨과 구연산의 탈부착 효과)

  • Trisodium phosphate 12 hydrate and citric acid monohydrate mixture showed the strong anti-sticking effect on Streptococcus mutans, Streptococcus mitis, and Streptococcus salivarius, which are adhered to glass beads. Each Streptococcus species was shaking-cultured in brain heart infusion broth containing three glass beads. After 18 hr, glass beads were slightly washed into normal saline by three-pin-pointed pincette. Each three glass-beads set was put into reagent -containing tubes, which have 40 mg of bits of weighing paper for gaining brushing effect as similar as brushing one's teeth. The tubes were shaken by vortex mixer for 10 min except non-oral microbe, Streptococcus agalactiae (5 min). The samples were colony-counted by serial agar dilution method. Experiment was repeated three times for each Streptococcus species. The relative ratios of bacterial de-adherence by reagents were calculated in comparison with normal saline control. The de-adherence degree of citric acid-trisodium phosphate-saline mixture (CTS, pH 6.0) against Streptococcus mutans came to an average of 12.5 times compared with normal saline control. Trisodium-saline (TS, pH 8.4) showed the average of 7.5 times, and citric acid-saline (CS, pH 4.6) showed 6.0 times compared to the control group. The bacterial de-adherence degree against Streptococcus salivarius was each 7.2,2.6 and 2.8 times in above reagent sequence in comparison with saline control. CTS and TS showed 2.4 and 3.4 times of anti-sticking effect on Streptococcus mitis respectively, but CS had no anti-sticking effect on this bacterium. CTS, TS and CS showed 0.7, 0.6, and 0.6 times on non-oral microbe, Streptococcus agalactiae, separately compared with saline control. These results show that oral Streptococcus mutans, Streptococcus salivarius, and Streptococcus mitis, which are causative of dental caries or subacute endocarditis, may be easily removed from oral cavity by CTS mixture. It is conceivable that our experimental results will enable the development of a new conceptive toothpaste to prevent dental caries or subacute endocarditis after drawing teeth.

인공 산성 빗물로 인한 잎으로부터의 양이온 이론

  • After spraying simulated sulphuric acid rain (SSAR) to the leaves of Glycine max, Quercus aliena var. pellucida and Pinus rigida, the leachates were consecutively collected and quantitatively determined for the concentration of K and Ca. The amount of the leached ion decreased with subsequent acid treatment for all plants. But as the pH of SSAR decreased, the amount of the leached ion increased. The cumulative quantities of K leached from each species were 1.04 to 1.46 times higher at pH 4.0, 1.09 to 1.58 times higher at pH 3.5 and 1.24 to 2.03 times higher at pH 3.0 compared with pH 5. 6 treatments. The cumulative quantities of Ca leached from each species were 1.23 to 1.47 times higher at pH 4.0, 1.50 to 1.92 times higher at pH 3.5, and 2.45 to 3.30 times higher at pH 3.0 compared with pH 5.6 treatments. The Ca /K ratio in 1000 ml leachate was 1.10 to 2.91 for Q. aliena var. pellucida and 1.68 to 2.98 for P. rigida, but 0.66 to 0.91 for G, max. The Ca /K ratio in 1000 rnl leachate increased for all three species, as the pH of SSAR decreased. Foliage analysis after acid rain treatment showed leaching effect at pH 3.0.
